の生成物が高収率で生産されるように安定して保存され
る強い調節エレメント(プロモーター及びターミネータ
ー)を含有するベクター。 2)異種遺伝子により提供される指令を正確に実行し
て、天然生成物と同一の生成物の生産を可能とし、コマ
ーシャルレベルでの培養に好適である[すなわち、耐性
であり、高密度に増殖でき、栄養因子に関する要求があ
まり過酷でなく、安全(すなわち毒性の汚染物を生産し
ない)である]宿主。
に当たりバシラス・サチリス(Bacillus subtilis)(以
降、B.サチリスと表示する)が注目されている。実
際、B.サチリスは、完全な非病原性であり、培地中に
遺伝子発現生成物を分泌でき、大規模な生育が容易であ
るため、バイオテクノロジーの見地から特に注目される
微生物である。医薬品工業及び食品工業にとって興味深
い異質タンパク質及びポリペプチドの発現用宿主として
B.サチリスを使用することは、これら生成物の生産方
法の許可を受けるための重要な要因である。しかしなが
ら、微生物の使用に関する禁忌の1つは、ある種の生理
的生育条件下における胞子形成の能力による。事実、化
学的−物理的試薬に対する高い抵抗性によって特徴づけ
られる胞子は、最も一般的な環境条件下で生残る大きい
可能性を有する。医薬及び食品の分野で興味深い生成物
の製法においてB.サチリスの組換え菌株を使用するこ
とは、胞子が外部環境に分散される可能性が低いことを
示さない場合には、法律上の規制を受けることになる。
B.サチリスにおける内生胞子の形成は、細胞の生理系
及び超微細構造における変化(生育栄養源が限定される
条件に対する応答の結果として)によって生ずる細胞分
化過程である。胞子形成(平均して37℃において6ない
し8時間で行われる)の間に、細胞は一連の良好に限定
された形態学的段階を受ける。この段階は胞子嚢内での
増殖形への交代細胞形である内生胞子の形成によって終
わる。これらの段階(慣例的に段階0−VIIとして定義
される)は、異なる遺伝子(spo遺伝子)によってコー
ド付けられる一連の物質を要求する。当分野では、化学
試薬又は物理試剤によるspo遺伝子の突然変異により、
又はインビトロでの変異誘発技術の利用により胞子を生
産しないB.サチリスの菌株(無胞子性菌株)を調製す
ることが知られている。胞子の形成を生じなくなった突
然変異体は、理論的には、異質タンパク質の生産に使用
される。しかしながら、突然変異体のいくつかのもの
は、spo-表現型の不安定性、発現ベクターを安定に保存
できないこと、菌株内に存在するベクターのコピー数が
少ないことにより、B.サチリスの無胞子性発現用の系
の開発に関して完全には満足できる宿主ではないことが
明らかになっている。
サチリスの無胞子性突然変異体を新たに単離した。この
突然変異体(SMS275として公知)は1990年10月5日付け
でCentraalbureau Voor Schimmelculturesに寄託してあ
り、その受託番号はCBS 432.90である。このように、本
発明の目的は、無胞子性菌株バシラス・サチリス SMS27
5を提供することにある。本発明の他の目的は、異質生
成物の生産用宿主−ベクター系における宿主としての該
菌株の使用にある。本発明の他の目的は、異質生成物を
コード付ける遺伝子を含有する発現ベクターによる無胞
子性菌株バシラス・サチリス SMS275の形質転換、好適
な培地中での形質転換菌株バシラス・サチリス SMS275
の生育及びこのようにして生産された遺伝子発現生成物
の分離及び精製を包含する所望の異質生成物の製法にあ
る。特に、本発明による無胞子性菌株バシラス・サチリ
ス SMS275は、遺伝子マーカー spoII:D-、Leu(ロイシ
ンの不存在下では最小培地中で生育しない)、pyrD1
(ウラシルの不存在下では最小培地中で生育しない)、a
pr-及びnpr-(セリンプロテアーゼ及びニュートラルプ
ロテアーゼを生育しない)によって特徴づけられる。該
菌株はspo-表現型を安定して保存でき(実際のところ、
胞子形成性への逆戻りの頻度は約10-8である)、複製可
能な発現ベクターの多数のコピーを安定に保存できる。
たり使用できる方法は、B.サチリスの胞子形成菌株に
ついてトランスポゾンにより突然変異を生じさせ、この
ようにして生成した無胞子性突然変異体を単離すること
からなる。トランスポゾンは、移動可能で、ゲノム内の
異なった位置に挿入され、宿主の菌株に新たに遺伝性特
性を付与するDNAのエレメントである。事実、ゲノム内
