Abstract
Description
1.ビフィドバクテリウム・ロンガムN714株(NITE BP-03004)
本発明のビフィズス菌はビフィドバクテリウム・ロンガム(Bifidobacterium longum)である。本発明におけるN714の記号は、日清食品ホールディングス株式会社で独自に菌株に付与した番号である。本発明の菌株は、乳児糞便より本発明者によって初めて分離されたものである。

本発明のビフィドバクテリウム・ロンガムN714株は、後述する実験例に示すように、高いIL-17産生抑制能を有する 。IL-17産生抑制能の確認については以下の試験方法によって行った。
IL-17産生抑制試験に用いた試料は、ビフィズス菌を表3に示すGAM培地(ニッスイ)で37℃ 、24時間培養することにより得た。次いで、遠心分離機を用いて増殖させた菌体を集菌した。集菌した菌体を滅菌水で洗浄し、遠心分離機で集菌を行った。洗浄と集菌を繰り返し3回行った。その後、加熱殺菌を施し、凍結乾燥機を用いて加熱殺菌体を凍結乾燥し、乾燥菌体粉末を得た。
IL-17産生抑制試験では、細胞としてヒト末梢血単核球(hPBMC、CTL)を使用した。10%ウシ胎児血清、4 mM GlutaMAX(Gibco)を加えたIMDM培地(Sigma)を用いて、細胞を1.0×106 cells/well、上記で得た試料を10 ng/wellとなるように96wellプレートに播種した。ここへ、Th17細胞分化刺激剤として25 ng/mL rhIL-6(BioLegend)、12.5 ng/mL rhIL-23(BioLegend)、25 ng/mL rhIL-21(BioLegend)、2 ng/mL rhTGF-β1(BioLegend)、25 ng/mL rhIL-1β(BioLegend)を加え、37℃、5%CO2条件下で96時間培養した。培養後、hPBMCの培養上清を回収し、上清中のIL-17Fを市販のイムノアッセイキット(R&D)で測定した。
3.飲食品
本発明のビフィズス菌は飲食品に含有せしめて使用することができる。本発明のビフィズス菌は特に飲料に好適に用いることができるが、例えば、発酵乳及び乳酸菌飲料が考えられる。現行の乳及び乳製品の成分規格等に関する省令では、成分規格として発酵乳(無脂乳固形分8.0%以上のもの)や乳製品乳酸菌飲料(無脂乳固形分3.0%以上のもの)であれば1.0×107 cfu/mL以上、乳酸菌飲料(無脂乳固形分3.0%未満のもの)であれば1.0×106 cfu/mL以上必要とされるが、乳などの発酵液中で増殖させたり、最終製品の形態で増殖させたりすることによって上記の菌数を実現することができる。また、ビフィズス菌入りの発酵乳及び乳酸菌飲料以外にも、バター等の乳製品、マヨネーズ等の卵加工品、バターケーキ等の菓子パン類等にも利用することができる。また、即席麺やクッキー等の加工食品にも好適に利用することができる。上記の他、本発明の食品は、前記ビフィズス菌と共に、必要に応じて適当な担体及び添加剤を添加して製剤化された形態(例えば、粉末、顆粒、カプセル、錠剤等)であってもよい。
<試験例1>IL-17産生抑制試験
本発明のビフィドバクテリウム・ロンガムN714株と、自社保有のビフィドバクテリウム・ロンガム比較株についてIL-17の産生抑制試験を実施した。
次に、本発明のビフィドバクテリウム・ロンガムN714株の優位性を確認するため、基準株のビフィドバクテリウム・ロンガムJCM1217との比較で評価を行った。試験は試験例1と同様の方法で3回行い、その平均値を用いて刺激と比較を行った。結果を図2に示す。

- 前記ビフィズス菌は、ビフィドバクテリウム・ロンガムN714株(NITE BP-03004)である、請求項1記載のビフィズス菌。
- 請求項1または2に記載のビフィズス菌を含有する飲食品。
