Description
即ち、本発明は、下記一般式(1)で表されるフルオレン誘導体と下記一般式(2)で表される有機金属化合物及び塩基とを触媒存在下に反応させることを特徴とする下記一般式(3)で表されるアリールアミン誘導体の製造方法に関するものである。
該フルオレン誘導体の合成方法については、特に制限はないが、例えば、下図に示すようなルートで合成可能である。具体的には、9,9’−ビス(4−ヒドロキシフェニル)−2,7−ジブロモフルオレン誘導体を原料に、メトキシエチルメトキシ基で保護、相当するボロン酸、グリニャール試薬、アミン化合物又はアルコキシ化合物にてカップリングし最後にルイス酸等で脱保護したのちトリフルオロメタンスルホン酸無水物と反応させることで合成可能である。
上記一般式(1)で表されるフルオレン誘導体中のR1〜R4のアルキル基としては、炭素数1〜18の直鎖,分岐若しくは環状のアルキル基が挙げられ、具体的には、メチル基、エチル基、プロピル基、イソプロピル基、ブチル基、sec−ブチル基、tert−ブチル基、ペンチル基、ヘキシル基、ヘプチル基、オクチル基、ステアリル基、トリクロロメチル基、トリフロロメチル基、シクロプロピル基、シクロヘキシル基、1,3−シクロヘキサジエニル基、2−シクロペンテン−1−イル基等を挙げることができる。
R7〜R12の具体例としては、前記のR1〜R4、Ar1、Ar2と同様の置換基を挙げることができる。前記一般式(9a)〜(9e)のアリーレン基のうち、(9a)、特にフェニレン基が合成的観点から原料が入手し易く特に好ましい。
窒素雰囲気下、キシレン90mlに4−ブロモクロロベンゼン10.4g(54.6mmol)、3−メチルジフェニルアミン10.0g(54.6mmol)、ナトリウム−tert−ブトキシド6.3g(65.5mmol)を懸濁させた。更に、酢酸パラジウム5.8mg、トリ−tert−ブチルホスフィン21mgを添加し、125℃に加熱した。同温度で2時間熟成した後、反応液を室温まで冷却した。水20mlで洗浄後、減圧下で溶媒を留去した。残査をシリカゲルカラムクロマトグラフィー及び再結晶により精製し、無色針状結晶を14.3g(収率89%)得た。NMR及び質量分析(FDMS)から目的物であることを確認した。

合成例2 4−(3−メチルジフェニルアミノ)ブロモベンゼンの合成
窒素雰囲気下、トルエン20mlに4−ブロモヨードベンゼン4.74g(16.81mmol)、3−メチルジフェニルアミン3.38g(18.46mmol)、塩化銅83mg(0.84mmol)、フェナントロリン166mg(0.84mmol)、水酸化カリウム7.53g(134.22mmol)を加え、加熱還流下15時間反応させた。反応液を室温まで冷却、トルエン70mlを加えた後、ろ過して不溶分を取り除いた。ろ液を飽和塩化アンモニウム水溶液、水で洗浄し、減圧下にて溶媒を留去した。シリカゲルクロマトグラフィー及び再結晶により精製し、無色針状結晶を得た。NMR及び質量分析(FDMS)から目的物であることを確認した。

合成例3 4−(3−メチルジフェニルアミノ)フェニルボロン酸の合成(合成法1)
窒素雰囲気下、テトラヒドロフラン2ml、マグネシウム0.4gに合成例1で得た4−(3−メチルジフェニルアミノ)クロロベンゼン4.0g(13.6mmol)のテトラヒドロフラン溶液4mlを滴下し、加熱還流下10時間反応させた。反応液を−78℃に冷却した後、ホウ酸トリメチル1.4g(13.6mmol)のテトラヒドロフラン溶液を滴下し、更に室温で12時間攪拌した。ここに10%塩酸水溶液を添加、有機層と水層を分離し、水層はジエチルエーテルで抽出した。抽出液を減圧下で濃縮した後、ヘキサンを添加し、ろ過すると白色粉末2.6g(収率63%)が得られた。NMR及びIRから目的物であることを確認した。

