Description
加齢黄班変性(AMD)は、60歳を超える高齢者人口の失明の第1の原因である。これは、罹患した人々の中心視力を破壊し、読書や運転などの日常生活に必要な活動を行う能力を奪う、深刻な疾患である(Bressler et al., 1988; Evans, 2001; Gottlieb, 2002)。ある研究にでは、75歳以上の人々におけるAMDの有病率は7.8%と報告された(Klein et al., 1992)。
本発明は、加齢黄班変性(AMD)を含む網膜の変性状態をスクリーニングし処置するための、新規な方法および組成物、ならびに、治療化合物および方法を試験するために有用な動物モデルを提供する。本発明は遺伝子発見戦略の成果であり、1)AMDに罹患したものと正常の眼組織において、および2)RPE細胞による外節(OS)の食作用の過程の間に、差次的発現を示す遺伝子の単離をもたらす。OS食作用は、RPE細胞の重要な機能であり、複雑な多段階プロセスが関与し、この副産物はRPE細胞における活性酸素種の生成とリポフスチン蓄積の原因となる。
上記の戦略により発見されたAMDP遺伝子の例は、膜型マトリックスメタロプロテアーゼ1(MT1−MMP)(配列番号15)である。MT1−MMPは、例えば、プロゲラチナーゼAを特異的に活性化させることにより、細胞外基質の再構築に関与するプロテアーゼをコードする遺伝子である。ゲラチナーゼAは、基底膜の主要な構成成分であるIV型コラーゲンの特異的な開裂に関与する、主要なメタロプロテアーゼである。MT1−MMPはまた、他の細胞外基質成分に対する活性も示す。
1)MT1−MMPは、AMD患者の眼において、AMDのサルモデルにおいて、およびRPEによるOS食作用に欠点を有する網膜変性のモデルであるRCSラットにおいて、RPEおよび光受容体中で上方制御される;
2)MT1−MMPは、RPE細胞による食作用のメカニズムに直接関与する;
3)RCSラットにおける網膜変性の進行は、抗MT1−MMP抗体を用いて、網膜下のスペースに存在する活性化されたMT1−MMPをブロックすることにより、大幅に低減される;
4)mRNAのスプライス変異体を産生することができて短縮タンパク質をもたらすMT1−MMPの同義多型(すなわち、P259P)、および、タンパク質の触媒ドメインに影響するMT1−MMPのミスセンス多型(すなわち、D273N)が、AMD患者(54.8%vs31.6%)および家族性黄班変性症患者(68.2%vs31.6%)のDNAにより高い頻度で見出される。
方法は、網膜もしくは脈絡膜変性疾患もしくは状態を遅延させるか、または疾患もしくは状態を逆転させることができる。
方法の実施において接触させる細胞型は、光受容体、RPE細胞、ミュラー細胞、または、内皮細胞、平滑筋細胞、白血球、マクロファージ、メラノサイトもしくは繊維芽細胞を含む、脈絡膜の細胞型である。
本発明の幾つかの態様において、剤は、AMDP関連または食作用関連遺伝子の核酸またはアミノ酸配列の発現を下方制御する。好ましい態様において、標的化遺伝子は、MT1−MMP、プロスタグランジンD2合成酵素およびAMDP−3を含み、これら遺伝子は、AMDにおいて過剰発現されることが本明細書で示される。剤は、オリゴヌクレオチドであってよく、例えば、リボザイム、アンチセンスRNA、干渉RNA(RNAi)分子、または三重らせん形成性分子(triple helix forming molecule)である。
他の態様において、MT1−MMP、プロスタグランジンD2合成酵素またはAMDP−3の発現を下方制御する剤は、小分子であることができる。
方法においてスクリーニングされる多型は、目的遺伝子のイントロン、エクソンまたはプロモーター領域内にあることができる。
方法の特に好ましい態様において、多型は、ヒトMT1−MMP遺伝子のエクソン5の285bp断片内にある。この領域内で、多型は、D273Nミスセンス多型およびP259P同義多型を含むことができる。
