Description
本発明は、消毒の分野に関する。
例えば、皮膚、加圧滅菌不可の医療機器、病棟、手術室、壁、手摺り、空調用ダクト等の表面の消毒は、依然として感染コントロールに関し、最も問題となる領域の1つである。
第1態様によれば、本発明は、1.5MHzより上になるように選択した周波数の超音波エネルギーを、少なくとも1つの界面活性剤と組み合わせた消毒薬を含む液体調合物に印加して、噴霧化された消毒薬生成物を生成するステップを特徴とする消毒方法にある。
ここで、本発明は、好適な実施態様を参照しつつ実例で説明する。
超音波および音響振動は、エーロゾルを生成することが知られている。超音波を用いて液体を霧状化する機構は、液体/空気界面近くでのキャビテーションバブルの微小噴出から成り、バブルが壊れることで液体が散乱される。ポンプで空気を送り込むことによるか、またはベルヌーイ効果によるかの何れかにより発生させた空気流を使用して、液滴のミストを液体バルクから分離して、対象物上へ向けて送出することができる。
(実施例1)
有効性のデータは、以下の消毒薬に関して得られた。
A.6%w/w 過酸化水素(pH=3)、94%w/w 水
B.6%w/w 過酸化水素+15%w/w n−プロパノール+0.3%w/w イルガサン(Irgasan) DP300+0.02%w/w PVPK15+0.5%w/w STPP(pH=7)+2%w/w LAS+2%w/w テリック(Teric)12A3
C.蒸留水で1:50で希釈した5%w/w ペルオキシ酢酸
D.蒸留水中、2%w/w グルコン酸クロルヘキシジン+15%w/w n−プロパノール
(装置)
ネブライザの操作原理は、他で(例えば、1936年Trans.Farady Soc.v.32,p1532でケー・ソルナー(K.Sollner)により)説明されている。超音波ネブライザの主な要素は、高周波発生器、ピエゾセラミックトランスデューサ、および噴霧化する溶液用の貯蔵器である。微細なエーロゾルの生成では、共鳴周波数を印加することによって、トランスデューサに機械的に振動させることを必要とする。高周波振動は、溶液の表面に近い部分に集中させて、「超音波源泉」を作り出す。一旦、エネルギーがある一定の閾値を超えると、液滴は、細かく割れて、空気の流れにより貯蔵器外へ強制排出される。
増殖性の緑膿菌(ATCC(米国の菌株保存機関)15442)、ミコバクテリウムテラ(Mycobacterium terrae)(ATCC15755)、大腸菌(ATCC8739)、および黄色ブドウ球菌(S.aureus)の播種材料は、一晩培養してから調製し、約108乃至109cfu/mL含有させた。
播種材料の20マイクロリットルは、滅菌済み(180Cのオーブンで3時間)の10×20mmガラスプレート上に置き、36℃のインキュベータ内で40分間乾燥した。滅菌済みの(180℃で3時間)ガラスペニシリンダは、10分間播種材料中に浸して、次に、40分間、36℃のインキュベータ内に入れる。
「試験無し」 試験せず
「合格」 10個のレプリカ中少なくとも10までについて、試験した有機体の完全殺菌が達成されて、生き残りは皆無であった。
「増殖」 生存試験有機体を担持していたキャリヤの数。
密閉系(図2)を使用してアルギナート歯の印象に対する消毒薬の有効性を評価する。
音波処理済み、および音波処理無しの過酸化水素溶液の殺菌の有効性を比較するために、以下の実験を行った。緑膿菌および増殖性枯草菌の0.1mL播種材料を、ガラスプレートの面積20×15mmに亘って一様に広げて、40分間乾燥し、次に、0.05mLの4%過酸化水素を用いて、2分間処理した。残っている微生物は、実施例1で説明したように、トリプトンソーヤ培養液の中へ移して、培養基で培養した。同じ汚染されたプレートは、同じ4%過酸化水素溶液の噴霧化されたミストを用いて、15秒間処理し、次に、1分45秒間放置した。各プレート上に凝縮された過酸化水素の合計量は、0.01mLより下であった(すなわち、引用の実験より少なくとも10倍少なかった)。結果は、以下の通りであった。バルク溶液を用いた実験では、観察された生き残りレベルは、4×103cfu/mLであり、噴霧化した過酸化水素では、全細菌を死滅させて、トリプトンソーヤ培養液のあるペトリ皿上または試験管内の何れでも、生き残りは検出されなかった。
1%次亜塩素酸塩消毒溶液を使用して、実施例2で説明した同じモデルから作られた下顎の歯の印象を消毒した。3通りの異なるモードの消毒薬供給量を比較した。
