Description
本発明は、正常な形質膜組織化の摂動および改変を受けている細胞、すなわち、細胞死を実行中の細胞、アポトーシス細胞または活性化された血小板に選択的に結合する化合物に関する。本発明は、さらに、この化合物を、医療行為において、診断用および治療用目的のために利用する方法を提供する。
無傷の真核細胞の形質膜(外膜)は、高度に組織化された構造を特徴とする。この高レベルの膜組織化は、とりわけ、この膜を構成する特定の脂質の分子構造;膜を構成する種々の脂質種の比率;この膜の外側と内側リーフレット間でのリン脂質の分布によって;および膜タンパク質構成要素によって決定される。
本発明の一つの態様において、形質膜の正常な組織化の摂動を受けている細胞(PNOM細胞)に選択的に結合し得るが、形質膜の正常な組織化を維持している細胞(これを、以下、「正常細胞」と定義する)には、より低程度で結合する化合物を提供する。PNOM細胞は、本発明の一つの実施形態において、死亡過程を実行中の細胞である。本発明の一つの実施形態ではこの細胞はアポトーシス細胞であり、別の実施形態では、この細胞は、活性化された血小板であり得る。本発明は、さらに、PNOM細胞に選択的に結合するこれらの化合物を使用することによりPNOM細胞を検出する方法に関する。本発明の別の実施形態では、式I〜XIVに示す構造で表される化合物を提供する。
本発明の別の実施形態において、PNOMを選択的に標的とし、式(II):
本発明は、形質膜の正常な組織化の摂動を受けている細胞(PNOM細胞)に対して選択的な結合を行うことができるが、形質膜の正常な組織化を維持している細胞には、より低程度で結合する化合物に関する。PNOM細胞は、死亡過程を実行中の細胞、アポトーシス細胞および活性化された血小板から選択される。本発明は、さらに、PNOM細胞に選択的に結合する化合物を用いることによりPNOM細胞を検出する方法に関する。
本発明の別の実施形態において、PNOM細胞を選択的に標的とする化合物であって、式(II):
と示す、すべての化合物が共有するモジュールの活性を、少なくともある程度含む。本発明の一つの実施形態において、Rはブチルである。
(i) 膜リン脂質の無秩序化(scrambling)(ホスファチジルエタノールアミン(PE)および負に帯電したホスファチジルセリン(PS)の細胞表面上に露出した状態で)。細胞表面上でのPSの露出により、負の表面電位がもたらされ、プロトンの吸引によって膜界面に大きな塊(bulk)が形成される;
(ii)膜の外側リーフレット内におけるアミノリン脂質(PEおよびPS)の割合の増加の結果、膜の外側リーフレットの界面におけるプロトン電流(界面プロトン電流、IPC)の増強がもたらされる。この増強は、PEおよびPSが外膜リーフレット内のホスファチジルコリン(PC)およびスフィンゴミエリン(sphingomeylin)(SM)と置き換わるにつれて、プロトン転移反応に関与しやすい官能基の数の実質的な増加によるものである。PEは第一級アミンを含み、一方、PSは第一級アミンとカルボキシル基を含む。対照的に、PCおよびSMは、各々、永久的な正電荷を有する第四級アンモニウムを含み、したがって、プロトン転移反応には関与し得ない;
(iii)加えて、アポトーシス膜は、膜構成要素のパッキングレベルの低下および膜流動性の増加を特徴とする;

(i)酸−アニオン対が形成され、このとき、例外的に、強い水素結合が、プロトン付加されたカルボキシル基とプロトン付加されていないカルボキシル基との間に形成される。この水素結合は強力であり、対称的で、共鳴および互変異性化(tautamerization)によって安定化されている。
(ii)これは、4個のカルボキシル原子の上部に負電荷の分布をもたらす、すなわち、部分的に非局在化させる。
