2、前記抗体は、
(1)配列番号1若しくは2に示される重鎖相補性決定領域1(CDR-H1)、及び/または配列番号3若しくは4に示される重鎖相補性決定領域2(CDR-H2)、及び/または配列番号5若しくは6に示される重鎖相補性決定領域3(CDR-H3)を含む重鎖可変領域を含む抗体;
(2)配列番号7若しくは8に示される軽鎖相補性決定領域1(CDR-L1)、及び/または配列番号9若しくは10に示される軽鎖相補性決定領域2(CDR-L2)、及び/または配列番号11若しくは12に示される軽鎖相補性決定領域3(CDR-L3)を含む軽鎖可変領域を含む抗体;
(3)(1)に記載の抗体の重鎖可変領域及び(2)に記載の抗体の軽鎖可変領域を含む抗体;
(4)(1)~(3)のいずれか1項に記載の抗体のバリアントであり、(1)~(3)のいずれか1項に記載の抗体と同等または類似の活性を有する抗体
から選択されるいずれかである、項1に記載のモノクローナル抗体。

(1)配列番号13もしくは14示されるアミノ酸配列または前記配列のバリアントを含む軽鎖可変領域を含む抗体;
(2)配列番号15もしくは16示されるアミノ酸配列または前記配列のバリアントを含む重鎖可変領域を含む抗体;
(3)(1)に記載の抗体の重鎖可変領域及び(2)に記載の抗体の軽鎖可変領域を含む抗体
から選択されるいずれかであることを特徴とする、項1または2に記載の抗体。

原材料の調製
本実施例では、BCMA抗原とそのリガンドをACRO Biosystemsから購入した。同時に、GlaxoSmithKline(GSK)の陽性抗体GSK2857916を米国特許出願(US20140105915A)に従って調製した。なお、調製された上記陽性抗体は毒素分子にカップリングしていないが、他の可能な機能には影響はない。
本願では、human-BCMA-Fc、human-BCMA-His、cyno-BCMA-Fc、及びcyno-BCMA-Hisの4つの抗原が使用され、いずれもACRO Biosystemsから購入したものであり、カタログ番号はそれぞれBC7-H5254、BCA-H522y、BCA-C5253、BCA-C52H7である。さらに、その活性はGSK2857916と相互検証されており、米国特許出願US20140105915Aに開示されているものと一致しており、結果を図2Aに示す。
本願で使用されるリガンドは、human-BAFF-Fcとhuman-APRIL-Fcの2種類が使用され、どちらも、ACRO Biosystemsから購入され、カタログ番号はそれぞれBAF-H4268、APL-H5267である。さらに、その活性は、human-BCMAとの受容体リガンド結合について検証されており、米国特許出願US20140105915Aに開示されているものと一致しており、結果を図2Bおよび2Cに示す。
本願では、陽性対照抗体GSK2857916は、一過性トランスフェクションシステム(ExpiCHO)を使用して発現させ、使用した主な材料は、Gibco培地(カタログ番号A29100-01)、Gibcoトランスフェクションキット(カタログ番号A29129)を含む。まず、GSK2857916の軽鎖配列(配列番号17の配列に示される)および重鎖配列(配列番号18の配列に示される)を、米国特許出願US20140105915Aに開示されている配列に従って合成し、完全なGSK2857916抗体軽鎖および重鎖遺伝子を含むプラスミドを分子クローニングによって構築した。GSK2857916抗体の軽鎖を含むプラスミドと重鎖を含むプラスミドを2:1の質量比で混合し、25mLの発現系で、上記のプラスミド混合物(25μg)を標準的な手順に従ってトランスフェクション試薬を混合して25mLのExpiCHO細胞発現システムに滴下した。完全に混合した後、37℃の細胞インキュベーター内で18~22時間発現させた。続いて、上記のトランスフェクション混合物に補充培地を添加し、32℃の細胞インキュベーターに入れて培養を続けた。トランスフェクション後5日目に、2回目の補充フィードを追加し、細胞を32℃の細胞インキュベーターにさらに10~12日間培養した。そして、発現した細胞懸濁液を高速遠心分離し、上澄みを取り、得られた上澄みを0.22μmフィルターメンブレンでろ過し、Protein A/Gアフィニティークロマトグラフィーカラムアフィニティー法で精製した。精製後、目的タンパク質を100mMグリシネート(pH3.0)で溶出し、濃縮、置換、分注し、SDS-PAGE同定及び活性同定に合格した後、凍結保存のために倉庫に保存した。
