Abstract
Description
本願は、2018年8月20日出願の米国仮出願第62/719,938号、カズンズ(Cousins)等210−97−PROVの利益を主張し、此れは其の全体が参照に依って此処に組み込まれる。
本発明は、国立衛生研究所に依って授与された助成金第EY029185−01号に依る政府の支援に依ってなされた。政府は本発明に或る種の権利を有する。
本願に於いて提供される「背景技術」の記載は、本開示の文脈を一般的に提示する目的の為である。其れが此の背景技術の項に記載される限りに於いて、現在記名されている発明者の成果、及び然もなければ出願時に於ける先行技術としての資格を有し得ない記載の側面は、本開示に対する先行技術として明示的にも暗黙的にも承認されない。
次の用語は当業者に依って良く理解されると信じられるが、次の定義は本開示の主題の説明を容易化する為に提出される。
新規の低分子阻害剤化合物の開発
STO−609/CaMKK2の結晶構造の計算モデリングを用いて、計算化学に基づくCaMMK2の低分子阻害剤(SMIC)のライブラリーを開発した。特に、STO−609/CaMKK2の結晶構造が得られた。受容体結合に干渉する事が予測されないが、細胞透過に対する有害な効果なしに改善された可溶性(計算されたLogP〜3)を有する筈であるツール化合物の部分を改変した(トータルの極性表面積<140が好都合と見做された)。5つの(5)初期化合物をMonomerChem(RTP、ノースカロライナ)に依って合成し、本願のデータを生成する為に用いた。
CaMKK2に対する生化学的活性
化合物EY301〜EY305の予備的なスクリーニングをインビトロでHEK293細胞に依って行った(全て10μMで試験された)。手短には、HEK293細胞をRPMI−40完全培地及び1%胎児ウシ血清(FBS)に依ってサブコンフルエンス迄培養し、37℃に於いて5%CO2でインキュベーションした。其れから、培養された細胞を実験の開始に先行して血清不含培地に一晩切り換えた。其れから、細胞を5つのSMIC(EY301、EY302、EY303、EY304、及びEY305、全て10μM濃度)の1つ、STO−609(10μM)、又は基剤対照に依って2時間に渡って前処置し、37℃に於いて5%CO2でインキュベーションした。其れから、細胞をカルシウムイオノフォアのイオノマイシン(0.5μg/mL)に依って15分に渡って刺激し、此れの後に、細胞を洗浄し、収穫し、トータルタンパク質の回収の為に標準的な方法を用いてリシスした。其れから、標準的なタンパク質ゲル電気泳動後に、各条件についてリン酸化CaMKK2(図4A)又はリン酸化AMPK(図4B)何方かのウエスタンブロットを行った。研究は三重で行った。デンシトメトリー分析は、EY301、EY303、EY304、及びEY305についてCaMKK2自己リン酸化のロバストな阻害(図4C)(EY301及びEY304はSTO−609と比較してより有効に見えた)と、EY303、EY304、及びEY305についてリン酸化AMPKの部分的阻害(図4D)とを実証した。此れ等の全てはSTO−609に匹敵した。
化合物1〜5の機能スクリーニング
EY301〜EY305の機能的な阻害能力をスクリーニングする為に、1方向混合リンパ球反応(MLR)(図5)が用いられるであろう。C57BL/6マウスの脾臓細胞が応答細胞として用いられ、BALB/cマウスの脾臓細胞(Co60、3000radに依って照射される)が刺激細胞として用いられるであろう。其れから、2つの型の細胞は等しい体積及び濃度で混合され、1)基剤対照;2〜6)5つのSMIC(EY301〜EY305)の各1つ、又は7)STO−609(各SMIC又はSTO−609については、様々な濃度10μM、3μM、1μM、0.3μM、及び0.1μMで)の存在下に於いて、37℃に於いて5%CO2で低血清条件下で24時間に渡ってインキュベーションされる。各濃度/条件は三重で行われるであろう。其れから、浸潤免疫細胞に依る破壊的なダメージを媒介すると考えられている幾つかの鍵のT細胞及びマクロファージ由来のサイトカイン及びエフェクター分子(例えば、TNF−α、IL−2、TGF−β、及びIFN−γ)の其々の酵素結合免疫吸着アッセイ(ELISA)に依る分析の為に上清が回収され、濃縮されるであろう。其れから、目当ての各サイトカインについての各化合物のIC50の計算に依って、EY301〜EY305の機能的な阻害活性が決定されるであろう。其れから、免疫細胞エフェクター機能についての各SMICの相対的な効力及び力価を評価する為に、化合物は目当ての各サイトカインについてのIC50濃度に依って順位付けされるであろう。
