Description
本発明により使用されるラクトバチルス菌株の選択は次の2段階方式において実施することができる:
a)ラクトバチルス菌株によるS.ムタンスの阻止効果の評価
阻止効果を測定するために使用する菌株の1例はスタフィロコッカス・ムタンス、ATCC25175(The American Type Culture Collection,Manassas,VA,USAから得られる)である。分離株は、0.5%酵母エキス(Difco)を補足したトリプティカーゼ大豆ブロス(Difco、Detroit,USA)(TSBY)において増殖される。細胞は対数増殖期に1000xgでの遠心分離によって収穫し、PBSにより2回洗浄し、そして同じバッファーに再懸濁する。細胞懸濁液を低度の超音波装置にかけて細菌の凝集塊を分散させる。
ブタの胃からのムチン(Sigma,St.Louis,MO,USA)を濃度0.1mg/mlにおいてpH9.7の炭酸バッファーに懸濁する。この溶液200μlをミクロタイターウェル中にピペットで採取し、37℃で約3時間、コーティングのために放置する。ウェルを200μlPBS1%Tween20の添加によって室温で1時間ブロックし、そしてPBS0.5%Tween20(PBST)で3回洗浄する。ラクトバチルスの評価菌株を、イン・ビボの増殖条件を模倣して0.01%ブタ・胃ムチン(Sigma,M1778)を補足したMRSブロスにおいて37℃で一夜増殖させ、次いでPBSTで洗浄し、再懸濁する。細菌細胞の光学濃度(OD)をBeckman DU650分光光度計で600nmにおいて測定し、OD0.5に調節する。100μlの細胞懸濁液を各ウェルに添加し、4℃で一夜インキュベートする。ウェルをPBSTで洗浄し、そして結合を倒立顕微鏡により検査する。バッファーを注ぎ出し、ウェルを乾燥した後、OD405をELISAプレートリーダーにおいて測定する。結合の結果を0〜3のスケールでスコアするが、ここで0は結合なしであり、3はムチンに対する強い結合である。

この研究においては、前記ラクトバチルス菌株を、本発明にしたがってS.ムタンスの阻止およびムチンへの粘着という選択基準を用いて評価するために選ぶ。実施例1に記述した方法を使用する。もっとも適当な菌株であるとして選択された菌株は、ラクトバチルス菌株細胞のS.ムタンス阻止効果およびムチンへの粘着の組み合わせに基づいている。
実施例2のリストにしたがって添加した評価ラクトバチルス株を用いて発酵させた乳(試験ヨーグルト)、およびプラセボ発酵乳(プラセボ・ヨーグルト)のムタンス連鎖球菌の口中保菌(carriage)に及ぼす効果を検討する。試験ヨーグルトは次のとおりである:
−試験ヨーグルト1.、L.ロイテリSD2112含有
−試験ヨーグルト2.、L.ロイテリDSM20016含有
−試験ヨーグルト3.、L.ロイテリMM2−3含有
−試験ヨーグルト4.、L.ロイテリCF2−7F含有
−試験ヨーグルト5.、L.ロイテリMF2−3含有
−試験ヨーグルト6.、L.ロイテリMF14−C含有
−試験ヨーグルト7.、L.ロイテリMF52−1F含有
−試験ヨーグルト8.、L.サリバリウスLS1含有
−試験ヨーグルト9.、L.ラムノサスGG含有
−試験ヨーグルト10.プラセボ、以下参照
200人の健康な女性被験者(年齢;20±2才)を10群に分ける。全被験者はいかなる活性なう食病巣、歯肉炎もしくは歯根膜疾患のいずれの症候も有してはならない。第1群の被験者は、2週間の間、毎日昼食時(12:00〜13:00)にプラセボ・ヨーグルト1カップ(95g)を食し、次いでその後の2週間の間昼食時に試験ヨーグルトNo.1の1カップを食す。第2群の被験者は、2週間の間、毎日昼食時(12:00〜13:00)にプラセボ・ヨーグルト1カップ(95g)を食し、次いでその後の2週間の間1日の昼食時に試験ヨーグルトNo.2の1カップを食し、そしてすべて9試験群については第1群と同じようにする。試験群No.10は両期間プラセボ・ヨーグルトを食する。各種ヨーグルトの摂食前後に、ムタンス連鎖球菌の口中保菌レベルを次のように決定する。未刺激での総唾液約5mlを15:00〜16:00に氷上の容器中に収集する。次いで、ムタンス連鎖球菌の口中保菌を慣用の生存菌数によって決定する。プラセボ・ヨーグルトは、発酵乳製品において広く用いられるL.ブルガリスとS.サーモフィルスからなり、このプラセボ・ヨーグルトを80℃で10分間加熱して微生物を殺菌し、試験ヨーグルトNo.10として調製さる。被験者および研究者は共に、研究中どのヨーグルトがどの試験微生物を含有していたかを知らない。ラクトバチルスもしくは他の製薬学的乳酸菌を含有する他の生産物の使用は、本介入前1週間および介入中は禁止される。データは統計学的に解析される。
この実施例では、L.ロイテリCF2−7F(指定された場合付加されるATTC番号)が、標準的ヨーグルトに菌株を添加するために、S.ムタンス阻止とムチン結合に関して上記方法を用いて選択された。L.ロイテリ菌株を、酪農産業においてラクトバチルスを増殖させる標準的方法を用いて、増殖させ、凍結乾燥した。次いでこの培養物を伝統的なヨーグルト培養物を用いて前以て発酵した乳に、107CFU/ヨーグルト1gのレベルにおいて添加し、そしてこのヨーグルトを虫歯を予防する方法としてヒトによって使用した。
