Description

一態様において、本発明は、水溶性ポリマーと結合したIL−18ポリペプチドを含む生物学的に活性な組成物に関し、ここにおいて、前記ポリペプチドはヒトIL−18、その置換突然変異体、またはそのフラグメントである。
第二の態様において、本発明は、治療的に有効な量の、水溶性ポリマーに共有結合したIL−18ポリペプチド生物学的に活性な組成物を投与することにより、患者における癌を治療する方法に関し、ここにおいて、前記ポリペプチドはヒトIL−18またはその置換突然変異体である。もう一つ別の態様において、本発明は:腎臓細胞癌、黒色腫、他のIL−18反応性腫瘍タイプ(例えば、骨髄腫およびリンパ腫)、および黒色腫からなる群から選択される免疫反応性腫瘍を治療するための方法に関する。

本発明は、ヒトIL−18ポリペプチドを含む組成物を提供し、ここにおいて、該ポリペプチドは水溶性ポリマーに結合している。本発明の結合したポリペプチドは、対応する未結合ポリペプチドと比較して予想外の生物学的性質を示す。
免疫療法は腫瘍学治療手段に、副作用が減少した抗腫瘍効果を示し、患者の生活の質を改善する可能性という有益な相加である。インターロイキン−18(IL−18)は現在、腎臓細胞癌および黒色腫の腫瘍免疫療法の新規形態として研究されている。これらの腫瘍タイプはどちらも免疫感受性であると考えられ、単一の薬剤としてIL−18に反応する。IL−18と細胞毒性剤および他の生物学的製剤の組み合わせは、IL−18の臨床用途を異なるタイプの血液学的および充実性腫瘍に拡大する。
本発明において有用なポリペプチドは、硫酸アンモニウムまたはエタノール沈殿、酸抽出、アニオンまたはカチオン交換クロマトグラフィー、ホスホセルロースクロマトグラフィー、疎水性相互作用クロマトグラフィー、アフィニティークロマトグラフィー、ヒドロキシアパタイトクロマトグラフィーおよびレシチンクロマトグラフィーをはじめとする周知の方法により組み換え細胞培養物から回収し、精製することができる。最も好ましくは、精製するために高性能液体クロマトグラフィーを用いることができる。単離および精製中にポリペプチドが変性する場合、活性構造を再生するためにタンパク質のリホールディングのための周知の技術を用いることができる。
本発明の組成物は、水溶性ポリマーがポリエチレングリコールホモポリマーであるものである。本発明における使用に好適なポリマーは、分岐鎖、非分岐鎖、星状または直鎖である。本発明の一実施形態において、このような組成物は直鎖ポリエチレングリコールホモポリマーを含む。本発明の別の実施形態において、このような組成物は、分岐鎖ポリエチレングリコールホモポリマーを含む。本発明における使用に好適なポリマーは、次の特許、特許出願および刊行物に開示されている:米国特許第4,097,470号、第4,847,325号、第5,037,883号、第5,252,714号、第5,580,853号、第5,643,575号、第5,672,662号、第5,739,208号、第5,747,446号、第5,824,784号、第5,846,951号、第5,880,255号、第5,919,455号、第5,919,758号、第5,932,462号、第5,985,263号、第5,951,974号、第5,990,237号、第6,042,822号、第6,046,30号、第6,107,272号および第6,113,906号;国際特許公開番号WO92/16555;欧州特許公開番号EP727,437、EP727,438、EP439,508およびEP714,402;Zalipsky、S.(1995)Bioconjugate Chem 6:150−165;Gregoriadis、G.ら、(1999) Pharma Sciences 9:61−66(それぞれは本発明の一部として参照される)。さらに、ポリペプチドおよび他の生物学的物質との結合を促進するために修飾された誘導化または官能化されたポリマーが本発明における使用に好適である。例えば、遊離アミノ基(例えばリシン残基でのイプシロンアミノ基またはN末端での遊離アミノ基)、システイン残基上の遊離スルフヒドリル基、または炭水化物部分による結合を促進するためのポリマーの修飾が望ましい。有用なポリマーは、ポリエチレングリコールのモノメトキシ誘導体(mPEG)も含む。本発明において用いることができる官能化ポリマーとしては、これらに限定されないが:メトキシポリエチレングリコールスクシニミジルプロピオネート;メトキシポリエチレングリコールスクシニミジルブタノエート;カルボキシメチル化メトキシポリエチレングリコールのスクシニミジルエステル;メトキシポリエチレングリコールアルデヒド;メトキシポリエチレングリコールヒドラジド、メトキシポリエチレングリコールヨードアセトアミド;メトキシポリエチレングリコールマレイミド;およびメトキシポリエチレングリコールトレシレートが挙げられる。
