Description
本発明の健康補助食品は、上記本発明のβグルカン組成物からなるものである。好ましくは上記本発明のβグルカン組成物としての必須成分(A)〜(C)の合計質量として0.1〜100質量%、より好ましくは0.5〜50質量%、更に好ましくは2.0〜20質量%であるとよい。
本発明の健康食品は、上記本発明のβグルカン組成物を配合した健康食品である。即ち、上記本発明のβグルカン組成物を食品素材と混合して得ることができるものである。
〔実施例1、比較例1〜9〕
表1に示した配合のβグルカン組成物を調製し、ラットから採取した免疫系細胞に対するTNF−α産生誘導作用を評価した。試験方法は以下の通りである。結果を表1に示す。
ラットの腹腔にプロテオースペプトンを注入し免疫系の細胞を誘導し、腹腔に誘導された免疫系の細胞を採取してin vitro培養後、本発明のβグルカン組成物及び比較のためのβグルカン組成物を用いた被験サンプルを添加し、TNF−α産生誘導能を測定した。
鹿角霊芝乾燥原体粉末(抽出加工品、株式会社坂本バイオ製「鹿角霊芝 王角 抽出処理粉末」:βグルカン総量50質量%、以下「原料A」という)、大麦由来βグルカン粉末(旭電化工業株式会社製「大麦ベータグルカンC60」:β1,3−1,4グルカン量65質量%、以下「原料B」という)及び黒酵母由来βグルカン(旭電化工業株式会社製「発酵ベータグルカン」:β1,3−1,6グルカン量90質量%、以下「原料C」という)をそれぞれ1mg/mlとなるよう5%牛胎児血清含有RPMI1640培地に溶解させた(以下、この原料Aの溶液を「溶液A」、原料Bの溶液を「溶液B」、原料Cの溶液を「溶液C」という)。
被験サンプル1で使用した溶液Aを、原料Aの濃度として100μg/mlとなるよう、5%牛胎児血清含有RPMI1640培地で希釈して被験サンプル2とした。
被験サンプル1で使用した溶液Bを、原料Bの濃度として100μg/mlとなるよう、5%牛胎児血清含有RPMI1640培地で希釈して被験サンプル3とした。
被験サンプル1で使用した溶液Cを、原料Cの濃度として100μg/mlとなるよう、5%牛胎児血清含有RPMI1640培地で希釈して被験サンプル4とした。
霊芝(傘状形態のマンネンタケ)乾燥原体粉末(株式会社坂本バイオ試験生産品、βグルカン総量20質量%、以下「原料D」という)を1mg/mlとなるよう5%牛胎児血清含有RPMI1640培地に溶解させた(以下、この原料Dの溶液を「溶液D」という)。
被験サンプル1で使用した溶液Aと溶液Bを1:1の容積比で混合した後、原料A、原料Bの合計量の濃度として100μg/mlとなるよう、同培地で希釈して被験サンプル6とした。
被験サンプル1で使用した溶液Aと溶液Cを1:1の容積比で混合した後、原料A、原料Cの合計量の濃度として100μg/mlとなるよう、同培地で希釈して被験サンプル7とした。
被験サンプル1で使用した溶液Bと溶液Cを1:1の容積比で混合した後、原料B、原料Cの合計量の濃度として100μg/mlとなるよう、同培地で希釈して被験サンプル8とした。
被験サンプル5で使用した溶液Dと、被験サンプル1で使用した溶液Bと溶液Cを、溶液D:溶液B:溶液C=5:5:1の容積比で混合した後、原料D、原料B、原料Cの合計量の濃度として100μg/mlとなるよう、同培地で希釈して被験サンプル9とした。
(免疫系の細胞の採取)
Wistarラット(8週齢、雄)を固定器に固定し、腹部をアルコールで消毒し、滅菌済みのバクトペプトン(DIFCO社製)10%水溶液を注射針が臓器に刺さらないように注意しながらラットの腹腔に10ml注入した。その後3日間飼育し、免疫系の細胞を誘導した。
採取した腹腔浸出液に対し1000rpmで5分間の遠心沈降操作を行った。上清を捨て、赤血球除去用塩化アンモニウム溶液(0.017M Tris−HCl pH7.65、に塩化アンモニウムを0.75質量%加え滅菌した溶液)を9ml加え、ピペッティングでペレットをほぐし、室温で7分間放置した。
本発明の健康補助食品の例として、上記の被験サンプル1で使用した原料A、原料B、原料Cを、原料A:原料B:原料C=5:5:1の質量比で混合して実施例2のβグルカン組成物を得、これをそのまま本発明の健康補助食品1とした。
〔官能試験方法〕
上記健康補助食品1〜3それぞれのサンプルについて、10名のパネラーにより、茶さじ1/5程度の量を口に含んだ際に感じられる苦味および粘着感を評価した。
5 食品としてそのままでは許容しがたい(非常に強い)
4 食品としてかなり強い苦味であり受容できる限度に近い(強い)
3 食品としてはやや強いが、許容できる(普通)
2 若干の苦味を感じるが食品としてまったく問題のない程度である(弱い)
1 ほとんど苦味を感じない(ほとんどない)
5 非常に強い
4 強い
3 普通
2 弱い
1 ほとんどない
Claims (2)
- 必須の成分として、
(A)鹿角霊芝由来のβグルカンと、
(B)大麦由来のβ1,3−1,4グルカンと、
(C)黒酵母由来のβ1,3−1,6グルカンと、
を含有するβグルカン組成物からなることを特徴とする免疫増強剤。 - 前記(A)成分100質量部に対して、前記(B)成分が50〜200質量部、前記(C)成分が10〜100質量部である請求項1に記載の免疫増強剤。