他の側面において、本発明は、ベクターを用いて、食作用関連またはAMDP関連遺伝子産物の所望の形態を、これを必要とする対象に送達することにより、網膜または脈絡膜変性疾患または状態を処置する方法を提供する。ベクターは、食作用関連またはAMDP関連遺伝子の野生型または多型変異体のどちらかをコードする核酸配列を含む。
さらに本発明の範囲であるのは、ベクターを含有する、対象における網膜または脈絡膜変性疾患または状態の予防または処置のための組成物である。種々の態様において、ベクターは、食作用関連またはAMDP関連mRNAまたはタンパク質の発現を下方制御または阻害する剤をコードする核酸、または、食作用関連またはAMDP関連タンパク質の野生型または多型変異体をコードする核酸を含むことができる。好ましい態様において、食作用関連またはAMDP関連遺伝子は、MT1−MMP、プロスタグランジンD2合成酵素およびAMDP−3を含む。特に好ましい態様において、遺伝子はMT1−MMPである。
ポリトランスジェニック動物の好ましい態様において、第1遺伝子は、MT1−MMPであり、第2遺伝子は、ABCR、アポリポタンパク質E、C−Cケモカイン受容体2、シスタチンC、ヘミセンチン/FIBL−6、マンガンスーパーオキシドジスムターゼ、C−Cケモカインリガンド/単球走化性因子タンパク質1(monocyte chemoattractant protein 1)、およびパラオキソナーゼから選択される。
本発明のトランスジェニック動物の特に好ましい態様はマウスであり、これは、比較的短い寿命の利点を提供し、サルなどの他のより長命の動物モデルに比べて、これを加齢疾患の研究に好適なものとしている。
本発明はさらに、複数の単離されたオリゴヌクレオチド配列を含み、該配列は、天然のヒトAMDP関連または食作用関連遺伝子のイントロン、エクソンまたはプロモーター配列内に位置する、遺伝子アレイを提供する。アレイ内にオリゴヌクレオチド配列により表わされる遺伝子は、本明細書において配列番号1〜17および配列番号62〜69で表わされる核酸配列を含むcDNAをコードする。
遺伝子アレイはさらに、1つまたは2つ以上のAMD関連遺伝子の少なくとも1つの多型変異体を含む少なくとも1つのオリゴヌクレオチド配列を、MT1−MMPに加えて含むことができる。多型変異体配列は、ABCR(D217N;G1961E)、マンガンスーパーオキシドジスムターゼ(V47A)、アポリポタンパク質E(C130、R176CおよびC130R、R176)、シスタチンC(A25T)およびパラオキソナーゼ(Q192R、L54M)を含むことができる。

前述の発見に基づき、本発明は、AMDおよび/またはRPE細胞による食作用に関連する新規な遺伝子、AMDおよび他の網膜変性状態を検出し処置するための方法および組成物、並びに、とりわけAMDの治療組成物および処置プロトコルを試験するために有用な、食作用関連および/またはAMDP関連遺伝子に基づく動物モデルを提供する。以下に記載する好ましい態様は、これらの組成物および方法の適応を説明する。しかしなお、これらの態様の記載から、以下に提供される記載に基づき本発明の他の側面を作製し、および/または実行することが可能である。
従来の分子生物学的技法が関与する方法を、本明細書に記載する。かかる技法は、当分野に一般に知られており、以下のような方法論の専門書に詳細に記載されている:Molecular Cloning: A Laboratory Manual, 2nd ed., vol. 1-3, ed. 本発明を導いた研究は、RPE細胞によるOS食作用に関与する遺伝子であって、摂動された場合に、RPEにおけるストレスおよび機能障害をもたらし得るものを同定するために行われた。かかるストレスは、黄班、網膜または脈絡膜疾患に関連する1つまたは2つ以上の望ましくない変化、例えば、リポフスチン蓄積の増加、ドルーゼン形成、または新生血管膜の発生などをもたらし得る。