Description
糖が結合していないか、または糖結合の不足した異常な糖結合のムチン類の過剰産生が、乳癌、子宮癌、膵癌、結腸癌、肺癌、前立腺癌及びその他の分泌組織の腫瘍の特徴である。ヒト ムチン類MUC1乃至MUC7のそれぞれの蛋白質核のcDNA配列がクローンされて、特徴付られており、VNTR'sとして知られる特定アミノ酸の主要素の反復数が異なる高度に反復性の中心部が見出されている。例として挙げると、MUC1は特異なアミノ基及びカルボキシ基末端配列から成り、20種のアミノ酸主要部の40乃至80個の縦列主要素を有している。MUC1乃至MUC7のVNTRを以下に示す:

ペプチド合成は、例えばMUC1の5つのサブユニットの反復の様な約100 個に及ぶアミノ酸を含有するポリペプチドに用いられる。通常、約20個以上のアミノ酸を含むポリペプチドには、好ましい生成方法は宿主細胞中、好ましくは原核宿主細胞、更に好ましくは細菌性宿主細胞中の組換え発現である。しかしながら、先に考察した様に、真核細胞系も用いられる。宿主細胞中の組換え蛋白質の発現方法は充分に確立されており、例えばSambrookらを参照のこと(Sambrook, et al., Molecular Cloning: A Laboratory Manual, Cold Spring Harbor Press, ColdSpring Harbor, 1989)。

CD3 、CD4 およびCD8 に対する抗体が欠如しているか、または CD3+ 、CD 4+およびCD 8+ T-細胞がブロックされているかを確認する目的で、残留細胞の血清学的分析を、CD3 、CD4 及びCD8 に対する抗体を用いて実施した。脾臓およびリンパ節細胞を、正常および処置BALB/cマウスより摘除し、リンパ球を処置し、DME 中で洗浄し、37℃で5分間、0.83% 塩化アンモニウム中で赤血球を溶血する為に反応させた。血清学的テストは、抗-CD3、抗-CD4及び抗-CD8腹水の1:500 希釈液に、マウスの2 x 105 脾臓及びリンパ節細胞を添加して行なった。充分に洗浄した後に、細胞をマウス胸腺細胞を吸着したラット抗マウスIgG と、氷冷しながら30分間反応させた。抗-CD3、抗-CD4、抗-CD8または抗-CD4+ CD8 で処理したマウスを、これらの抗体に就いてそれぞれテストした。CD 3+ 細胞が欠如し、 CD4+ びCD8+ 細胞がブロックされていることが見出された。
MUC1 cDNA のVNTR領域から60塩基対主要部を5回よりやや多く反復コードした309 塩基対の挿入(FDF9.3)(Siddiqui, J., Abe, M., Hayes, D., Shani, E.,Yunis, E. & Kufe, D., Proc. Natl. Acad. Sci., 85: 2320-2323,1988) を、正確に読み取って位置付けした細菌性発現ベクトルpGEX-3X 中にサブクローンした(Smith, D. B. & Johnson, K. S., Gene, 67: 31-40, 1988)。グルタチオン-S-トランスフェラーゼ(GST、26Kd) およびMUC1 VNTR (12kD)を含有する融合蛋白質(FP)を、0.1mM IPTGで誘発した(Smith, D. B. & Johnson, K. S., Gene, 67: 31-40, 1988)。細胞を遠心分離で集め、洗浄し、1%(v/v)Triton X-100 を含有する緩衝液中で超音波分解処理した。可溶性FPを含有する上清を硫黄結合グルタチオン- アガロース ビーズ(Sigma社、St. Louis)と混合し、遠心分離して採取した。FP((C-PAHGVTSAPDTRPAPGSTAP)x 5-GST) を還元型グルタチオン5mMを含有する緩衝液で溶出し、リン酸緩衝生理食塩液に対して透析し、SDS-PAGEで分析した。
分析すべき試料はSDS 試料緩衝液と混合し、5分間煮沸し、12.5% SDS-PAGEゲルに負荷した。ゲルは0.2% クーマシー ブルー(Coomassie brilliant blue)で染色後、7%酢酸で脱色するか、または銀染色した (Apostolopoulos, V., Xing,P. X., Trapani, J. A. and McKenzie, I. F. C., Br. J. Cancer, 67(4):713-720,1993)。分子量マーカーとして、200,000 ミオシン、116,000 β- ガラクトシダーゼ、92,500 ホスホリラーゼ b、66,200 ウシ血清アルブミン(BSA)、43,000 鶏卵の卵白アルブミン、31,000 ウシ炭酸脱水酵素、21,500 大豆トリプシン阻害剤、および14,400 鶏卵の卵白リゾチームを使用した。