(iii)強酸−アニオン水素結合はこの分子を剛性化し、バルキーで剛性の平坦な6員環を生成する。これは、部分的に非局在化した負電荷を有し、カルボキシル二重(bouble)結合の上部にπ電子雲を含む。かかる成分(element)は、本発明の化合物の作用機序の非限定的な仮定によれば、膜界面への比較的有利な浸透を行ない得る。しかしながら、そのバルキーな剛性構造は、緩くパッキングされた膜(emebrane)、すなわち、アポトーシス膜(memebrane)に選択的に結合し、高度にパッキングされた膜(例えば、健常細胞の形質膜)への結合を排除するよう指令する。したがって、このような立体構造の特徴は、アポトーシス膜への結合における選択性を促進する。
A:この化合物は水溶液中に存在し、したがって、両カルボキシル基は脱プロトン化されている、すなわち負に帯電しており、この化合物は高度に可溶性である。
(iii)「クリアランスアプローチ」と称する本発明の第3のアプローチによれば、PNOM細胞に対する本発明の化合物の選択的な結合は、この化合物の固相支持体との結合を介して、体液(例えば、血液)を、その凝血促進性により潜在的に有害であり得るPNOM細胞から浄化するために利用される。

本発明の医薬組成物、ならびに診断用組成物は、任意の公知の経路、とりわけ、経口、静脈内、腹腔内、筋肉内、皮下、舌下、眼内、鼻腔内または局所投与、または大脳内投与によって投与され得る。担体は、所望の投与様式に応じて選択するのがよく、任意の公知の成分、例えば、溶媒;乳化剤(emulgator)、賦形剤、タルク;香料;着色料などを含み得る。医薬組成物は、所望により、他の医薬的に活性な化合物(これは、疾患を治療するため、副作用を排除するため、または活性成分の活性を増大させるために使用される)もまた含み得る。
本発明を理解するため、およびこれを実際にどのように行い得るかがわかるように、以下の実施例:本発明の化合物の合成に関する実施例;および死亡過程を実行中の細胞に対する選択的な結合における本発明の化合物の性能に関する実施例を記載する。本発明の化合物の検出を可能にするため、これをトリチウムで放射標識し、損傷を受けた領域内への取込みを測定することにより、またはオートラジオグラフィー法によって検出した。一部の実施例では、化合物を蛍光標識、すなわちダンシルアミド基への結合によって標識し、蛍光顕微鏡検査によって検出した。アポトーシス細胞に対するこの化合物の結合の選択性は、インビトロで組織培養物において、およびインビボで細胞死を中大脳動脈の閉塞によって誘導した脳卒中のマウスモデル、腎虚血性または毒性傷害のマウスモデル、黒色腫のマウスモデル、結腸癌のマウスモデル、および実験的自己免疫性脳脊髄炎(EAE)において、多発性硬化症に関連するマウスモデルにおいて示された。
(参考例1)
ジ−t−ブチルマロネート(5mL)を、1当量のNaHを含むジメチルホルムアミド(DMF)で脱プロトン化し、1当量のヨウ化n−ブチルを、水素発生が止まった後、添加した。反応混合物を50℃まで14時間加熱した。5.8gのジ−t−ブチル、ブチルマロネート(2)を95%収率で、カラムクロマトグラフィーを用いて得た。2(3.8g)を、NaOCH3(0.05当量、触媒量)およびアクロレイン(1.1当量)を含むトルエンで処理し、1.26gのアルデヒド(3)を30%収率で得た。化合物3を、次いで、NaBH4(1.05当量)を含むエタノール/水(8:1v/v)の混合物で2時間反応させた。処理(work−up)およびフラッシュクロマトグラフィー後、純粋なアルコール4を得た(93%)。得られた生成物を、1.1当量の塩化メタンスルホニル(MsCl)および2.2当量のトリエチルアミン(Et3N)で処理し、メシラート化合物5を97%収率で得た。この生成物は本質的に純粋であり、さらに精製を行なわずに次の反応に直接持ち越した。
