Description
本開発の実施例としてラピッドプロトタイピング(RP;rapid prototyping)装備を利用した任意形状製作(SFF;solid freeform fabrication)方式で1層、1層積層して形状を製作する3Dプリンティング技術を利用して培養容器を製作し、滅菌は、電子ビーム加速器を使用して行った。
本発明の3次元細胞培養のための培養容器を利用した3次元細胞培養の効果を測定するための実験として次のような条件で実験を実施した。
-培養期間:7日間培養
-細胞計数:0、3、7日目細胞計数
-アルギネート混合:1.3%濃度
第2細胞は、脂肪由来間葉系幹細胞(Adipose derived mesenchymal stem cell,ADMSC)であって、第2細胞が接種された3次元構造体をビーズ形態に製作した。
支持体は、ポリカプロラクトンを利用して製作した。まず、ポリカプロラクトンをチューブに投入して90℃下で650~730kPaの圧力を加えた後には、エアコントローラーの吸入機能を利用して3次元支持体を製作し、電子ビーム加速度を利用して滅菌工程を処理した。
第2細胞が含まれた支持体を利用した3次元細胞培養は、第2細胞を25μlずつ4個のドロップ(drop)支持体上に接種した後、1時間の間培養し、1ml培地添加し、37℃、5%二酸化炭素の条件下で培養することによって、前記第2細胞が接種された支持体形態の3次元構造体を製造した。

- 3次元細胞培養のための培養容器において、
前記培養容器の底面上に位置する中空の柱のそれぞれの内部に形成されたウェル(well)と、前記ウェルの内部に3つまたは4つが互いに離隔して形成され、前記柱から突出した複数の支持部とを含み、
前記複数の支持部は、その最上端が前記柱の20~60%の高さに位置し、細胞が接種された3次元構造体に対する支持が可能な3次元細胞培養のための培養容器。 - 前記支持部は、前記培養容器の底面から離隔して位置することを特徴とする請求項1に記載の3次元細胞培養のための培養容器。
- 前記支持部は、前記培養容器の底面に接する形態であって、前記柱の側面に沿って支持部が形成されることを特徴とする請求項1に記載の3次元細胞培養のための培養容器。
- 前記ウェルの断面部を基準として、前記柱から前記支持部の終端までの距離は、前記ウェルの直径対比15~30%であることを特徴とする請求項1に記載の3次元細胞培養のための培養容器。
- 前記支持部と接する前記柱領域のうち少なくとも一部と前記柱領域に接する前記支持部領域のうち少なくとも一部が除去されたことを特徴とする請求項3に記載の3次元細胞培養のための培養容器。

前記複数の支持部は、その上端表面のうち少なくとも一部が凹状に掘られた形態で構成され、その最上端が前記柱の20~60%の高さに位置し、細胞が接種された3次元構造体に対する支持が可能な3次元細胞培養のための培養容器。 - 前記支持部の上端表面は、端部領域と中央部領域に区分され、前記中央部領域は、凹状に掘られた形態であり、前記端部領域は、フラットにしたり突出方向に凸状に突出した形態であり、前記支持部の上端表面で、前記端部領域は、10~30%、前記中央部領域は、70~90%の面積を占めることを特徴とする請求項6に記載の3次元細胞培養のための培養容器。

第2細胞が接種された3次元構造体を前記支持部上に位置させて共培養する共培養段階と、を含み、
前記準備段階で、前記支持部の最上端は、前記柱の20~60%の高さに位置する3次元細胞共培養方法。 - 前記共培養段階の後に、1~5日間隔で前記培養液を交替する交換段階;をさらに含むことを特徴とする請求項9に記載の3次元細胞共培養方法。