マンノースを含有する多糖類であるマンナン 14 mgを、0.1Mリン酸緩衝液1ml、pH 6.0乃至9.0 中で、リン酸緩衝液中0.1M 過ヨウ素酸ナトリウム 100μl を使用して4℃ 1時間で酸化してポリアルデヒドにした。エタンジオール10μl を添加して更に30分間、4℃で反応させた後、反応混合物をPD-10 カラムに通過させて、マンナン分画を採取した。酸化マンナンにMUC1 FP 230 μg を添加し、一夜室温で反応させ、以降の研究に使用した。
雌性8週齢BALB/cマウスを、リン酸緩衝生理食塩液(PBS)中のFP量に相当するマンナン-FP 5 μg 、FPおよびFPと非複合マンナン+FP との混合物で毎週1回3週間腹腔内に免疫処置した。予めFP単独で免疫処置したマウスは、抗体産生の対照として下記の様に用いた。腫瘍注射の前に、マウスを飼育し、血清はFP(抗マンナン抗体)に対する抗体産生に就いてELISA で下記の様に検査した。
ras遺伝子とともにMUC1cDNA膜通過型をBALB/cマウス繊維芽細胞株3T3にトランスフェクトして、MUC1+3T3細胞株を得た(イスラエル、Tel Aviv大学、Dr. D. Wreschnerより入手)。マウスにPBS 中の適量の腫瘍細胞 0.2mlを皮下注射して、以後の腫瘍の成長を測定した。測定のすべてはダイアルゲージ カリパス(Schnelltaster 、H.C. Kroplin、Hessen、ドイツ)で行い、腫瘍の大きさは腫瘍面積(cm2、直径×直径)で現わした。ネズミのDBA/2 肥満細胞腫株P815、及びMUC1の膜固定型のcDNAを含むMUC1+ P815は、Dr. B. Acres(Trangene社、Strasboug、フランス)より入手した。
ペプチドC-p13-32(C-PAHGVTSAPDTRPAPGSTAP)(MUC1 VNTR)及びT4N1(KTLVLGKEQESAELPCEY) (マウスCD4 N-末端領域ペプチド)はアップライド バイオシステムズ社 モデル430A(Applied Biosystems Model 430A)自動ペプチド合成器を用いて合成した。HMFGはヒト乳から単離して前記した様に調製した(Xing, P. X., Tjandra, J. J., Stacker, S. A., Teh, J. G., Thompson, C. H., McLaughlin, P. J., and McKenzie, I. F. C., Immunol. Cell Biol., 67:183-185, 1989)。
(a) 抗融合蛋白質抗体の測定:
微量測定プレートのウェル中でコートした FP 20μg/ml、2%ウシ血清アルブミン(BSA)でブロックした非特異的結合、およびFP並びにマンナン-FP 免疫処置マウスの血清50μl を室温で2時間添加して、ELISA 検出を行なった(Xing, P. X., Tjandra, J. J., Stacker, S. A., Teh, J. G., Thompson, C. H., McLaughlin, P. J., and McKenzie, I. F. C., Immunol. Cell Biol., 67:183-185, 1989)。正常マウス血清(NMS) を陰性対照として用いた。洗浄後、西洋ワサビ パーオキシダーゼ複合体に結合したヒツジ抗マウス免疫グロブリン(Amersham社、英国)を添加し、室温で反応させ、プレートは0.03% 2,2'- アジノ- ジ (3-エチルベンズチアゾリンスルホネート(Amersham社、英国)、0.1Mクエン酸緩衝液、pH 4.0中の0.02% H2O2(100容量、Ajax Chemical社)で発色させ、所望の濃度に達する迄室温で10乃至15分反応させた。吸光度はプレート読み取り器で 405nmで測定した。
(b) マンナンに縮合した融合蛋白質の活性の測定 :
上記した方法でELISA 検出は以下の変更を加えておこなった: FP 20μg/ml、マンナン-FP 及びマンナンはプレート上でコートし、使用した一次抗体はVA1 及びVA2(抗FP Mabs)とした。
