一般:[Nle8,18,Tyr34]bPTH(1−34)アミド(PTH(1−34))、[Nle8,18,Tyr34]bPTH(3−34)アミド(PTH(3−34))及び[Nle8,18,Tyr34]bPTH(7−34)アミド(PTH(7−34))はBachem Fine Chemicals, Torrance, CAから購入した;[Tyr36]PTHrP(1−36)アミド(PTHrP(1−36))は記載(Keutman et al., Endocrinology 117:1230,1985)されているように,Applied Biosystems Synthesizer 420Aを用いて合成した。ダルベッコ変法イーグル培地(DMEM)、EDTA/トリプシン及びゲンタマイシンはGIBCO(Grand Island, NY)から;牛胎児血清(FBS)はHiclone Laboratory, Logan, UTから入手した。ヒト腎臓の総RNAはスエーデン、ウプサラ、大学病院のPer Hellmann氏によって提供された。オリゴヌクレオチドプライマーはApplied Biosystems 380B DNA Synthesizerを用いて合成した。制限酵素類、クレノウ酵素、T4ポリヌクレオチドキナーゼとT4DNAリガーゼは、New England Biolabs,Beverly, MAから購入した。子牛アルカリホスファターゼは、Boehringer Mannheim, ドイツから購入した。すべての他の試薬は入手可能の最高純度のものであった。
使用された細胞には、COS細胞、OK細胞、SaOS−2細胞、CHO細胞、AtT20細胞、LLC−PK1細胞及びUMR−106細胞が含まれ、Americann Type Culture Collection(Rockland, Maryland)、受託番号、夫々、CRL1650、CRL6551、HTB85、CCL61、CCL89、CL101及びCRL1161を含む種々の供給源から入手した。ROS17/2及びROS 17/2.8はGideon Rodan博士(Merck Laboratories, West Point, PA)を含むいくつかの供給源から入手した。MC−3T3細胞はマウスの骨細胞から誘導されるが、滋野長平博士(医化学、医学部、京都大学、京都、日本)を含む種々の供給源からも入手出来る。
ラット及びオポッサムのPTH/PTHrPレセプターをコードしているcDNAクローンの分離:
総RNAは、最初にラットの骨肉腫(ROS)細胞(ROS 17/2.8)とオポッサムの腎臓(OK)細胞から、グアニジンイソチオシアネートを用いる標準法(Ullrich et al.,Science 196: 1313,1977; Chirgwin et al., Biochemistry 24: 5294,1979)と塩化セシウム密度勾配遠心法(Gilsen et al., Biochemistry 13: 2633,1974)によって分離された。次いでポリA+RNAs(mRNAs)は、AvivとLeder(Proc. Natl. Acad. Sci.USA 69:14087, 1972)の方法により、総RNAsをオリゴdTカラム(Pharmacia,Piscataway, NJ)を通した後回収された。ROS 17/2.8 mRNAからのcDNAライブラリーは、ポリA+RNAからGublerとHoffman(Gene(Amst.) 25: 263, 1983)の方法を用いて作られた。オリゴdTをプライマーとする、及びランダムプライマーをプライマーとするcDNAは、夫々、ポリA+ROS 17/2.8及びOK細胞のmRNAから合成された(AvivとLeder,前記)。cDNAはBstX1リンカー(Invitrogen,San Diego, CA)に連結され、5−20%の酢酸カリウム密度勾配遠心(3時間、55,000xg)によってサイズ選別が行われた。サイズ選別されたcDNAは、次いで、非自己アニーリングBstX1制限部位を用いてプラスミドベクター、pcDNAI(Invitrogen)に挿入された。