Description
−例えば比色定量、蛍光又は発光によって検出可能なシグナルを発する、ホースラディッシュペルオキシダーゼ、アルカリフォスファターゼ、α−ガラクトシダーゼ又はグルコース−6−リン酸デヒドロゲナーゼ等の酵素、
−蛍光、発光又は染料の化合物等の発色団、
−32P、35S又は125I等の放射性物質、及び、
−アレクサ(alexa)又はフィコシアニン等の蛍光性物質
を挙げることができるが、これらに限定されない。
・図1Aは、MCF−7、T47D、MDA−MB−231及びBT20がん細胞がNGFを分泌していること、コントロールとしての顎下腺細胞がNGFを分泌していること、並びに、NBEC正常細胞(正常な胸部上皮細胞)がNGFを分泌していないことを示す、RT−PCR後の電気泳動ゲルを示す。
・図1Bは、MCF−7、T47D、MDA−MB−231及びBT20がん細胞がNGFを分泌していること、顎下腺細胞がNGFを分泌していること、並びに、NBEC細胞がNGFを分泌していないことを示す、イムノブロッティング後の電気泳動ゲルを示す。
・図1Cは、ELISAで調べた各細胞(MCF−7、T47D、MDA−MB−231及びBT20がん細胞、顎下腺細胞並びにNBEC細胞)によって分泌されたNGFの量を示すグラフである。
・図2Aは、NGF存在下で増殖するPC12細胞(1)とがん細胞(2)との共培養の実験スキームを示す。
・図2Bは、コントロールとしてのPC12細胞の培養(写真2)、NGFの影響下におけるPC12細胞の分化(写真3)、MCF−7がん細胞の存在下におけるNGFに感受性のある細胞の共培養(写真4)、抗NGF抗体の存在下におけるMCF−7がん細胞の培養(写真5)、阻害剤K252aの存在下におけるMCF−7がん細胞の培養(写真6)、及び、NBEC正常乳房細胞の培養(写真7)を示す写真である。
・図4Aは、腫瘍増殖阻害剤がない状態(コントロール)において、又は、抗NGF抗体MAB−256の存在下において、又は、阻害剤K−252aの存在下において、MCF−7細胞の増殖を細胞数(104個/35mmディスク)として示すグラフである。
・図4B及び4Cは、MCF−7細胞の数をMAB−256抗体の投与量に対して(図4B)、又は、阻害剤K−252aの投与量に対して(図4C)示すグラフである。
1.1 試験する細胞
ヒト乳がん細胞株4株、すなわち、ATCC collection(メリーランド州ロックヴィル)から得たMCF−7、T47D、MDA−MB−231及びBT20細胞について、それぞれHTB−22、HTB−133、HTB−26及びHBT−29と番号をつけて試験した。
Rneasyミニキット(Qiagen社、フランス)を使用して、がん細胞株、NBEC細胞及びヒト顎下腺(重量で20mg)細胞から総RNAを分離した。Rotor−Statorホモジナイザー(Bribolyser、Hybaid社)を使用して、顎下腺を破裂してホモジナイズした。
センスプローブ:5’−GACAGTGTCAGCGTGTGGGTT−3’
アンチセンスプローブ:5’−CCCAACACCATCACCTCCTT−3’
95℃で3分間処理してcDNAを変性させた後、94℃1分間、57℃2分間及び72℃3分間のサイクルを30回実施した。最終サイクル終了後、PCRチューブを72℃で更に10分間インキュベートしてcDNA断片を伸長させた。
2.1 試験する細胞
試験する細胞は上記1.1中で示すものである。
上記1.1中で示すように培養してサブコンフルエントにした細胞を洗浄し、フィブロネクチン及びトランスフェリンを含む非血清培地中に24時間配置した。細胞から栄養素を除去した後、PBSバッファー(リン酸緩衝生理食塩水)中でこすり取ることによって細胞を回収し、遠心分離(1000g、5分間)した。
3.1 試験する細胞
試験する細胞は上記1.1中で示したものである。
この試験を実施するために、ヒトβ−NGFについてtwo−siteアッセイを使用した。簡潔に説明する。希釈バッファー(BSA0.1%、Tris緩衝生理食塩水中にTween20を0.005%溶解したもの、pH7.3)中に希釈したモノクローナル捕獲抗体(R&D Systems社、英国)1μg/mlでイムノプレートをコートした。コントロールのウェルは、同量のマウスIgG(R&D Systems社)でコートした。各ウェルに上記希釈液100μlを注入してプレートを密閉し、室温で一晩インキュベートした。
この培養を実施するために、NGFの存在下で増殖する細胞、すなわちPC12細胞を、上記1.1に記載するようにMCF−7乳がん細胞と、又は、上記1.1に記載するようにNBEC細胞と(コントロール)、共培養した。
−図2Aは実験スキームを示すが、このスキーム中の(2)はがん細胞又は非がん細胞であり、(1)はPC12細胞である。
・写真2及び3は、コントロールとしてPC12細胞の培養をNGF非存在下及びNGF存在下においてそれぞれ示しており、分化による神経突起の伸長が観察される。