本明細書に記載された遺伝子の発見は、RPEによる食作用における機能障害が、かかるAMD関連の変化をもたらす鍵となる要因であるとの仮定に基づいていた。RPE細胞は、光受容体の恒常性を維持する重要な機能を行う。この難しい課題には、とりわけ食作用の毎日のプロセスおよび、毎日更新され光受容体のOSの先から脱離するOS膜の消化を含む(Young and Bok, 1969)。以下にさらに記載されるように、食作用のプロセスは、OS膜の結合、摂取および消化のステップを含む。正常な状況の下では、RPE細胞は分裂しない細胞である。従って、個人の寿命の間を通して、OS食作用の毎日のプロセスは、これらの細胞に対する多量の代謝負荷を表わすだけでなく、これら細胞内への未消化物質、特にリポフスチンの蓄積に寄与し、リポフスチンとは、A2Eなどの光受容体由来の毒性物質を含む、細胞老廃物の複合した混合物である。
他の側面において、本発明は、AMDと関連すると以前に知られていなかった遺伝子の、核酸およびタンパク質配列を提供する。AMD関連遺伝子を得るために、10,000個のRPE発現遺伝子のCHANGEアレイを、上記と同様に「+/−」プローブの対を用いて繰り返しスクリーニングした。AMD関連遺伝子を同定するために用いた+/−プローブは、AMDに罹患および非罹患したヒト提供者の眼のRPE/脈絡膜から抽出した、および、AMDのサルモデルの、年齢を整合させた正常および罹患した眼から抽出した、全RNAから作製した。アレイの遺伝子は、さらなる解析のために、加齢の正常な対照眼と比較して、AMDにおいて差次的(すなわち、増加または減少)発現を示すことに基づき選択した。CHANGEにより検出される変化した遺伝子発現の基準に基づき、約200個のAMD関連遺伝子が同定された。
AMD関連食作用遺伝子(「AMDP遺伝子」)を同定するために、上記の2つのCHANGEスクリーニングからのデータを比較して、RPE細胞によるOS食作用およびAMDの両方において差次的に発現されたRPE遺伝子のサブセットを同定した。上記のように、食作用CHANGEスクリーニングにより約60個の食作用遺伝子がもたらされ、推定AMD関連遺伝子は約200個となった。2つのデータベースの初めの比較により、食作用およびAMDの両方で発現変化を示す6個の遺伝子のサブセットがもたらされた(表2)。これらの遺伝子は、本明細書において、「AMD関連食作用遺伝子」または「AMD/食作用遺伝子」と指定され、「AMDP」と略される。
上記のように、本発明は、RPE食作用の間におよび/またはAMDにおいて発現の変化を示すことが、差次的クローニング戦略(CHANGE)により発見された遺伝子に関連する、核酸およびアミノ酸配列を提供する。1つの態様において、本発明は、この戦略により単離された、新規な精製された核酸(ポリヌクレオチド)を提供する。本発明の、以前に知られていなかった核酸は、本明細書においてPHG−1(配列番号1);PHG−4(配列番号4);PHG−5(配列番号5);PHG−13(配列番号12);およびAMDP−3(配列番号17)と同定された核酸配列を含む。これらの核酸は、それぞれ、本明細書において配列番号71〜79;82〜84;85;92〜98;および103〜121として同定されたアミノ酸配列を有するポリペプチドをコードする。
本発明の核酸分子は、RNAの形態またはDNAの形態(例えば、cDNA、ゲノムDNA、および合成DNA)であることができる。本発明の好ましい核酸分子は、それぞれの天然のポリヌクレオチドであり、本明細書において配列番号1、2、3、4、5、6、7、8、9、10、11、12、13、14、15、16、および17と示されたヌクレオチド配列を含有する。
他の側面において、本発明は、対象の網膜または脈絡膜疾患または変性状態を発症するリスクを決定するための方法を提供する。本明細書において、「網膜または脈絡膜疾患または変性状態」とは、限定なく、眼の網膜または脈絡膜の任意の状態であって、光受容体、RPE細胞または網膜の他の細胞型の損傷または死をもたらすもの、または、脈絡膜細胞型であって、限定なく、内皮細胞、メラノサイト、平滑筋細胞、繊維芽細胞、リンパ球、好中球、好酸球、巨核球、単核球、マクロファージおよびマスト細胞を含むものの損傷、死または異常な増殖をもたらすものである。