4−ブロモ−1−ブタノール(1)3gを、1.5当量の3,4−ジヒドロ−2H−ピランおよび0.1当量のp−トルエンスルホン酸ピリジニウム(PPTS)を含む135mLのCH2Cl2で処理した。処理および精製後、1.45g(33%)の生成物2を得た。1.0当量のジエチルメチルマロネートを、1当量のNaHで脱プロトン化し、1.0当量のブロミド2を、触媒量のKIとともに50℃で添加した。完全な変換が10時間に観察され、90%収率が得られた。テトラヒドロピラン(THP)のPPTS含有エタノールでの脱保護は、55℃で順調に進行した。処理後、定量的収率のアルコール4が得られ、メシル化反応に直接使用した(前述と同様)。得られたメシラート5を用い、クリプトフィックスの反応を、上記のようにして適用した。化合物6を68%収率で得た。化合物6(233mg)を、2N NaOH/EtOH(30mL/5mL)で50℃にて処理し、NST−ML−Fを約99%収率(190mg)で得た。この化合物のNMRデータは、以下のとおりである。1HNMR(300MHz,CDC13)艪P1.89(bs,2H),4.46(dt,Jt=5.9Hz,Jd=47.2Hz,2H),1.99−1.92(m,2H),1.82−1.64(m,2H),1.50(s,3H),1.50−1.40(m,2H),13CNMR(75MHz,CDC13)艪P78.6,85.1,82.9,54.2,35.5,31.0,30.8,20.8,20.7,20.2;19FNMR(282MHz,CDC13)艨|219.0,MS(EI).
実験手順
培養ジャーカット細胞(ヒト成人T細胞白血病細胞)を、10%ウシ胎仔血清(FCS)、2mMのL−グルタミン、lmMのピルビン酸ナトリウム、1mM HEPESおよび抗生物質(100単位/mlペニシリン;100μg/mlストレプトマイシンおよび12.5単位/mlのナイスタチン)を加えたRPMI培地(Beit−Haemek,Israel)中の懸濁液中で成長させた。アポトーシスの誘導前に、培地をHBSバッファー(10mM HEPES;140mM NaCl、1mM CaCl)と交換した。次いで、アポトーシスを、CD95(0.1ug/107細胞/ml)での処置によって誘発した。その結果、著しい割合の細胞がアポトーシスとなった。非処置細胞を対照とした。次いで、対照細胞およびアポトーシス細胞の両方を40分間、室温でインキュベートした後、30分間氷上で、(2μCi/107細胞/0.5ml)トリチウム標識NST 200およびNST 205とともにインキュベートした。
図2Aから明白にわかるように、アポトーシス細胞は、非アポトーシス細胞と比べ、著しく高いNST200の取込み(8倍を超える)を示した。この実験は、NST200が、アポトーシス細胞に選択的に結合でき、その検出のためのマーカーとしての機能を果たし得ることを示す。同様に、図2Bからわかるように、アポトーシス細胞は、非アポトーシス細胞と比べ、高いNST205の取込みを示し、この効果は、カスパーゼ阻害剤であるZVAD(フルオロメチルケトンペプチド(peptid)(V−バリン、A−アラニン、D−アスパラギン酸塩)カスパーゼ阻害剤)を添加している間に完全に逆転した。
(参考例4)
HeLa S3 細胞(ATCC CCL−2.2)を、2mMのL−グルタミン;100単位/mlのペニシリン;100μg/mlのストレプトマイシン;12.5単位/mlのナイスタチンおよび10%のウシ胎仔血清(FCS)を加えたダルベッコ改変イーグル培地(DMEM)中で成長させた。細胞を、5×106細胞/プレートの密度で10cm3 培養プレート上に、10mlの容量で播種し、成長培地を交換することなく96時間、培養物をインキュベートすることにより熟成させた。その結果、著しい割合の細胞がアポトーシスとなった。細胞を、細胞スクレーパーを用いて回収し、18Gニードルを有するシリンジに通すことにより単一の細胞に分離し、106細胞/mlの密度でPBSバッファー(pH=7.4)中に再懸濁した。これは、単一の細胞の蛍光(flouresence)の可視化を可能にするダンシルアミド基を含むNST 203の前に見られた。アポトーシス細胞に対するNST203の選択的な結合を図3Aに示す。図は、アポトーシス細胞集団内へのNST 203の取込みを示す(緑、照り輝く(glowing)色の細胞の代表を矢印で示す)。一方、非アポトーシス細胞では蛍光観察されない(青、照り輝いていない細胞、矢じりで示す)。