その結果得られたプラスミドライブラリーは、次に、より大きなヘルパープラスミド、p3を持つE.coli(MC1061/P3、Invitrogen)を形質転換するのに用いられた。p3プラスミドは、2つの遺伝子にアンバ−変異を持ち、アンピシリン及びテトラサイクリン耐性をコードしている。アンピシリン及びテトラサイクリンを用いると、p3とpcDNA I内に含まれているtRNAサプレッサー遺伝子の両方を持つ細胞のみが成長することが出来た。変異した細菌は、次いで集密状態まで培養され、プラスミドcDNAは標準技法(例えば、Ausebel et al., Current Protocols in Molecular Biology、 John Wiley Sons, New York、 1989を参照のこと)により分離された。これらDNAは、次いでDEAE−デキストラン溶液に取り込まれ、予め“サイドフラスコ”(Nunc、 デンマーク)中で75%の集密度にまで培養されたアフリカミドリザルの腎臓(COS)細胞に移入するのに用いられた。
ラムダGT10中のヒト腎臓のオリゴdTをプライマーとするcDNAライブラリー(1.7x106 独立クローン)及びEMBL3(Sp6/T7)(Clontech, Palo Alto, CA)中のヒト胎盤DNAのゲノムライブラリー(2.5x106 独立クローン)は、32P−標識(ランダムプライマーをプライマーとする標識キット、ベーリンガー、マンハイム、ドイツ)されたBamHI/NotI 1.8kbの制限酵素断片で、ラットの骨のPTH/PTHrPレセプターをコーディング配列の大部分をコードしているものとのプラークハイブリッド形成法(Sambrook et al., Molecular Cloning:A Laboratory Manual,2版、108−113頁、ColdSpring Harbor Laboratory, Cold Spring Harbor, NY、 1989)によってスクリーニングされた(図3)。ニトロセルロースフィルターは、50%ホルムアミド、4xクエン酸ナトリウム生理食塩水(SSC; 1xSSC: 300mMNaCl,30mMクエン酸ナトリウム、pH 7.0)、2xデンハルト溶液、10%硫酸デキストラン、100μg/mlのサケ精子DNA(最終濃度)を含む前ハイブリッド形成溶液中、42℃で4時間インキュベートした。ハイブリッド形成は、同じ溶液中で42℃で18−24時間行われた。フィルターは、2xSSC/0.1%SDSで室温で30分間、次いで、 1xSSC/0.1%SDSで45℃で30分間洗浄した。フイルムは、増強スクリーンを用い、−80℃で18−24時間、露出した。
3μgのヒト腎臓由来のポリ(A)+RNA(Clontech,Palo Alto, CA)を73.5μlの水の中で100℃で30秒インキュベートし、氷冷して反応を静め、20μlの5xRT緩衝液(1xRT緩衝液:40mMトリス塩酸、pH8.2、40mMKCl、6.6mMMgCl2、10mMジチオトレイトールと各5mMのデオキシヌクレオシド三リン酸(dNTP))、2μl(4単位)のRNasin(Promega Biotec, Madison, WI)、1μl(80pmol/μl)のヒトcDNAプライマーH12(5´−AGATGAGGCTGTGCAGGT−3´;配列番号:14)と80単位のトリ骨髄芽球炎ウイルスの逆転写酵素(Life Sciences, St. Pertersburg,FL)に加えた。反応混合物を42℃で40分間インキュベートした。次いで、10分の1容の最初の鎖を合成する反応混合物は、PCRにより、3mMMgSO4 、200μMのdNTP、2単位のVentポリメラーゼ(New England Biolab,Beverly, MA)と各2μMの正方向と逆方向のプライマー(PCRの条件:変性、1分間、94℃;アニーリング、1分間、50℃;延長、72℃で3分間;40サイクル)を含む最終容量100μlの中で増幅された。
2つの独立したPCRは、以下の2つの異なった正方向のプライマーを用いて行われた:i)2つの以前にクローン化されたPTH/PTHrPレセプター

