Description
本発明は、アポトーシスの効果から哺乳動物を保護するためのNF−κBのインデューサの使用に関する。より詳細には、本発明は、放射線治療および癌治療等のストレスへの曝露から哺乳動物を保護するためのNF−κBのインデューサの使用に関する。
正常細胞から腫瘍細胞への進行は、増殖阻害刺激に対する耐性ならびに増殖因子およびホルモンに対する依存性の欠如を含む、増殖調節の負の機構の喪失を含む。放射線または細胞傷害性薬剤に基づく伝統的な癌治療は、正常細胞および悪性細胞の増殖制御の差異による。伝統的な癌治療は、激しい遺伝子毒性ストレスへ細胞を供する。これらの条件下で、正常細胞の大部分は、拘束されそしてしたがって確保され、一方、腫瘍細胞は、分裂しそして死滅し続ける。
本明細書中において提供されるのは、アポトーシスを誘発する1以上の条件または治療から哺乳動物を保護する方法である。哺乳動物に、式:

を有する薬学的に許容される量の化合物を含む組成物が投与され得る。
種々のストレスによって引き起こされるアポトーシスから正常な細胞および組織を保護する方法が、本明細書中において提供される。アポトーシスは、通常、傷ついたまたは遺伝子的に損傷した細胞を組織から「取り除く」ように機能し、一方、サイトカインは、ストレスに対する生物の防衛システムを動員する。しかし、重傷の条件下で、両方のストレス応答機構は、死の原因として作用し得る。例えば、放射線からの死亡率は、造血系、免疫系および消化器系において生じる大量のアポトーシスから生じ得る。
本明細書中において使用される専門用語は、特定の実施形態を記載するためだけであり、そして限定的であるようには意図されないことが理解される。本明細書および添付の特許請求の範囲中において使用される場合、文脈が明確にそうではないように指示しない限り、単数形「ア(a)」、「アン(an)」および「ザ(the)」は、複数形指示対象を含むことが、注意されなければならない。
リポペプチドは、NF−κB活性を誘発するための薬剤として使用され得る。リポペプチドは、グラム陰性菌、グラム陽性菌、およびマイコプラスマの外膜の一部である。細菌性リポペプチドは、配列相同性を共有しないが、2または3の脂肪酸によってアシル化されている普通ではないN末端アミノ酸S−(2,3−ジヒドロキシプロピル)−L−システインを特徴とする。細菌性リポペプチドは、TLR2-TLR1またはTLR2-TLR6ヘテロダイマーを介してのシグナル伝達によって感染後に早期宿主応答を活性化させ、NF−κBおよびサイトカイン産生の活性化へ導く、強力な免疫モジュレータである。天然リポペプチドのN末端リポペプチドの合成アナログは、TLRおよびNF−κBの強力なアクチベータであり、同様に、インビボおよびインビトロで免疫アジュバントである。
NF−κB活性を誘発する薬剤は、細胞ストレスを生じさせ、それによってアポトーシスを誘発する条件または治療から正常細胞を保護するために使用され得る。条件または治療の代表的な例としては、癌治療、例えば、放射線療法または化学療法;温度衝撃;有害な線量の放射線への曝露、例えば、原子力発電所、防衛産業または放射線医薬品製造における労働者、あるいは兵士;細胞老化;損傷;中毒;および感染が挙げられる。
前記条件は、構成的に活性なNF−κB癌であり得る。NF−κB活性を誘発する薬剤は、化学療法または放射線療法等の癌治療と組み合わせて投与され得る。
前記条件はまた、構成的に活性なNF−κB癌の治療に起因する正常組織への損傷であり得る。NF−κB活性を誘発する薬剤は、上述の癌治療と組み合わせて投与され得る。
前記条件はまた細胞老化であり得る。
前記条件はまた、放射線への曝露であり得る。電離放射線(IR)への曝露は、短時間または長時間であり得、それは、全身へまたは局所的に、単回線量または複数回線量として適用され得る。したがって、原発事故または軍事攻撃は、単回高線量の全身照射への曝露(場合によっては放射性同位体での長期間の中毒が続く)を含み得る。同様に、単回線量の放射線は、それらから宿主血液前駆体を「取り除く」ことによってドナー骨髄についての宿主の造血器官を調製することが必要である場合、骨髄移植患者の前処置のために一般的に使用される。