(参考例5)
実験手順
マウス(c57/black;8週齢雄マウス)に、マウス黒色腫由来B16−F10細胞(ATCC CRL−6475;105細胞/マウスを100μlの容量で)を、脇腹に左右対称に皮下注射した。注射の前、この細胞株は、4mMのL−グルタミン;100単位/mlのペニシリン;100μg/mlのストレプトマイシン;12.5単位/mlのナイスタチンおよび10%のウシ胎仔血清(FCS)を加えたダルベッコ改変イーグル培地(DMEM)中で、培養状態で維持した。10日後、腫瘍直径が5〜7mmの大きさに達したとき、マウスを、化学療法処置(Taxol 20mg/Kgをシクロホスファミド(Cyclophosphomide)300mg/Kgとともに、200μlの容量で腹腔内注射)に供した。24時間後、NST−203を、2.8mg/マウス(10%発色団含有トリス系バッファー中)の用量で静脈内注射した。2時間後、マウスを致死させ、腫瘍および他の器官を回収し、直ちに液体窒素中で凍結させた。腫瘍または他の器官によるNST−203の取込みを、各組織由来の凍結切片の蛍光顕微鏡検査によって評価した。
図4Aは、腫瘍の蛍光顕微鏡検査を示す。アポトーシスを実行中の数多くの腫瘍細胞に対するNST203の広範な結合を観察することができる。また、この化合物の細胞内蓄積(写真の右側)および高レベルの選択性も示されており、アポトーシス細胞内への著しい取込みを反映しているが、周囲の腫瘍生細胞は、非染色のままである(写真の左上側)。

結腸癌モデルを、Balb/c 雄マウスの腹部において確立した。第12〜14日に、腫瘍の大きさが直径6〜8mmになったとき、結腸癌腫瘍を、2回用量のドキソルビシン(20mg/kg、72時間空ける)の点滴で処置した後、腫瘍内へのNST200の取込みを測定した。化学療法の48時間後、対照非処置動物および化学療法処置動物の両方に、放射標識したNST200(80μCi/動物)を静脈内注射した。4時間後、動物を致死させた。10μm凍結切片を調製し、風乾し、トリチウム感受性フィルムに露光した。フィルムを7週間の期間露光し、現像し、デンシトメトリー測定によって解析した。3H−DDC密度(光学密度/mm2)の測定を、虚血中心対反対側の半球とでのシグナルにおいて行い、TINAソフトウェアによって評価した。シグナルを、Microscaleオートラジオグラフィー標準にしたがって変換し、nCi/mg単位で示した。
非処置腫瘍はNST200の取込みを示さず、したがって、オートラジオグラフィーによって画像は検出され得なかった(図5A)。しかしながら、化学療法により細胞死を誘導すると、NST 200取込みの劇的な増加が起こり、これは、照射誘導性細胞死の大規模プロセスを示す(図5BおよびC)。NST 200取込みは、腫瘍の表面上の多くの病巣において、暗くて強い斑の形態で検出され得る。NST−200標識化は、腫瘍内の特定のアポトーシスの病巣領域に局在し、腫瘍全体に拡散していなかった。処置腫瘍の他の領域は標識されず、これは、腫瘍組織の生存を示す。この観察結果により、NST 200が、死滅中の細胞に選択的に大量に蓄積され、生細胞には蓄積されない分子を標的化するのに好都合であることが強調された(図5B)。同じ腫瘍内であっても治療に対する応答が不均一な性質が図5Cに示されている。NST200の取込みは、特に、細胞死の増大を示した腫瘍の大きな一領域に蓄積しているが、化学療法に対する応答を示さなかった他の腫瘍領域には蓄積されていない。
(実施例7)
実験手順
結腸癌モデル
マウス結腸癌細胞(CT−26)(ATCC CRL−2638)を、RPMI(Gibco, UK)、2mMのL−グルタミン;100単位/mlのペニシリン;100μg/mlのストレプトマイシン;12.5単位/mlナイスタチン;および10%加熱不活化FCS中で維持した。試験は、8〜10週齢の成体雄Balb/Cマウス(体重20〜25g)において行なった。
