Description
本発明は、容器本体と、該容器本体の底面に配置され細胞を3次元的に付着させて培養可能な3次元培養基材と、該3次元培養基材の上面を被覆し、細胞を2次元的に付着させて培養可能であり、かつ、培養温度より低い温度で液状化する温度応答性の平坦な膜状の2次元培養基材とを備える培養容器を提供する。
このように構成することで、3次元培養基材の内部にまで細胞を付着させることができ、より広い範囲で細胞を培養することができる。
このように構成することで、容器本体内の限られた範囲で3次元培養基材の表面積を大きくすることができ、より広い範囲で細胞を培養することができる。
本実施形態に係る培養容器100は、図1に示すように、細胞50(図3参照)を収容する容器本体1と、容器本体1を密閉する蓋5とを備えている。培養する細胞50としては、例えば、皮膚細胞、軟骨細胞、骨髄由来幹細胞、脂肪由来幹細胞等、接着性を有する生体組織由来の様々な細胞が挙げられる。
培養容器100を用いて細胞50を培養するには、図3に示すように、少数の細胞50を容器本体1の温度感受性シート2の表面全面に略均等に間隔をあけて播種する(細胞播種工程)。温度感受性シート2上に細胞50が播種されたら、容器本体1に蓋5をして所定の培養温度で保存する。この状態で、所定の細胞数まで細胞50を培養する(第1の培養工程)。

Claims (4)
- 容器本体と、
該容器本体の底面に配置され細胞を3次元的に付着させて培養可能な3次元培養基材と、
該3次元培養基材の上面を被覆し、細胞を2次元的に付着させて培養可能であり、かつ、培養温度より低い温度で液状化する温度応答性の平坦な膜状の2次元培養基材と
を備える培養容器。 - 前記3次元培養基材が、ハニカム構造または多孔質構造を有している請求項1に記載の培養容器。
- 前記3次元培養基材が、プリーツ構造を有する請求項1に記載の培養容器。
- 3次元培養基材の上面を被覆し培養温度より低い温度で液状化する2次元培養基材に細胞を2次元的に播種する細胞播種工程と、
前記培養温度に設定して前記2次元培養基材上の前記細胞を培養する第1の培養工程と、
前記培養温度より低い温度に冷却して前記2次元培養基材を液状化させ、該2次元培養基材に付着していた前記細胞を前記3次元培養基材に3次元的に付着させる継代工程と、
前記所定の培養温度に設定して前記3次元培養基材に付着した前記細胞を培養する第2の培養工程と
を備える細胞培養方法。