(上述)コーディング末端に基づく変性プライマーRK−1(5´−GGAATTCCATGGGAGCGGCCCGGAT−3´;配列番号:15)、及び、ii)ヒトゲノムクローンHPG1の5´−非コーディング領域に基づくプライマーRK−2(5´−CGGGATCCCGCGGCCCTAGGCGGT−3´;配列番号:16)。両PCR反応はヒトのPTH/PTHrPレセプターのコーディング領域の713〜731番目のヌクレオチドを現す逆プライマーH26(5´−AGTATAGCGTCCTTGACGA−3´)を用いた(図4)。PCRの生成物はクレノー酵素により末端を平滑にされ、EcoRVで切断されて、脱リン酸されたpcDNAIにクローン化された。
COS細胞に発現されたクローン化レセプターの機能的諸性質を特徴づける試験には以下のものが含まれた:
I)PTHとPTHrPの断片及び類似体の結合、
II)PTHとPTHrPの断片及び類似体によるサイクリックAMP蓄積の刺激、
III)PTHとPTHrPの断片及び類似体による細胞内の遊離カルシウムの増大、及び
IV)PTHとPTHrPの断片及び類似体によるイノシトールリン酸代謝の活性化。方法論は次の通りである:
[Nle8 ,Nle18,Tyr34]bPTH−(1−34)アミド(NlePTH)及び[Tyr36]PTHrP(1−36)アミド(PTHrP)は Na 125I(担体なし、New England Nuclear, Boston, MA)により既報(Segre et al., J.Biol.Chem. 254:6980, 1979)に従ってヨード化され、逆相HPLCによって精製された。簡略に述べると、標識されたペプチドは0.1%のトリフルオロ酢酸(TFA)に溶かされ、C18Sep−pakカートリッジ(Waters Associates, Inc., Milford, MA)にかけられ、0.1%TFA中60%アセトニトリル溶液によって溶出された。凍結乾燥後、ラジオリガンドは、次いで、C18−μBondapakカラム(3.9mmx30cm。Waters Associates)にかけられ、0.1%TFA中30−50%のアセトニトリルの直線密度勾配を用い、2ml/分の流速で30分かけて溶出した。ラジオリガンドは、2つのピ−クに別れて溶出した;最初のピークは約38%のアセトニトリルのところで溶出し、より高い総結合と特異的結合を示したので、この研究に用いられた。比活性は500±75mCi/mgで、これは平均のヨード−ペプチド比1に相当する。
細胞内cAMPの蓄積は既述(Abou−Samra et al., J.Biol.Chem.262:1129, 1986)のようにして測定された。24穴プレート内の細胞を0.1%BSAと2mMIBMXを含む培地で洗浄した。次いで、細胞はPTHまたはPTHrPと15分間、37℃でインキュベートした。上清を除き、細胞は直ちにプレート全体をドライアイス粉末中に置くことによって凍結した。細胞内cAMPは1mlの50mMHCl中で細胞を融かすことによって抽出され、抗cAMP抗体(例、Sigma, St.Louis, MO)を用い、特異的ラジオイムノアッセイによって分析された。cAMPの代わりにトレーサーとして用いられたcAMP類似体(2´−O−モノサクシニル−アデノシン3´:5´−サイクリックモノリン酸トシルメチルエステル、Sigmaより購入)は、クロラミンT法によりヨウ素化された。遊離のヨウ素は、ヨウ素化されたcAMP類似体をC18Sep−pakカートリッジ(Waters, Milford, MA)に吸着することによって除去した。
dH2Oで洗浄後、ヨウ化cAMP類似体はSep−pakカートリッジから40%アセトニトリル(ACN)と0.1%トリフルオロ酢酸(TFA)によって溶出された。ヨウ化cAMP類似体は凍結乾燥され、1mlの0.1%TFAにもどし、C18逆相FPLCカラム(Waters)に注入された。カラムを0.10%ACNの1%TFA溶液で平衡化し、0.1%TFA中、10−30%ACNの密度勾配で溶出した。この操作によりモノヨウ化cAMP類似体を、非ヨウ化cAMP類似体から分離することが可能となる。トレーサーは−20℃で保存する時は、4ヶ月までは安定である。アッセイに用いられた標準品、アデノシン3´,5´りん酸はSigmaから購入した。