Description
(A)エタノール 5〜50質量%
(B)カテキン類 0.00625〜0.2質量%
を含有する抗菌組成物を提供するものである。
当該製茶された茶葉には、(1)煎茶、番茶、玉露、てん茶、釜煎り茶などの緑茶類;(2)総称して烏龍茶と呼ばれる鉄観音、色種、黄金桂、武夷岩茶などの半発酵茶;(3)紅茶と呼ばれるダージリン、ウバ、キーマンなどの発酵茶が含まれる。
抽出助剤としては、アスコルビン酸ナトリウム等の有機酸又はこれら有機酸塩類が挙げられる
例えば、当該茶抽出物(茶カテキンともいう。)は、特開昭59-219384号、特開平4-20589号、特開平5-260907号、特開平5-306279号等に詳細に例示されている方法で調製することができる。また、市販品を用いることもでき、斯かる市販品としては、三井農林(株)「ポリフェノン」、(株)伊藤園「テアフラン」、太陽化学(株)「サンフェノン」、DSMニュートリショナル・プロダクツ「テアビゴ」、サントリー(株)「サンウーロン」等が挙げられる。
また、茶葉中のカテキン類の大部分はエピ体カテキン類として存在しており、このエピ体カテキン類を用いて熱や酸やアルカリ等の処理により立体異性体である非エピ体に変化させることができる。従って、非エピ体カテキン類を使用する場合には、緑茶類、半発酵茶類又は発酵茶類からの抽出液や茶抽出液の濃縮物を水溶液にして、例えば40〜140℃、0.1分〜120時間加熱処理して得ることができる。また非エピカテキン類含有量の高い茶抽出液の濃縮物を使用してもよい。それらは単独又は併用してもよい。
本発明の組成物が有効な細菌としては、グラム陽性菌又はグラム陰性菌に限定されない。
例えば、グラム陰性菌としては、S. dysenteria(赤痢菌A亜群),S. flexneri(赤痢菌B亜群),S. boydii(赤痢菌C亜群),S. sonnei(赤痢菌D亜群)等の赤痢属(Shigella)細菌:ブルセラ属(Brucella)細菌:E. coli.O157等の大腸菌群(Escherichia coli):S. typhi(チフス菌),S. paratyphi A(パラチフスA菌),S. paratyphi B(パラチフスB菌),S. Typhimurium (ネズミチフス菌)S. Enteritidis(ゲルトネル菌)等のサルモネラ属(Salmonella)細菌:V. cholerae(コレラ菌),V. parahaemolyticus(腸炎ビブリオ)等のビブリオ属(Vibrio)細菌:緑膿菌(P. aeruginosa)等のシュードモナス属(Pseudomonas)細菌等が挙げられる。
また、例えば、グラム陽性菌としては、枯草菌(B. subtilis)、炭疽菌(B. anthracis)、セレウス菌(B. cereus)等のバシラス属(Bacillus)細菌:リステリア・モノサイトゲネス菌(L. monocytogenes)リステリア・イバノヴィ菌(L. ivanovii)、リステリア・シーリゲリー菌(L. seeligeri)等のリステリア属(Listeria)細菌:Al. acidoterrestris(旧B. acidoterrestris)等のアリシクロバチルス属(Alicyclobacillus)細菌:S. aureus(黄色ブドウ球菌),S. pyogenes等のブドウ球菌属(Staphylococcus)細菌:C. botulinum(ボツリヌス菌),C. perfringens(ウェルシュ菌),C. difficile, C. sporogens等のクロストリジウム属(Clostridium)細菌:Leuconostoc mesenteroides等のリューコノストック属(Leuconostoc)細菌:Desulfotomaculum nigrificans等のデスルフォマクルム属(Desulfotomaculum)細菌:Enterococcus faecalis等のエンテロコッカス属(Enterococcus)細菌等が挙げられる。