OGVHDのマウスモデル
T細胞及びマクロファージが鍵の病原性の役割を演ずる事が公知である疾患の眼移植片対宿主病(OGVHD)は、モデル炎症性眼疾患に相当する。何故なら、A)其れは他のより普通の炎症性眼疾患(即ち涙液減少症、マイボーム腺機能不全、ぶどう膜疾患、他)とオーバーラップする病理学的及び臨床的特徴を共有する事が公知であり;B)其れは血液学的悪性物、自己免疫性疾患、及び他の障害について同種骨髄移植を受ける個体で起こるので、リスク集団は良く定義されているからである。因って、OGVHDの実験モデルは、眼科疾患についてCaMKK2を標的化及び阻害する事の治療ポテンシャルを研究する為の魅力的な系に相当する。脾臓T細胞の補充を伴うC57BL/6からBALB/cマウスへの骨髄移植(「BMT」)に基づくマウスモデルは、眼移植片対宿主病(OGVHD)の鍵の病理学的特徴を再現し、此れは主要組織適合性ミスマッチに相当する(図6)。手短には、C57BL/6及びBALB/c血統の2〜3月齢マウスが此のモデルでは用いられる。C57BL/6マウスが屠殺され、此れの後に、RPMI−1640培地が回収された脛骨及び大腿骨両方の骨幹部管にフラッシュされる。其れから、骨髄がホモジナイゼーション及び濾過され、赤血球を欠く未精製の骨髄(BM)細胞がドナーマウスから回収される。加えて、ドナー脾臓が無菌的に収穫され、小片に切られ、スパチュラに依ってすり潰され、単離された脾臓由来T細胞の単細胞懸濁液へと濾過及び単離される。レシピエントBALB/cマウスは、照射される950cGyのドーズでの全身致死線量照射を受ける(セシウム137線源に依る)。此れの後に、ドナーC57BL/6BM細胞及び脾臓由来T細胞が尾静脈注射に依って照射されたレシピエントマウスに移植される。
CaMKK2阻害剤の局所投与
例4に記載されているOGVHDモデルを用いて、ツール化合物CaMKK2阻害剤STO−609(1.5mg/mL)の局所眼投与が実験的OGVHDの重症度を防止又は改善し得るかどうかを試験した。BMT+T細胞移入後に、実験手続きの開始に先行して、レシピエントBALB/cマウスが14日に渡って回復する事を許した。BMT+T細胞移入後の第14日に始まって、STO−609又は基剤を2週間に渡って毎日投与し、定期検査が処置期間を通して行われた。其れから、処置期間の終わりに、眼を(マスク化された様式で)OGVHDの徴候についてグレード分類し、改変された臨床グレード分類システムに従ってスコアリングした(33、34)。其れから、OGVHD(即ち、免疫細胞浸潤、結膜瘢痕、杯細胞喪失、マイボーム腺瘢痕、及び涙腺破綻/浸潤/瘢痕)の病理学的評価を伴う眼付属器組織(瞼、結膜)及び前部(結膜、強膜、角膜)の組織学的分析の為に、マウスを屠殺し、眼を回収した。
インビトロのT細胞及び/又はマクロファージ活性を抑制するSMICの同定
我々は、20個のNCESMICのライブラリーから、インビトロのT細胞及び/又はマクロファージ活性を抑制し、OGVHDのマウスモデルに於いて効力がある少なくとも1つの(1)SMICを同定するであろう。此れはインビトロで高濃度に於いて抑制活性を有さない化合物の初期排除を可能にするであろう。マクロファージ及びT細胞株を用いて、ウエスタンブロットデンシトメトリー分析を用いて、候補分子が、CaMKK2活性、CaMKK2基質CaMKIV及びAMPKのリン酸化並びにCaMKK2の自己リン酸化を阻害する能力が評価されるであろう。手短には、15分に渡る0.5μg/mLのイオノマイシン及び10ng/mLのPMAに依る処置に先行して、基剤、STO−609、候補SMIC、又は基剤対照の存在下に於いて、細胞が低血清条件下で培養されるであろう。其れから、細胞はトータルタンパク質の為に回収及びリシスされ、pCaMKK2、pAMPK、pCaMKIVについてウエスタンブロット分析が行われるであろう。デンシトメトリー分析がアクチンに対して正規化され、SMIC阻害能力がSTO−609及び基剤と比較される。