マウスでDTH を誘導する為に、シクロフォスファミド(cyclophosphamide、Endoxan-Asta、Mead Johnson)200 mg/kg 体重の腹膜腔内投与を、マンナン-FP5μg の腹腔内投与2日前に行なった。6日後、後肢足蹠に凍結と融解を5回反復した105 個の3T3 またはMUC1+ 3T3 の20μl 、50μg のHMFG、FP、グルタールアルデヒドを用いてスカシガイのヘモシアニンに結合したC-p13-32、関連性のないペプチドT4N1、マンナン-FP 、GST 及びマンナン並びに等量のPBS を注射した。DTH 反応は48時間後に、足蹠の幅と厚さを測定して、その積を算出した。足蹠測定のすべてはダイアルゲージ カリパス(Schnelltaster 、H.C. Kroplin、Hessen、ドイツ)で行った。
マンナン-FP で免疫処置したBALB/cマウスを屠殺して脾臓細胞を採取し、2%ウシ胎仔血清/PBSで洗浄した。標的細胞であるP815及びMUC1+ P815細胞は、標準的51Cr放出検定前に、フェニル N- アセチル -α-D- ガラクトサミニド(pagal)5mMを用いて、O-結合グリコシル化阻害の為に2日間反応の有無双方を行なった(Sigma、St. Louis 、米国)。106 個の標的腫瘍細胞は、 100μCi Na2 51CrO4(AmershamCorp., Arlington Heights)で、37℃、60分間放射性標識し、充分洗浄した。脾臓細胞および標的細胞は培養基中に再度懸濁させ、次いで96ウェル U- 型底プレート(Costar Corp.)中で種々の作用剤- 標的比で混合した。プレートは100 x g 、3 分間遠心して、細胞の接触を開始させ、10% CO2 中、37℃で4時間反応させた。反応後、上清を採取して放射活性をγ- 計数器(Beckman Instrument)で測定した。
[(実験値- 自然放出値)/( 最大- 自然放出値)] x 100% で求めた。
M-FPで免疫処置したマウスを屠殺し、脾臓細胞を採取し、2%ウシ胎仔血清/PBSで洗浄し、赤血球を0.14% NH4Cl で溶血させ、100 μl 培養基中の 5x105脾臓細胞の重複培養(duplicate culture)は96- ウェル プレートに接種した。脾臓細胞は以下の各種の 100μl で刺激した:10μg - T4N1、GST 、マンナン、HMFG、Cp13-32 、FP、MFP 、及び105 個のpagal 処理有無双方の、乳癌細胞3T3 、MUC1+ T3、P815、MUC1+ P815、及び105 個ヒト乳癌細胞株-T47D 、MCF7、及びZR15。すべての腫瘍細胞はmitomycin-C (Sigma 社、オーストラリア、Victoria)25μg/mlで、腫瘍細胞の成長を阻止する為に37℃で2時間処置した。培養は5%CO2中、37℃、36時間行なった。3[H]-TdR(Amersham社、英国)の6.7 Ci/mmol 取り込み測定は培養の最終4時間に行なった(1μCi/ ウェル)。
試験管内のMUC1+ 3T3 細胞は、通常の3T3 細胞と外観および成長の特徴が同一であるので異なっているとは見えなかった。血清学的分析では、MUC1+ 3T3 細胞は高濃度のMUC1を示し、H-2d+であった。MUC1 VNTR ペプチドに対する抗体は、ヒト乳癌細胞株T47DおよびMCF7と同様に、MUC1+ 3T3 およびMUC1+ P815と反応し、抗-HMFG 類別:BC2 抗体、抗- 融合蛋白質:VA1 及びVA2 抗体、並びに抗-MUC1 ペプチド抗体:BCP7、BCP8、BCP9及びBCP10 であった。しかしながら、ネズミ腫瘍はヒト腫瘍とは異なって糖と結合し、MUC1+ 3T3 及びMUC1+ P815細胞は、抗- 炭水化物(3E1.2 )抗体(エピトープ:glycolylsialyl-Tn )と反応性だが、その他の炭水化物に対する抗体(CC5-エピトープ:血液群Lca )とは反応性ではなかった。このことは蛋白質抗原は変化しないが、糖結合が変化することを示す。マウスとヒトとが異なったグリコシル トランスフェラーゼを有しており、従って糖結合に異なった形態を示すのは驚くことではない。しかしながら、pagal処理で糖が離脱した後は、先には細胞株との反応性が微弱または無く、非-APDTR反応性抗体であるMUC1 VNTR に対する抗体が、そのエピトープが露出したので反応性となった。(AP)DTR(PA) の反応性抗体に就いて差は認められなかった。炭水化物反応性抗体(3E1.2) との反応性には大きな差が存し、pagal 処理後には染色が微弱または失われ、3E1.2 のエピトープがムチンの蛋白質核に酸素を介して結合しているので、pagal が実際にO-結合糖を除去することを示した(Devine, P.L., Clark, B. A., Birrell, G. W., Layton, G. T., Ward, B. G, Alewood, P.F. and McKenzie, I. F. C., Cancer Res., 51:5826-5836, 1991) 。類別を別の機会に行なったが、同一結果が得られ、フェノタイプの安定性が示された(記載なし)。