試料(1−10μlのHCl抽出液)あるいは標準品(0.04−100fmol/チュ−ブ)を50mM酢酸ナトリウム(pH5.5)で希釈し、トリエチルアミンと無水酢酸の混合物(容量比 2:1)10μlでアセチル化した。アセチル化後、cAMP抗血清を、PBS(pH7.4)、5mMEDTA及び1%正常ウサギ血清中に作られた原液(1:4000)から加えた。トレーサーは、0.1%BSAを含むPBS(pH7.4)で希釈して加えられた(20,000cpm/チューブ)。アッセイは4℃で一晩インキュベートした。結合したトレーサーは100μlのヤギの抗ウサギ抗血清(1:20、PBS中)及び1mlの7%ポリエチレングリコール(分子量5000−6000)を加えて沈殿させ、2000rpmで30分間、4℃で遠心した。上清を除き、結合した放射能をγ−カウンター(Micromedic)で測定した。標準曲線はMicromedicから提供された4−パラメーターRIAプログラムを用いて算定した。典型的な例では、アッセイ感度は0.1fmol/チューブで、トレーサーの50%を置換する標準濃度は5fmol/チューブである。
PTH/PTHrPレセプターcDNAを移入された細胞の細胞内カルシウムレベルの測定は、Fura−2 AM(Fura−2のアセトメトキシエステル、Molecular Probes Inc., Eugene, OR)を負荷した細胞を用いて行われた。方法論の詳細は以下の通りである:
COS細胞を塗布したカバーガラスを、Fura−2 AMを負荷し、以下のものを含む(mM濃度)緩衝液中でインキュベートした:HEPES(N−[2−ヒドロキシエチル]ピペラジン−N´−[2−エタンスルホン酸]),20;CaCl2 ,1; KCl,5; NaCl,145; MgSO4 ,0.5;NaHCO3 ,25; K2HPO4,1.4; グルコース,10;およびFura−2 AM(1−[2−(5´−カルボキシオキサゾール−2´−イル)−6−アミノベンゾフラン−5−オキシ]−2−(2´−アミノ−5´−メチルフェノキシ)エタン−N,N,N´,N´−四酢酸ペンタアセトキシメチルエステル),0.5; 37℃でpH7.4、95%空気と5%CO2 とを45分間通した。Fura−2 AMを負荷した細胞は、次いで、以下のものを含む(mM濃度で)修正クレブス・ヘンゼライト(KH)緩衝液で洗浄した: HEPES、 20; CaCl2 , 1; KCl, 5; NaCl, 145;MgSO4 、 0.5; Na2HPO4 , 1; グルコース、 5; pH7.4。エステルの開裂が起きたことをチェックするために、Fura−2 AMインキュベーション後、時間を変えて励起スペクトルを測定した。インキュベーション開始後5分で、励起スペクトルは約360nmにピークがあり、Fura−2 AMの不完全な加水分解を反映していたが、30分を越えると、励起スペクトルはFura−2の特徴である約345nmにピークがあった。
個々の細胞の蛍光を測定するために、カバーガラスを顕微鏡の組織チャンバー(Biophysica Technologies, Inc., MD)に置いた。チャンバーは、シリコンラバーシールを持つテフロン製の浅い、傾斜のついたコンパートメントからなっていた。カバーガラスはチャンバーの底の役割をした。カバーガラスを所定の場所に密着させるシリコンラバーには、ヒーター/クーラーリングが封じ込められていた。温度はヒーターに0−7.4ボルトを加えることによって、22℃〜37℃の間で変動させた。温度を特定しない限り、実験は37℃で施行された。チャンバーは倒立顕微鏡(Zeiss IM−35,Thornwood, NY)の載物台に載せた。Fura−2の蛍光のために、コンデンサー(Photon Technologies Intenational(PTI) Inc., NJ)の焦点におかれた75ワットのキセノンアーク灯で励起された。鏡板が透過光分割器と交番する回転チョッパーによって交番される回折格子単色光分光器が、波長選定に使用された。単色光分光器の出力は結集されて一本の共通の光路を形成し、調節可能な虹彩を通して発光源のハウジングを励起した。次いで、光は石英レンズを、そして100倍のニコン蛍光対物レンズを通して二色性の鏡を通過した。光量子計算PMT装置による検出が光の出力測定に使用された。データの分析は、MS−DOS操作系を用いるIBM互換性AT/286コンピュータにPTIソフトウエアを使用して行った。データは圧縮二者択一性形式に保存、操作された。