他の側面において、本発明は、食作用関連および/またはAMDP関連遺伝子のmRNAまたはタンパク質の発現レベルを調節する剤を提供する。下方制御用の標的とするのに好ましい遺伝子/タンパク質は、AMDおよび関連疾患において発現の増加を示すものであり、本明細書に示されるように、プロスタグランジンD2合成酵素、PD2S(それぞれの核酸およびアミノ酸配列:配列番号2および80)、MT1−MMP(配列番号15および101)、およびAMDP−3(配列番号17および103〜121)を含む。上方制御用に標的とするのに好ましい遺伝子/タンパク質は、AMDおよび関連疾患において発現の低下を示すものであり、本明細書に示されるように、SWI/SNF関連/OSA−1核タンパク質(配列番号16および102)、カゼインキナーゼ1エプシロン(配列番号9および89)、フェリチン重ポリペプチド1(配列番号10および101)を含む。
発現の下方制御に用いられる阻害剤は、例えば、アンチセンスRNA分子、リボザイム、低分子干渉RNA(RNAi)分子および三重らせん構造を含むことができる。かかる剤の好ましい態様は、PD2S(配列番号2)、MT1−MMP(配列番号15)、およびAMDP−3(配列番号17)、またはこれらの変異体に対するものである。阻害剤はまた、過剰発現された食作用関連および/またはAMDP関連タンパク質、例えばPD2S、MT1−MMPまたはAMDP−3に、選択的に結合する抗体分子も含むことができる。
低分子阻害RNA分子(small inhibitory RNA molecule)は、細胞内でタンパク質複合体に結合することにより作動し、これは、RNA誘導サイレンシング複合体(RISC)と呼ばれ、これはヘリカーゼ活性およびエンドヌクレアーゼ活性を含む。ヘリカーゼ活性は、RNA分子の2本鎖を解き、siRNAのアンチセンス鎖を標的RNA分子に結合させる(Zamore, 2002; Vickers et al., 2003)。エンドヌクレアーゼ活性は、標的RNAをアンチセンス鎖が結合した部位において加水分解する。
他の側面において、本発明は、食作用関連および/またはAMDP関連遺伝子をコードする天然もしくは合成核酸、またはこれら遺伝子の発現もしくは活性を調節する剤の送達を提供する。「遺伝子療法」は、遺伝によるかまたは後天性の疾患の、細胞内への遺伝的情報の導入および発現による処置と定義できる。遺伝子療法ベクターが関与する方法および組成物は、本明細書に記載されている。かかる技法は一般に当分野に知られており、方法論の参考文献、例えばViral Vectors, eds. 幾つかの非ウィルス的方法が、食作用関連および/またはAMDP関連核酸、または細胞内でかかる核酸の発現もしくは活性を調節する剤を導入するために知られている。非ウィルス的方法の概説は、例えば、Nishikawa and Huang, Human Gene Ther. 12:861-870, 2001を参照のこと。食作用関連および/またはAMDP関連核酸、または細胞内に発現される食作用関連および/またはAMDP関連核酸の発現を調節する剤を細胞内に導入するために、プラスミドDNAを用いる種々の技法が、本発明に従って提供される。かかる技法は一般に当分野で知られており、例えばIlan, Y., Curr. Opin. Mol. Ther. 1:116-120 (1999)およびWolff, J.A., Neuromuscular Disord. 7:314-318 (1997)などの文献に記載されている。