治療有効量のNF−κBのインデューサを含む組成物もまた、本明細書中において提供される。組成物は、当該分野において周知の方法を使用して製造され得る、薬学的組成物であり得る。上記に記載されるように、NF−κBのインデューサを含む組成物は、放射線への曝露、癌治療の副作用、ストレスおよび細胞老化が挙げられるがこれらに限定されない、アポトーシスに関連する条件の治療のために、哺乳動物へ投与され得る。組成物はまた、放射線防護剤または化学療法薬が挙げられるがこれらに限定されない追加の薬剤を含み得る。
本明細書中において提供される組成物は、経口、非経口、舌下、経皮、経直腸、経粘膜、局所的、吸入、口腔投与、またはそれらの組み合わせが挙げられるがこれらに限定されない任意の様式で投与され得る。非経口投与としては、静脈内、動脈内、腹腔内、皮下、筋内、髄腔内、および関節内が挙げられるが、これらに限定されない。獣医学的用途について、前記組成物は、通常の獣医学的実務に従って、適切に許容される製剤として投与され得る。獣医は、特定の動物について最も適切である投与計画および投与経路を容易に決定し得る。
本明細書中において提供される組成物は、従来の様式で製剤化される錠剤またはロゼンジの形態であり得る。例えば、経口投与のための錠剤およびカプセル剤は、結合剤、充填剤、滑沢剤、崩壊剤および湿潤剤が挙げられるがこれらに限定されない従来の賦形剤を含有し得る。結合剤としては、シロップ、アカシア(accacia)、ゼラチン、ソルビトール、トラガカント、スターチの粘液、およびポリビニルピロリドンが挙げられるが、これらに限定されない。充填剤としては、乳糖、砂糖、微結晶性セルロース、トウモロコシデンプン、リン酸カルシウム、およびソルビトールが挙げられるが、これらに限定されない。滑沢剤としては、ステアリン酸マグネシウム、ステアリン酸、タルク、ポリエチレングリコール、およびシリカが挙げられるが、これらに限定されない。崩壊剤としては、ジャガイモデンプン、およびデンプングリコール酸ナトリウムが挙げられるが、これらに限定されない。湿潤剤としては、ラウリル硫酸ナトリウムが挙げられるが、これに限定されない。錠剤は、当該分野において周知の方法に従ってコーティングされ得る。
治療において使用されるために必要とされる前記薬剤の治療有効量は、治療される条件の性質、NF−κB活性の誘発が望まれる時間の長さ、ならびに患者の年齢および条件により変化し、そして最終的に主治医によって決定される。しかし、一般的に、成人治療に使用される用量は、典型的に、1日当たり0.001mg/kg〜約200mg/kgの範囲内である。用量は、1日当たり約1μg/kg〜約100μg/kgであり得る。所望の用量が、単回用量で、あるいは、好適な間隔で、例えば、1日当たり2、3、4回またはそれ以上の副用量として投与される複数回用量として、投与され得る。複数回用量が、しばしば望ましいか、または必要とされ、何故ならば、いったん前記薬剤が投与されなくなると、正常細胞におけるNF−κB活性が低下され得るためである。
本明細書中において提供される方法はまた、NF−κB活性を誘発する薬剤を同定する方法に関する。NF−κB活性化発現系へNF−κB活性の疑われるインデューサを添加すること、NF−κB活性化発現レベルをコントロールと比較し、それによって、NF−κB活性化発現系のレベルを増加させる能力によってNF−κB活性のインデューサが同定されることを含む方法によって、NF−κB活性を誘発する薬剤が同定され得る。
p53欠損はマウスにおけるGI症候群の進行を促進する