PTH/PTHrPレセプターを移入されたCOS細胞におけるイノシトールりん酸の代謝レベルは既報の方法を用いて測定された(Bonventre et al., J.Biol.Chem. 265: 4934, 1990)。
分子的性質
2つの別個のクローン(OK−H及びOK−O)は、ともにOK細胞のcDNAライブラリーから分離されたが、およそ2キロベースの長さを持っていた。これらのクローンの決定されたヌクレオチド配列と予想されるアミノ酸配列は、夫々、図1(配列番号:1)及び図2(配列番号:2)に示されている。ROS細胞のcDNAライブラリーから分離されたR15Bクローンは、およそ4キロベースの長さであった。ラットの骨のPTH/PTHrPレセプターの決定されたヌクレオチド配列と予想されるアミノ酸配列は図3(配列番号:3)に示されている。
オポッサムとラットのPTH/PTHrPレセプターの生物学的性質の機能的特性の検討は一過性に移入されたCOS細胞において、放射性リガンドとして 125I−PTHrPと 125I−NlePTHの両方を用いるラヂオレセプターアッセイ法により、また、リガンドによるcAMPの蓄積、細胞内遊離カルシウムの増加、及びイノシトールりん酸代謝の促進を測定するバイオアッセイにより、上記の方法によって行われた。
ヒトのPTH/PTHrPレセプターのアミノ酸配列は、ラット、真獣類動物からの骨PTH/PTHrPレセプターと比べて、非常に高度の保存を示すが、オポッサム、有袋動物のPTH/PTHrPレセプターとの配列の相同性はそれほど著明ではない。オポッサムの腎臓及びラットの骨のレセプターのように、ヒトの腎臓のレセプターは、PTHまたはPTHrPで刺激された時、細胞内のcAMPと遊離のカルシウムの増加を誘引する。ヒトのPTH/PTHrPレセプターとオポッサム及びラットの同族体間の高度の相同性にも拘らず、一過性に発現されたヒトのレセプターは、ラットの骨のレセプターとは異なった機能的特徴を持っている。これには、PTH(1−34)に対するやや高い親和性とPTHrP(1−36)に対する著しく低い親和性が含まれる。より高い親和性はPTH(3−34)と特にPTH(7−34)に対して認められ、そのヒトのレセプターに対する親和性はラット骨レセプターと比較して約35倍も高かった。これらの発見は、PTH/PTHrP類自体の将来の開発に重要な意味を持つかも知れない、それは種得異的な組織は、インビトロでアンタゴニスト(とアゴニスト)の有効性検査に対する適当な組織であることを予言するからである。
PTHとPTHrPレセプターの生化学的諸性質は、これらがG−蛋白質連携の膜レセプターとして知られている膜レセプター分子群のメンバーであることを示唆している。よく調べられているG−蛋白質レセプターの構造的特徴は、それらがすべて、数個の連続した疎水性アミノ酸からなる少なくとも7つの領域を持ち、これらの領域の各々が原形質膜を貫通するのに十分な長さのものであることである。
特許手続きの目的のための微生物寄託についての国際的承認に関するブダペスト条約の約条の下に、cDNA発現プラスミドR15B、OK−O及びOK−H;ファージHPG1;及びヒトのクローンの一部を含むプラスミド(名称8A6)が、American Type Culture Collection (ATCC)に寄託され、そこでは、夫々の受託番号ATCC No.68571,68572,68573,40998,及び68570を付けられている。申請者の指定代理人、The General Hospital Corporationは、ATCCがこの寄託の永続性と、もし、特許が許可された場合には、その容易な分譲を提供する寄託所であることを申し立てる。このように寄託物質の分譲に関するすべての制限は、特許の許可と共に改変されることなく撤去される。物質は特許申請の未決期間中は、37 CFR 1.14及び35 U.S.C.122の下、コミッショナーによって資格ありと決定された者に分譲される。寄託された物質はあらゆる必要な注意を払って維持され、寄託されたプラスミドの試料提供に対する最も新しい要請後少なくとも5年間、そして、いかなる場合でも、寄託の日付後、少なくとも30年の期間、あるいは特許の実行期間、いずれか長い方の期間、生存状態で、汚染されないように保存を継続する。申請者の指定代理人は、もし、寄託物の状態のために、要請があっても寄託所が試料を提供出来ないならば、寄託物を取り替える責任のあることを認める。
