Description
1.この方法は、動物の心外膜からなるのが好ましい生物組織を、アルデヒドのようなどれか適切な物質により既知の方法で化学的に固定することからはじまる。アルデヒドを使用する場合は、グルタルアルデヒドを、例えば、0.625%の濃度で用いるのが好ましい。そのような化学的固定により生物組織に、非抗原性と、化学的、生物学的、物理的安定性と、特に温度と機械的ストレスの変化に対する耐性とを与える。
・ 超融解の危険(局部再水和に到る水の融解)を回避するため、運動は、非常に遅く、例えば、1時間当たり2℃と1時間当たり8℃との間であるべきである。
・ 脱水の度合いは少なくとも75%に等しく、約78%から80%までが好ましい。脱水度が低すぎると組織のある種の再水和、よって繊維のある種の可動性を許容し、これは可撓性の損失と、(短くても)保存段階後の再水和不良に反映される。脱水が過剰すぎると、その脱水としては、生物学的材料を変性させ、かなり収縮させる。事実、生物学的材料は、その材料の実際の構成部分であり、よって除去する必要がない結合水(心外膜に対し10%の固体と10%の結合水)からなる。
Claims (7)
- エラストマである可撓性合成基質と、
生体過敏反応を阻止するために化学的に固定(fixed)された動物の生物組織とを備え、
上記の合成基質の構成物質と同じ物質があらかじめ溶媒に分散されてなる血液適合性癒着剤であって上記の生物組織に含侵させ得る血液適合性癒着剤を用いて、上記の合成基質と上記の生物組織とを密に相互連結していることを特徴とする血液適合性複合材料。 - 上記の生物組織が動物の心外膜であることを特徴とする請求項1に記載の血液適合性複合材料。
- 上記のエラストマがポリウレタン・エラストマあるいはシリコン・エラストマであることを特徴とする請求項1に記載の血液適合性複合材料。
- 上記の請求項1から3のいずれか1項に記載の血液適合性複合材料の製造方法であって、
上記の動物の生物組織を化学的に固定した後、
− 上記の生物組織はグリコールで数日間処理し、このグリコールを上記生物組織の繊維同士間に導入し、
− このように処理した生物組織は、少なくとも75%に等しい脱水度に達するまでゆっくりフリーズドライされ、
− 上記の生物組織は、上記の基質の構成物質の溶媒への分散により上記の基質に結合され、
− 上記の溶媒が除去され、
− こうして得られた材料が再水和される、
ことを特徴とする方法。 - 用いられたグリコールがポリエチレン・グリコールであることを特徴とする請求項4に記載の血液適合性複合材料の製造方法。
- 上記の溶媒は、遅い熱抽出、および/または真空下での抽出および/または生理塩水での抽出により除去されることを特徴とする請求項4又は5に記載の血液適合性複合材料の製造方法。
- フリーズドライ率が1時間当たり2℃と8℃との間であることを特徴とする請求項4から6のいずれか1項に記載の血液適合性複合材料の製造方法。