BALB/cマウスに5x106 個のMUC1+ 3T3 または3T3 細胞を皮下注射して、その後の成長を測定した。3T3細胞は進行的に成長して、BALB/cマウスに予測される様に拒絶されなかった。これに反して、MUC1+ 3T3 細胞は第10日迄は進行的に成長したが、その後縮小を始め、第18日迄には徐々に消失した。従って、ヒトMUC1+遺伝子生成物は3T3 細胞に免疫原性を与えて拒絶に至る様である。この点はその後の5 x 106 個のMUC1+ 3T3 または3T3 細胞の接種が免疫されたマウスではMUC1+ 3T3 細胞の成長に完全な抵抗性を示し、一方、3T3 細胞は成長して、免疫原性がMUC1+ を有する腫瘍にのみ見出され、この抗原に特異的であった。特異性及び記憶はMUC1+ への免疫反応と、MUC1+ が3T3 の成長性に影響する様な、その他の効果のないことを示す。生体内で経過した腫瘍を用いた反復実験の数週間後に、すべてのマウスがその腫瘍を拒絶するのではなく、30% に至るMUC1+ 腫瘍は成長を続けた。これらの腫瘍を摘除して、MUC1+ を血清学的に測定すると、腫瘍細胞の若干はMUC1- であり、MUC1+ 感染細胞の若干が生体内でそのMUC1+ 発現能力を失うが、本発明者らは遺伝子が未だ存するか否かは確認しなかった。この様な観察は別途にラット腫瘍(Hareuveni, M., Cautier, C., Kieny, M. P., Wreschner, D., Chambon, P. and Lathe, R., Proc. Natl. Acad. Sci. USA, 87:9498-9502, 1990)で報告され、恐らくMUC1の不安定発現に依るのであろう。その後の研究では、本発明者らは使用した腫瘍が100%MUC1+ であることを、抗-HMFG 抗体BC2を伴うMUC1+ 発現を血清学的にテストすることに依って確証した。
細胞性免疫はマウスの同種腫瘍移植の拒絶の、最も普遍的な形態であるので、最も妥当な拒絶の形態の様である。この点は抗CD3 抗体の能力が完全に免疫を失ったことで確認された。CD 4+ またはCD 8+ 細胞が拒絶に関与していたか否かを決定する為に、マウスに抗-CD4または抗-CD8抗体を、これらがその他のモデルでは免疫抑制を生じることが既知であるので複数回投与した(Pierres, A., Naquet, P., VanAgthoven, A., Bekkhoucha, F., Denizot, F., Mishal, Z., Schmitt-Verhulst, A-M. and Pierres, M., J. Immunol., 132:2775-2782, 1984、Miller, R. A. and Stutman, O., Nature, 296:76-78, 1982) 。ブロックに依る機能的CD4 細胞喪失は、腫瘍成長に一過性の影響を与え、腫瘍は無処置マウスと同様な経過で拒絶された。これに反して、抗-CD8投与は腫瘍成長の延長をもたらした。本発明者らは CD3+ 細胞が拒絶に深く関与し、CD 4+ 細胞は僅かな作用を有し、CD 8+ 細胞は移植拒絶に主要効果を有していると結論した。抗-CD8投与マウスに於ては、腫瘍は抗-CD3を投与したマウスより小さく、明かに抗-CD8抗体は抗-CD3抗体に依る完全なT-細胞排除程には効果がないことが注目された。CD4 細胞が僅かな作用を有するのは、抗-CD4及び抗-CD8の複合作用が、抗-CD8単独より優れていないので、可能性はありそうもない。しかしながら、本発明者らは単独では無効である。抗- γ- インターフェロン(γIFN )治療を抗-CD8と併用すると、抗-CD3と同様の作用を現わし、従ってγIFN は腫瘍移植拒絶に役割を果たし、CD 8+ 細胞及びγIFN に依って介在されることに注目した。
明らかに免疫反応は細胞性であり、CD 8+ 細胞に依る。これが通常CD4+ 細胞が介在すると考えられるDTH 反応、または通常CD 8+ である細胞毒性T-細胞反応を含むかを決定する為に、マウスをMUC1+ 3T3 細胞で免疫処置し、DTH を種々の抗原を後肢足蹠に注射することで行なった。
