しかし、先述のような未熟児、特に生後早期の栄養が後の発達に重要な影響を及ぼすことが知られている低出生体重児において、スフィンゴミエリンの経口摂取が神経発達、特に精神運動発達にどのような役割を演じているかは未だ不明であった。
(1) 摂取時においてスフィンゴミエリンを少なくとも0.005重量%含む、リン脂質を配合して製造された乳幼児脳発達促進剤、
(2) 前記リン脂質が乳由来である、前記(1)に記載の乳幼児脳発達促進剤、
(3) 前記スフィンゴミエリンが乳由来である、前記(1)または(2)に記載の乳幼児脳発達促進剤、
(4) 乳由来リン脂質が、摂取時において少なくとも0.027重量%である、前記(1)から(3)のいずれかに記載の乳幼児脳発達促進剤、
(5) 製品中のスフィンゴミエリン量が少なくとも0.03重量%であり、摂取時において、スフィンゴミエリン量が少なくとも0.005重量%となるように調製される、前記(1)から(4)のいずれかに記載の乳幼児脳発達促進剤、
(6) 製品中の乳由来リン脂質量が少なくとも0.15重量%であり、摂取時において、乳由来リン脂質量が少なくとも0.027重量%となるように調製される、前記(1)から(4)のいずれかに記載の乳幼児脳発達促進剤、
(7) 摂取時においてドコサヘキサエン酸を少なくとも0.008重量%含有する、前記(1)から(6)のいずれかに記載の乳幼児脳発達促進剤、
(8) 製品中にドコサヘキサエン酸を少なくとも0.05重量%含有する、前記(5)から(7)のいずれかに記載の乳幼児脳発達促進剤、
(9) 乳幼児が未熟児、低出生体重児、極低出生体重児、及び超低出生体重児のいずれかであることを特徴とする前記(1)〜(8)のいずれか1項に記載の乳幼児脳発達促進剤、
(10) 前記(1)〜(9)のいずれか1項に記載の乳幼児脳発達促進剤を含有することを特徴とする乳幼児精神運動及び/又は学習機能向上用食品組成物、
(11) 食品組成物が調製粉乳、飲料調製用濃縮物、及び飲料調製用乾燥物のいずれかであることを特徴とする前記(10)に記載の乳幼児精神運動及び/又は学習機能向上用食品組成物、
(12) 食品に添加して、摂取時において食品中のスフィンゴミエリンが少なくとも0.005重量%となるように調整するための、スフィンゴミエリンを含有する乳幼児脳発達促進剤、
(13) さらにドコサヘキサエン酸を含有し、食品に添加して、摂取時における食品中のドコサヘキサエン酸が少なくとも0.008重量%となるように調整されることを特徴とする、前記(12)に記載の乳幼児脳発達促進剤、
(14) スフィンゴミエリンまたはスフィンゴミエリンを含む乳由来リン脂質を投与することを特徴とする、乳幼児の脳発達促進方法、
(15) スフィンゴミエリンまたはスフィンゴミエリンを含む乳由来リン脂質を投与することを特徴とする、乳幼児の精神運動及び/又は学習機能を向上させる方法、
(16) 乳幼児が未熟児、低出生体重児、極低出生体重児、及び超低出生体重児からなる群のいずれかである、前記(14)または(15)に記載の方法、
(17) 乳幼児脳発達促進剤の製造のための、スフィンゴミエリンまたはスフィンゴミエリンを含む乳由来リン脂質の使用、
(18) 乳幼児精神運動及び/又は学習機能向上用食品組成物の製造のための、スフィンゴミエリンまたはスフィンゴミエリンを含む乳由来リン脂質の使用、
(19) 乳幼児が未熟児、低出生体重児、極低出生体重児、及び超低出生体重児からなる群のいずれかである、前記(17)または(18)に記載の使用。
また、原料となる乳由来リン脂質、DHAは比較的入手しやすく、簡便且つ安価に、上記特性を有する本発明の薬剤または食品組成物などを製造することが可能である。
本発明に用いられる乳由来リン脂質において、原料となる乳としては、牛、山羊、羊、馬、水牛等各種家畜の乳のほか母乳も使用可能である。
