本明細書では、化合物Y

メチル(R)-4-((6-クロロ-1-(ヒドロキシメチル)-1,2,3,4-テトラヒドロナフタレン-1-イル)メトキシ)-3-ニトロベンゾエート(化合物Y2、式中、R1はCO2Meである):EasyMax反応容器(ガラス製、100mL)に、[6-クロロ-1-(ヒドロキシメチル)テトラリン-1-イル]メタノール(THN-DIOL、3.00g、13.2mmol,1.0当量)、メチル4-フルオロ-3-ニトロベンゾエート(2.64g、13.2mmol、1.0当量、ここで、LgはFであり、R1はメチルである)及び(1R,2S)-N-エチル-1-ヒドロキシ-N,N-ジメチル-1-フェニルプロパン-2-アミニウムブロミド(11、1.43g、3.31mmol、0.25当量)を順次投入した。容器をEasyMax Advanced Synthesis Workstationに入れ、オーバーヘッド撹拌機、窒素(N2)入口ライン、温度プローブ、及び混合を助けるためのバッフルを備えた6ポート反応器ヘッドで密閉した。反応器にトルエン(82.5mL、27.5容積、THN-DIOLに対して0.15M)を入れ、残りのポートはゴム隔膜で封止した。反応混合物を撹拌し(オーバーヘッド、800rpm)、これによりオフホワイト色のわずかに不均一な溶液(微粒子状)を得た。反応混合物を、内部温度が-17.0℃に達するまで冷却した(ジャケットの温度は-20.0℃であった)。この時、炭酸セシウム水溶液(4.54mL、13.2mmol、1.0当量、50wt%の水溶液)を、隔壁を介してシリンジにより反応混合物に添加した(滴下、2分)。塩基を添加すると、反応混合物は淡黄色に変化した。この溶液を16時間撹拌した(ジャケットの温度は-20.0℃であった)。この後、撹拌を止め、混合物を温め(5℃)、黄色の反応混合物を分液漏斗(250mL)に移した。この溶液のアリコートをLC分析用に取り出した。水(30mL、10容積)を分離漏斗に加え、混合物を穏やかに振とうし、通気させた。二相性混合物を沈降させ(10分間)、相を分離した。両相のLC分析後、有機層をロータリーエバポレータで濃縮し、濃い黄色の油状物を得た。この粗反応混合物をシリカゲル上で自動カラムクロマトグラフィー(Biotage、340gカラム、ヘプタン勾配中の5~40%EtOAc、16カラム容量)で精製し、R1がCO2Meである化合物Y2をわずかに黄色のガラス状固体(0.930g、収率17.3%、54%ee)として得た。
メチル(S)-4-((6-クロロ-1-(ヒドロキシメチル)-1,2,3,4-テトラヒドロナフタレン-1-イル)メトキシ)-3-ニトロベンゾエート(化合物Y1、ここでR1はCO2Meである):バイアルに、N-[4-(トリフルオロメチル)ベンジル]シンコニウムブロミド(0.10当量、6.7mg)、[6-クロロ-1-(ヒドロキシメチル)テトラリン-1-イル]メタノール(THN-DIOL,1.0当量、23mg)、メチル4-フルオロ-3-ニトロベンゾエート(1.0当量、20mg、LgはFであり、R1はメチルである)及びCHCl3(167μL)を投入した。バイアルを+1℃に冷却し、炭酸セシウム(50wt%溶液、1当量)を投入した。バイアルを16時間振とうし、1N HCl(50μL)でクエンチした。HPLC分析は、表題化合物(Y1)が32%の変換率、32%eeで形成されることを示した。

- Ar3はフェニルである、請求項24に記載の方法。
- Zは、ハライド、トリフレート、メシレート、トシレート、又はノシレートである、請求項12~28のいずれか一項に記載の方法。
- Zはクロリド又はブロミドである、請求項29に記載の方法。
- 前記溶媒は無機塩基を含む、請求項1~30のいずれか一項に記載の方法。
- 前記塩基は、Cs2CO3、K2CO3、Rb2CO3、Na2CO3、Na2CO3、Li2CO3、CaCO3、MgCO3、K3PO4、Na3PO4、Li3PO4、K2HPO4、Na2HPO4、Li2HPO4、NaHCO3、LiHCO3、又はKHCO3を含む、請求項31に記載の方法。
- 前記塩基は、Cs2CO3を含む、請求項32に記載の方法。
- 前記無機塩基は、化合物(I)の1.0モル当量を基準として、0.95~6モル当量で存在する、請求項31~33のいずれか一項に記載の方法。
- 前記二相性溶媒系は非プロトン性有機溶媒及び水を含む、請求項1~34のいずれか一項に記載の方法。
- 前記非プロトン性有機溶媒は、ジクロロメタン、テトラヒドロフラン、トルエン、ベンゼン、シクロペンチルメチルエーテル、tert-ブチルメチルエーテル、2-メチルテトラヒドロフラン、アニソール、キシレン、ベンゾトリフルオリド、1,2-ジクロロエタン、メチルイソブチルケトン、酢酸エチル、酢酸イソプロピル、又はこれらの組み合わせを含む、請求項35に記載の方法。
- 前記非プロトン性有機溶媒は、トルエンを含む、請求項36に記載の方法。
- 前記トルエンは、化合物(I)の重量を基準として、3L/kg~30L/kgの濃度で存在する、請求項37に記載の方法。
- 前記塩基を添加する前に、化合物(I)、化合物(II)及び前記触媒を前記非プロトン性有機溶媒中で混和することを含む、請求項35~38のいずれか一項に記載の方法。
- 前記混和は、-40℃~30℃の温度で行う、請求項1~39のいずれか一項に記載の方法。
- 前記混和は、-15℃~-25℃の温度で行う、請求項40に記載の方法。
- 前記混和は、1時間~72時間にわたり行う、請求項1~41のいずれか一項に記載の方法。
- 前記混和は、14時間~18時間にわたり行う、請求項42に記載の方法。
- 化合物(II)は、化合物(I)の1.0モル当量を基準として、0.9~2モル当量で存在する、請求項1~43のいずれか一項に記載の方法。
- 化合物(II)は、化合物(I)の1.0モル当量を基準として、1.0モル当量で存在する、請求項44に記載の方法。
- 前記不斉触媒は、化合物(I)の1.0モル当量を基準として、0.005~1.50モル当量で存在する、請求項1~45のいずれか一項に記載の方法。
- 前記不斉触媒は、化合物(I)の1.0モル当量を基準として、0.25モル当量で存在する、請求項46に記載の方法。
- 化合物Yは、30%以上のエナンチオマー過剰率で生成される、請求項1~47のいずれか一項に記載の方法。
- 化合物Yは、40%以上のエナンチオマー過剰率で生成される、請求項48に記載の方法。
- 化合物Yは、50%以上のエナンチオマー過剰率で生成される、請求項49に記載の方法。
