Description
ある実施形態では、本発明は、対象者における認知機能を向上させるための、対象者の知能を向上させるための、又は対象者における脳内のシナプスのサイズ又は数を増加させるための、幼児用調整乳を製造するための、(a)オメガ3脂肪酸、オメガ6脂肪酸、又はそれらの組み合わせ、(b)ウリジン、そのアシル誘導体、ウリジン・リン酸又はCDPコリン、及び、(c)コリン塩の使用であって、前記幼児用調整乳が前記対象者に治療に効果的な量で投入されると、神経細胞又は脳細胞のシナプス膜に関連するリン脂質の量が増加し、対象者の知能が向上し、又は対象者における脳内のシナプスのサイズ又は数が増加することを特徴とする使用を提供する。
ある実施形態では、本発明は、(a)ウリジン、そのアシル誘導体、又はウリジン・リン酸、(b)オメガ6脂肪酸、及び(c)コリン又はコリン塩を含有する幼児用調整乳を提供する。
他の実施形態では、この用語は、10〜24ヵ月を意味する。他の実施形態では、この用語は、13〜24ヵ月を意味する。他の実施形態では、この用語は、14〜24ヵ月を意味する。他の実施形態では、この用語は、16〜24ヵ月を意味する。他の実施形態では、この用語は、18〜24ヵ月を意味する。他の実施形態では、この用語は、12〜18ヵ月を意味する。他の実施形態では、この用語は、13〜18ヵ月を意味する。他の実施形態では、この用語は、15〜18ヵ月を意味する。他の実施形態では、この用語は、12〜20ヵ月を意味する。他の実施形態では、この用語は、14〜20ヵ月を意味する。他の実施形態では、この用語は、離乳しているが、20ヶ月以下の子供を意味する。他の実施形態では、この用語は、離乳しているが、24ヶ月以下の子供を意味する。各可能な形態は、本発明の異なる実施形態に相当する。
1晩インキュベートし、TBST緩衝液で5回洗浄した後に、ブロットをペルオキシダーゼに結合した適切な2次抗体と1時間インキュベートした。次に、ブロットをTBST緩衝液で5回洗浄し、ECLシステム(Amersham Biosciences、iscataway、NJ、米国)及びKodak X-ARフィルムを使用してタンパク質抗体の複合体を検出及び可視化した。透過アダプタを有するSupervista S-12スキャナー(UMAX Technologies、Freemont、CA、米国)を使用して膜をデジタル化した。Public Domain NIH Imageプログラムを使用することで、密度測定で免疫反応バンドを比較した。吸光度曲線の下の領域は、同じブロットにおいて対照群で生じた領域のパーセンテージとして標準化した。タンパク質レベルは、対照群のタンパク質量のパーセンテージとして表した。
実施例9に記載したように、妊娠及び授乳している母ラットにDHA及び/又はウリジンを投与した。子ラットを、生後の20日目に屠殺し、脳の試料を用意し、リン脂質の測定を行った。DHAのみの投与により、細胞(DNA)当たりの脳のPC、PI、PE、及びスフィンゴミエリン(SM)がそれぞれ36、166、38、及び78%増加した。UMPにより、DHAの効果は、対照に対してそれぞれ66、210、68、及び99%にまでに顕著に増加した(表9)。これらの増加率は、成体のラットで観察された増加率よりも大きかった。値をタンパク質に標準化しても、同様な結果が得られた(表8)。したがって、DHA及び/又はウリジンを妊娠及び授乳している母ラットに投与することで、子ラットのリン脂質レベルを増加させることができる。
ラットの海馬細胞をNeurobasal+B27培地で成熟するまでに(3週間)培養した。実験の日に、細胞を、DHAを含む又は含まないDMEM+コリンで培養した。14Cコリンを加え、細胞をさらに2時間培養し、新たに形成された14C標識のPCを実施例1に記載したように抽出し、測定した。
Claims (23)
- 対象者における認知機能を向上させるための幼児用調整乳を製造するための、
(a)オメガ3脂肪酸、オメガ6脂肪酸、又はそれらの組み合わせ、
(b)ウリジン、そのアシル誘導体、ウリジン・リン酸又はCDPコリン、及び、
(c)コリン塩の使用であって、
前記幼児用調整乳が前記対象者に治療に効果的な量で投入されると、神経細胞又は脳細胞のシナプス膜に関連するリン脂質の量が増加することを特徴とする使用。 - 請求項1に記載の使用であって、
前記リン脂質は、ホスファチジルコリン、ホスファチジルエタノールアミン、ホスファチジルイノシトール、ホスファチジルセリン又はスフィンゴミエリンであることを特徴とする使用。 - 請求項1に記載の使用であって、
前記オメガ3脂肪酸は、ドコサヘキサエン酸、エイコサペンタエン酸、ドコサペンタエン酸又はαリノレン酸であることを特徴とする使用。 - 請求項1に記載の使用であって、
前記オメガ3脂肪酸で、前記神経細胞又は脳細胞のリン脂質の合成を増加させることによって、前記リン脂質のレベルを増加させるようにしたことを特徴とする使用。 - 請求項1に記載の使用であって、
前記ウリジン・リン酸は、ウリジン5´一リン酸(UMP)、ウリジン5´二リン酸(UDP)、ウリジン5´三リン酸(UTP)、又はUMP、UDP、若しくはUTPの塩であることを特徴とする使用。 - 請求項1に記載の使用であって、
前記コリン塩は、塩化コリン、酒石酸水素コリン、コリンクエン酸塩、コリン酒石酸塩、又は鉄−コリンクエン酸塩錯体であることを特徴とする使用。 - 請求項1に記載の使用であって、
前記対象者は、生後0〜6ヶ月であることを特徴とする使用。 - 請求項1に記載の使用であって、
前記対象者は、生後7〜24ヶ月であることを特徴とする使用。 - 対象者の知能を向上させるための幼児用調整乳を製造するための(a)オメガ3脂肪酸、オメガ6脂肪酸、又はその組み合わせ、(b)ウリジン、そのアシル誘導体、ウリジン・リン酸又はCDPコリン、及び、(c)コリン塩の使用であって、
前記対象者に、前記幼児用調整乳が治療に効果的な量で投入されると、対象者の知能が向上することを特徴とする使用。 - 請求項9に記載の使用であって、

前記コリン塩は、塩化コリン、酒石酸水素コリン、コリンクエン酸塩、コリン酒石酸塩、又は鉄−コリンクエン酸塩錯体であることを特徴とする幼児用調整乳。 - 請求項21に記載の幼児用調整乳であって、
前記オメガ6脂肪酸は、アラキドン酸又はリノール酸であることを特徴とする幼児用調整乳。