細胞株の調製
本実施例では、BCMAを発現するヒト骨髄腫細胞株H929を購入し、ヒトBCMAを過剰発現する細胞株BCMA-HEK293細胞およびサルBCMAを過剰発現する細胞株BCMA-CHO細胞を構築した。
本願では、H929は北京協和セルバンクから購入され、カタログ番号は3111C0001CCC000360であり、これは、ヒトBCMAを自然に高度に発現する骨髄腫細胞株である。
2.2.1 全長サルBCMAを発現するプラスミドの構築
カニクイザルのBCMAタンパク質を含むDNAフラグメントを、遺伝子合成技術によって合成し、発現ベクターにクローニングした。形質転換法により大腸菌に導入し、大腸菌の単一クローンを採取して配列決定し、正しいプラスミドクローンを得た後、プラスミドを抽出し、再度配列決定して確認した。
CHO-s細胞は、GibcoのCD-CHO無血清培地(カタログ番号10743029)を使用して培養した。エレクトロポレーションの1日前に細胞を5×106/mLで継代し、翌日、Invitrogenのエレクトロポレーションキット(カタログ番号:MPK10096)およびエレクトロポレーター(カタログ番号:MP922947)を使用して、構築されたプラスミドをCHO-s細胞に導入した。エレクトロポレーション後の細胞をCD-CHO培地に移し、37℃の細胞インキュベーターにおいて48時間培養した。
エレクトロポレーションしたCHO-s細胞を2000細胞/ウェルで96ウェルプレートにプレーティングし、最終濃度30μM MSX(Millipore、GSS-1015-F)およびGSサプリメント(Sigma、58672C-100ml)を添加し、37℃の二酸化炭素インキュベーター内で培養し、10日後に30μM MSXおよび1×GSサプリメントを含む培地を補充した。
96ウェルプレートで増殖した単一細胞クローンを選び、24ウェル培養プレートに移してさらに増殖させた後、サルBCMAが安定的にトランスフェクトされた細胞株をFACSで同定した。
2.3.1 全長ヒトBCMAを発現するプラスミドの構築
ヒトBCMAタンパク質を含むDNAフラグメントを、遺伝子合成技術によって合成し、発現ベクターにクローニングした。形質転換法により大腸菌に導入し、大腸菌の単一クローンを採取して配列決定し、正しいプラスミドクローンを得た後、プラスミドを抽出し、再度配列決定して確認した。
HEK293細胞は、GibcoのDMEM無血清培地(カタログ番号12634010)を使用して培養した。エレクトロポレーションの1日前に細胞を2×105/mLで継代し、翌日、Invitrogenのエレクトロポレーションキット(カタログ番号:MPK10096)およびエレクトロポレーター(カタログ番号:MP922947)を使用して、構築されたプラスミドをHEK293細胞に導入した。エレクトロポレーション後の細胞をDMEM培地に移し、37℃の細胞インキュベーターにおいて48時間培養した。
エレクトロポレーションしたHEK293細胞を1000細胞/ウェルで96ウェルプレートにプレーティングし、最終濃度2μg/mlのpuromycinを添加し、37℃の二酸化炭素インキュベーター内で培養し、14日後に2μg/mlのpuromycinを含む培地を補充した。
96ウェルプレートで増殖した単一細胞クローンを選び、24ウェル培養プレートに移してさらに増殖させた後、ヒトBCMA安定的にトランスフェクトされた細胞株をFACSで同定した。
動物の免疫
本実施例では、動物の免疫化にはいくつかの計画が含まれ、同時に実行される。免疫化プロセス中に、力価検出のためにマウス血清を採取し、免疫化の効果を評価し、最後にライブラリーを構築するマウスの順番を血清力価に従って決定した。
3.1.1. 免疫計画1