本発明によれば、ポリペプチドには、図1−3及び6に夫々、示されているオポッサム、ラット及びヒトの副甲状腺ホルモンレセプター、及びいかなる他の自然界に存在するレセプターにせよ、これらのレセプターをクローン化して発現するのに用いられたものに類似の方法により、あるいは、ここに記述されている配列の1つの全体または1部をプローブとして利用する方法によって生成され得るものを含む。さらに、副甲状腺ホルモン、または副甲状腺ホルモン関連蛋白質に対する結合能のあるPTHレセプターの類似体あるいは断片も本発明の範囲内にある。

これらの断片はKeutmannらの方法(Ednocrinology 117: 1230, 1984)によって合成され,HPLCによって精製された。
本発明によるポリペプチドは、本発明の細胞レセプターの一部または全体をコードしている配列を持つ組換え核酸から、なにか適切な発現系、例えば、適当な宿主細胞(原核あるいは真核)の、適当な発現伝達体(例、pcDNAI)内の組換え核酸による突然変異を用いて、発現することにより生成することが可能である。正確にいかなる細胞を用いるかは、本発明にとって重要ではない。しかしながら、本発明のポペプチドがPTH/PTHrPレセプターの全体または一部を含む場合には、以下の宿主細胞が好適である:COS細胞、LLC−PK1細胞、OK細胞、AtT20細胞、及びCHO細胞。移入の方法及び発現伝達体の選択は選ばれた宿主系に依存する。哺乳類細胞の移入の方法は、例えば、Ausubel et al.(Current Protocols in Molecular Biology, John Willey & Sons, New York, 1989)に記載されており、発現伝達体は、例えば、Cloning Vectors: A Laboratory Manual (P.H. Pouwels et al., 1985, 補遺、 1987)に論じられているものから選んでもよい。安定に移入された細胞は、レセプターDNAを宿主細胞の染色体に組み込むことによって作ることが出来る。適当なDNAは、pcDNA,pcDNAI−Neo、あるいは他の適切なプラスミドに挿入され、次いで、細胞は,このプラスミドにより、psV−2−Neo、あるいはpsV−2−DHFRとの共同移入をして、あるいはしないで、標準電気が孔、りん酸カルシウム、及び/あるいはDEAE/デキストラン技法によって移入される。移入された細胞の選択は、累進的に上昇するレベルのG418(Geneticin, GIBCO)、そしてもし必要ならば、メトトレキサートを用いて行われる。
1)図3に示されるようなラットの骨のPTH/PTHrPレセプターの、推定リーダー配列を含む、アミノ酸1−192からなるアミノ末端部分。
2)図3に示されるようなラットの骨のPTH/PTHrPレセプターの、推定リーダー配列を除外した、アミノ酸27−192からなるアミノ酸末端部分。 3)図3に示されるようなラットの骨の完全長のPTH/PTHrPレセプター。
4)PR−1(上記のような)。
5)PR−2(上記のような)。