結合を行うための反応条件は、タンパク質の反応基がシステイン上の遊離チオール基またはメチオニン上のチオエーテル基であるならば、pH約6〜9、さらに好ましくはpH6〜7で前記結合反応を行うことをさらに含む。前記方法を用いて、タンパク質をポリマーに添加された少なくとも1つの末端チオール反応性基により結合させる。これらのチオール反応性基としては、これらに限定されないが:ハロアセチル、マレイミド、ピリジルジスルフィド誘導体、アジリジン、アクリロイル誘導体、アリール化剤が挙げられる。用いられる未反応活性化ポリマーの量は、通常、モノマーまたはマルチマー(好ましくはダイマー)形態であるタンパク質よりも1〜10倍過剰の活性化ポリマーである。一般に、反応プロセスは、活性化ポリマーをタンパク質と2:1(ポリマー:ポリマー)の比で反応させることを含む。典型的には、反応はリン酸塩緩衝液pH6.2、100mM NaCl中、4℃で約1時間〜約10時間行われる。結合後、所望の結合タンパク質を液体クロマトグラフィーなどにより回収し、精製する。
さらに別の態様において、本発明は、治療的に有効な量の本発明の組成物を、治療的に許容される担体または賦形剤との組み合わせにおいて含む医薬組成物を提供する。このような担体としては、これらに限定されないが、食塩水、緩衝食塩水、デキストロース、水、グリセロール、エタノール、およびその組み合わせが挙げられる。本発明はさらに、本発明の前記組成の1以上の成分を充填された1以上の容器を含む医薬パックおよびキットに関する。本発明の組成物は、単独または他の化合物、例えば治療化合物との組み合わせにおいて用いることができる。
これらに限定されないが、本明細書において言及し、または本出願が優先権を主張する特許および特許出願を包含する全ての刊行物は、個々の出版物が具体的かつ個別にあたかも完全に表示されているように出典明示により本発明の一部として参照される。
実施例1:ヒトおよびネズミIL−18結晶構造に基づくPEG化部位の予想:
Cys38およびCys68(2つの天然の表面が露出したシステイン)でのヒトIL−18(配列番号1)のPEG化により、低活性を有する分子が産生された。従って、本発明者らは所望の生物学的活性と緩衝することなくCysと突然変異形成し、その後PEG化するためのヒトIL−18における部位を同定することを試みた。
マウスIL−18の結晶構造は解明されている(米国特許出願番号10/640,524、2003年8月13日提出)。IL−18およびIL−1βはそのコア構造が非常に類似しているが、その表面ループは異なることなることが明らかである。ヒトIL−18の構造は、高い配列相同性(65%同一)のためにネズミIL−18構造と非常に類似している。ネズミIL−18の結晶構造を用いて、表面およびループ残基をヒトIL−18上にマップすることができる。
ネズミIL−18結晶構造に基づいて(米国特許出願番号10/640,524、2003年8月13日提出)、また後に天然のヒトIL−18結晶構造により確認されるように(PCT出願番号WO03/089653、2003年10月30日提出)、生物学的活性に対する負の効果を最小限に抑えるためにモノPEGを予想されるレセプター結合領域から離れた遊離システインと結合させるPEG化法を考案した。天然のヒトIL−18は、図1(配列番号1)に示される成熟ヒトIL−18のアミノ酸配列において下線で示される4つのシステイン、C38、C68、C76、およびC127を含有する。これらの内の2つ、C38およびC68は、結晶構造により予想されるようにPEG化に利用可能である。PEG化された天然のヒトIL−18は、C38およびC68の両方で二重PEG化されており、減少したインビトロ生物学的活性を示す(表1)。モノPEG化は、天然のシステインの1つを非反応性アミノ酸で置換し、残りの反応性システインをPEG化するか、または両方の天然のシステインを置換し、分子中の他の場所の遊離システインを置換することにより達成される。