Babl/cマウス(上海霊暢バイオテクノロジー株式会社、雌、6~8週間、n=2)を皮下注射および腹腔内注射により免疫し、材料は上記のヒトBCMA-FcおよびサルBCMA-Fc抗原タンパク質(ACRO Biosystems)を含み、交差免疫を使用した。最初の免疫化用量は100μg/匹で、CFA(フロイント完全アジュバント)を添加し、次に免疫化用量を50μg/匹に減らし、IFA(フロイント不完全アジュバント)を添加し、両方とも2週間ごとに交差免疫を1回した。対応するマウスには、Mouse 1およびMouse 2の番号が付けられた。
Babl/cマウス(マウスは計画1と同じバッチ、n=2)を皮下注射および腹腔内注射によって免疫し、材料は上記のヒトBCMA-Fc、サルBCMA-Fc抗原タンパク質(ACRO Biosystems)、ヒトBCMAを高度に発現するHEK293細胞、およびサルBCMAを高度に発現するCHO細胞を含み、交差免疫を使用した。タンパク質免疫の用量は20μg/匹であり、細胞免疫の用量は1×107/匹であり、腹腔内注射し、1週間ごとに交差免疫を1回した。対応するマウスには、Mouse 3とMouse 4の番号が付けられた。
3.2.1 抗原のコーティングとブロッキング
ELISAプレート上で、ヒトBCMAおよびサルBCMAを1日前にそれぞれコーティングし、コーティング濃度は2μg/mL、30μL/ウェルであり、4℃で一晩した。免疫力価測定の日に、プレートをPBSTで3回洗浄し、5%脱脂乳で室温、2時間ブロックし、次にプレートをPBSTで3回洗浄した。
各血清サンプルを最初にストック(原液)に基づいて500倍に希釈し、次に3倍の勾配で希釈し、一次抗体としてELISAプレートに添加し、室温、1時間インキュベートし、プレートをPBSTで3回洗浄した後、二次抗体(Goat-anti-mouse-IgG-Fab-HRP、Sigma、M4115)を添加し、室温、1時間インキュベートした。
インキュベーション後、プレートをPBSTで6回洗浄し、TMB(SurModics、TMBS-1000-01)を加えて発色させた。発色結果に応じて、2M HClを加えて反応を終止させた。プレートは、マイクロプレートリーダー(Molecular Devices、SpectreMax190)によってOD450で読み取った。血清力価の結果は、免疫化されたマウスがヒトBCMAおよびサルBCMAに対して高い力価を示し、次の抗体ライブラリー構築のステップに使用できることを示している。結果を以下の表1-1および1-2に示す。
本実施例では、実施例3で最高の力価を有するマウスを好ましく選択し、マウス脾臓中のB細胞の抗体遺伝子を、ファージディスプレイ技術を使用してファージディスプレイベクターにクローン化し、抗体ライブラリーを構築した。スクリーニング抗原としてヒトBCMAおよびサルBCMAを使用し、ライブラリー大規模選択、モノクローナル一次スクリーニングおよび配列決定などのプロセスを通じて、ヒトおよびサルBCMA抗原に結合活性を有する17の抗体分子を最終的に得た。
免疫化後、安楽死のための標準的な手順に従ってマウスを処理した。マウスの脾臓を採取し、粉砕し、濾過して脾臓細胞を収集し、1mLのTRIzol(商標)試薬(Thermo Fisher、15596026)を加えて脾臓細胞を溶解し、フェノール・クロロホルム法によって全RNAを抽出した。抽出したRNAを逆転写キット(TaKaRa、6210A)によってcDNAに逆転写した。次に、cDNAをPCRテンプレートとして使用し、マウス抗体配列の増幅に特定のプライマーを使用して、抗体の軽鎖遺伝子と重鎖遺伝子をそれぞれ増幅した。最後に、NcoI+NotI二重消化とT4リガーゼを介して、抗体遺伝子フラグメントをファージディスプレイベクターに挿入し、ライゲーション産物をDNA回収キット(Omega、D6492-02)で回収し、最後にエレクトロポレーター(Bio-Rad、MicroPulser)によってコンピテントな大腸菌SS320(Lucigen、MC1061F)に形質転換し、アンピシリンとテトラサイクリンを含む2-YT(C+/K+2-YT)固体プレートに広げて、正しく形質転換された抗体プラスミドのSS320株を増幅し、最後にFabフラグメント抗体配列を含むライブラリーを構築した。
4.2.1 磁気ビーズ法によるファージディスプレイ抗体遺伝子ライブラリーのスクリーニング
