Description
Claims (14)
- 複数の血小板を有する血液試料中の血小板を計数するための方法であって、
被分析試料を静止状態に保つように構成された分析チャンバに前記試料を入れるステップであって、前記チャンバが第1のパネルと第2のパネルとにより画定され、それらのパネルは双方とも透明である、ステップと、
着色剤を前記試料と混和するステップであって、この着色剤が、1つ又は複数の所定の第1の光の波長に曝露すると血小板に蛍光を発生させるように機能する、ステップと、
血小板を含む前記試料の少なくとも一部分を前記第1の波長で照明するステップと、
前記試料の前記少なくとも一部分を撮像するステップであって、ある強度を有する血小板からの蛍光発光を示す画像信号の生成を含む、ステップと、
前記画像信号を用いて、血小板をそれらの蛍光発光により同定するステップと、
前記試料中に同定された個々の血小板の平均蛍光発光強度値を決定するステップと、
前記試料中の血小板凝集塊を、それらの蛍光発光、面積、形状、及び粒状性のうちの1つ又は複数を用いて同定するステップと、
前記試料中の個々の血小板について決定された前記平均蛍光発光強度値を用いて、各血小板凝集塊中の血小板を計数するステップと、
を含む、方法。 - 前記血液試料が実質的に無希釈である、請求項1に記載の方法。
- 前記血液試料が全血である、請求項1に記載の方法。
- 前記試料中に個々に存在する前記血小板を計数し、各血小板凝集塊中の血小板と前記試料中に個々に存在する血小板との合計として前記試料中の血小板の総数を決定するステップをさらに含む、請求項1に記載の方法。
- 前記試料中の巨大血小板を同定するステップをさらに含む、請求項1に記載の方法。
- 前記試料中の前記巨大血小板が、蛍光発光、面積、及び形状のうちの1つ又は複数によって同定される、請求項5に記載の方法。
- 前記試料中の前記巨大血小板のうちの少なくとも1つの面積を決定するステップをさらに含む、請求項5に記載の方法。
- 前記試料中に同定された個々の血小板についての平均面積を決定し、前記少なくとも1つの巨大血小板の面積を前記平均血小板面積と比較するステップをさらに含む、請求項7に記載の方法。
- 前記試料中に同定された個々の血小板についての平均面積と前記個々の血小板の面積の統計的偏差とを決定し、前記少なくとも1つの巨大血小板の面積を、個々の血小板面積の前記統計的偏差に基づく値と比較することにより、前記少なくとも1つの巨大血小板を同定するステップをさらに含む、請求項7に記載の方法。
- 前記少なくとも1つの巨大血小板の面積を、所定の値と比較することにより前記少なくとも1つの巨大血小板を同定するステップをさらに含む、請求項7に記載の方法。
- 前記試料中に同定された個々の血小板についての平均面積を決定するステップをさらに含む、請求項7に記載の方法であって、前記各血小板凝集塊中の血小板を計数するステップが、前記試料中の個々の血小板についての平均面積を用いることをさらに含む、方法。
- 前記血小板を含む試料の少なくとも一部分を、1つ又は複数の第2の波長で照明するステップと、
前記試料の少なくとも一部分を撮像するステップであって、前記試料中の1つ又は複数の構成成分の光学濃度を示す画像信号の生成を含む、ステップと、
前記画像信号を用いて、前記試料中の1つ又は複数の構成成分の光学濃度を決定するステップと、
前記光学濃度を用いて、血小板と1つ又は複数の構成成分とを互いに識別するステップと、
をさらに含む、請求項1に記載の方法。 - 前記試料の少なくとも一部分を、ヘモグロビンにより吸収される1つ又は複数の第2の波長の光で照明することにより、ヘモグロビンを含む前記試料中の構成成分と血小板を識別するステップをさらに含む、請求項1に記載の方法。
- 血液試料中の血小板を計数するための装置であって、
第1のパネルと第2のパネルとにより画定された、それらのパネルが双方とも透明な、被分析試料を静止状態に保つように構成された分析チャンバであって、ある量の着色剤が前記チャンバ内に入れられ、前記着色剤が、1つ又は複数の所定の第1の光の波長に曝露すると血小板に蛍光を発生させるように機能する、チャンバと、
前記血小板を含む試料の少なくとも一部分を前記第1の波長で照明し、
照明器と解像装置とを含む撮像ユニットであって、前記試料の少なくとも一部分を撮像し、且つある強度を有する血小板からの蛍光発光を示す画像信号を生成するように機能するユニットと、
前記画像信号を用いて、血小板をそれらの蛍光発光により同定し、及び前記試料中に同定された個々の血小板についての平均蛍光発光強度値を決定し、及び前記試料中の血小板凝集塊を、それらの蛍光発光、面積、形状、及び粒状性のうちの1つ又は複数を用いて同定し、及び前記試料中の個々の血小板について決定された前記平均蛍光発光強度値を用いて、各血小板凝集塊中の血小板を計数するように構成されたプログラム可能分析器と、
を含む、装置。