ウィルスおよび非ウィルスに基づく両方の成分を含む方法も、本発明に従って用いることができる。Epstein Barr ウィルス(EBV)に基づく治療的遺伝子送達用のプラスミドは、Cui et al., Gene Therapy 8: 1508-1515, 2001に記載されている。アデノウィルスに結合したDNA/リガンド/ポリカチオン付加物(polycationic adjunct)を含む方法は、Curiel, D.T., Nat. Immun. 13:141-164 (1994)に記載されている。
本発明の幾つかの態様は、スクリーニング法、網膜変性の動物モデル、および膜型マトリックスメタロプロテアーゼ1(MT1−MMP)(配列番号15)に基づく処置方法である。上記にリストしたAMDP遺伝子の中で、1つの遺伝子、すなわちMT1−MMP(ここではまた、PHG−16およびAMDP−6とも指定する)が初めに選択されて、AMD療法の候補標的としてさらに評価された。以下の例に示されるように、種々の確認解析の結果は、MT1−MMPが食作用遺伝子であることを、以下の事項により明確に示した:1)発現の日周パターン(diurnal pattern):ここではin vivoでの最大のOS脱離(shedding)および食作用の時である早朝にピークを示す(図7);2)ラットおよびヒトの眼における、OSの先端への局在化(図8、9);および3)ラットの眼への網膜下注射後にin vitro(図10)およびin vivoの両方における、MT1−MMPに対する抗体によるOS食作用の阻害(図11)。
侵入的な腫瘍細胞上で発現される、MT1−MMPの以前に認識されていた機能は、プロゲラチナーゼAを活性化させ、種々のECM成分を消化する能力を含む(Sato et al., 1994; Cao et al., 1995: Pei and Weiss, 1996)。本明細書に記載された発見に基づき、この遺伝子が、AMD並びに他の網膜および脈絡膜変性疾患を治療的に標的とする、新しい魅力的な候補遺伝子を提供することが明らかである。
他の側面において、本発明は、AMDおよび他の網膜および脈絡膜の変性状態の動物モデルとして用いるのに好適な、非ヒトトランスジェニック動物(例えばマウス)を含む。従来、AMDの治療化合物および処置方法の試験は、加齢変化を実際的に追跡可能な、好適に短命な疾患の動物モデルが欠如していることによって妨げられていた。少なくとも3つのAMD/食作用遺伝子、すなわち、PD2S(配列番号2)、MT1−MMP(配列番号15)およびAMDP−3(配列番号17)の、AMDの眼における過剰発現の発見に基づき、および、AMDを有するヒト、AMDを有するサル、および遺伝性の網膜変性を有するRCSラットの網膜における、MT1−MMP mRNAおよびタンパク質の過剰発現の実証に基づき、本発明は、好ましい態様として、PD2S、MT1−MMPおよびAMDP−3の少なくとも1つを過剰発現するトランスジェニック動物を提供する。
本発明を、以下の特定の例によりさらに説明するが、これらはどのような方法でも本発明の範囲および内容を限定するものと解釈されるべきではない。
以下に記載するのは、本発明の経過中に開発された研究ツールであり、1)多数の遺伝子における発現をハイブリダイゼーションにより同時に測定する、簡単で手頃な方法;および2)食作用RPE細胞株および食作用のバイタルアッセイ(vital assay)に基づく、食作用関連遺伝子の同定用のツール、を含む。
図1を参照すると、遺伝子発現の比較ハイブリダイゼーション解析(CHANGE)と呼ばれるマイクロアレイ技術を開発した。λgt11 cDNAライブラリを、分子生物学分野の当業者に知られた技法を用いて、ラットのRPE/脈絡膜RNAおよびヒト網膜RNAから作製した。ライブラリに用いたラットRNAは、周期的な照明(12時間明るい:12時間暗い)下で飼育し、日周期の間の種々の時点で犠牲にした、約2〜3月齢の動物のRPE/脈絡膜から得た。ライブラリから約10,000個のクローンを個別に取り出してプレート上で増幅し、アレイとしてブロットに移した。