磁気ビーズスクリーニングは、Biotin標識抗原タンパク質とAvidin結合磁気ビーズの結合に基づいて、抗原結合磁気ビーズとライブラリーのインキュベーション、洗浄、溶出のプロセスを通じて、2~4ラウンドの大規模選択後、最後に抗原に対する特異的モノクローナル抗体を濃縮するプロセスである。本実施例では、Biotinで標識されたヒトBCMA抗原とサルBCMA抗原の交差選択の原理を使用し、1回目と3回目はヒトBCMA選択を使用し、2回目はサルBCMA選択を使用し、合計3回の大規模選択をした。次に、濃縮された抗体配列の混合物を、ヒトおよびサルのBCMAの一次モノクローナルスクリーニングに供した。
まず、Avidin結合磁気ビーズをBiotin標識ヒトBCMAタンパク質とインキュベートして、ヒトBCMAタンパク質を磁気ビーズに結合させた。BCMA抗原と結合した磁気ビーズおよび構築されたファージライブラリーを室温、2時間インキュベートした。PBSTで6~8回洗浄した後、非特異的に吸着したファージを除去し、Trypsin(Gibco、25200072)を加えて軽く混合し、20分間反応させて、特異的に結合した抗体ディスプレイファージを溶出した。続いて、対数期SS320菌体(Lucigen、MC1061 F)を、溶出したファージに感染させ、30分間静置した後、220rpmで1時間インキュベートした後、VSCM13ヘルパーファージを添加して30分間静置した。続いて、220rpmで1時間培養し、遠心分離してC+/K+2-YT培地に置換し、最終的に得られたファージを次の大規模選択に使用した。
免疫試験管スクリーニングの原理は、BCMAタンパク質を高い吸着力を有する免疫試験管の表面にコーティングし、ファージディスプレイ抗体ライブラリーを免疫試験管に加え、免疫試験管の表面に吸着した抗原タンパク質とインキュベート、洗浄、溶出する大規模選択プロセスによって、2~4ラウンドの大規模選択後、最終的に抗原に対する特異的モノクローナル抗体を濃縮するプロセスである。免疫試験管法と磁気ビーズ法の目的は、抗原に対する特異的抗体を濃縮することであり、これらは2つの補完的な実験方法である。本実施例では、ヒトBCMA抗原とサルBCMA抗原の交差選択の原理を使用し、1ラウンド目と3ラウンド目はヒトBCMAによる選択を使用し、2ラウンド目はサルBCMAによる選択を使用し、合計3ラウンドの選択を行った。そして、濃縮された抗体配列の混合物を、ヒトおよびサルのBCMAの一次モノクローナルスクリーニングに使用した。具体的な実施方法は、スクリーニング用の磁気ビーズ法と同様である。
3ラウンドのスクリーニングの後、第3ラウンドのプールからいくつかのモノクローナルクローンを選択して、ヒトBCMAおよびサルBCMAへの結合を含むELISA検出をした。最終的に、1344個のクローンからヒトとサルBCMAの両方に結合できる合計93個の陽性クローンを選択した。配列決定分析と、ヒトおよびサルのBCMAによるアフィニティーシーケンス結果の分析の後、最後に、次の実験のために全長を構築するために、17個のクローンの配列を選択した。
全長抗体の構築、発現および精製
この実施例では、実施例4で得られた17のヒト-サル交差Fab抗体をヒトIgG1タイプに構築し、ここで、軽鎖はいずれもKappaであり、抗体タイプはヒト-マウスキメラ抗体である。
スクリーニングで得られた配列の重鎖配列をヒトIgG1 Fcセグメントと融合して構築し、軽鎖をヒトKappa定常領域と融合して構築した。重鎖と軽鎖のプラスミドをExpiCHO細胞に形質転換し、発現を誘導して全長抗体を得た。
本願では、抗体はExpiCHO一過性発現システムを採用し、培地は(Gibco、A29100-01)、トランスフェクションキットは(Gibco、A29129)である。具体的な方法は次のとおりである。トランスフェクションの1日前にExpiCHO細胞を継代し、構築したプラスミド25μgとトランスフェクション試薬を25mLシステムで混合し、25mL ExpiCHO細胞に滴下し、十分に混合した後、37℃細胞インキュベーターで18~22時間発現させた。続いて、上記のトランスフェクション混合物に補充培地を添加し、32℃の細胞インキュベーターに入れて培養を続けた。トランスフェクション後5日目に、2回目の補充フィードを追加し、細胞を32℃の細胞インキュベーターにさらに10~12日間培養した。そして、発現した細胞懸濁液を高速遠心分離し、上澄みを取り、得られた上澄みを0.22μmフィルターメンブレンでろ過し、Protein A/G アフィニティークロマトグラフィーカラムアフィニティー法で精製した。精製後、目的タンパク質を100mMグリシネート(pH3.0)で溶出し、濃縮、置換、分注し、SDS-PAGE同定、SEC純度測定、及び活性同定を経て、凍結保存のために倉庫に保存した。
候補抗体ELISAレベルの親和性ブロッキング効果の検出
本実施例では、ELISAの方法により、ヒトおよびサルBCMAに対する候補抗体の親和性効果を検証し、ELISAの方法により、BAFFまたはAPRILへのBCMAの結合のブロッキングに対する候補抗体の効果を検証した。
96ウェルELISAプレート上で、ヒトBCMAとサルBCMAをそれぞれ2μg/mL、30μL/ウェルでコーティングし、4度で一晩放置した。翌日、ウェルプレートをPBSTで3回洗浄し、5%脱脂乳で2時間ブロックした。プレートをPBSTで3回洗浄した後、段階希釈した候補抗体と陽性対照抗体(GSK2857916)を添加し、1時間インキュベートした。その後、PBSTで3回洗浄した後、二次抗体(anti-human-IgG-Kappa-HRP、abcam、ab79115)を添加し、1時間インキュベートした。インキュベーション後、プレートをPBSTで6回洗浄し、TMB(SurModics、TMBS-1000-01)を加えて発色させた。発色結果によって、反応を終止するために2M HClを添加し、マイクロプレートリーダー(Molecular Devices、SpectreMax 190)によってプレートをOD450で読み取った。その結果を図4Aと図4Bに示す。その結果は、候補抗体が、ヒトBCMAおよびサルBCMAに対するGSK2857916抗体に近いまたはそれ以上の親和性を有していることを示している。

候補抗体のFACSレベルのヒトおよびサルBCMA発現細胞株との親和性効果の検出
本実施例では、ヒトおよびサルBCMAを発現する細胞に対する候補抗体の親和性効果をFACS法に基づいて検証した。
指数増殖期のヒトBCMA-HEK293細胞を収集し、300gで遠心分離して上澄みを除去し、細胞を調製したFACSバッファーに再懸濁してカウントし、細胞懸濁液の密度を2×106/mLに調整した。続いて、BCMA-HEK293細胞を96ウェル丸底プレートにウェルあたり100μLで添加し、300gで遠心分離して上澄みを除去した。候補抗体の段階希釈液と陽性対照抗体希釈液を対応するウェルに加え、マルチチャンネルピペットで細胞を均一にブローし、4℃で30分間インキュベートした。インキュベートした細胞混合液を300gで遠心分離して上澄みを除去し、200μLのFACS緩衝液を対応するウェルに加え、マルチチャンネルピペットを使用して細胞を再懸濁した。2回繰り返し、300gで遠心分離して上澄みを除去した。PE標識されたanti-human-IgG-Fcフローサイトメトリー用抗体(Abeam、98596)を添加し、細胞をマルチチャンネルピペットで均一にブローし、4℃で30分間インキュベートし、300gで遠心分離して上澄みを除去した。続いて、FACS緩衝液を添加し、細胞を再懸濁した。2回繰り返した後、FACS緩衝液をウェルに200μL/ウェルで添加し、細胞を再懸濁した。最後に、フローサイトメーター(Beckman、CytoFLEX AOO-1-1102)によって検出された。本実施例では、結果を図3Aに示す。その結果は、抗体SY14-3rd-5-6-7のヒトBCMA-HEK293細胞に対する親和性が、陽性対照抗体(GSK2857916)よりも優れていることを示している。
本実施例では、操作プロセスは、細胞がサルBCMA-CHO細胞である以外、7.1とまったく同じである。その結果を図3Bに示す。その結果は、抗体SY14-3rd-5-6-7のサルBCMA-CHO細胞に対する親和性が、陽性対照抗体(GSK2857916)の親和性よりも優れていることを示している。また、ヒトとサルのシグナルの違いは、BCMA-HEK293またはBCMA-CHOでのBCMAの発現の違いが原因であると推測される。
候補抗体のエンドサイトーシス効果の検出
本実施例では、ヒト骨髄腫細胞株H929細胞に対する抗体のエンドサイトーシス効果を検出する方法システムを開発し、それに応じて候補抗体のエンドサイトーシス効果をテストした。その基本原理は、細胞毒性によって抗体のエンドサイトーシス効果を検出することである。Fab-ZAP(Atsbio、IT-51-100)は、タンパク質合成を阻害して細胞死を引き起こし得るリボソーム阻害剤であるsaporin(サポニン)に結合したFabフラグメントである。本実施例では、Fab-ZAPはヒト抗体Fcに結合できるFabフラグメントである。Fab-ZAPと抗BCMA抗体を共培養した後、抗体に毒素がロードされる。抗BCMA抗体がH929細胞によってエンドサイトーシスされると、毒素は抗体とともに細胞に入り、細胞を死滅させる。したがって、抗体のエンドサイトーシス効果は、MTSキット(Promega、G3580)によって細胞の活性を検出することで検出できる。
これまでのところ、候補抗体のすべての検出結果を図7にまとめている。
抗体のヒト化操作
本実施例では、マウス抗体の重鎖および軽鎖V領域のフレームワークに対してアミノ酸点突然変異を実行して、それらをヒトGermlineに近づけた。それらの中で、操作された候補抗体は、SY14-3rd-5-6-7(5-6-7または5-6-7-WTとも呼ばれる)およびSY14-3rd-5-6-32(5-6-32または5-6-32-WTとも呼ばれる)を含み、操作された好ましい候補抗体は、それぞれ5-6-7-hu-2および5-6-32-hu-2と呼ばれる。
ヒト化抗体の機能検証
10.1 ヒト化前後のヒトおよびサルBCMAに対する2つの候補抗体の親和性のELISAによる検出
具体的な操作方法は実施例6を参照されたい。結果を図8A、図8B、図9A、図9Bに示す。その結果は、ヒト化前後の抗体が、ヒトおよびサルBCMA結合活性と基本的に同じであることを示している。
具体的な操作については、実施例6を参照されたい。結果を図10Aおよび11Aに示す。その結果は、BCMAおよびBAFFの結合をブロッキングするヒト化抗体の効果が陽性抗体の効果よりもわずかに悪いことを示している。

本実施例では、2つの候補抗体のヒト化前後のヒト骨髄腫細胞株H929細胞に対するエンドサイトーシス効果もテストされた。具体的な操作については、実施例8を参照されたい。結果を図13Aおよび13Bに示す。その結果は、ヒト化抗体5-6-7-huV2のエンドサイトーシス効果がヒト化前(5-6-7-WT)と比較して部分的に改善され、ヒト化前後の他の抗体のヒト骨髄腫細胞株H929細胞に対するエンドサイトーシス効果が基本的に同じであり、すべて陽性対照抗体(GSK2857916)よりも優れていることを示している。
ヒト化前後の候補抗体のDSF検出
本実施例では、ヒト化前後の2つの候補抗体と陽性対照抗体(GSK2857916)の熱安定性データをテストした。具体的なプロセスは次のとおりである。抗体溶液を調製し、0.25mg/mL、19μL/ウェルで、各テストサンプルに3つの並列ウェルを設定し、PBSとIPIを参照として使用した。次に、1μLのSYPRO оrange色素を100倍の濃度で各ウェルに加え、ロードの準備をした。テストにはABI7500 FAST RT-PCR装置を使用し、テストタイプとして融解曲線を選択し、連続モードを使用し、スキャン温度範囲を25~95℃、加熱速度を1%、25℃で5分間平衡した。加熱プロセス中にデータを収集し、レポーター基をROX、クエンチャー基をNone、反応用量を20μLにし、融解曲線の一次導関数の最初のピークと谷に対応する温度を、候補抗体の変性温度として確定した。結果を表1-3に示す。
ヒト化前後の候補抗体の親和性検出
本実施例では、Fortebio Octet RED96機器を使用して、ヒトBCMAおよびサルBCMAに対するヒト化前後の5-6-7および5-6-32の候補抗体の親和性を検出した。
1gのBSAを量り、500μLのTween 20を測定し、1000mLの1×PBSに加えてよく均一に混合した。フィルタリング後、分注して保存した。0.1mLの0.1Mグリシン溶液(pH=2.0)をピペットして0.9mLの超純水に加え、均一に混合した。抗体をKB緩衝液で10μg/mLに希釈し、抗原をKB緩衝液で希釈して、200、50、12.5、および0nMの一連の濃度勾配をこの順で希釈した。
センサー(sensor)(Anti-human Fba-CH1 2nd Generation、FAB2G)を少なくとも10分間遮光予湿した後、サンプルプレート(GreinierBio、PN655209)をテストし、テストが正しく行われた後、予定のプログラムに従って続行した。ここで、200μL/ウェルKB緩衝液をサンプルプレート1の第1、10、12カラムに添加し、0.01M pH2.0グリシン溶液を第11カラムに添加し、調製したサンプル溶液を第2~8カラムに添加し(1つのサンプルに4つのウェルを追加、つまり1つのカラムに2つのサンプル)、第9カラムに、高濃度から低濃度までの順にヒトBCMA-Fcを添加し、すなわち、1番目と5番目のウェルに200nMの抗原溶液を添加し、2番目と6番目のウェルに50nMの抗原溶液を添加し、3番目と7番目のウェルに12.5nMの抗原溶液を添加し、4番目と8番目のウェルに0nMの抗原溶液を添加した。サンプルプレート2の調製は、第9カラムの抗原がサルBCMAタンパク質に置き換えられたことを除いて、変更されなかった。データの結果を表1-4に示す。
上記の実施例に基づいて5-6-7-hu-2および5-6-32-hu-2を選択し、分析および配列決定を行った。IMGTデータベース(http://www.imgt.org/)に基づいてヒト抗体配列の可変領域を定義し、本発明の抗体の軽鎖および重鎖可変領域の配列(配列番号13~16)を確定し、可変領域配列を分析し、CDRをAbMによって定義する方法によって、抗体の重鎖および軽鎖の相補性決定領域配列(配列番号1~12)を確定した。

Claims (23)
- ヒトBCMAとサルBCMAの両方に結合できる、B細胞成熟抗原(BCMA)を標的とする単離されたモノクローナル抗体。
- 前記抗体は、
(1)配列番号1若しくは2に示される重鎖相補性決定領域1(CDR-H1)、及び/または配列番号3若しくは4に示される重鎖相補性決定領域2(CDR-H2)、及び/または配列番号5若しくは6に示される重鎖相補性決定領域3(CDR-H3)を含む重鎖可変領域を含む抗体;
(2)配列番号7若しくは8に示される軽鎖相補性決定領域1(CDR-L1)、及び/または配列番号9若しくは10に示される軽鎖相補性決定領域2(CDR-L2)、及び/または配列番号11若しくは12に示される軽鎖相補性決定領域3(CDR-L3)を含む軽鎖可変領域を含む抗体;
(3)(1)に記載の抗体の重鎖可変領域及び(2)に記載の抗体の軽鎖可変領域を含む抗体;
(4)(1)~(3)のいずれか1項に記載の抗体のバリアントであり、(1)~(3)のいずれか1項に記載の抗体と同等または類似の活性を有する抗体
から選択されるいずれかである、請求項1に記載のモノクローナル抗体。 - 前記抗体は、
(1)配列番号13もしくは14示されるアミノ酸配列または前記配列のバリアントを含む軽鎖可変領域を含む抗体;
(2)配列番号15もしくは16示されるアミノ酸配列または前記配列のバリアントを含む重鎖可変領域を含む抗体;
(3)(1)に記載の抗体の重鎖可変領域及び(2)に記載の抗体の軽鎖可変領域を含む抗体
から選択されるいずれかであることを特徴とする、請求項1または2に記載の抗体。 - 前記抗体の重鎖可変領域が、配列番号1に示されるCDR-H1、配列番号3に示されるCDR-H2、及び配列番号5に示されるCDR-H3を含むことを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 前記抗体の重鎖可変領域が、配列番号2に示されるCDR-H1、配列番号4に示されるCDR-H2、及び配列番号6に示されるCDR-H3を含むことを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 前記抗体の軽鎖可変領域が、配列番号7に示されるCDR-L1、配列番号9に示されるCDR-L2、及び配列番号11に示されるCDR-L3を含むことを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 前記抗体の軽鎖可変領域が、配列番号8に示されるCDR-L1、配列番号10に示されるCDR-L2、及び配列番号12に示されるCDR-L3を含むことを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 前記抗体の軽鎖可変領域が、配列番号13または14に示される配列を有するか、または前記配列のいずれか1つと少なくとも80%、例えば、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%の類似性を有する配列を有することを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 前記抗体の重鎖可変領域が、配列番号15または16に示される配列を有するか、または前記配列のいずれか1つと少なくとも80%、例えば、85%、90%、91%、92%、93%、94%、95%、96%、97%、98%、99%の類似性を有する配列を有することを特徴とする、請求項1~3のいずれか1項に記載の抗体。
- 請求項1~9のいずれか1項に記載の抗体と同じエピトープを認識することを特徴とする、B細胞成熟抗原(BCMA)を標的とするモノクローナル抗体。
- 請求項1~9のいずれか1項に記載の抗体と競合してB細胞成熟抗原(BCMA)に結合することを特徴とする、B細胞成熟抗原(BCMA)を標的とするモノクローナル抗体。
- 請求項1~11のいずれか1項に記載の抗体をコードする核酸。
- 請求項12に記載の核酸を含む発現ベクター。
- 請求項13に記載の発現ベクターを含み、またはそのゲノムに請求項12に記載の核酸が組み込まれた、宿主細胞。
- 請求項14に記載の宿主細胞を培養することによって請求項1~11のいずれか1項に記載のモノクローナル抗体を産生する、モノクローナル抗体を産生する方法。
- 請求項1~11のいずれか1項に記載のモノクローナル抗体及び薬学的に許容される担体を含む、医薬組成物。
- 請求項1~11のいずれか1項に記載のモノクローナル抗体または請求項16に記載の医薬組成物を含む、キットまたは製品。
- 請求項1~11のいずれか1項に記載のモノクローナル抗体または請求項16に記載の医薬組成物または請求項17に記載のキット若しくは製品を、それを必要とする被験者に投与することを含む、
BCMAの発現に関連する疾患を治療するための方法。 - 前記疾患が、B細胞急性リンパ球性白血病、T細胞急性リンパ球性白血病、急性リンパ球性白血病、慢性骨髄性白血病、慢性リンパ球性白血病、B細胞前リンパ球性白血病、芽球性形質細胞様樹状細胞腫瘍、バーキットリンパ腫、びまん性大細胞型B細胞リンパ腫、濾胞性リンパ腫、有毛細胞白血病、小細胞または大細胞濾胞性リンパ腫、悪性リンパ腫、悪性リンパ球増殖性疾患、MALTリンパ腫、マントル細胞リンパ腫、辺縁帯リンパ腫、多発性骨髄腫、骨髄異形成及び骨髄異形成症候群、非ホジキンリンパ腫、形質芽球性リンパ腫、形質細胞様樹状細胞腫瘍、ワルデンストレームマクログロブリン血症、骨髄腫、MGUS、形質細胞腫、全身性アミロイド軽鎖アミロイドーシス及びPOEMS症候群から選択される、請求項18に記載の方法。
- 前記疾患が多発性骨髄腫である、請求項18または19に記載の方法。
- BCMAの発現に関連する疾患を治療するための薬剤の調製における、請求項1~11のいずれか1項に記載のモノクローナル抗体の使用。
- 前記疾患が、B細胞急性リンパ球性白血病、T細胞急性リンパ球性白血病、急性リンパ球性白血病、慢性骨髄性白血病、慢性リンパ球性白血病、B細胞前リンパ球性白血病、芽球性形質細胞様樹状細胞腫瘍、バーキットリンパ腫、びまん性大細胞型B細胞リンパ腫、濾胞性リンパ腫、有毛細胞白血病、小細胞または大細胞濾胞性リンパ腫、悪性リンパ腫、悪性リンパ球増殖性疾患、MALTリンパ腫、マントル細胞リンパ腫、辺縁帯リンパ腫、多発性骨髄腫、骨髄異形成及び骨髄異形成症候群、非ホジキンリンパ腫、形質芽球性リンパ腫、形質細胞様樹状細胞腫瘍、ワルデンストレームマクログロブリン血症、骨髄腫、MGUS、形質細胞腫、全身性アミロイド軽鎖アミロイドーシス及びPOEMS症候群から選択される、請求項21に記載の使用。