・写真4はMCF−7及びPC12細胞の共培養に関するものであり、PC12細胞が培養48時間後に分化していることを示す。
・写真5は抗NGF抗体の存在下でのMCF−7及びPC12細胞の共培養に関するものであり、この抗体によってPC12細胞の分化が強く阻害されることを示す。
・写真6は阻害剤K−252aの存在下でのMCF−7及びPC12細胞の共培養に関するものであり、この阻害剤によってPC12細胞の分化が強く阻害されることを示す。
・写真7は正常なNBEC細胞及びPC12細胞の共培養に関するものであり、PC12細胞が分化していないことを実証する。
5.1 生検の採取及び固定
手術直後に、人間の乳がん組織又は正常な非がん組織(コントロール)の生検を、10%ホルモル溶液で24時間固定した。アルコール溶液を(30〜100%)増やして脱水した後、試料をパラフィン中に包埋した。そして、切片をミクロトームで切断した後(5μmの薄い切片)、3−アミノプロピルトリエトキシシラン(TESPA、Dako社、フランス)でコートしたスライドグラス上に固定した。
下記工程に従ってスライドグラスを浴中に浸漬することによってパラフィンを除去した。
−トルエン浴二回(10分間)、
−100%アルコール浴一回(10分間)、
−95%アルコール浴一回(10分間)、
−70%アルコール浴一回(10分間)、
−水浴一回(10分間)。
その後、上記スライドグラスを+4℃で保存した。
切片をTBS−0.1%Tween(TBS:NaClを17.54g、Tris塩基2.42g、水qs2L、pH7.4)で10分間震盪しながら洗浄した。0.01Mクエン酸バッファー(クエン酸一水化物1.05g、蒸留水qs500ml、pH6(NaOH))を使用して95℃の湯浴中で5分間ずつ二回、抗原の再活性化を実施した。上記スライドグラスを3%過酸化水素水溶液/TBS中で震盪しながら5分間インキュベートし、TBS/3%BSA中に室温で1時間浸漬した。
培養皿の底をこすり取ることによってMCF−7細胞を剥がした。その後、定着剤(Coulter社)100μlを添加し、室温で15分間インキュベートした。その後、細胞をPBSで三回洗浄し、1900rpmで5分間遠心分離した。
MCF−7細胞の増殖に及ぼすブロッキング抗体MAB−256(上記)及び阻害剤K−252aの影響を調べた。これを調べるために、2×104個/mlの割合で35mmディッシュ中の完全培地中に細胞を播種した。40%コンフルエントに達した後、細胞を二回洗浄し、フィブロネクチン(2μg/ml)及びトランスフェリン(30μg/ml)を含む非血清培地中で栄養分を与えずに3時間保持した。その後、様々な濃度の阻害剤K−252a又は抗体MAB−26を含む上記と同様の培地2mlで培地を置き換えて、薬理的阻害剤又はブロッキング阻害剤が増殖基準値に及ぼす影響を調べた。
−図4Aは、基本培地中のMCF−7細胞の増殖(正方形)が、抗体の存在下(三角形)及び阻害剤の存在下(菱形)で減少することを示す。
−図4B及び4Cは、抗体及び阻害剤はいずれも投与量に応じて、NGFのがん細胞増殖効果に影響を及ぼすことを示す。
従って、NGFは治療の際のターゲットとして確かに使用できる。
MDA−MB−231乳がん細胞の増殖を以下のように調べた。
SCIDマウス(劇症合併免疫不全症)30匹の脇腹に、in vitro培養由来のMDA−MB−231細胞をマウス一匹当たり3×106個/PBS50μLの割合で時間0(D0)において皮下注射した。マウスは、標準的な繁殖コロニー条件において室温で昼/夜を交互にした状態で、食物及び飲料を自由に摂取させた。

表中、線を引いた柱状グラフは抗NGF抗体で処置していないマウス(コントロール)を示し、白抜きの柱状グラフは上記抗体で処置したマウスを示す。
Claims (14)
- 乳がんの疑いのある患者から採取した体液中のNGFの存在を決定することからなる
ことを特徴とするin vitroにおける乳がんの検出方法。 - NGFの存在は前記体液中のNGFの直接検出によって決定する
ことを特徴とする請求項1に記載の方法。 - NGFの検出はイムノアッセイ又は質量分析によって実施する
ことを特徴とする請求項2に記載の方法。 - 前記体液は、前処理して前記体液中に含まれている循環腫瘍細胞を分離した体液からなる
ことを特徴とする請求項2又は3に記載の方法。 - 前記循環腫瘍細胞をその後、NGFを分泌する条件で培養する
ことを特徴とする請求項4に記載の方法。 - 前記循環腫瘍細胞を、前記に加えて、前記循環細胞内部のNGFを阻害した条件でも培養する
ことを特徴とする請求項4又は5に記載の方法。 - NGFの検出は、NGFに感受性のある細胞を前記体液の存在下で培養することによって決定する
ことを特徴とする請求項1に記載の方法。 - 前記体液は、前処理して前記体液中に含まれている循環腫瘍細胞を分離した体液からなる
ことを特徴とする請求項7に記載の方法。 - 前記循環腫瘍細胞をその後、NGFを分泌する条件で培養する
ことを特徴とする請求項8に記載の方法。 - NGFの検出は、前記体液中のNGFmRNAの検出によって実施する