の部位に挿入された後、トランスポゾン(抗生物質に対
する耐性をコード付ける遺伝子を含有する)は遺伝子の
配列を中断する(表現型突然変異によって表示される)
と共に、宿主に特殊な抗生物質に対する耐性を付与す
る。本発明の1具体例によれば、トランスポゾンTN917
(Tomich及びClewell,J.Bacteriol.,(1980),141:1
366−1574)(中でも、抗生物質のエリスロマイシン(E
m)に対する耐性をコード付ける)を使用する。トラン
スポゾンの挿入による突然変異は、公知技術による接合
又は形質転換によって行われる。特に、本発明による無
胞子性菌株は、野生形(胞子形成性)の菌株B.サチリ
ス SMS118を、バシラス属菌において複製されないもの
であって、トランスポゾンTN917を含有するプラスミド
で形質転換させ、エリスロマイシンを添加した培地で突
然変異菌株を選別することによって調製される。実際の
ところ、理論上では、トランスポゾンTN917が胞子形成
性菌株の染色体DNA内に一体化されたクローンのみがこ
の培地で生育できる。
pTV1TS、pTV32TS、又はpTV51TS(Youngmannら,Regulat
ion of Prokaryotic development,I.Smith,R.A.Sl
epacky及びP.Settlow編,American Soc.for Microbio
logy,p65−87,1989)である。一方、トランスポゾン
TN917は、プラスミドpAD2[Tomich及びClewell,J.Bac
teriol.,(1980),141:1366−1574]から単離され、
B.サチリス内で複製されないプラスミドに導入され
る。エリスロマイシン耐性クローンの非胞子形成特性
を、光学顕微鏡での分析及び胞子形成培地上での直接表
示(胞子を形成するコロニーは数日後には褐色に着色す
るが、無胞子性のコロニーは白色のままであり、溶菌す
る傾向がある)の両方によってテストした。分析したい
くつかのEm耐性(Emr)形質転換体はspo-表現型を示し
た。spoII:D遺伝子内における変異を含むspo-形質転換
体の1つ(SMS275と称す)を、さらにleu、pyrD1、apr-
及びnpr-遺伝子型の安定性をチェックするためテストに
供した。Leu及びpyrD1マーカーの分析を、ロイシン及び
ウラシルの不存在下及び存在下、最小培地で菌株SMS275
を生育することによって行った。両化合物を含有する培
地でのみ生育する菌株の能力はマーカーの安定性を示す
ものである。また、apr-及びnpr-マーカーの分析を、カ
ゼインを濃度1%で含有する最大培地(たとえばVY培地
又はTBAB培地(DIFCO)など)で菌株SMS275を平板培養
することによって行った。SMS275の周りに暈輪が存在し
ないことは、該菌株が2種のプロテアーゼを生産しない
ことを表示する。最後に、菌株SMS275のspo-表現型の安
定性を、該菌株を高温で処理することにより検定した。
このテストは、胞子を生成し得ない菌株(spo-表現型)
は短時間の高温処理に対する抵抗性が低下するとの事実
に基づくものである。熱処理は細胞を破壊するが、胞子
を破壊しないため、平面培地上で生育するコロニーは、
液体培地中での生育の間に生成され、最大培地上で平板
培養される際に発芽し得る胞子に由来するものである。
この目的のため、菌株SMS275を胞子形成培地において37
℃で約24時間生育させ、ついで80℃で約10分間熱処理し
た。ついで、培養物の好適な希釈物を、カナマイシン及
びクロラムフェニコールを含有する最大培地で平板培養
し、生きている細胞(CFU)を計数した。データの分析
結果は、菌株SMS275がspo+表現型に逆戻りする頻度は1
×10-8より小であることを示した。このように、菌株
B.サチリス SMS275は、所望の異質生成物の生産用宿
主−ベクター系における宿主としての使用に特に適して
いると思われる。
株を、所望の異質生成物をコード付ける遺伝子を含有す
る複製可能な発現ベクターで形質転換させ、このように
して形質転換した無胞子性菌株を好適な条件下で生育さ
せ、最後に、得られた遺伝子発現生成物を単離し、精製
することからなる。好適なベクターは、各種実験室及び
コレクションセンターから利用できるB.サチリスにお
いて複製可能なプラスミドの中から選択される。これら
ベクターによるB.サチリス SMS275細胞の形質転換
は、常法の1つを利用して行われる。無胞子性菌株B.