我々は、操作されていない2〜3月齢の野生型BALB/cマウスに於いて外用投与された上位(5)SMICの非GLP毒性学研究を行い、臨床評価及び組織病理に依って局所毒性を評価するであろう。手短には、水系溶媒中の15mMストック濃度が各候補SMICについて用いられ、予備的なドーズ範囲研究が、3倍希釈に依って(3ドーズ、投薬群当たりn=3、5つの候補化合物)外用的に毎日2回、14日に渡って、基剤対照(n=5)と比較して行われるであろう。局所毒性は2つのメトリクス:(1)臨床所見(結膜出血、ケモーシス、角膜ヘイズ、ACの炎症細胞)及び(2)組織学(角膜、結膜、強膜、水晶体、網膜、脈絡膜)に依って評価されるであろう。臨床用生体顕微鏡眼検後に、組織学の為に、マウスは屠殺され、眼が回収されるであろう。眼組織の標準的なH&E切片が炎症の証拠又は毒性を示唆する組織形態学の変化についてマスク化された様式で検査されるであろう。全身毒性の外面的な評価が行われ、生育不全及び死亡率を評価するであろう。「僅かに異常」よりも多大な何れかの所見は其の化合物濃度を失格とするであろう。此れ等のインビボ忍容性研究のゴールは、外用に依る眼毒性の欠如を有する少なくとも2つの(2)SMICを同定する事であろう。
OGVHDに依る概念実証
上位2つの(2)SMIC候補の外用の臨床効力を実証する為に、我々はOGVHDのマウスモデルに於ける前臨床の概念実証研究を行うであろう。
a)対照群:同時のT細胞なしのBL/6ドナーBM(n=5)がBALB/cレシピエントに移植される(n=10)。OGVHDを発生しない対照群。如何なる薬物も投与されない。
b)実験群1:BL/6ドナーBM及びT細胞(n=8)がBALB/cレシピエントに移植される(n=15)。基剤対照。
c)実験群2:BL/6ドナーBM及びT細胞(n=8)がBALB/cレシピエントに移植される(n=15)。STO−609。
d)実験群3:BL/6ドナーBM及びT細胞(n=8)がBALB/cレシピエントに移植される(n=15)。SMICのNo.1。
e)実験群4:BL/6ドナーBM及びT細胞(n=8)がBALB/cレシピエントに移植される(n=15)。SMICのNo.2。
インビトロの細胞のCaMKK2活性を抑制するSMICの同定
合成されたSMICの初期セットが表1に於いて参照されている。此のセットから、6つの候補SMIC(EY1003.A001.B001、EY1005.A001.B001、EY1006.A001.B001、EY1007.A001.B001、EY1008.A001.B001、EY1001.A002.B001)をHEK293細胞に於ける細胞系の生化学スクリーニングアッセイの為に選択した。CaMKK2の公知の活性化因子の1mMイオノマイシンに依る刺激に先行して、HEK−293細胞を次の1つに依って処置した:STO−609、EY1003.A001.B001、EY1005.A001.B001、EY1006.A001.B001、EY1007.A001.B001、EY1008.A001.B001、EY1001.A002.B001(全て30μM)、又は基剤対照。其れから、細胞ライセートをCaMKK2及びAMPKに対するリン酸化特異的抗体に依るウエスタンブロット分析に依って探査して、其々CaMKK2の自己リン酸化活性及び基質リン酸化活性に対する候補SMICの効果を評価した(図12)。Bアクチンをローディング対照として包含した。グラフは相対的なデンシトメトリーの定量を示しており、シグナルはサンプル間のタンパク質のローディングの変動を補正する為にBアクチンに対して正規化されている。此の分析に基づくと、候補SMICのEY1006.A001.B001及びEY1001.A002.B001はインビトロのCaMKK2阻害活性について類似の力価を実証した。
SMICのインビボ忍容性
局所投与された候補SMICのEY1006.A001.B001及びEY1001.A002.B001の非GLP毒性学研究を2〜3月齢野生型BALB/cマウスに於いて評価し、臨床評価及び組織病理に依って局所毒性を評価した。両方の化合物を14日に渡って1.5mg/mLで毎日2回外用投与した。臨床用生体顕微鏡眼検査又は死後組織学的評価に依って、何れの薬物についても、局所毒性の如何なる徴候も観察されなかった。加えて、何れの化合物も何れかの明らかな全身毒性には結び付けられなかった(即ち、生育不全又は死亡率の如何なる証拠もなし)。
OGVHDに於ける概念実証