a.IL−18のシステイン置換突然変異体の精製
実施例2において前述した突然変異体の全ては、イー・コリにおいて、発現レベルの検出と精製の便宜のためのN末端ヘキサヒス標識を有する可溶性代用形として発現された。ヘキサヒス/proIL−18(配列番号3)を発現するイー・コリ細胞を、10ml/g細胞で溶解緩衝液中に懸濁させた。溶液緩衝液は、50mM Trisl HCl(pH8.0)、500mM NaCl、5%グリセロール、10mM2−メルカプトエタノール(緩衝液A)、1μg/mlペプスタチンAおよび0.4mMフェニルメチルスルホニルフルオリドを含有していた。細胞を溶解緩衝液中に均質化させ、ミクロフルイダイザー(M110−Y、Microfluidics)に12000psiで2回通すことにより溶解させた。細胞溶解物を30000gで30分間遠心分離して、細胞片を分離し、上清をNiNTAアガロースカラムにかけ、これを3カラム体積の緩衝液Aで洗浄した。カラムをさらに3カラム体積の、30mMイミダゾールを含有する緩衝液Aで洗浄して非特異性結合不純物を除去し、ヘキサヒス/proIL−18を緩衝液A中300mMイミダゾールで溶出した。プールを100mM NaClおよび10mM 2−メルカプトエタノールを含有する25mM HEPES pH7.5(緩衝液B)に対して透析した。緩衝液Bはカスパーゼ反応について最適の緩衝液であった。緩衝液B中のプールにカスパーゼ5を1:100w/w(カスパーゼ5対proIL−18)を添加し、一夜室温でインキュベートして、開裂反応を完了させた。反応混合物を0.5M NaClに調節し、これをNiNTAアガロースカラムにかけた。成熟IL−18をカラムに通し、その間、ヘキサヒス/プロドメイン結合をカラムに戻した。第一NiNTAアガロースカラムに結合し、溶出される、あるイー・コリタンパク質(不純物)をカラムに再結合させ、さらに純度の高い成熟IL−18を得た。未結合タンパク質を25mM DTTに調節し、1時間インキュベートして、還元状態で全てのシステインを回復し、精製中に形成されたBME付加物およびジスルフィド結合を還元した。システイン酸化を回避するために2Mリン酸を添加することにより還元された成熟IL−18溶液をpH6.0に調節し、YM10膜で濃縮した。緩衝液を交換するために、濃縮されたサンプルを、0.15M NaClおよび1mM EDTAを含有する10mMリン酸ナトリウムpH6.0で平衡化されたSuperdex75カラムにかけて、凝集物を除去し、インビボ研究に適したエンドトキシンを除去した。成熟IL−18をモノマーとしてSuperdex75カラムから溶出させ、LC/MS分析による分子量は、cDNAから計算されたモノマー形態から予想されるとおりであった。DTNB滴定により、4つのシステインすべてが還元形態であることが示された。
PEG化IL−18、遊離PEG、および未修飾IL−18を含有するPEG化反応混合物を、反応直後に、等体積の25mM MES中2M(NH4)2SO4(pH6.2)(緩衝液C)で希釈した。希釈された反応混合物をSource15フェニル(Pharmnacia)カラムにかけ、これを1.5カラム体積の緩衝液C中1M (NH4)2SO4で洗浄した。カラムを5カラム体積の緩衝液C中0.5M (NH4)2SO4への直線的勾配で溶出させた。遊離IL−18がまず溶出され、続いて遊離PEGおよびPEG化IL−18が最後に溶出される。PEG化IL−18を含有するフラクションをプールし、YM10膜で濃縮し、あらかじめ平衡化させた寸法排除カラムSuperdex200調製等級(Pharmacia)にかけ、緩衝液Bで溶出させた。Superdex200は任意の残存する遊離IL−18、遊離PEG、任意の凝集物、およびエンドトキシンを除去する。PEG化IL−18を含有するフラクションをプールし、〜5−6mg/mlに濃縮した。