細胞をトリプシン処理し、HBS(140mM NaCl;0. 5M Hepes、PH 7.4)で2回洗浄し、次いで(than)遠心分離し(5分間、1000rpm、4℃)、(2%)メチルセルロースと生理食塩水(1:3)の混合物中2×106細胞/mlに濃縮した。0.2mlの容量の上記の溶液(4×105細胞/用量を含有)をマウス腹部の両側に皮下注射し、この間、マウスには麻酔した。麻酔用ストック溶液は、0.85mlケタミン(lOOmg/ml)+0.15mlキシラジン(2%)を1:10に希釈して調製した。重量10gあたりO.lmlの希釈溶液を、(i.p.)注射した。
マウスを、毎日、触診可能な腫瘍形成について検査した。腫瘍注射の7〜10日間後、小さな腫瘍が現れた。
腫瘍内への3H−NST200の取込みを、2回用量のドキソルビシン(20mg/kg、72時間空ける)の点滴での結腸癌腫瘍の処置後に測定した。2回目の投与の48時間後、マウスに3H−NST200(10μCi/動物)を静脈内注射した。4時間後、腫瘍を回収し、重量計測し、組織を処理した。腫瘍の溶解を、SOLVABLE(商標)試薬(GNE9100, Packard Bioscience)を、150mgの腫瘍組織に対して試薬1mlの比率で用い、60℃にて、20ml容シンチレーションガラスバイアル内で行なった。2〜4時間後、各組織抽出物の1mlをガラスシンチレーションバイアルに移した。着色消光の問題を低減するため、試料を0.4mlの30%H202で0.066M EDTAの存在下で処理した。15分間の室温でのインキュベーション時間後、抽出物を1時間60℃でインキュベートした後、さらに15分間室温でインキュベーションした。10mlのシンチレーション液(Ultima gold、6013329、Packard Bioscience)を各バイアルに添加した。バイアルを1時間室温でインキュベートし、次いで、βカウンター(TRI−CARB 2100TR、液体シンチレーションアナライザー、Packard Bioscience)において解析した。すべての試料は、3連で測定した。注射した用量のパーセントの値(%ID/g(組織))を各試料について計算した。
ドキソルビシン処置した結腸癌腫瘍対非処置対照腫瘍における3H−NST200蓄積の定量分析により、処置の48時間後に3H−NST200の大量蓄積が示された。一方、対照群では、すべての腫瘍に、同様だが少量の3H−NST200が蓄積し、化学療法処置腫瘍では、蓄積値は、腫瘍によってばらつきがあり、対照群と比べ、取込みの40〜50倍の増加を示した。処置群における取込みの平均値は1.28%ID/gであり、これは、more of 対照群の平均値より12.1倍大きかった(図6参照)。この広域の3H−NST200蓄積値は、種々の腫瘍の抗癌処置に対する個々の応答を反映する。上記の実験は、3H−NST200が、癌を検出する機能および癌細胞に対して細胞傷害剤の効果を検出する機能を果たし得ることを明白に示す。
(実施例8)
実験手順
結腸癌モデルを、Balb/c雄マウスの腹部において、前述の実施例に記載のようにして確立し、3H−NST200(10μCi)を、ドキソルビシンで処理した動物に注射した。4時間後、動物を致死させ、種々の器官/組織を回収し、処理し、これらの内部での3H−NST200の蓄積を測定した。各器官内における取込みを、注射した用量(ID)に対する%で示し、腫瘍と他の器官との取込みの比率を計算し、これを表に添付の図7として示す。
ドキソルビシンによって誘導された腫瘍組織に対する障害は、実際、他の非標的器官(心臓および小腸を含む)に対する障害よりも大きかった。腫瘍対すべての器官における取込みの比率は、表において>1であり、これは、腫瘍におけるアポトーシスの増加および標的対非標的組織における3H−NST200の選択的蓄積を示す。