予備研究はシクロホスファミドが存在しないと、測定可能なDTH 反応は生じなかった。DTH 反応は、凍結、解凍を5 回反復した死滅MUC1+ 3T3 細胞を足蹠に注射して検出し、HMFG、融合蛋白質-GSTおよびCp13-32-KLH のいずれかを試験投与した際に、微弱な反応を検出した。これらの反応は3T3細胞が反応を生じなかったので、明らかに特異的であった。DTH反応がCD 4+ またはCD 8+ 細胞に依って介在されるか否かを決定する為に、マウスに抗-CD4及び抗-CD8抗体を注射して、DTH 反応を測定した。
MUC1+ 3T3 細胞、Cp13-32 、HMFG及び融合蛋白質を投与した際に、抗-CD4は完全にDTH 反応をブロックし、抗-CD8はDTH 反応を部分的にブロックしたが、程度は低かった。従って、ヒトMUC1(CD8) に対する効果的な免疫反応を生じる細胞は、DTH 反応を生じる細胞とは異なっているが、CD 8+ 細胞は確実にDTH に関与していた。
細胞毒性の検定を実施し、MUC1+ 3T3 細胞の免疫処置後に、pagal 処理MUC1+P815標的の60% に及ぶ分解が生じた。無処理
MUC1+ P815標的及び非感染P815標的は分解されなかった。Pagal 処理及び無処理3T3及びMUC1+ 3T3 標的も分解せず、恐らくは3T3細胞がTc検定には好ましくない標的であったからであろう。Tc、抗-CD4及び抗-CD8抗体の表現型を決定する為に、ブロック研究に使用した、抗-CD8試剤(53-6.7)はCD8+細胞に依るT-細胞分解のブロックが可能であることは既知である。これらの研究で抗-CD8がTcをブロックできる状態であることが証明されたが、抗-CD4及び対照抗体の作用は僅かであった。Pagal 処理MUC1+ P815標的にはTcのみが見出され、非APDTR 反応性の抗-MUC1 抗体(VA1、BCP7、BCP9及びBCP10)がpagal 処理したMUC1+ 3T3 、MUC1+ P815、T47D及びMCF7細胞と反応性になったので、抗体反応性及びT-細胞反応性エピトープが隠されて、ともにpagal 処理後に曝露されることが明かである。
細胞性免疫が効果的であり、抗体産生が僅かであることが先に示されたが、抗体の役割は不明確である。更に、マウスは通常抗体産生が少なく、補体の動きが乏しいので、a )ポリクローナルまたはモノクローナルであろうと、充分な抗体の供給、b )充分な補体の供給、c )高密度の表面抗原等の特定の条件を満たさない限り、抗体介在性の移植破壊の研究に選択される選択種ではない。MUC1+ 抗原密度が高いので、追加の抗体及び補体が供給される。更に、マウスは細胞性免疫のすべての成分を除く様に、CD3 に依って免疫抑制された(Tomonari, K., Immunogenetics, 28:455-458, 1988)。材料と方法の項で記述した様に、皮膚の同種及び異種移植が可能な様に、大量の抗体及び補体にも拘らず、腫瘍は進行的に、事実抗体を投与されなかったマウスと同様な速度で増殖した。従って、抗体及び補体はMUC1+ 3T3 細胞の拒絶を起こし得ない。
前項は3T3 細胞に感染させたヒトMUC1に対するネズミ免疫反応のモデルを定義し、細胞性MUC1に依って産生されたと同様に、合成材料に依る同様もしくはより強力な免疫反応を生じる為に、種々の免疫原使用の基礎を形成した。本目的は免疫処置後に腫瘍成長を大きく低下させるにある。免疫原として、天然ムチン(HMFG)、VNTR二量体から成るMUC1ペプチド合成産物、及び5 x VNTR反復融合蛋白質を使用した。予め免疫処置なしには、腫瘍は18日後には拒絶され、この様なマウスは以後の接種に抵抗性と成る。従って、腫瘍が拒絶される迄には約18日間の"窓" が開いている。そこで、この期間の免疫処置は腫瘍の消失または大きさを縮小させられる。
従って、第6日に、ペプチドまたは融合蛋白質で免疫処置したマウスは、対照群に比して約25% の腫瘍を有しており、HMFGで免疫処置した群では、効果はより低く、腫瘍は対照群の約60% であった。しかしながら、MUC1+ 3T3 細胞 5x106個を移植した際には、対照群に比して腫瘍の大きさに若干差が生じたが、少量の移植では明瞭さが低下した。その後の腫瘍の移植では予想される様に、腫瘍を拒絶したペプチドで免疫処置したマウスは、今や腫瘍に抵抗性となった。従って、ペプチド、融合蛋白質またはHMFGで免疫処置して、少量のMUC1+3T3細胞を移植したマウスは、若干の抗腫瘍効果の上昇を示した。ペプチド、融合蛋白質及びHMFGを含有するVNTRは、若干保護作用を得たが、MUC1のN-またはC-末端ペプチドで免疫処置したマウスは有意な保護を示さず、これらのペプチドがMUC1に免疫を誘発しないことを示し、免疫処置自体が無効であることを示した。免疫反応の種々の変数を測定する為に、本発明者らは(a) MUC1抗体産生、(b) 遅延型過敏性及び(c)細胞毒性T-リンパ球を検討した。