PEG化IL−18の調製の一般的手順を以下に記載する。適当な量の平均分子量20000、30000または40000ダルトンを有するメトキシポリエチレングリコールマレイミド(MAL MPEG)を、リン酸塩緩液pH6.0〜6.5中IL−18の2.5mg/mL溶液に添加した(固体またはあらかじめ水性または有機溶媒中に溶解させるかのいずれかとして)。MAL MPEGを、過剰モル量のMAL MPEGでタンパク質溶液に添加した。反応を5℃で1〜12時間進行させた。反応の最後に、過剰量(例えば20倍)のシステイン(0.5M)を添加して反応を停止した。この段階で、野生型ヒトIL−18をPEG化に使用する場合、反応混合物は主にモノ、ジおよび非PEG化IL−18からなることが判明した。ヒトIL−18置換突然変異体を使用する場合(前章において議論するように)、反応混合物は主にモノ−PEG化および非PEG化IL−18タンパク質からなることが判明した(図11参照)。
各サンプルを特徴づけるために4種の分析:(1)SDS−PAGE、(2)逆相液体クロマトグラフィー(RP−HPLC)、(3)分子量測定(LC/MS)および(4)ペプチドマッピングを行った。
b.PEG化度
PEG化の程度(すなわち、1つのタンパク質に結合するPEG分子の数)を、ドデシル硫酸ナトリウムポリアクリルアミドゲル電気泳動またはRP−HPLCにより分析した。ヒトIL−18またはネズミIL−18PEG化反応混合物のサンプルを還元条件下、レーンあたり10.0μgのロードで、4〜12%ビス−トリスポリアクリルアミド勾配プレキャストゲル上にかけた。タンパク質を検出し、クーマシーR−250で染色した後に定量化した。定量化はレーザーデンシトメトリーにより行った。
PEG化の程度をさらに、RP−HPLC法を用いて分析した。反応混合物のサンプルを1mL/分の流速で、8.6%/分の移動相B(移動相A:0.1%水中トリフルオロ酢酸、移動相B:80%アセトニトリル/水中0.1%TFA)の勾配で、POROS R2/Hカラム(カラム温度40℃)で流した。
タンパク質上の正確な位置に対するPEGの結合の位置を証明するために、精製PEG化IL−18サンプルをペプチドマッピングにより分析した。IL−18のトリプシンマッピングは従来法を変更し(J.Bongersら、J.Pharm.Biomed Anal.、21:1099−1128(2000))、IL−18におけるシステイン(遊離スルフヒトリル)の直接S−カルボキシメチル化を用いる:サンプル(2.5mg)を真空下で蒸発乾固させ、6Mのグアニジン・HCl中0.5mLの新たに調製された20mMヨード酢酸ナトリウム中に溶解させ、室温で暗所に40分間置き、その後直ちにBio−Gel P−6DGゲルカラム(BioRad Econ−Pac10DG)上で50mM Tris/HCl、1mM CaCl2(pH8.1)消化緩衝液中に緩衝液交換した。トリプシン(TPCK−処理Worthington)、Lys−C(Wako)、およびGlu−C(Worthington)でのタンパク質分解的消化は全て2mg/mL CM−CysIL−18基質について100/1wt:wtS/E、37℃で2時間であった。RP−HPLCマッピング:Vydac 218MS52 C18カラム(2.1x250mm);55℃;0〜32% CH3CN(0.05%TFA)60分;0.2mL/分、215/280nmw/ダイオード−アレイおよびAgilent G1946A MS検出器(MSD)w/sourceで検出。より小さなサンプル(50−300μg)を、Bio−Rad Micro Bio−Spin Bio−Gel P−6DGゲルカラムを用いて前記手順のセミミクロバージョンにより行った。
モノPEG化野生型hIL−18産物の図14におけるトリプシンマップデータは、56分に溶出する14−39ペプチドピークについて相対的ピーク面積対対照における減少に基づいて、それぞれほぼ65および35±5モル%の程度までの排他的に溶媒の影響を受けやすいシステイン38および68でのPEG化を示す。マップにおける71−79および113−129トリプシンペプチドの定量的回収により証明されるように、埋め込まれたシステイン76および127で検出可能なPEG化は起こらなかった。これらの結果は、後に溶出する疎水性ピーク含有PEG化ペプチドプールが同時に出現する、モノPEG化hIL−18マップ対非PEG化hIL−18対照マップにおけるシステイン含有トリプシンペプチドの相対的ピーク面積において観察される減少、またはその喪失に基づく。システイン38および68上のPEGの約65〜35モル%分布は、さらにLC/MC/MS−MSデータからの抽出されたイオン電流トレースにより確認した。