サチリス SMS275は、たとえば酵素(α−アミラーゼ、
β−アミラーゼ等)の如き原核性ポリペプチド、又はた
とえばインターロイキン、インターフェロン、ヒト生長
ホルモン、又はこれらの前駆体の如き真核性ポリペプチ
ドをコード付ける遺伝子の発現用宿主として有用であ
る。本発明の1具体例によれば、ヨーロッパ特許公開第
321940号に開示された如く、ヒト生長ホルモンの前駆体
をコード付けるDNA配列を含有するプラスミド pSM274で
菌株B.サチリス SMS275を形質転換させる。ついで、
形質転換された菌株を、炭素源、窒素源及びミネラル塩
を含有する培地において、光学密度(波長600nmで測
定)約3−4まで生育させる。
S275(pSM274)のhGH前駆体の生産能力を測定する。そ
の結果は、プラスミド pSM274が安定であること(多数
のコピーが細胞中に存在する)を示した(図1)。さら
に、菌株から抽出された総可溶性タンパク質の電気泳動
分析では、ヒト生長ホルモンの前駆体に相当するバンド
の存在を示した。菌株B.サチリス SMS275(pSM274)
の生存度を、グリセリン中に6カ月間維持した培地1ml
当たりの生存細胞の数(CFU/ml)の測定によって評価
した。得られた結果は、これらが保持された条件下にお
いて細胞の生存度が良好であること及びspo-表現型が安
定であることを示した。プラスミド pSM274を含有する
無胞子性菌株B.サチリス SMS275を1990年10月5日付
けでCentraalbureau Voor Schimmelcuturesに寄託して
あり、受託番号はCBS 433.90である。
ス spo-菌株をプラスミド pSM274で形質転換させる例を
開示する。プラスミドの安定性及びペアーの数について
及びspo-表現型の安定性について分析を行った際、これ
らの菌株は下記の結果を示した。 −spoOF変異を有する菌株SMS268内では、プラスミド pS
M274は構造的には不安定である。 −spoIIA1変異を有する菌株SMS270内では、野生形菌株S
MS118内におけるよりも少ないプラスミド pSM274のコピ
ーが存在する。 −spoIIF96変異を有する菌株SMS272内では、プラスミド
は安定であり、コピーの数は野生形菌株SMS118内におけ
るものに匹敵する。しかしながら、顕微鏡観察によって
行った評価では、この菌株はかなり高い頻度でspo+表現
型を要求する傾向があった。 さらに、菌株SMS272は、たとえばグリセリン中に維持さ
れる際には、菌株SMS275(pSM274)よりも生存度が低い
ことを示す。事実、グリセリンの添加後、培地1ml当た
り生存細胞の数は3.7×107 CFU/mlであり、7日後、こ
の値は73%に低下し、19日後では68%に、40日後では約
54%に低下した。これらの理由から、菌株SMS272は、工
業的発酵処理における使用には適していない。

Hind IIIで消化したpSM274コントロール、3:菌株SMS2
75(pSM274)から抽出したプラスミドDNA(未消化)、
4:菌株SMS275(pSM274)から抽出し、EcoRI及びHind
IIIで消化したプラスミドDNA、5:分子量基準である。
図2において、1−4:菌株spoIID SMS275(pSM274)
の2つのクローンから抽出されたプラスミドDNAであっ
て、1及び3は未消化のDNA、2及び4はEcoRI及びHind
IIIで消化したDNAであり;5−8;菌株spoIIA1 SMS27
0(pSM274)の2つのクローンから抽出されたプラスミ
ドDNAであって、5及び7は未消化のDNA、6及び8はEc
oRI及びHind IIIで消化したDNAであり;9は分子量基準
であり;10−13:菌株spoIIF96 SMS272(pSM274)の2
つのクローンから抽出されたプラスミドDNAであって、1
0及び12は未消化のDNA、11及び13はEcoRI及びHind III
で消化したDNAであり;14及び15:菌株spoOF SMS228(p