ペプチド、融合蛋白質またはHMFGで免疫処置したマウスは、腫瘍移植の前後の双方で高レベルの抗-MUC1 抗体値を有していた。従って、免疫処置は高レベルの抗体を生じるが、腫瘍に対する僅かな効果に依って示す様に見かけの細胞性免疫は僅かである。対照ペプチドT4N1で免疫処置して、腫瘍を拒絶したマウスは、MUC1に対する抗体を産生せず、ペプチド及びその他の免疫原で免疫処置したマウスは腫瘍拒絶後に抗体値が増加しなかったことは興味深い。
HMFG 、Cp13-22及び融合蛋白質-GSTで免疫処置したマウスは種々のMUC1抗原に対してDTH 反応を示し、CD4 抗体で完全に、CD8 抗体で部分的に阻害された。従って、この3 試剤に依る免疫処置は、細胞性免疫を若干増加させるが、腫瘍成長を大きく阻害するには不十分であった。
免疫処置したマウスの脾臓及びリンパ節細胞からTc検定を実施して、細胞毒性の細胞は検出されなかった。従って、種々の免疫処置方法は細胞性免疫より、抗体産生に対する免疫反応に偏向を生じる様である。

MUC1 FP は材料と方法の項に記載した様に、過ヨウソ酸塩を用いてマンナンと結合させた。FPのアミノ基は酸化されたマンナンのアルデヒド残基と反応して不安定なシッフ塩基を形成する(図12)。遊離マンナン及びFPは複合マンナンから分離しなかった。ゲル濾過クロマトグラフィーに依って得られた125I-FP 及び125I-M-FP の溶出状況はマンナン- 融合蛋白質が、201 Kd及び73 Kd と2つのピークとしてして溶出されることを示した。2 つのピークとして溶出されたFPは38 Kd 及び20 Kd を示し、低いピークはFPの開裂に依るGSTに依るのであろう。125I-FP及び125I-M-FPのオートラジオグラフィー分析は大部分のFPがマンナンと結合していることを示した。
BALB/cマウスに皮下注射したMUC1+ 3T3 細胞の 5 x 106個は、第10日迄進行的に成長したが、その後縮小し始め、第18日迄に消失したが、3T3 細胞は実施例2に示されたBALB/cマウスから予測された様に拒絶されなかった(図1)。従って、ヒトMUC1+ 遺伝子生成物は3T3 細胞に免疫原性を与えて、拒絶をもたらし、その様なマウスは以後の接種に完全に抵抗性であった。細胞性免疫は、抗-CD3及び抗-CD8抗体が完全に免疫を失わせたので、拒絶の形態である。
マンナン-FP の免疫原性を検討する為に、BALB/cマウス群をFP 5μg に相当するマンナン-FP 5μg で免疫処置し、MUC1+ 3T3 細胞105 乃至5 x 107 個を接種した。免疫処置したマウス(図2A)では非免疫処置マウス(図2B)に比して、明瞭な腫瘍成長がなかった。マンナン-FP 複合体が特異的抗腫瘍免疫を生じ、マンナンまたはFP単独では作用のないことを示す為に、マウスを等用量の複合体中では7 mg/ml に相当するマンナン、マンナンとFPとの混合物で免疫処置し、また免疫処置しなかったマウス群に、MUC1+3T3細胞106 個を接種した。マンナン-FP 複合体で免疫処置したマウスでは、腫瘍の成長が認められず、一方マンナン単独及びマンナンとFPとの混合物で免疫処置したマウスでは、非免疫処置マウスに比して成長に差はなかった(図3A、3B及び3C)。従って腫瘍成長の保護は、本複合物に特異的であり、マンナン及びFP単独には効果がなかった。マンナン-FP 免疫処置マウスに、3T3 細胞106 個を接種し、3T3 細胞は免疫処置の有無双方のマウスで同様な成長を示し(図4)、免疫処置生成物に非特異的作用のないことを示した。
抗-CD3抗体はM-FPで免疫処置したマウスに於ける免疫を完全に失わせ(図5)、抗-CD4または抗-CD8抗体を投与したマウスは以下の効果を示した:CD4 免疫抑制処置は、腫瘍成長には効果が少なく(図5)、これに反して抗-CD8処置は腫瘍成長の延長をもたらした。従って、CD 3+ 及びCD 8+ 細胞は免疫及び腫瘍保護に完全に作用を示し、CD 4+ 細胞は最少の作用を有していた(図5)。従って、M-FPは3T3 細胞に担持されたMUC1に対して免疫作用を示し、CD 3+ 及びCD 8+ 胞に依って発現する細胞性免疫を生じるが、CD 3+ 及びCD 4+ に依っては発現しない。免疫反応の種々の変数を測定する為に、本発明者らは(a) 遅延型過敏性、(b)細胞毒性T-リンパ球、(c)T- 細胞増殖及び(d)抗体産生を検討した。