in vivoでRPE食作用に関連する遺伝子を同定する好ましいアプローチは、外節(OS)食作用の機能をin vivo系で起こるのと同様の同調的(synchronous)様式で行うin vitro系において、RPE遺伝子発現を解析することである。げっ歯類および他の哺乳類において、OSの脱離および食作用は、日周性リズムに従う。光受容体によるピークの脱離およびRPE細胞による大規模な摂取は、光が発生する直前に始まり、数時間の間にわたって起こることが知られている(LaVail, 1976)。OS食作用の経過中に、培養RPE細胞における差次的発現に基づき食作用遺伝子を成功裡に同定するには、食作用プロセスの動態が培養物にわたり均一であり、特に、それらの隣接物について非同調的食作用を示す細胞からの「ノイズ」を最小化することが好ましい。一次RPE培養物は一般にこの目的に適さないが、これは、一次培養物中のRPE細胞の表現型の顕著な不均一性(heterogeneity)、および、異なる表現型の細胞によって示される、食作用の動態における対応する不均一性のためである。
従って、例えばBPEI−1などの好適なRPE細胞株の細胞は、高密度でプレートし(例えば、6ウェルマルチウェルプレートの1つのウェル当たり約106細胞)、例えば前に記載した培地中(McLaren et al., 1993c; McLaren, 1996)で1〜2日間培養する。
食作用の異なるステージにおいて発現された食作用遺伝子を単離するために、ステージ特異的プローブを、OSを与えた後の種々の時点(例えば、0、1、6、12、18および30時間)でRPE細胞培養物から抽出した全RNAから、および同じ時点において、OSを与えていない対照培養物から抽出した全RNAから、調整した。全RNAの逆転写による「+/−OS」食作用プローブの調製の後に、かかるプローブの対を本明細書のCHANGE解析に用いて、遺伝子アレイ、例えば、本明細書の約10,000個のRPE発現遺伝子のアレイをスクリーニングして、RPE細胞によるOS食作用の間に差次的に発現された遺伝子を同定する。OS食作用の間に発現の変化を示す遺伝子は、続いてDNA配列解析により、標準の技法を用いて同定し、GenBankなどのデータベースの配列と比較する。
この例は、RPE食作用の間に変化した発現を示す遺伝子の、上記方法を用いた単離について記載する。
約10,000個のRPE由来cDNAを含むアレイをスクリーニングする、「+/−OS」プローブを用いたCHANGE解析から、約60個の差次的に発現された推定遺伝子が最初に得られた。差次的発現のノーザンブロット解析による確認を含む、さらなる詳細な解析により、16個の確認された食作用関連遺伝子の最初のサブセットが、さらなる検討のために提供された。上の表1は、本明細書に記載のCHANGE技法により単離された、確認された食作用遺伝子の識別リストおよび配列リストの表示(すなわち、核酸:配列番号1〜15およびアミノ酸:配列番号71〜101)を提供する。
本明細書に記載するのは、推定AMD遺伝子のCHANGEによる単離に用いる手順、およびこれらのAMDとの関係を確認するための方法である。
例2に記載されたものと同様のアプローチにおいて、CHANGE技法を用いて、AMDの病因に役割を果たす遺伝子が疾患の経過の間に発現の変化を示すとの仮定に基づき、AMDに関連する遺伝子を同定した。ヒト提供者の眼は、地方のアイバンクから得た。一般に、死後3時間以内に摘出され、12時間以内に処理に利用可能であった眼が受け入れられた。死亡時刻および処理までに経過した時間に関わらず、組織の実際の質は、肉眼的検査による外観、組織切片の微視的評価、並びにノーザンブロット解析およびRT−PCRにより評価される、得られたRNAの量および質を含む、幾つかの基準により評価した。