SM274)の1つのクローンから抽出されたプラスミドDNA
であって、14は未消化のDNA、15はEcoRI及びHindIIIで
消化したDNAである。図3は、菌株SMS275(pSM274)か
ら抽出されたタンパク質の12.5%ナトリウムドデシルポ
リアクリルアミドゲル(SDS−PAGE)における電気泳動
の結果を示す写真である。図中、Aは標準(部分的に精
製したhGH前駆体)であり;BはhGH前駆体をコード付け
る配列を含有しないコントロールプラスミド pSM214に
よって形質転換された菌株275から抽出されたタンパク
質であり;C−F:プラスミド pSM274によって形質転
換された菌株SMS275の4つのクローンから抽出されたタ
ンパク質である。以下の実施例は本発明を説明するもの
であり、これらに限定されない。
etics,(1979),167:251−258)に従い、トランスポゾ
ンTn917を含有するプラスミドDNA pTV5TS(1μg)を
使用してコンピテントB.サチリス SMS118細胞を形質
転換させた。ついで、細胞をVY最大培地(veal infusio
n)(veal infusion broth(DIFCO)25g/リットル、酵母エ
キス5g/リットル及び寒天(DIFCO)20g/リットル)20ml上
で平板培養し、該プレート上にエリスロマイシン 125μ
g/mlを含有するソフト寒天5mlを注加し、プレートを
37℃でさらに18時間インキュベートすることによって形
質転換体を選別した。エリスロマイシン耐性(Emr)形
質転換体(すなわち、染色体レベルで同種の組換えが行
われたもの)を、胞子を形成しない能力に関するテスト
に供した。この目的のため、Emrコロニーを、下記組成
を有するSchaeffer胞子形成培地(pH7.0)のプレート上
に移し、37℃で生育させた。 普通ブロス(DIFCO) 8.0 g/リットル KCl 1.0 g/リットル MgSO4 1.25×10-1g/リットル 寒天(DIFCO) 16.0 g/リットル MnCl2・4H2O 0.98×10-3 g/リットル FeSO4・7H2O 2.78×10-4 g/リットル Na2SO4 1.42×10-1 g/リットル H2O 1.0 リットル 数日後、光学顕微鏡での観察、及びコロニーの形態的変
成の実施によって胞子の形成を測定した。胞子を形成し
たコロニーは実際に象徴的な褐色に着色し、一方、無胞
子性のコロニーは透明のままであり、溶菌する傾向を示
した。上述の如く得られた結果から、無胞子性表現型
(spo-)を有するいくつかのEmr形質転換体を単離する
ことができた。spoII:D遺伝子内に変異を有するこれら
形質転換体の1つをSMS275と表示した。
r-、npr-、leu、pyrD1及びspo-の安定性をチェックする
ため検定を行った。詳述すれば、(i)生育培地におけ
る菌株のプロテアーゼ活性が低いこと(セリンプロテア
ーゼ遺伝子(apr)及びニュートラルプロテアーゼ遺伝
子(npr)が不活性化されたことを示す)、(ii)ロイシ
ン及びウラシルの非存在下、最小培地において菌株が生
育しないこと及び、(iii)非胞子形性能をチェックする
ためテストを行った。カゼイン1%を含有するVY最大培
地に菌株SMS272を平板培養することによりプロテアーゼ
活性を測定した(Tomaら、J.Bacteriol.,(1986),14
7:740−743)。セリンプロテアーゼ及びニュートラル
プロテアーゼの作用によるコロニーの周囲における特徴
的な加水分解暈輪の不存在は、これら2つの酵素が分泌
されていないことを示す。菌株SMS275から単離されたコ
ロニーはいずれも加水分解暈輪をもたない。しかしなが
ら、ロイシン(leu)及びウラシル(pyrD1)に関する栄
養要求については、P.YoungmanによりPlasmids:a pra
ctical approach,K.G.Hardy編,IRL Press,(198
6),79−103に記載された組成を有する最小培地(生育