合成例4 4−(3−メチルジフェニルアミノ)フェニルボロン酸の合成(合成法2)
窒素雰囲気下、テトラヒドロフラン40mlに合成例2で得た4−(3−メチルジフェニルアミノ)ブロモベンゼン3.0g(8.9mmol)を溶解させた。−78℃に冷却し、n−ブチルリチウムのヘキサン溶液(2.6mol/L)を4.1ml添加した後、0℃で2時間反応させた。反応液を再度−78℃に冷却し、ホウ酸トリメチル0.9g(8.9mmol)のテトラヒドロフラン溶液10mlを滴下、更に室温で12時間攪拌した。ここに10%塩酸水溶液を添加、有機層と水層を分離し、水層はジエチルエーテルで抽出した。抽出液を減圧下で濃縮した後、ヘキサンを添加し、ろ過すると白色粉末が得られた。NMR及びIRから目的物であることを確認した。
窒素雰囲気下、9,9−ビス[4−(トリフルオロメタンスルホニルオキシ)フェニル]フルオレン1.23g(2.0mmol)をテトラヒドロフラン6mlに溶解させ、20%炭酸ナトリウム水溶液4.7g、4−(3−メチルジフェニルアミノ)フェニルボロン酸1.27g(4.2mmol)、テトラキス(トリフェニルホスフィン)パラジウム23mg(0.02mmol)を加えた。65℃で2時間反応させた後、室温まで冷却し、分液した。有機層を飽和塩化アンモニウム水溶液及び飽和食塩水で洗浄し、減圧下にて溶媒を留去した。残査をシリカゲルクロマトグラフィーに付し、淡黄色粉末1.27g(収率84%、LC純度=99.7%)を得た。元素分析及び質量分析(FDMS)から目的物であることを確認した。

比較例1 9,9−ビス[4−(3−メチルジフェニルアミノ)−1,1’−ビフェニル]フルオレン(化合物1)の合成
50ml三つ口フラスコに9,9−ビス(4−クロロ−1,1’−ビフェニル)フルオレン1.0g(1.8mmol)、N−(3−トリル)アニリン0.68g(3.7mmol)ナトリウム−tert−ブトキシド0.54g(5.6mmol)及びキシレン20mlを窒素雰囲気下仕込み、触媒として酢酸パラジウム4mg、トリ−tert−ブチルホス10mgを添加して120℃で6時間反応させた。反応後、水を添加して有機相を分離した。有機層を濃縮した後ショートカラムにて精製し化合物1を1.3g単離した。LC純度は97.6%であった。
合成例3に準じて得られた4−(3−メチルジフェニルアミノ)フェニルマグネシウムクロリドのグリニャール溶液(4.2mmol)と9,9−ビス[4−(トリフルオロメタンスルホニルオキシ)フェニル]フルオレン(2.0mmol)とをテトラキス(トリフェニルホスフィン)パラジウム触媒存在下に2時間還流させた。後処理を実施例1に準じて、目的とする化合物1を64%で単離した。LC純度を表1に示す。
4−(3−メチルジフェニルアミノ)フェニルボロン酸の代わりにトリフェニルアミンボロン酸(4.2mmol)を用いた以外は、実施例1に準じて化合物2を88%で単離した。FDMSにより目的物であることを確認した。LC純度を表1に示す。
4−(3−メチルジフェニルアミノ)フェニルボロン酸の代わりに4−(N−カルバゾリル)フェニルボロン酸(4.2mmol)を用いた以外は、実施例1に準じて化合物3を81%で単離した。FDMS、1H及び13C−NMRにより目的物を確認した。LC純度を表1に示す。

実施例5(化合物4の合成)
9,9−ビス[4−(トリフルオロメタンスルホニルオキシ)フェニル]フルオレンの代わりに2,7−ビス(4−ビフェニル)−9,9−ビス[4−(トリフルオロメタンスルホニルオキシ)フェニル]フルオレンを用いた以外は実施例1に準じて目的とする化合物4を単離した。FDMSにより目的物であることを確認した。LC純度を表1に示す。

Claims (8)
- 下記一般式(1)で表されるフルオレン誘導体と下記一般式(2)で表される有機金属化合物及び塩基とを触媒存在下に反応させることを特徴とする下記一般式(3)で表されるアリールアミン誘導体の製造方法。
- 上記一般式(2)において、Ar3がフェニレン基であることを特徴とする請求項1に記載のアリールアミン誘導体の製造方法。
- 上記一般式(2)においてMが上記一般式(4)又は(5)のいずれかであることを特徴とする請求項1乃至2のいずれかに記載のアリールアミン誘導体の製造方法。
- 上記一般式(2)においてMが上記一般式(5)であることを特徴とする請求項3記載のアリールアミン誘導体の製造方法。
- 触媒が0価又は2価のパラジウム触媒であることを特徴とする請求項1乃至4のいずれかに記載のアリールアミン誘導体の製造方法。
- 触媒が2価のパラジウム触媒であることを特徴とする請求項5記載のアリールアミン誘導体の製造方法。
- 触媒がジクロロビス(トリフェニルホスフィン)パラジウムであることを特徴とする請求項5に記載のアリールアミン誘導体の製造方法。
- 触媒がパラジウム炭素であることを特徴とする請求項5に記載のアリールアミン誘導体の製造方法。