OGVHDのマウスモデルに於ける前臨床の概念実証研究を行って、例6及び7に於いて同定された2つの最も有望な候補SMICのEY1006.A001.B001及びEY1001.A002.B001の外用投与の臨床効力を評価した。先の実験はCaMKK2のツール化合物阻害剤STO−609が此のゴールを達成するという事を実証し、此の研究の目的はリード候補NCESMICが匹敵する効力を達成するかどうかを評価する事であった。手短には、2〜3月齢マウスC57BL/6Jドナーマウスからの骨髄及び脾臓T細胞の単離と、レシピエント骨髄のアブレーション後のレシピエント2〜3月齢BALB/cマウスへの単離された骨髄及びT細胞の移植とに依って、OGVHDマウスモデルを開始した。其れから、更なる実験手続きの開始前に、レシピエントBALB/cマウスが凡そ14日に渡って回復する事を許した。其れから、BMT後の第14日に始まって、2週間に渡って連続して、マウスを次の1つに依る毎日の外用処置に依って処置した(実験群、n=5):STO−609酢酸塩(1.5mg/mL)、リードSMIC化合物EY1006.A001.B001及びEY1001.A002.B001(両方とも1.5mg/mL)、並びに1%プレドニゾロン酢酸エステル。
全ての出発材料がイミン中間体に変換される迄(ca.1.5h)、EtOH(4mL)中の対応するアルデヒド及びフェニレンジアミン(1mmol)の等モル溶液を還流下に於いて加熱した。DDQ(395mg、1.1mmol)の追加後に、イミン中間体が完全にベンゾイミダゾール生成物に変換される迄(典型的には30〜60min)、反応混合物を還流下に於いて加熱した。其れから、反応混合物を室温に冷却し、飽和NaHCO3水溶液に依って希釈し、生成物を酢酸エチルに依って2回抽出した。組み合わせた有機抽出液を無水MgSO4に依って乾燥し、濾過し、減圧濃縮した。クルードな生成物をシリカに依るフラッシュカラムクロマトグラフィーに依って精製して、対応する純粋なベンゾイミダゾールを与えた。
150mLのメタノール:酢酸エチル中のニトロ置換ベンゾイミダゾール中間体(2g、7mmol)の溶液を常圧下の室温に於いて活性炭担持Pd触媒(600mg、5wt%Pd)の存在下で水素化した。反応の完了後に(TLC)、触媒をセライトのパッドに依って濾別し、濾液を減圧濃縮し、所望の生成物を黄色固体として提供した。
THF(100mL)中の出発材料アニリン(7.5mmol)の溶液にトリホスゲンを追加した(7.5mmol。混合物を10minに渡ってソニケーションし、齎された懸濁液を55〜60℃で1hに渡って加熱した。混合物を濃縮・乾燥した。残渣に氷水及び飽和重炭酸ナトリウムを追加した。形成された固体を濾過した。固体を水に依って洗浄し、乾燥し、其れを次の反応に用いた。
THF5mL中の出発材料尿素(1.36mmol)、トリフェニルホスフィン(6mmol)、及びアルコール(l50mL、過剰)の懸濁液を5minに渡ってソニケーションし、アゾジカルボン酸ジイソプロピル(DIAD)1.4mL(7mmol)を追加した。混合物を30minに渡ってソニケーションした。混合物を室温で一晩攪拌した。混合物を濃縮・乾燥し、残渣をジエチルエーテルに依って希釈した。沈澱した固体を濾過し、固体を乾燥エーテルに依って洗浄した(4×50mL)。固体を乾燥し、フラッシュクロマトグラフィー(ヘキサン−EtOAc、0〜50%)に依って精製して所望の化合物を与えた。
エタノール5mL中のニトロ化合物(1mmol)の溶液に、Zn粉末(5mmol)及び飽和NH4Cl溶液2mLを追加した。混合物を2hに渡ってソニケーションした。混合物を酢酸エチルに依って抽出した(3×30mL)。有機層を水で洗浄し、乾燥し(Na2SO4)、濃縮して褐色固体を手に入れた。クルードな固体をカラムクロマトグラフィー(ヘキサン−EtOAc、0〜100%)に依って精製して、所望の生成物を与えた。

式中、各R2及びR3が独立して水素(H)、C1〜C10アルキル、−OR7、−OCH2CH2OR7、−OCH2CH2NR5R6、−OCH2CH2COOR7、又は−OCH2CH2PO3Hであり;
式中、R4が水素(H)、C1〜C10アルキル、−CH2CN、−CH2C(O)NH2、−CH2COOH、−CH2SO2CH3、−CH2CH2NH2、−CH2CH2OH、−CH2チエニル、−CH2フラニル、−CH2CH2複素環、−CH2CH2シクロアルキル、−CH2CH2ヘテロシクロアルキル、−CH2CHOCH2OH、又は−(CH2)nZ(CH2)mCH3であり、式中、nが1から5の整数であり、mが0から5の整数であり、Zが酸素(O)又は硫黄(S)であり;