1.微細スプレイハンドポンプ(AC Colmack Ltd)を用いて霧状化した。消毒薬は、印象上に噴霧して、10分間放置した。
2.3分間、40KHzマイクロミスト超音波アトマイザ(Misonix Inc)を用いて霧状化し、次に、もう8分間放置した。合計接触時間は、10分である。
3.3分間、2.64MHzMousson超音波ネブライザを用いて霧状化し、次に、7分間、噴霧化処理室(密閉系)内に放置した。合計接触時間は、10分である。
界面活性剤含有および含有無しの音波処理を受けた消毒薬の殺菌の有効性は、以下のとおり比較した。
水溶液:
CL: 0.5%茨亜塩素酸ナトリウム
CLA: 0.5%次亜塩素酸ナトリウム+0.5%LAS
CLN: 0.5%次亜塩素酸ナトリウム+0.5%PEG6200(BASF)
HP: 1%過酸化水素
HPA: 1%過酸化水素+0.5%LAS
HPN: 1%過酸化水素+0.5%PEG6200
HPE: 1%過酸化水素+5%エタノール
上記水溶液を閉じられた処理室において(Musson−1 2.64MHz超音波ネブライザを使用して)、ガラスプレート上に、緑膿菌(P.aeroginosa)(109cfu/mL)および増殖期枯草菌の乾燥した播種材料を、凝縮された噴霧剤で一様に覆われるまで噴霧化した。次に、ガラスプレートは、生き残りの微生物を定量するために、実施例1で説明したように、トリプトンソーヤ培養液の中へ移した。凝縮された消毒薬の合計量は化学天秤を使用して重さを量り、プレートを噴霧剤で一様に覆うのに要した時間を記録した。
Claims (16)
- 1.5MHzより上の周波数の超音波エネルギーを、消毒薬と界面活性剤とを含む、水溶液である液体調合物に印加して、霧状液滴を生成することと、
前記霧状液滴を、消毒される晒された表面と接触させて消毒を行うことと、
を含む消毒方法であって、
噴霧化される前記調合物が、
過酸化水素、過酢酸、過硫酸塩、過炭酸塩、及び、それらの混合物から成るグループから選択される消毒薬と、
エトキシル化アルコール、ドデシルベンゼンスルホン酸塩、並びに、アルコールとエチレンオキシドおよび酸化プロピレンのブロック共重合体から成るグループから選択される少なくとも1つの界面活性剤であって、液滴の90%が0.8〜2.0マイクロメートルのマスメジアン空気動力学的直径を有する前記霧状液滴を生成するよう前記調合物の表面張力を変えるために十分な量の前記少なくとも1つの界面活性剤と、
を含む水溶液である、消毒方法。
- 前記超音波エネルギーが、噴霧化室で前記液体調合物に印加される、請求項1に記載の方法。
- 前記超音波エネルギーが、噴霧化持続期間の間、組み合わせて選択された超音波周波数で印加されて、前記霧状液滴に晒された対象物の消毒を行う、請求項1または2に記載の方法。
- 前記超音波周波数が、2MHzよりも上である、請求項1〜3の何れか1項に記載の方法。
- 前記消毒が40℃より下で行われる、請求項1〜4の何れか1項に記載の方法。
- 前記界面活性剤がエトキシル化アルコールである、請求項1〜5の何れか1項に記載の消毒方法。
- 密閉された消毒処理室に存在するかあるいは同処理室と連通する密閉された超音波処理室内で超音波の印加により、前記液体調合物の噴霧化を行う、請求項1〜6の何れか1項に記載の方法。
- 前記霧状液滴が、消毒処理室内で生成する、請求項1〜7の何れか1項に記載の方法。
- 前記消毒される物品が、吹き付け乾燥される、請求項1〜8の何れか1項に記載の方法。
- 前記液体調合物が、水に比して蒸気圧が高い少なくとも1つの液体物質を含む、請求項1〜9の何れか1項に記載の方法。
- 蒸気圧が高い前記少なくとも1つの液体物質が、乾燥時間を短くできるように選択される、請求項10に記載の方法。
- 蒸気圧が高い前記少なくとも1つの液体物質が、アルコール、エーテル、炭化水素、及び、エステルから成るグループから選択される、請求項10に記載の方法。
- 前記消毒ステップに続いて、中和剤を用いて前記消毒薬を中和するステップを更に含む、請求項1〜12の何れか1項に記載の方法。
- 前記中和剤が、噴霧化された形態で用いられる、請求項13に記載の方法。
- 前記中和剤が、ペルオキシダーゼ酵素またはチオ硫酸ナトリウムである、請求項13に記載の方法。
- 消毒する物品が歯の印象形態である、請求項1〜15の何れか1項に記載の方法。