哺乳動物の電離放射線(IR)からの死の根本原因は、放射線線量に依存する。9〜10Gyの線量で、致死骨髄枯渇、即ち造血(HP)症候群から、マウスが12〜20日後に死ぬ。この線量で、照射されたマウスは、骨髄移植によって致死性から救われ得る。>13〜15Gyを受けた動物は、小腸への損傷の合併症、即ち、胃腸管(GI)症候群から、治療後7〜12日で(造血症候群がそれらを殺し得る前に)死ぬ。小腸の上皮細胞の細胞増殖は、幹細胞および初期増殖性前駆細胞(early proliferating progenitors)が局在される陰窩に限定されることは周知である。2〜3回の細胞分裂後、陰窩幹細胞の既に分化された子孫は、絨毛へ移動し、絨毛先端(villar tip)で発せられる。マウスの小腸において、細胞の全体の「旅」(即ち、増殖性区画から絨毛の先端へ)は、通常、3〜5日かかる。ガンマ線に対する小腸の反応を病理形態学的レベルで試験したが、初期事象を含む、GI致死性の正確な原因は、依然として不明のままである。死は、上皮陰窩細胞への損傷、続いての流体および電解質不均衡、菌血症、ならびに内毒血症へ導く絨毛の露出の直接的な結果として生じ得る。炎症および間質応答に加えて、内皮機能不全は、致死性に寄与する重要な因子であるようである。
実施例2
リポペプチドは全身ガンマ線照射によって引き起こされるマウス死を遅延させる
リポペプチドはNF−κBの強力なアクチベータであり、そして、それ自体は、アポトーシス死の阻害剤として作用し得る。リポペプチドが放射線防護剤として機能するかどうかを決定するために、種々のリポペプチドを最初に試験し、最大耐量(MTD)を決定した。次いで、それぞれ、10Gyまたは13〜15Gyの全身ガンマ線への曝露後、致死造血またはnd(or nd)胃腸症候群に対するNIH Swissマウスにおけるそれらの防護効果を測定するために、種々のリポペプチドを試験した。リポペプチド(0.3〜10μg/マウス)を、照射30分前に皮下投与した。放射線防護を提供した試験したリポペプチドを表3に記載する。
実施例3
全身ガンマ線照射に対する単回用量のCBLB601の放射線防護有効性
C−SKKKK(配列番号8)からなるペプチド部分を有するR−Pam2−リポペプチドであるCBLB601を、NF−κBを活性化するその能力およびNIH−SWISSマウスにおける放射線防護についての予備インビボデータ(実施例2を参照のこと)に基づいて、放射線防護剤としてのより詳細なキャラクタライゼーションのために選択した。この研究の目的は、プロテクターとして役立つCBLB601の最適な用量、投与経路および投与時間を決定することである。10〜15週齢のICR雌性マウスを、1グループまたは条件当たり10〜15匹の動物で使用した。
実施例4
CBLB601によって致死線量TBIから救われたマウスの免疫状態
CBLB601の投与によって10GyのTBIから救われたマウスは、損なわれた免疫系を有し得、したがって、「正常な」生活を有していないかもしれない。それらの免疫系の状態を確認するために、それらに3μgのCBLB601/マウスまたはPBSを注射してから24時間後に、ICRマウスを致死(10Gy)または非致死(6Gy)線量のTBIへ曝露した。各条件からの5匹のマウスの群をまさに3日で犠牲にし、そしてそれらの脾臓および胸腺を除去しそして重さを測定した。脾臓の重量を対応の動物の体重へ標準化し、そして結果を図15に示す。6Gyの放射線へ曝露されたPBS処置マウスは、それらの脾臓が正常重量へ回復するまでに約13〜14日かかり、一方、6Gyの放射線へ曝露されたCBLB601処置マウスは、8日目までに正常脾臓重量を有した。PBS注射した動物は10GyのTBIを生き延びず、一方、CBLB601注射したマウスは、致死用量の放射線を生き延びただけでなく、照射後13〜14日目までにそれらの脾臓重量を完全に回復させた。正常な重量へ胸腺を回復させるためにより長い時間(約30日)を要した(示さず)。
実施例5
他のリポペプチドによって提供されるNF−κBの活性化および放射線防護
さらなるリポペプチドを合成し、そしてNF−κBの活性化および致死線量の放射線からマウスを防護するそれらの能力について試験した。化合物の名称およびそれらの重要な構成要素を表4に示す。前記化合物のいくつかについて、対応の遊離ペプチドをまた合成しそして試験した。