ファクターを含まない)、leu(50μg/ml)のみ含有す
る最小培地、ウラシル(50μg/ml)のみ含有する最小
培地、及び両方を含む最小培地で菌株を平板培養するこ
とによってコントロールテストを行った。両方の栄養フ
ァクターを含有する培地でのみ生育が観察された。これ
より、遺伝子性マーカー leu及びpyrD1の安定性が確認
された。最後に、細胞の総量に関連して生成する胞子の
量を測定し、同時に、エリスロマイシンによる選択的圧
力の不存在下におけるspo-表現型の安定性をチェックし
た。該分析は、胞子を生成し得ない菌株は短時間の高温
処理に対する抵抗性が小さいとの事実に基づくものであ
る。それぞれSchaeffer液状胞子形成培地10mlを収容す
るフラスコ(100ml)2個に、それぞれ菌株SMS275及び
胞子形成性菌株SMS181(コントロール)を接種し、初め
に37℃で24時間、ついで80℃で10分間インキュベートし
た。ついで、培養物の一定量を適当に希釈し、各希釈物
0.1mlを、抗生物質を補充したVY最大培地で平板培養し
た。室温で生育後、生存する細胞(CFU)を80℃での処
理の前後で計数した。
しないため、平板培地上で生育したコロニーは、37℃に
おける液体培地中での生育の間に生成し、最大培地上で
平板培養する際に発芽し得る胞子に由来するものであ
る。表1は上記テストの結果(CFU/mlで表示)を示
す。
に存在する細胞の数は、それぞれ1.9×108 CFU/ml及び
1×108 CFU/mlである。 ii)菌株SMS275がspo+表現型に逆戻りする頻度は1×10
-8より小であり、熱処理に対して生残るSMS275及びSMS1
18の百分率の間の比率は0.001%より小である。 iii)37℃、24時間後に生成され、最大培地で発芽し得
る胞子の量は、胞子形成性菌株SMS118については1.4×1
05であり、非胞子形成性菌株SMS275については0であ
る。 液状培地中、37℃での培養をエリスロマイシンの不存在
下で行っているのであるから、80℃における熱処理後に
SMS275コロニーが全く存在しないことは、菌株SMS275に
よって獲得されたspo-特性が非常に安定であることを示
す。実際、細胞はトランスポゾンを失っていると思われ
るが、無胞子性表現型を保持している。
M274(10ng)で形質転換させ、ついで、カナマイシン5
μg/ml及びクロラムフェニコール5μg/mlを含有す
るVY最大培地のプレート上、37℃で18時間培養して形質
転換体を選別した。Recombinant DNA Techniques:an i
ntroduction[Rodriguez及びTait編,Addison−Wesley
Publishing Company,(1983),164]に記載された急速
抽出法によって耐性コロニーの1つから単離したプラス
ミドDNAの一定量を、制限酵素EcoRI及びHind III(BR
L)により、該酵素の供給者の情報に従って消化した。
ついで、消化したプラスミドDNA 2μリットル及び未消化の
同じDNA 2μリットルを0.8%アーガロースゲル上に負荷
し、同様にプラスミド pSM274(コントロール)(2μリット
ル)(前記のものと同じ酵素で処理したもの及び処理して
いないもの)及びいくつかの分子量基準を負荷した。図
1に示す結果は、B.サチリス SMS275クローンから単
離し、制限酵素で消化したプラスミド pSM274が予想さ
れる移動速度を示し、pSM274のものと一致する消化パタ
ーンを示すことを表した。実際、6700bp及び800bpの長
さ(それぞれプラスミドベクター及びヒト生長ホルモン
をコード付けるインサートに相当)を有する2つのフラ
グメントに相当する2つのバンドを、脱色後、臭化エチ
ジウムで溶解させた。菌株B.サチリス SMS275(pSM27
4)のhGH前駆体を生成する能力を、該菌株をVY培地10ml
中、37℃で18時間生育させ、溶解させた細胞から抽出さ
れた可溶性の総タンパク質を12.5%SDS−PAGE上で電気
