Description
市場で成功した前駆体プロテアーゼのほとんどが、サブチリシン309(又はサビナーゼ(サビナーゼ)(登録商標))である。サビナーゼのタンパク質設計は1898年に国際公開第89/06279号において初めて公開された。その後、サビナーゼのタンパク質設計に関連する多数の特許出願が、出願人及び他の企業、例えばジェネンコア・インターナショナル社、プロクター・アンド・ギャンブル、ユニリーバ(Unilever NV)等により出願がなされてきた。また、サビナーゼ変異体も、ノボザイムス社(Novozymes A/S)及びジェネンコア・インターナショナル社により市販されてきた。
改変Y167A+R170S+A194Pを含んでなる特定のサビナーゼ変異体は、国際公開第98/20115号で開示された。本出願では、この変異体をサブチリシンKLと呼ぶ。
配列比較は、GCGパッケージversion9.1のGAPルーティンにより、以下のパラメータを用い変異体に番号をつけることにより得られる。ギャップ生成ペナルティ(gap creation penalty)=8及びギャップ延長ペナルティ(gap extension penalty)=8、並びに他のパラメータを初期値に保つ。
従って、多くの欠失及び挿入はBASBPN(配列番号1)との関係で定義される。遺伝子操作によるポリペプチドに導入された改変の命名の詳細な説明については、国際公開第00/71691号、7-12ページを引用する。これは参照することにより本明細書に組み込まれる。
アミノ酸位置/残基の番号付け−別途記載しない限り本明細書におけるアミノ酸番号づけは、サブチラーゼBPN'(BASBPN)配列に対応する。さらなるBPN'配列の説明は、Siezen等によるProtein Engng. 4 (1991 ) 719-737を参照されたい。
酵素
以下の実施例においては、商業的に利用可能な酵素を使用する。Alcalase(登録商標)とサビナーゼ(登録商標)を比較標準として使用する。
サブチリシンKLはサビナーゼのY167A+R170S+A194P変異体(BPN'番号付けを使用)である。
プロテアーゼ適合性:
酵素のプロテアーゼ適合性は、各実施例に指示される洗剤組成物の調製、及び当該実施例に指示される期間経過後の他の酵素の残存活性の測定から決定する。
酵素活性:
酵素活性は、周知の標準手法を用いて測定する。
実施例において用いられる洗剤組成物は、以下に提示する組成物に従うモデル洗剤であるか、又は市販の液体洗濯洗剤のいずれかであり、商業用洗剤の例としてはTide、Era、Gain、Cheer、Wisk、All、Purex、Arm & Hammer、Sun、Great Value、Ariel、Persil、Total、Skip、Dash、Dixan、Ava、又は当該液体洗剤の任意の他のブランド拡充版もしくは用濃縮版が挙げられる。使用する市販の洗濯洗剤が酵素を含む場合は、使用前に85℃で5分間電子レジにて当該洗剤を熱することで不活性化した。モデル洗剤組成物Aは洗剤実施例1である。
Claims (13)
- 液体又はゲル洗剤組成物であって、サブチリシンKL(subtilisin KL)又はその変異体と、少なくとも1のリパーゼ;アミラーゼ;セルラーゼ;又はマンナナーゼとの組み合わせを含んでなり、
リパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼの含有量に対する、サブチリシンKL又はその変異体の含有量の重量比が、0.001から100である、前記組成物。 - リパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼの含有量に対する、サブチリシンKL又はその変異体の含有量の重量比が、0.01から10である、請求項1記載の洗剤組成物。
- リパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼの含有量に対する、サブチリシンKL又はその変異体の含有量の重量比が、0.5から5である、請求項1記載の洗剤組成物。
- リパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼの含有量に対する、サブチリシンKL又はその変異体の含有量の重量比が、1から3である、請求項1記載の洗剤組成物。
- 前記リパーゼが、フミコラ(Humicola)(サーモマイシス(Thermomyces))由来のリパーゼ、シュードモナス(Pseudomonas)リパーゼ、バチルス(Bacillus)リパーゼ及びこれらの化学的変異体又はタンパク質設計された変異体からなる群から選択される、請求項1から4のいずれか1項記載の洗剤組成物。
- 前記リパーゼが、H.ラヌギノサ(H. lanuginosa)(T.ラヌギノサス(T. lanuginosus))又はH.インソレンス(H. insolens)由来のリパーゼ、P.アルカリゲネス(P. alcaligenes)、P.シュードアルカリゲネス(P. pseudoalcaligenes)、P.セパシア(P. cepacia)、P.スツッツェリ(P. stutzeri)、P.フルオレセンス(P. fluorescens)、シュードモナス種SD705株又はP.ウィスコンシネンシス(P. wisconsinensis)由来のリパーゼ、B.サブチリス(B. subtilis)、B.ステアロサーモフィルス(B. stearothermophilus)又はB.プミルス(B. pumilus)由来のリパーゼ及びこれらの化学的変異体又はタンパク質設計された変異体からなる群から選択される、請求項5記載の洗剤組成物。
- 前記アミラーゼが、バチルス(Bacillus)由来のアミラーゼを含んでなる群から選択される、請求項1から6のいずれか1項記載の洗剤組成物。
- 前記アミラーゼが、B.リケニフォルミス(B. licheniformis)由来のアミラーゼを含んでなる群から選択される、請求項7記載の洗剤組成物。
- 前記セルラーゼが、バチルス(Bacillus)属、シュードモナス(Pseudomonas)属、マイセリオフソラ(Myceliophthora)属、フミコラ(Humicola)属、フザリウム(Fusarium)属、チエラビア(Thielavia)属、アクレモニウム(Acremonium)属由来のセルラーゼを含んでなる群から選択される、請求項1から8のいずれか1項記載の洗剤組成物。
- 前記セルラーゼが、フミコラインソレンス(Humicola insolens)、マイセリオフソラサーモフィラ(Myceliophthora thermophila)及びフザリウムオキシスポラム(Fusarium oxysporum)由来のセルラーゼを含んでなる群から選択される、請求項9記載の洗剤組成物。
- サブチリシンKL又はその変異体の含有量が0.001から5重量%であり、リパーゼ、アミラーゼ、セルラーゼ、又はマンナナーゼが存在する場合は、各含有量が0.001から5重量%である、請求項1から10のいずれか1項記載の洗剤組成物。
- サブチリシンKL又はその変異体と、少なくとも1のリパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼとの組み合わせの使用であって、非プロテアーゼ酵素の安定性が向上した、液体又はゲル型の水性洗剤組成物の調製のための使用。
- プロテアーゼと少なくとも1の非プロテアーゼ酵素を含んでなる液体又はゲル洗剤組成物において、プロテアーゼ酵素と他の酵素の組み合わせにおける、非プロテアーゼ酵素の安定性を向上させる方法であって、サブチリシンKL又はその変異体をプロテアーゼ酵素として用いて液体又はゲル洗剤組成物を調製することを含んでなり、前記少なくとも1の非プロテアーゼ酵素が、リパーゼ、アミラーゼ、セルラーゼ又はマンナナーゼから選択される、前記方法。