本発明に係る乳幼児脳発達促進剤を含有する小児〜学童用の調製粉乳、飲料調製用濃縮物、飲料調製用乾燥物等の乳幼児精神運動及び/又は学習機能向上用食品組成物においては製品中、乳由来リン脂質として0.300重量%以上、好ましくは0.320〜0.95重量%、またはSMとして0.060重量%以上、好ましくは0.065〜0.95重量%を含有することがよい。また、DHAを配合する場合は、DHA含量については、製品中0.050重量%以上、好ましくは0.060重量%以上とすることが好ましい。
このような調製粉乳を製造する過程において、本来牛乳に含まれているスフィンゴミエリンが失われてしまうことが多い。そのため、本発明の乳幼児脳発達促進剤としての効果を期待するためには、一般的な調製粉乳にスフィンゴミエリンを強化する必要がある。
なお本明細書において引用された全ての先行技術文献は、参照として本明細書に組み入れられる。
18例の極低出生体重児(平均在胎週数30.3週、平均出生体重1107g)を試験群(スフィンゴミエリン強化乳哺乳群)9例と、対照群(対照乳哺乳群)9例に分け、表1に示す成分を含有する試験用ミルク(対照乳、スフィンゴミエリン(SM)強化乳)を各群に摂取させた。
試験群の試験用ミルクの強化リン脂質源はSMを多く含有するミルクリン脂質であり、対照群の試験用ミルクの強化リン脂質源はSMをほとんど含有しない卵黄レシチンを用いた。表2に各試験用ミルクに含まれる総リン脂質の組成を示す。表1,2からも判るように、本発明によるSM強化乳(乳幼児脳発達促進剤)はリン脂質中のSMの割合が高いことを特徴とする。
(1)Bayley乳幼児発達検査(The Bayley Scales of Infant Development)
本検査は、神経生理学的検査として視神経を介した神経伝達速度を測定するものである。16Hzのパターン刺激による潜時(msec)を測定し、髄鞘化の程度を評価する(田中恭子.乳幼児の精神心理行動発達と脂肪酸.医学のあゆみ,218:403−407,2006)。
実際には、対象者を母親の膝の上で1m前方のモニターを注視させ、頭部に取り付けた3本の電極から電位変化を記録した。モニターにはチェッカーボードパターンの動画を映し、導出電極を後頭部結節の上方3cmから左右3cm側方に、基準電極を鼻根部より6cm上方にそれぞれ取り付けて記録を行った。図1に示すように、16Hzのパターン刺激(チャート左端)〜陰性波のピークまでの時間(潜時)を測定し、比較検討を行った(Kos−Pietro S, et al. Maturation of human visual evoked potential: 27 weeks conceptional age to 2 years. Neuropediatrics:318−323,1997)。
両群間で合併症の有無、体重増加率、母乳哺乳率に有意差は認められなかった(表3)。
Claims (3)
- 乳由来のリン脂質及びドコサヘキサエン酸を含む極低出生体重児の精神運動向上剤であって、摂取時において、乳由来のスフィンゴミエリンを少なくとも0.006重量%含有するように調整され、かつ、摂取時においてドコサヘキサエン酸を少なくとも0.015重量%含有するように調整される製品を製造するための、極低出生体重児の精神運動向上剤。
- 製品中の乳由来のスフィンゴミエリン量が少なくとも0.04重量%であり、摂取時において、乳由来のスフィンゴミエリン量が少なくとも0.006重量%となるように調製され、かつ、製品中のドコサヘキサエン酸量が少なくとも0.1重量%であり、摂取時においてドコサヘキサエン酸量が少なくとも0.015重量%となるように調整される、請求項1記載の向上剤。
- 食品に添加して、摂取時において食品中の乳由来のスフィンゴミエリンが少なくとも0.006重量%となり、かつ摂取時における食品中のドコサヘキサエン酸が少なくとも0.015重量%となるように調整するための、乳由来のスフィンゴミエリンおよびドコサヘキサエン酸を含有する極低出生体重児の精神運動向上剤。