泳動させ、クマシーブルーで染色することによって分析
してチェックした。タンパク質抽出物中におけるヒト生
長ホルモン前駆体に相当するバンドの存在が図3に見ら
れる。
の形質転換 それぞれspoIIF96、spoIIA1及びspoOF変異を有する無胞
子性菌株B.サチリスSMS268、SMS270及びSMS272をプラ
スミド pSM274で形質転換させ、ついでプラスミドの安
定性、存在するプラスミドのコピーの数及びspo-表現型
の安定性をチェックするためテストを行った。図2から
見られるように、spoOF変異を有する菌株においてはプ
ラスミド pSM274は不安定であり、spoIIA1変異を有する
菌株内に存在するプラスミドのコピーの数は菌株SMS118
内よりも少ない。これに対して、spoIIF96菌株はプラス
ミドを安定状態で保持しており、そのコピー数は菌株SM
S118の場合に匹敵するものである。しかしながら、spo-
の安定性をチェックするためにテストした際、この菌株
は顕微鏡観察によって行った評価では、かなり高い頻度
でspo-表現型を要求する傾向を示した。
で4つの細胞ブランクを調製した。実際には、菌株SMS2
75(pSM274)及び菌株SMS272(pSM274)の予培養物(10
0ml)2つを下記処方のTYM培地中で調製した。 トリプトン 13g/リットル 酵母エキス 3g/リットル マルトース 40g/リットル カナマイシン 5mg/リットル クロラムフェニコール 5mg/リットル フラスコを220rpmで撹拌し、37℃で24時間インキュベー
トした。それぞれTYM培地1リットルを収容する2つの発酵
槽(2リットル)に、予培養物の1つを100ml接種した(初
期光学密度(O.D.):660nmにおいて0.190)。空気を0.
5v/v/分で充填しながら、800rpm、pH7.0で発酵を行っ
た。O.D.3.0まで15時間発酵を行った後、培養物を遠
心処理し、細胞を回収した。ついで、サンプル2mlにグ
リセリンを最終濃度15%となるまで補充し、−80℃に維
持した。調製の日から6カ月後、グリセリン内における
サンプル中の生存細胞(コロニー形成単位−CFU)の量
を測定した。このテストは、グリセリン中のサンプルの
一定量を培地で希釈し、直ちに希釈物0.1mlを寒天を含
有し、抗生物質(カナマイシン及びクロラムフェニコー
ル)を含有又は含有しないTBAB最大生育培地上で平板培
養することによって実施される。SMS275(pSM274)につ
いては、グリセリン中のサンプル1ml当たりの生存細胞
数は2.4×108 CFU/mlであった。該分析を抗生物質(カ
ナマイシン及びクロラムフェニコール)の存在下又は不
存在下で実施した。これらの結果は、利用した保存条件
下においてSMS275細胞の安定性及び生存率が良好である
ことを示した。しかしながら、SMS272(pSM274)につい
て得られたCFU値は生存率が低いことを示した。実際の
ところ、グリセリンの添加後では、3.7×107 CFU/mlの
値が測定された。7日後、この値は73%に低下し、19日
後では68%に、40日後では約54%に低下した。菌株SMS2
74(pSM274)の培養物についても、実施例2及び3に記
載の如く分析したところ、プラスミドは安定であり、hG
H前駆体をコード付ける遺伝子の配列が無傷であること
を示した。

らの前駆体を製造する方法において、バシラス属菌内で
発現可能であり、前記異質ポリペプチド又はタンパク質
又はこれらの前駆体をコード付ける遺伝子を含有するベ
クターによって形質転換させた請求項1記載の菌株バシ
ラス・サチリス SMS275を、炭素源、窒素源、ミネラル
塩、ロイシン及びウラシルを含有する培地中で生育さ
せ、遺伝子発現生成物を回収することを特徴とする、異
質ポリペプチド及びタンパク質又はこれらの前駆体の製
法。 - 【請求項8】請求項7記載の製法において、前記バシラ
ス・サチリスがCBS433.90である、異質ポリペプチド及
びタンパク質又はこれらの前駆体の製法。