このときの組成物又は製品のpHは、本発明の効果を阻害しない範囲であれば特に限定されないが、具体的には組成物又は製品を水溶液(20℃)とした時のpHを6〜9とするのが好ましく、6.5〜8とするのがより好ましい。
斯くして製造された本発明の組成物は、化粧品、医薬部外品、医薬品、又はハウスホールド製品の何れにも配合又は使用することができ、例えば、口紅、乳液、保湿パック、美容液、化粧水等の化粧品:液剤、ゲル剤、クリーム剤、パップ剤、エアゾール剤、ローション剤等の皮膚外用剤:洗顔料、メイク落とし、ボディシャンプー、ハンドソープ等の皮膚洗浄剤、シェービングフォーム、シャンプー等の毛髪洗浄剤、デオドラント剤等のトイレタリー用品:歯磨剤、洗口剤、口中清涼剤、義歯用品等のオーラルケア用品:台所用洗剤、衣類洗浄剤や柔軟剤、トイレ洗浄剤等の洗浄剤:殺虫剤、防虫剤、除湿剤、除菌剤、抗菌剤、消臭剤、芳香剤等の家庭用化学製品に配合することができる。また、紙製品、生理用品、紙おむつ、医療品等の衛生用品等には、例えば紙おむつ等に本発明の組成物を吹き付けることやシート状の紙・不職布製品に本発明の組成物を含む抗菌剤等を含浸することで使用できる。また、コンタクトレンズ洗浄・保存剤等に配合できる。
ここで、ハウスホールド製品とは、衣料・台所・住居用洗剤、柔軟仕上剤、掃除用紙製品、トイレタリー用品及びペット用品等広く一般家庭において使用される製品をいう
また、上記化粧品等の他、保存剤、防腐剤、抗菌剤や抗菌剤等として飲食品に使用することができる。
試験菌として、大腸菌:Escherichia coli O157:H7株、黄色ブドウ球菌:Staphylococcus aureus IFO3060株を用いた。
(2)試験用溶液調製
a)50%LB培地:組成:0.5%トリプトン、0.25%酵母エキス、pH6.5に調製後、オートクレーブ滅菌し、50%LB液体培地を作製した。
b)カテキン類溶液:カテキン類(「POLYPHENON 70A」カテキン製剤、三井農林(株)製)を1.0%(W/V)となるように純水に溶解し、更に10000, 5000, 2500, 1250, 625, 312.5, 156, 78,0ppmになるように各溶液を調製後、濾過滅菌(孔径0.2μm、関東化学社製)した。
c)エタノール溶液:エタノールを純水に溶解し、1, 5, 10, 50, 80質量%の濃度になるように調製後、濾過滅菌(孔径0.2μm、関東化学社製)した。
上記の50%LB培地(8mL)、カテキン類溶液(1mL)及びエタノール溶液(1mL)を混合した試料用溶液(10mL)に、105〜106CFU/mLに調製した菌液100μLを添加(終菌数103〜105CFU/mL)し、表1〜2に示すエタノール及びカテキン類の最終濃度並びにpHに調製した。この菌を接種した試料用溶液を、37℃で48時間静置(時々ボルテックス)した。静置後の生菌数を平板塗抹法により求めた。TSA培地上のコロニー数から生菌濃度を算出した。
TSA(Tryptic Soy Agar)寒天培地(Becton Dickinson 社):1.7%カゼイン分解物、0.3%大豆酵素分解物、0.25%デキストロース、0.5%塩化ナトリウム、1.5%寒天、pH7。
下記の表1〜2に示すように、大腸菌及び黄色ブドウ球菌共に、エタノール、カテキン類単独で抗菌作用が認められなかった低濃度でも、エタノール及びカテキン類を併用すると抗菌効果が相乗的に高まった。
Claims (2)
- 次の成分(A)及び(B):
(A)エタノール 5〜8質量%、
(B)カテキン類 0.0125〜0.1質量%、
を含有する大腸菌群(Escherichia coli)用抗菌剤組成物。 - 化粧品、医薬部外品、医薬品、及びハウスホールド製品から選ばれる製品に、次の成分(A)及び(B)を、製品中の含有量が(A)エタノール 5〜8質量%、及び(B)カテキン類 0.0125〜0.1質量%となるように配合することを特徴とする、前記製品に大腸菌群(Escherichia coli)に対する抗菌性を付与する方法。