RP−HPLCトリプシンマップにおける非PEG化ペプチドのピーク面積における対照に対する差によりPEG化の部位および程度を決定することに加えて、本発明者らはタンパク質から放出されるPEG化ペプチドフラグメントの直接分析も行った。PEG化ペプチドに関するこのような直接的データは、非PEG化ペプチドについてのピーク面積における差から得られる相補的間接的証拠に対する「質量バランス」により関連づけられる。先に記載したように、これらの異なるPEG化ペプチドはすべてRP−HPLCトリプシンマップにおいて、PEG化ペプチド種の未分割のプールを含有する単一の後で溶出するピークとして同時に溶出する。PEG化ペプチドはイオントラップまたは四極子質量分析器でのオンラインエレクトロスプレー質量分析の影響を受けにくいので、本発明者らはRP−HPLCトリプシンマップから集められたPEG化ペプチドのプールを分析するためにミクロ化学EdmanN末端シーケンシングを利用した。
トリプシンマッピングデータ(図13および15)は、ヒトIL−18置換突然変異体C38S、C68S、L144C(配列番号9)およびC38S、C68S、D157C(配列番号10)およびそれぞれの精製されたモノPEG化結合体の予想される化学構造を裏付ける。RP−HPLCデータは、それぞれ残基144および157での操作された表面システインでの>95%部位特異性定量的PEG化と一致する。そして、残基76および127での埋め込まれたシステインを含有するカルボキシメチル化トリプシンペプチドは、野生型ヒトIL−18標準の対照トリプシンマップに対してほぼ定量的収率で回収され、事実上他の顕著な化学的修飾(副反応)は最終生成物において観察されなかった。
ヒトIL−18の生物学的効果は、その細胞表面レセプターとの結合に関連し、おそらくは天然の拮抗物質、IL−18BPに対するその結合による。IL−18の結合検定およびレセプターのアルファ鎖、またはIL−18BPの何れかは、BIAcore装置、表面プラスモン共鳴ベースのバイオセンサーを用いて開発された。この技術は、センサーチップ上の1つの生体分子を固定化し、リアルタイムで溶液中の第二の成分とのその相互作用をモニターすることを含む。
IL−18の効果は、αおよびβ鎖からなるヘテロダイマー表面レセプターとの結合により媒介される。α鎖は単独でIL−18と結合するが、両サブユニットは、細胞内シグナリング経路を活性化し、標的細胞上のIL−18の生物学的効果を媒介することができる機能的高親和性IL−18レセプターを形成するために必要である。転写因子NF−κBはIL−18の免疫調節効果の重要なメディエーターである。IL−18レセプターシグナリングは、IKKタンパク質キナーゼの活性化を誘発し、これは次にNF−κB阻害剤IκBαをリン酸化する。リン酸化IκBαは分解し、核へ転位し、遺伝子転写を活性化する遊離NF−κBを放出させる。多IL−18ムテインをこの検定において評価した。これらの研究から、C38S、C68D、L144C(配列番号7)、C38S、C68S L144C(配列番号9)、C38S、C68D、D157C(配列番号8)、およびC38S、C68S、D157C(配列番号10)を含有するIL−18ムテインが最も有効であると確認された。
BALB/cをネズミIL−18を1回注射することにより治療すると、サイトカイン産生が増大した。最大の増加は、IFNγおよびGM−CSFに関して見いだされた。IFNγ応答は非常に迅速で大規模であるので、このバイオマーカーはPEG化IL−18の効力を天然のIL−18分子と比較するために使用した。ヒトIL−18(配列番号1)およびPEG化ヒトIL−18(配列番号1)をSC注射により10または100μgの投与量で投与した。治療後2、4、6、8、12および16時間に血清を集め、IFNγ産生に関して分析した。等モル量のPEG化IL−18は非PEG化IL−18よりも高いIFNγの循環レベルを誘発した。PEG化IL−18のIFNγのピークレベルは、4〜16時間の間に見いだされるが、非PEG化IL−18は2〜4時間の間にピークを生じさせた。IL−18α鎖レセプターについての親和性の減少(表3に示すとおり)、
