Description
ステント等のポリマー製埋込型医療装置の治療用コーティングにおける潜在的問題として、不十分な靭性、遅い分解速度、および悪い接着性を挙げることができる。
「ガラス転移温度」Tgは、大気圧において、ポリマーの非晶質ドメインが脆いガラス状態から、固体の、変形可能または延性のある状態あるいはゴム状に変化する温度である。言い換えれば、Tgは、ポリマー鎖内でセグメント運動が開始する温度に相当する。非晶質または半晶質ポリマーが温度の上昇に晒されると、ポリマーの膨張係数および熱容量の両方が温度上昇に伴って増加し、分子運動の増加を示す。温度上昇時には、試料中の実際の分子体積は一定に保たれるので、膨張係数が高くなるほど、系に伴う自由体積は増加し、したがって分子が運動する自由度は増す。増加する熱容量は、運動を介した熱放散の増加に相当する。所与のポリマーのTgは、加熱速度に依存し得るものであり、ポリマーの熱履歴に左右され得る。さらに、ポリマーの化学構造は、ガラス転移に多大な影響を与える。
[実施例]
実施例1.P(GA−co−CL)−b−PLLAコポリマーの合成
ステップ1:機械的攪拌棒を備えた1つの500mL反応槽を、高純度窒素が充填されたグローブボックス内に配置する。反応槽を予熱して全ての水分を除去する。
ステップ2:GA、CL、ドデカノール、キシレン、およびオクチル酸スズを反応槽に加える。この混合物を120℃で72時間攪拌する。
ステップ3:次いで、LLAを反応槽に加え、さらに72時間反応を継続させる。
ステップ4:次に、CHCl3100mLを反応槽に加えて、最終生成物を希釈する。生成物の溶液をメタノール600mLに沈殿、ろ過し、恒量となるまで、80℃で真空乾燥する。
実施例2.PLLA−b−P(GA−co−CL)−b−PLLAコポリマーの合成
ステップ1:機械的攪拌棒を備えた1つの500mL反応槽を、高純度窒素が充填されたグローブボックス内に配置する。反応槽を予熱して全ての水分を除去する。

- 埋込型医療装置であって、前記装置のポリマー表面の上方にコーティングを備え、
前記コーティングは、弾性ブロックとアンカーブロックとを含むブロックコポリマーを含み、
前記弾性ブロックは、ランダムコポリマーまたは交互コポリマーであるとともに、生理学的条件においてエラストマー挙動を呈し、
前記アンカーブロックは、前記ポリマー表面の表面ポリマーとの間で混和性を有し、
前記弾性ブロックは、カプロラクトン、テトラメチルカーボネート、4−ヒドロキシブチレート、ジオキサノン、およびそれらの組合せからなるグループから選択される単位を含み、
前記弾性ブロックの構成モノマーの割合を調節することにより、前記コーティングの分解速度が所望値に調節された、
埋込型医療装置。 - 前記装置がステントである、