以下に本発明の主な特徴と態様を列挙する。
1.ラクトバチルス・ロイテリ菌株CF2−7F(与えられるATCC番号)の生物学的に純粋な培養物。
2.ラクトバチルス・ロイテリ菌株MF2−3(与えられるATCC番号)の生物学的に純粋な培養物。
3.口のムチンへの良好な結合と組み合わせた阻止活性の選択アッセイを用いて、哺乳動物の虫歯の予防または処置のために口中のストレプトコッカス・ムタンスの数を低減する能力について選択されたラクトバチルスの生物学的に純粋な培養物。
4.口のムチンへの良好な結合と組み合わせて齲蝕原性細菌に対する阻止活性を有するよう選択されたラクトバチルス・ロイテリの少なくとも1菌株の細胞を含んでなる、虫歯細菌の増殖を阻止するための生産物。
5.少なくとも1つの菌株が、ラクトバチルス・ロイテリ菌株CF2−7F(与えられるATCC番号)およびラクトバチルス・ロイテリ菌株MF2−3(与えられるATCC番号)からなる群から選ばれる、4.に記載の生産物。
6.生産物が食用生産物である、4.に記載の生産物。
7.食用生産物が、ヨーグルト、ゼリー、プディング、チューインガム、キャンディー、チョコレート、ビスケット、クッキー、チーズ、ジュースおよびティーからなる群から選ばれる、6.に記載の生産物。
8.食用生産物が乳含有生産物である、6.に記載の生産物。
9.食用生産物がヨーグルトである、8.に記載の生産物。
10.生産物が歯科処置用生産物である、4.に記載の生産物。
11.生産物がマウスウォッシュである、10.に記載の生産物。
12.口のムチンへの良好な結合と組み合わせて齲蝕原性細菌に対する阻止活性を有するよう選択されたラクトバチルス・ロイテリの少なくとも1菌株の細胞を生産物に添加することを含んでなる、抗う食原性生産物を製造する方法。
13.少なくとも1つの菌株が、ラクトバチルス・ロイテリ菌株CF2−7F(与えられるATCC番号)およびラクトバチルス・ロイテリ菌株MF2−3(与えられるATCC番号)からなる群から選ばれる、12.に記載の方法。
14.生産物が食用生産物である、12.に記載の方法。
15.食用生産物が、ヨーグルト、ゼリー、プディング、チューインガム、キャンディー、チョコレート、ビスケット、クッキー、チーズ、ジュースおよびティーからなる群から選ばれる、14.に記載の方法。
16.食用生産物が乳含有生産物である、14.に記載の方法。
17.食用生産物がヨーグルトである、16.に記載の方法。
18.生産物が歯科処置用生産物である、12.に記載の生産物。

Claims (17)
- ラクトバチルス・ロイテリ菌株CF2−7F(ATCC PTA−4965)の生物学的に純粋な培養物。
- ラクトバチルス・ロイテリ菌株MF2−3(ATCC PTA−4964)の生物学的に純粋な培養物。
- 哺乳動物の虫歯の予防または処置のための抗齲蝕生産物の製造方法であって、
a.ラクトバチルス・ロイテリの菌株を選択しおよび培養し、
b.前記ラクトバチルス・ロイテリの菌株の、ストレプトコッカス・ムタンスに対する阻止活性および口のムチンへの粘着能力を評価し、ここで、前記菌株のストレプトコッカス・ムタンスに対する良好な阻止活性および口のムチンへの良好な結合を示すことの両方の能力が生産物に使用するラクトバチルス・ロイテリの菌株を決定するものであり、そして、
c.段階b.で選択したラクトバチルス・ロイテリの少なくとも1つの菌株の細胞を含む生産物を調整すること、
を含んでなる上記方法。 - ラクトバチルス・ロイテリ菌株ATCC PTA−4965およびラクトバチルス・ロイテリ菌株ATCC PTA−4964からなる群から選ばれるラクトバチルス・ロイテリの少なくとも1つの菌株の細胞を含んでなる、虫歯細菌の増殖を阻止するための生産物であって、前記菌株が口のムチンに対する良好な結合との組み合わせで虫歯菌に対する阻止活性を有する、上記生産物。
- 生産物が食用生産物である、請求項4に記載の生産物。
- 食用生産物が、ヨーグルト、ゼリー、プディング、チューインガム、キャンディー、チョコレート、ビスケット、クッキー、チーズ、ジュースおよびティーからなる群から選ばれる、請求項5に記載の生産物。
- 食用生産物が乳含有生産物である、請求項5に記載の生産物。
- 食用生産物がヨーグルトである、請求項7に記載の生産物。
- 生産物が歯科処置用生産物である、請求項4に記載の生産物。
- 生産物がマウスウォッシュである、請求項9に記載の生産物。
- 少なくとも1つの菌株が、ラクトバチルス・ロイテリ菌株ATCC PTA−4965およびラクトバチルス・ロイテリ菌株ATCC PTA−4964からなる群から選ばれ、前記菌株が口のムチンに対する良好な結合との組み合わせで虫歯菌に対する阻止活性を有する、請求項3に記載の方法。
- 生産物が食用生産物である、請求項3に記載の方法。