DTH 反応は通常CD4 + 細胞が介在すると考えられており、種々の抗原を後肢足蹠に注射することで現われる(図6)。DTH 反応は足蹠投与に依って凍結、解凍を5 回反復した死滅MUC1+ 3T3 細胞、HMFG、FP-GST、Cp13-32-KLH 、マンナン-FP 及びGST がFPの一部分であるのでGST に対する微弱な反応で検出される(図6)。これらの反応は死滅3T3 細胞、マンナン単独、関連のないT4N1ペプチドに明瞭に特異的であり、PBS は反応を生じなかった。DTH 反応がCD 4+ またはCD8 +細胞に依って介在されるか否かを決定する為に、マウスに抗-CD4及び抗-CD8抗体を注射し、DTH 反応を測定した。抗-CD4は完全にDTH 反応を阻害し、抗-CD8は阻害はしたが程度は低かった(図6)。従って、ヒトMUC1に効果的に免疫反応を生じさせる細胞は、DTH反応を生じさせるものとは異なっており、CD 8+ 細胞に就いては図5に示したが、確かにDTH に関与していた。
確立した腫瘍に対する治療剤としてのM-FPの有効性を測定する為に、MFP の注射は腫瘍が確立する迄遅らせた。MUC1+ P815細胞は(DBA/2 x BALB/c)F1マウスの皮下に移植すると、第16乃至20日迄進行的に成長し、その後縮小を開始し、通常第28日迄に消失した(図8A)。確立した腫瘍に対するM-FPの有効性を検討する為に、1群5匹の(DBA/2 x BALB/c)F1マウスにMUC1+ P815細胞5 x 106 個を注射し、13日後にFP 5μg に相当するM-FP 5μg を1回または隔日に注射した。PBS を投与した対照群マウスの腫瘍は、第28日迄に拒絶された。しかしながら、定期的に注射したマウスは直ちに腫瘍を拒絶し始め、腫瘍は第20日迄に急速に消失した。単回注射も、より速やかな拒絶を生じる傾向であった(図8A)。

マウス中有で産生されたCTL がMHC であるか、または実際にクラスIMHCに限定されたか否かを決定する為に、マウスをMFP 5 μg/週 x 3で免疫処置し、その脾臓細胞を採取して、種々の51Cr標識標的細胞に対してCTL として用いた。結果は以下の通りである:
a) 免疫処置したH-2d マウス(DBA/2、NZB 、BALB/cまたはB10.D2)はP815-Tm211(MUC1+ ) P815細胞に対してCTL を産生したが、P815-MUC1 細胞に対しては産生しなかった。
b) その他のH-2 ハプロ型マウスを免疫処置した場合、CTL はH-2d P815 MUC1+ 細胞テストでは見出されなかった、特にH-2h ;C57BL/6 ;129 及びBALB.B;H- 2k ;
CBA;H-2s;SJL 及びH-2w :NZW 。これらの研究で興味ある所見は:
BALB/c (H-2d ) は+)これらはH-2 のみが異なる類似遺伝子対である。
BALB.B (H-2b )は−)
B10.D2 (H-2d )は+)これらはH-2 のみが異なる類似遺伝子対である。
C57BL/6(H-2h )は−)
本図はH-2MIC複合体と反応性であることを示す。
c) 別個の研究では、H-2b ハプロ型マウスがH-2b (E3 MUC1+ 腫瘍細胞)に対して活性を有するが、その他のH-2 ハプロ型に対しては反応性ではなかった。従って、マウスのMFP に対するCTL 反応はH-2 (MHC)限定的である。
マウスを種々のMFP 5 μg/週 x 3で免疫処置し、増殖検定に於て異なったペプチド用量でテストした。本検定で、組織培養中の脾臓細胞に異なったペプチドを異なった量で添加して、48時間後に3H- チミジンを24時間添加した。細胞を採取して取り込まれた放射活性を測定した。本研究で以下の点が示された:
i)MFPはペプチドの存在下に免疫処置されたマウスのT-細胞の増殖を刺激する。
ii)用量反応性があるのでペプチドの至適用量は以下の通りである:

本エピトープは抗体エピトープAPDTR の様ではなく、p14-24は恐らくGSTAP である。
本実施例の目的は、先ず合成またはその他のMUC1剤がヒトに於て免疫原性であるかを決定し、次いで自己ペプチドに対する免疫反応が、MUC1を含有する正常組織に及ぼす反応に依って、何等かの有害作用を有するか否かを決定するにある第I相試験である。抗腫瘍作用はその後に検討する。