本発明の細胞レセプターあるいはレセプター断片は、専門家にはよく知られているなにか通常の方法によって、ポリクローナル抗体を産生する方法、及びモノクローナル抗体を産生する方法を含め、抗体を産生するのに用いることが出来る。例えば、PTHレセプターの推定アミノ酸配列は、他のG蛋白質連鎖レセプターに見出だされる領域に類似した細胞外と細胞内と思われる領域(図21を見よ)を点々配置した数個の貫膜(即ち、疎水性)ドメインを持つと思われる蛋白質構造を示している。この情報は、レセプター蛋白質の、細胞外に露出している見込みがあり、従ってin vivoで抗体に提示されるであろう領域の選択を手引きするために用いることが出来る。このような領域の1つ、あるいはさらに多くを表現する短いペプチドを合成し(例えば、化学的に、あるいは組換えDNA技法により)、ポリクローナル、あるいはモノクローナル抗体を産生するために動物(例えば、ウサギ、あるいはマウス)を免疫するのに用いることが出来る。例えば、上記のPTH/PTHrPレセプターのペプチドのあるもの(RP−1、RP−5、及びRP−6)は、標準の技法を用いて化学的に合成され、以下の手順によりウサギにポリクローナル抗体を産生するのに用いられた:
抗体価は2つの方法のいずれかを用いて評価される。第一に、抗体を1%正常ウサギ血清で連続希釈し、125I−標識PTH/PTHrPレセプター断片と常法(例えば、Segre et al., 上記)により24時間、4℃でインキュベーションする。結合した125I−標識PTH/PTHrPレセプター断片は、非結合のものから、100μlの第二抗体(抗ウサギIgG、 Sigma)を20倍に希釈したもの及び1mlの5%ポリエチレングリコールを加え、次いで、2000rpmで30分、4℃で遠心することによって分離される。上清を除去し、ペレットをγカウンターで、放射活性の分析をする。第二の方法では、自然型の(例えば、ROS 17/2.8、OK、SaOS−2細胞)あるいは組換え(R15B、OK−OあるいはOK−Hを移入されたCOS細胞あるいはCHO細胞)PTH/PTYHrPレセプターを発現している細胞系が、連続希釈された抗体と4℃、20℃、あるいは37℃で1−4時間インキュベートされる。細胞をPBSで(3回)洗浄し、 125I−標識(NEM、 Dupont)あるいはFITC−標識(Sigma)の第二抗体と2時間、4℃でインキュベートする。洗浄後(PBSで3回)、細胞は0.1MNaOHで溶解してγカウンターで測定されるか(もし、 125I−標識の第二抗体が用いられた時)、あるいは1%パラフォルムアルデヒドで固定し、蛍光顕微鏡法で検査された(もし、FITC−標識の第二抗体が用いられた時)。
本発明のポリペプチド及び抗体ならびに他の化合物は、PTH競合性、及びアンタゴニスト的あるいはアゴニスト的な性質について、ここに記載のアッセイを用いてスクリーニングすることが出来る。
本発明のポリペプチド、抗体、及び他の化合物は、本発明の細胞レセプターとその特異的リガンド間の相互作用に関連すると特徴づけられるような疾病の診断、分類、予後、及び/あるいは処置に有用である。例えば、高カルシウム血症及び低カルシウム血症のいくつかの型は、PTH及びPTHrPとPTH/PTHrPレセプター(複数)間の相互作用に関連している。高カルシウム血症は、血清のカルシウムレベルに異常な上昇のある状態である; それは、しばしば、副甲状腺機能亢進症、骨粗鬆症、乳腺、肺、及び前立腺の癌腫、食道の頭部及び頚部の類表皮癌、多発性骨髄腫、及び副腎腫を含む他の病気に関連している。低カルシウム血症は、血清カルシウムレベルが異常に低い状態であり、例えば、甲状腺手術後に起こる有効PTHの不足から生じることがある。
他の実施態様は、以下の特許請求の範囲内にある。例えば、本発明の核酸には、トリ、あるいは有袋類、げっ歯類、またはヒトのような哺乳類を含むいかなる脊椎動物種にせよ、これらから本来分離された遺伝子、あるいはcDNAあるいはRNAを包含する。オポッサム、ラット、及びヒトのような多様な動物種からのPTHレセプターに対して証明された高度の相同性は、ここに開示されたPTHレセプターの分離方法が、多様な動物種からの関連した細胞レセプターの分離に広く応用されることを示唆している。
Claims (2)
- 配列番号1、3及び4からなる群より選択される全長アミノ酸配列よりは少なく、且つ少なくとも10個のアミノ酸を含むポリペプチドであって、各々、配列番号1、3または4のアミノ酸配列からなるポリペプチドの抗原性を有するポリペプチド。
- 副甲状腺ホルモンまたは副甲状腺ホルモン関連蛋白質と結合し得るポリペプチドである、請求項1に記載のポリペプチド。