「+AMD」プローブを調製するために、ヒト提供者の眼および+3〜+5(中程度から重篤)のAMD変化を有するプールした複数の眼のRPE/脈絡膜から、全RNAを抽出した。年齢を整合させた、罹患していない眼のRPE/脈絡膜からプールされたRNAを用いて、「−AMD」対照プローブを調製した。+/−プローブは、上に記したように、差次的に発現される遺伝子をCHANGEにより同定するために用いた。罹患していない個人と比較して、AMDを有する対象において差次的発現を示す約200個のRPE発現遺伝子が、最初に同定された。
この例は、CHANGEにより食作用およびAMDの両方において差次的に発現されることが見出された例示の遺伝子(すなわち、「AMD関連食作用遺伝子」または「AMDP遺伝子」)である、MT1−MMP(配列番号15)の同定について記載する。
AMDおよびOS食作用の両方に関連する遺伝子を同定するために、2つのCHANGE解析の結果を上記のようにして比較した。両方のスクリーンにおいて差次的に発現された候補遺伝子の中で、クローン91−40が、メタロプロテイナーゼの比較的新しいタイプであるとして、すなわち、AMDへの関与が疑われる遺伝子の要求を合理的に満たすような機能を有するMT1−MMP(Sato et al., 1994)として、選ばれた。これらの機能は、OS食作用における役割(本明細書に記載のもの)、およびプロゲラチナーゼAの活性化および種々の細胞外基質成分に対する分解活性(degradative activity)(Sato et al., 1994; Cao et al., 1995; Pei and Weiss, 1996)を含む。
この例は、AMDおよび黄斑変性患者並びに正常対照集団の遺伝解析のための方法、およびAMDを含む黄斑変性と相関する、MT1−MMP多型の発見を示す結果について記載する。
AMDおよび他の黄斑疾患に罹患した高齢の患者および正常な高齢患者から、末梢血を採集した。白血球からDNAを抽出した。DNAは、地方のアイバンクの提供者の眼の網膜およびRPE/脈絡膜からも、抽出した。提供者の眼における病変の程度は、眼底撮影で記録し、例3に記載された基準を用いて顕微鏡的にグレード付けした。MT1−MMPにおける多型についてのスクリーニングを可能とするために、ヒトMT1−MMPの全10個のエクソンを、公開されたマウス遺伝子構造(Apte et al. 1997)から決定し、下の表3に示されたヒトのエクソン特異的アンプリマー(すなわち、配列番号18〜37)を用いてPCRにより増幅した。
この例は、動物(ラット)モデルにおける遺伝性の網膜変性の速度を、MT1−MMPタンパク質を中和する剤を用いて遅らせることを示す研究について、記載する。
上の例4に記載したように、MT1−MMPは、AMDを有するヒトの眼において、AMDのサルモデルにおいて、およびRPEに基づく遺伝性網膜変性の動物モデルであるRCSラットにおいて、過剰発現されることが見出された。MERTK遺伝子の突然変異によるRCSラットの突然変異体の表現型は、RPE細胞による食作用の摂取相における欠陥を特徴とする。別の研究において、MT1−MMPはCHANGE解析で再度単離され、ここで+/−プローブは突然変異体および年齢を整合させた対照RCSラットの網膜RNAから調製した。RCSラット網膜におけるMT1−MMP発現のノーザンブロット解析は、MT1−MMP mRNAの発現が、このモデルにおいて網膜変性の進行と共に増加したことを、明らかにした。この結果は、MT1−MMPが、網膜変性、特に第一にRPE細胞に影響すると考えられる欠陥に基づく疾患の多くの形態の病因において、共通の役割を果たす可能性があることを示唆する。
AMDの病因の研究は、例えば候補の治療化合物およびアプローチを試験するのに有用な、適切で実用的な動物モデルの欠如によって妨げられている。この例は、本明細書においてAMDで上方制御されることが示された遺伝子を過剰発現する、マウスにおける、AMDの動物モデルの作成について記載する。好ましい態様において、過剰発現される遺伝子は、プロスタグランジンD2合成酵素(PD2S)、MT1−MMP、およびAMDP−3であり、それぞれ本明細書において配列番号2、15、および17と同定されたcDNA配列を含む。幾つかの態様において、遺伝子は条件付きで過剰発現され、幾つかの種類において、遺伝子は、動物の光受容体、RPE細胞、および/または脈絡膜細胞においてのみ、過剰発現される。