(実施例9)
実験手順
結腸癌モデルを、Balb/c雄マウスの腹部において、実施例10に記載のようにして確立した。第12〜14日に、腫瘍の大きさが直径6〜8mmになったとき、マウスにドキソルビシンを静脈内注射した。合計2回のドキソルビシンを、3日間の間隔を空けて与えた(各用量は20mg/Kgとした)。2回目のドキソルビシン注射の2日後、マウスに、10μCiの3H−NST200を0.2mlの容量の生理食塩水にて点滴した。3H−NST200注射の4時間、マウスを、ペンタールの過剰投与により致死させた。腫瘍をエッペンドルフチューブ内に回収し、重量計測し、−20℃にて凍結させた。
ドキソルビシン(5日間持続)での処置の間、腫瘍塊は縮小しなかった(図8A参照)。対照的に、腫瘍は、成長し続け、その容積は、対照非処置腫瘍(処置腫瘍よりも5日間早く回収した)と比べ50%増加を示した。この増加は、非有意であることがわかった。腫瘍容積のかかる非有意な変化は、2回用量のドキソルビシン処置後に起こり、3H−NST200取込みの18.5倍という劇的な増加が検出され、これは、3H−NST200が、腫瘍塊の収縮が検出されない場合であっても、腫瘍内の細胞死を検知することできる感度のよいツールであることを示す(図8B参照)。
(実施例10)
実験手順
p−MCAを、側頭骨(subtemporal)からのアプローチにより、顕著な虚血性障害の転帰を有するBalb/Cマウスにおいて誘導した。p−MCAの24時間後、動物の神経学的スコアを評価し(0−臨床的症状なし〜3−片麻痺、回転行動および緊張病)、放射標識されたNST200(80μCi/動物)を、連続2時間静脈内注射した後、動物を致死させた。10μm凍結切片を調製し、風乾し、トリチウム感受性フィルム(Hyperfilm−3h、RPN535B Amersham−Pharmacia, Eu)に露光した。フィルムを7週間の期間露光し、現像し(GBX Developer & Fixer, Kodak, USA)、デンシトメトリー測定によって解析した。3H−DDC密度(光学密度/mm2)の測定を、虚血中心対反対側の半球とでのシグナルにおいて行い、TINAソフトウェアによって評価した。シグナルを、Microscaleオートラジオグラフィー標準(RPN510 Amersham−Pharmacia, Eu)にしたがって変換し、nCi/mg単位で示した。
3H−NST 200の蓄積(a)H&E染色(b)を示す図9からわかるように、オートラジオグラフィー画像解析により、アポトーシス(apoptotoc)細胞死/壊死性細胞死の領域内での3H−NST200による傷害の標的化の特異性が示された。この結果を、H&E染色によってさらに確認した。したがって、3H−NST200は、オートラジオグラフィー画像解析において、脳アポトーシス障害のマーカーとして使用することができる。
(実施例11)
実験手順
ケタミン(80mg/kg)とキシラジン(lOmg/kg)の組合せの腹腔内投与によって誘導した全身麻酔下のラットにおいて手術を行なった。腎虚血は、片側性左腎動脈を、小さな非外傷性血管用鉗子を用いて45分間締めることにより誘導した。同じ動物の反対側の未処置腎臓を、偽手術された(sham−operated)対照由来の腎臓とした。再潅流は、鉗子を外すことにより開始した。腎再潅流の期間は24時間とした。再潅流の過程の間、動物に100μCiの3H−NST205を静脈内注射し、1時間後、両方の腎臓を切除し、液体窒素中で凍結し、−70℃で使用まで保存した。10μm凍結切片を調製し、風乾し、トリチウム感受性フィルム(Hyperfilm−3h、RPN535B Amersham−Pharmacia, Eu)に露光した。フィルムを7週間の期間露光し、現像し(GBX Developer & Fixer, Kodak, USA)、デンシトメトリー測定によって解析した。3H−DDC密度(光学密度/mm2)の測定を、虚血性腎臓対反対側の腎臓とでのシグナルにおいて行い、TINAソフトウェアによって評価した。シグナルを、Microscaleオートラジオグラフィー標準(RPN510 Amersham−Pharmacia, Eu)にしたがって変換し、nCi/mg単位で示した。