またはNF−κB細胞ベースの検定における活性の低下(表4に示すとおり)にもかかわらず、PEG化ヒトIL−18(配列番号1)は、IL−18分子の重要なPDマーカーであるIFNγにおいて増大を示した(表5)。
IL−18は、NK細胞を活性化するTh1−優性サイトカインである。この研究において、本発明者らはヒトPEG化および非PEG化IL−18(配列番号1)に反応するNK細胞毒性を測定することを目的とした。この検定は、抗腫瘍活性の直接滴尺度である。
ネズミYAC−1細胞(NK感受性T細胞リンパ腫)を標的として、また処置された動物からのBALB/c脾臓細胞をエフェクターとして用いるヨーロピウム放出検定によりNK細胞活性を測定した。マウスにヒトPEG化および非PEG化IL−18(等モル)濃度を1回注射した。マウスを処置後18〜24時間に屠殺した。処置された動物(および対照)からの脾臓細胞をヨーロピウム標識されたYAC−1標的細胞と組み合わせ、ヨーロピウム放出について測定した。
IGEN系を用いてIL−18(配列番号1)とIL−18結合タンパク質(IL−18BP)間の複合体を検出するためのイムノアッセイを開発した。この検定は、ヒトIL−18に対して非中和mAb(16D10mAb、ルテニウム結合)およびヒトIL−18BPに対して非ブロッキングmAb(mAb36、ビオチニル化)を使用する。125ng/mlのヒトIL−18BPを125ng/mlの非PEG化IL−18またはPEG化IL−18と組み合わせた。
PEG化IL−18分子はIL−18BPと複合体形成する能力が低下し(表7)、これは複合体を形成する非PEG化IL−18と対照的である。
PEG化IL−18および非PEG化IL−18に応答して、PDマーカーをカニクイザルにおいて評価した。カニクイザル(1群あたり3匹のオス)に、1mg/kgの非PEG化IL−18またはPEG化IL−18のいずれかを静脈内注射を1回した。投薬前、および投薬後10日期間にわたって複数の時点で血液サンプルを集めた。血漿を薬剤およびネオプテリン濃度について分析し;白血球を次いでCD64マーカーの発現についてフローサイトメトリーにより分析した。
カニクイザル白血球をCD64平均蛍光強度(MFI)における変化について評価した。PEG化IL−18および非PEG化IL−18は投薬後1から3日の間にCD64発現を増大させた。全白血球上の高いCD64発現は3日に極大になり、10日まで存続した。白血球CD64MFIは、投薬後のどの時点でも非PEG化IL−18と比較してPEG化IL−18に対する応答が高かった(2倍まで)。全白血球において、CD64MFIは非PEG化IL−18よりもPEG化IL−18について統計的に有意に高かった。
PEG化IL−18および非PEG化IL−18は、投薬後1から4日の間にネオプテリン産生を誘発し、ピーク反応は24または48時間であった。折りたたみの誘発およびネオプテリンの群平均濃度は、一般に、非PEG化IL−18に対してよりも(2日に13μ/ml)PEG化IL−18に対して(2日に15〜21μg/ml)の方が高かった(表9)。しかしながら、差は統計的有意に達しなかった。
カニクイザルにおけるPEG化および非PEG化ヒトIL−18についてのPKデータを表10に示す。0〜8時間の間のPEG化IL−18についてのグラフ下の面積(AUC)は、全336時間の非PEG化IL−18のAUCよりも約7倍大きかった。PEG化IL−18について、0〜8時間の見かけのt1/2は約0.5時間であるが、非PEG化IL−18初期相(全AUSの約43%)のt1/2は約7分であった。PEG化IL−18のCmax値は、非PEG化IL−18のCmax値よりも約4倍高く、血漿区画中へまず分布する薬剤と一致した。
先の記載事項は、その別の実施形態を含む本発明を完全に開示する。本明細書において具体的に開示されている実施形態の変更および改善は以下の請求の範囲内に含まれる。さらに努力することなく、当業者は前記載事項を用いて本発明を最大限利用することができる。従って、本明細書において記載されている実施例は単に例示的であって、本発明の範囲を何ら制限するものではないと解釈されるべきである。本発明の排他的特性または優先権が主張されている実施形態は次のように定義される。

- 生物学的に活性な組成物を調製する方法であって:
