Abstract
Description
実施例1:ラクトバチルスサケイWIKIM31菌株培養液の準備
キムチから由来し、韓国微生物保存センターに受託番号KFCC11654P(国内寄託)及びKCCM12654P(国際寄託)として受託されたラクトバチルスサケイWIKIM31(Lactobacillus sakei WIKIM31)菌株を実験に使用した。ラクトバチルスサケイWIKIM31菌株は、MRS培地で30℃、24時間培養した後、菌体を8,000rpmで5分間遠心分離し、PBSで洗浄して、残っている培地成分を除去した。PBSを用いて1×1010CFU/mL菌数に定量化し、0.2ml(1×109CFU)ずつゾンデを用いて実験動物に週に5回経口投与し、陰性及び陽性対照群には滅菌PBSを投与した。
8週齢の雄マウス(C57BL/6)を一週間飼育室で適応させ、30日間ラクトバチルスサケイWIKIM31(Lactobacillus sakei WIKIM31,KFCC11654P)を経口投与した。その後、MPTP(1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine)を腹腔内注射してパーキンソン病を誘発した後、ラクトバチルスサケイWIKIM31の効果を確認し、図1及び図2に示した。このとき、図1及び図2で、Naiveは正常マウスモデルであり、MPTPは、PBSを経口投与した後にパーキンソン病を誘発したマウスモデルであり、WIKIM31は、ラクトバチルスサケイWIKIM31を経口投与した後にパーキンソン病を誘発したマウスモデルである。
3-1.正常動物モデルでのうつ病誘発
6週齢の雄ラット(Rat,Sprague Dawley)を一週間飼育室で適応させ、7週齢から慢性ストレスを与えたが、水又は飼料の奪取、空の水筒提示、夜昼周期の逆転、ちらつく明かり、周期的騒音、濡れた敷物提供、ケージを傾ける、及び束縛ストレスなどを毎日、不規則的な方法でランダムに実験動物に加えた。誘発期間は総6週であり、毎日ストレスを変え、初めの2週間はラクトバチルスサケイWIKIM31(Lactobacillus sakei WIKIM31)摂取無しでストレスだけを提供してうつ病を誘発し、次の4週間は、ストレスと共にラクトバチルスサケイWIKIM31(Lactobacillus sakei WIKIM31,KCCM12654P)を摂取させた後、うつ病改善の有無を測定した。
ストレス誘発後、投与薬物の抗うつ効果を評価するために、血中コルチコステロン濃度を測定した。血中コルチコステロンの濃度は、ELISAキット(Corticosterone Assay,Ca:KGE009,RND systems,U.S.A)で分析し、血液を採血して分析を行った。分析血液は、頸動脈から採取し、一度で300uLの全血を採血した後、遠心分離機を用いて4,500rpmで10分間遠心分離して血清を分離し、これを分析に使用した。
憂うつ不安症状を評価するために、うつ病を誘発したラットモデルにおけるオープンフィールド(open field)分析を行った。オープンフィールド分析は、逃げたり隠れたりできない広い新しい空間に実験動物を露出させた後に行動を観察し、これに基づいて実験動物の憂うつ・不安レベルを検査するために行う。結果は、50cm×50cm×50cmのボックスで個体当たり15分間測定し、全体移動距離を基本にして中央の25cm×25cmのサイズを中心部位(center zone)に設定し、中心部位移動距離を分析した。全体移動距離のうち、中心部位移動距離の比率が少ないほどうつ病に一層近いといえよう。このとき、動物の行動は、ビデオ追跡システム(SMART3.0,panlab.U.S.A)を用いて分析した。
直径12cm、高さ40cmの透明なアクリル円筒に30cmの高さで水を満たした後、10分間強制に泳がせた。実験済みの動物は30分間乾燥するように赤外線灯下に30分間置いた後、元のケージに戻した。10分間の本実験において動物の行動を不動姿勢時間(immobility time)で測定し、イメージ分析装備(Smart3.0,panlab.,USA)を用いて分析した。

Claims (13)
- ラクトバチルスサケイ(Lactobacillus sakei)、その培養物、その破砕物、その発酵物又はその抽出物を有効成分として含むうつ病又は不安障害の予防又は治療用薬学組成物。
- 前記薬学組成物は、ラクトバチルスサケイを1×102CFU/ml~1015CFU/mlで含み、前記薬学組成物を対象体(subject)に週に3回以上投与時にうつ病又は不安障害が予防又は改善されることを特徴とする、請求項1に記載の薬学組成物。
- 前記ラクトバチルスサケイは、受託番号KCCM12654Pとして寄託されたラクトバチルスサケイWIKIM31であることを特徴とする、請求項1に記載の薬学組成物。
- 前記うつ病又は不安障害は、退行性脳疾患を有する個体におけるうつ病又は不安障害であることを特徴とする、請求項1に記載の薬学組成物。
- 前記退行性脳疾患は、パーキンソン病(Parkinson’s disease)、アルツハイマー病(Alzheimer’s disease)、ハンチントン病(Huntington’s disease)、ルーゲーリック病(amyotrophic lateral sclerosis)、クロイツフェルトヤコブ病(Creutzgeldt-Jacob disease)、脳卒中(stroke)、多発性硬化症(multiple sclerosis)、学習障害(learning disorder)、認知障害(cognitive impairment)及び記憶力損傷からなる群から選ばれる一つ以上であることを特徴とする、請求項4に記載の薬学組成物。
- 前記薬学組成物は、経口投与、静脈内投与、腹腔内投与、筋肉内投与、皮下投与、血中投与、局所投与、鼻内投与、肺内投与及び直腸内投与からなる群から選ばれる投与経路で投与されることを特徴とする、請求項1に記載の薬学組成物。
- ラクトバチルスサケイ(Lactobacillus sakei)、その培養物、その破砕物、その発酵物又はその抽出物を有効成分として含むうつ病又は不安障害予防又は改善用食品組成物。
- 前記ラクトバチルスサケイは、受託番号KCCM12654Pとして寄託されたラクトバチルスサケイWIKIM31であることを特徴とする、請求項7に記載の食品組成物。
- 前記食品は、健康機能食品であることを特徴とする、請求項7に記載の食品組成物。
- 前記食品は、飲料、バー又は発酵乳であることを特徴とする、請求項7に記載の食品組成物。
- 前記うつ病又は不安障害は、退行性脳疾患を有する個体におけるうつ病又は不安障害であることを特徴とする、請求項7に記載の食品組成物。
- 前記退行性脳疾患は、パーキンソン病(Parkinson’s disease)、アルツハイマー病(Alzheimer’s disease)、ハンチントン病(Huntington’s disease)、ルーゲーリック病(amyotrophic lateral sclerosis)、クロイツフェルトヤコブ病(Creutzgeldt-Jacob disease)、脳卒中(stroke)、多発性硬化症(multiple sclerosis)、学習障害(learning disorder)、認知障害(cognitive impairment)及び記憶力損傷からなる群から選ばれる一つ以上であることを特徴とする、請求項11に記載の食品組成物。