この例は、食作用関連またはAMD関連遺伝子の1つまたは2つ以上の多型変異体を発現する、AMDおよび他の網膜変性のマウスモデルの作成について記載する。
上に示したように、MT1−MMPを含む遺伝子のある多型変異体は、AMDを有する患者のDNAにおいてより高い頻度で見出される。ヒトの状態をモデル化するために、例えばMT1−MMPなどのヒト遺伝子の多型および野生型を発現するトランスジェニックマウスモデルを、以下のように作成する。最初に、マウスMT1−MMP遺伝子のベースライン状態を好ましくは決定する。例えば、ヒトMT1−MMP DNA配列のD273N多型の位置に位置する野生型のアミノ酸残基は、ヒトおよびマウスにおいて保全されることが決定された。この残基における多型は、マウスにおいては示されていない(マウスゲノムプロジェクト)。野生型残基の存在は、トランスジェニック作成に用いられるマウスにおいて、尾の生検、DNA単離、および遺伝子型決定により確認される。
AMDおよび他の網膜変性の動物モデルの他の態様は、AMDと既知の関連を有する少なくとも2つの遺伝子の多型変異体を発現する、ポリトランスジェニックマウスである。好ましい態様において、動物は、食作用および/またはAMDと相関を示す少なくとも1つの他の多型遺伝子変異体と組み合わせて、MT1−MMPの多型変異体を発現する。
同様に、AMDの「集団(collective)」病因論を反映するポリトランスジェニックモデルは、重要な機能(例えばOS食作用)のメカニズムにおける既知の関与を有する遺伝子の多型変異体と、MT1−MMP(本明細書に開示された、実証された食作用関連遺伝子;野生型cDNA配列:配列番号15;野生型アミノ酸配列:配列番号100)の多型変異体を組み合わせる。かかる遺伝子は、例えば、本明細書に開示された(上の表1および2を参照)、食作用関連遺伝子PHG−1〜PHG−15(配列番号1〜14)並びにAMDP−2および3(配列番号16および17)の多型変異体を含む。
本明細書に引用した文献を便宜上以下にリストし、これらはその全体を、本明細書に参照として組み込む。
Claims (14)
- 対象における網膜または脈絡膜変性疾患または状態を、遅延または逆転させるための組成物であって、
膜型マトリックスメタロプロテアーゼ1(MT1−MMP)に特異的に結合する抗体、および/またはMT1−MMPを下方制御するオリゴヌクレオチドであって、リボザイム、アンチセンスRNA、干渉RNA(RNAi)分子および三重らせん形成性分子からなる群から選択されるもの
を含む、前記組成物。 - 抗体が、MT1−MMPの生物活性を中和する、請求項1に記載の組成物。
- 生物活性が、プロゲラチナーゼAの活性化または細胞外基質の分解である、請求項2に記載の組成物。
- MT1−MMPが、配列番号15で表わされるヌクレオチド配列によってコードされる、請求項1〜3のいずれか一項に記載の組成物。
- 網膜または脈絡膜変性疾患または状態が、加齢黄班変性(AMD)である、請求項1〜4のいずれか一項に記載の組成物。
- 対象がAMDを患っている、請求項5に記載の組成物。
- 対象が、AMDを発症するリスクを有する、請求項5に記載の組成物。
- 組成物が、網膜または脈絡膜変性疾患または状態を遅延させる、請求項1に記載の組成物。
- 組成物が、網膜または脈絡膜変性疾患または状態を逆転させる、請求項1に記載の組成物。
- 抗体が、網膜または脈絡膜細胞と接触する、請求項1〜9のいずれか一項に記載の組成物。
- 細胞が、光受容体、RPE細胞、ミュラー細胞、または、内皮細胞、平滑筋細胞、白血球、マクロファージ、メラノサイトおよび繊維芽細胞からなる群から選択される脈絡膜の細胞型である、請求項10に記載の組成物。