試験に参加の条件として、患者は組織学的に確認された乳癌を有さねばならず、また転移病巣を考慮される患者に限り、特に治験前4週間に細胞毒性の化学療法を受けていない患者に限定した。患者は書面に依る告知済み承諾書を提出せねばならず、授乳中及び妊娠中の患者は本試験には不適格とした。全患者は臨床的状態を記録し、基礎血液試料をMSA 及びCASA分析に採取した。実施された本試験は通常の第I相試験であり、病歴及び臨床検査、全血検査、毒性記録、クレアチニン及びアミラーゼ分析の採血を含む。注射された物質に対する免疫反応の為の特定テストは数種の方法で実施した:
a) MSA 及びCASAテストを実施した。これらのテストは血中MUC1を検出し、抗原に対する免疫反応に就いてあらゆる変更を行ない、MSA は特異的な炭水化物抗原を、CASAはAPDTR ペプチドを検出する。
b) 試料はELISA テストに依って抗-MUC1 抗体を、またジフテリア毒素、融合蛋白質、GST 、p13-32合成ペプチド、HMFG及びその他のペプチドに就いて別個にテストした(STPAはCD46の配列から誘導され、被特異的陰性対照として用いた)。 c) T-細胞反応は3 種の反応で測定した:
i) 遅延型過敏性反応(DTH ) - 本反応試験はジフテリアまたは破傷風、注射物質(DT- ペプチド)に対する反応、また別個にヒト血清アルブミンの様なその他の担体に結合したペプチドに対する患者の反応を測定する為に標準的抗原を用いて実施した。
ii) T-細胞増殖 - 本テストは患者から採血し、末梢リンパ球(PBL)を分離して、適当な抗原と共に組織培養で確立して(下記参照)、48〜72時間後に3H- チミジンを添加するか、あるいは24時間後に増殖を測定した。
iii) T-細胞毒性 - 患者の増殖血球を分離してネズミMUC1+ 3T3 及びP815細胞並びにヒトMUC1(T47D、BT20)を発現するヒト腫瘍細胞から成る51Crで標識した標的細胞と4時間反応させた。
MUC1縦列反復の多様な変数(VNTR)の配列から誘導したペプチドC-p13-32
(CPAHGVTSAPDTRPAPGSTAP)を、ABI ペプチド合成器(Foster City 、California州、米国)を用いて合成した。ヒトCD46のセリン、スレオニン、及びプロリンに富んだ領域を現わすペプチドSTP-Aを、陰性対照として用いた。ペプチドC-p13-32は、グルタールアルデヒドを用いてジフテリア毒素(DT)(特別試料、CSL 、Melbourne 、オーストラリア)と複合体を形成させた。ペプチドC-p13-32 10mg を、0.25% グルタールアルデヒド 5 ml の存在下に、1250Lf DT と室温で6時間反応させ、リン酸緩衝生理食塩液に対して透析した。複合体DT-C-p13-32 を層流フード中で濾過(0.22 mm 、Millipore 社)した。DT-C-p13-32 の活性は抗-MUC1抗体BC2 でテストした。無菌度および発熱性物質テストはメルボルン大学、薬理学部及びAustin病院、微生物学部、オーストラリア)で実施した。
C-p13-32に対するヒト抗体をテストする為に、FP、DT及びHMFGを含む種々の抗原をPVC プレート(Costar)上に、0.05M 炭酸緩衝液中、pH 9.6で、2時間37℃でコートし、非特異的結合部位を2% BSAで1時間37℃でブロックした。PBS で洗浄後、0.05% 間隔で20段階希釈した血清試料を、各ウェルに注ぎ、4 ℃で一夜培養した。プレートを十分洗浄後、セイヨウワサビ パーオキシダーゼ(Silenus社、メルボルン、オーストラリア)で標識したヒツジ抗ヒト免疫グロブリンをプレートに添加し、2 時間室温で培養した。プレートを洗浄し、結合したヒト抗体は、0.02% H2O2含有0.1M クエン酸緩衝液中、pH 4で0.03% 2,2-アジノジ(3-エチルベンゾ -チアゾリンサルフェート) を添加して検出した。吸光度はELISA 検出器(Bioteck 社、EL312e)を用いて405 nmで測定した。本検定に用いた抗原は、a)ヒト乳脂肪小球(HMFG)、b) MUC1 のVNTRを5 回反復とグルタチオン-S- トランスフェラーゼを含み、P-GEX ベクトルを用いて調製しした融合蛋白質、c) DT 、d) C-p13-32 、及び e) 陰性対照ペプチドSTP-A とした。

- 抗原と、マンノースユニットとアルデヒド基とを含む酸化されたマンナンとの間の複合体と、薬学上許容される担体からなる免疫組成物。