図10に示すように、ラット腎臓の虚血−再潅流傷害の間、アポトーシスが、皮髄領域と外髄(OM)の遠位細管において観察され、一方、重篤な壊死が、(OM)の近位直細管において見られた。皮質では障害はあまり観察されなかった。反対側の偽腎臓では、障害細管は観察されなかった。オートラジオグラフィー画像解析により、アポトーシス細胞死/壊死性細胞死の領域内での3H−NST205による傷害の標的化の特異性が示され、形態素解析によって確認した。対照的に、反対側の腎臓内への3H−NST205取込みは示されず、傷害腎臓内のみへの3H−NST205取込みの特異性が強調される。
(参考例12)
実験手順
選択的髄質低酸素細管障害を有する腎障害を、インドメタシン(Sigma Chemical Co.)10mg/kg(i.v.)、Nωニトロ−L−アルギニンメチルエステル(L−NAME、Sigma Chemical Co.)10mg/kg(i.v.)、および放射線造影剤であるイオタラム酸ナトリウム80%(Angio−Conray, Mallinckrodt Inc)6mL/kg(i.a.)の併用投与によって誘導した。別のラットにビヒクルを注射し、対照とした。損傷を与えた24時間後、対照動物と実験動物の両方に、100μCiの3H−NST205を静脈内注射し、1時間後、両方の腎臓を切除し、液体窒素中で凍結した。
図11からわかるように、腎臓の形態の解析により、広範囲の程度の髄質(すなわち、外髄の外側および内側細片)障害が明らかになった。3H−NST205のホーミングは、主に、外髄内に限定された。形態の損傷のない特定の領域では、3H−NST205取込みは観察されなかった。
(実施例13)
実験手順
EAEを、C3H.SW/C57/bl雌マウス(6〜8週齢)の免疫処置により誘導した。動物は、ラットミエリン希突起膠細胞糖タンパク質(MOG)のアミノ酸35〜55を包含するペプチドで免疫処置した。このペプチドは、固相手法を用いてペプチド合成装置にて合成した。マウスの脇腹の一部位に、75μlのMOGペプチド含有完全フロイントアジュバント(CFA)および200μgのヒト結核菌(mycobacterium tuberculosis)を含むエマルジョン200μlを皮下注射した。同一のブースター(buster)を、1週間後に、他方の脇腹の一部位に注射した。脳炎誘発性抗原刺激の後、マウスを毎日観察し、EAEの臨床的兆候をスコアリングした(0=臨床的症状なし〜5=四肢の完全麻痺)。疾患の選択した病期において(症状発現前または末期)、1時間のインキュベーションのために、動物に、放射標識したNST200(100μCi/動物)を静脈内注射し、その後、動物を致死させた。10μm凍結切片を調製し、風乾し、トリチウム感受性フィルムに露光した。フィルムを7〜9週間の期間露光し、現像し、デンシトメトリー測定によって解析した。3H−DDC密度(光学密度/mm2)の測定を、虚血中心対反対側の半球とでのシグナルにおいて行い、TINAソフトウェアによって評価した。
多発性硬化症疾患のEAE動物モデルは、中枢神経系の白質を標的とする慢性障害性(disabling)自己免疫神経障害を擬態した。実験動物における白質の重篤な障害は、図12に示すオートラジオグラフィーにおいて観察された。脳レベルでは、冠状部切片は、対照の非免疫処置マウスでは明るい染色であるのと比べ、脳の底部の錐体路の暗い染色、および脳全体で過剰の放射能−リガンド蓄積を示した。また、放射性同位元素標識した3H−200の印象的な蓄積が、脊髄レベルでも観察された。脊髄の縦断面は、対照未処置動物の脊髄と比べ、免疫処置マウスの白質の側方路(lateral tract)の高レベルの標識化を示した。
