本明細書に開示する抗CD63抗体は、抗体が由来する生殖系列配列と比較して、重鎖可変ドメインのフレームワーク領域、及び/または、CDR領域に1つ以上のアミノ酸置換、挿入、及び/または、欠失を含み得る。
抗体、免疫グロブリン、抗体結合フラグメント、または、Fc含有タンパク質の、例えば、細胞表面タンパク質、または、そのフラグメントなどの所定の抗原に対する結合に関する用語「結合」とは、一般的には、抗体−抗原相互作用などの、最低でも2つの実体、または、分子構造間の相互作用または会合のことを指す。
抗CD63抗体及びその抗原結合フラグメント(抗CD63抗体、または、その抗原結合フラグメントを含む、多重特異性抗原結合分子、及び、マルチドメイン治療タンパク質を含む)は、個々の抗原結合ドメインが由来した対応する生殖系列配列と比較して、重鎖及び軽鎖可変ドメインのフレームワーク、及び/または、CDR領域に1つ以上のアミノ酸置換、挿入、及び/または、欠失を含み得る。このような変異は、本明細書に開示した例示的アミノ酸配列を、例えば、公共の抗体配列データベースから入手可能な生殖系列配列と比較することで、容易に確認することができる。本発明の抗原結合分子は、本明細書に開示するアミノ酸配列のいずれかに由来する抗原結合フラグメントを含み、1つ以上のフレームワーク、及び/または、CDR領域内の1つ以上のアミノ酸は、抗体が由来した生殖系列配列の対応する残基(複数可)、または、別のヒト生殖系列配列の対応する残基(複数可)、または、対応する生殖系列残基(複数可)の保存的アミノ酸置換へ変異する(このような配列変化を、本明細書では「生殖系列変異」と総称する)。当業者であれば、本明細書に開示する重鎖可変領域配列及び軽鎖可変領域配列から出発して、1つ以上の個々の生殖系列変異、または、これらの組み合わせを含む数多くの抗体、及び、抗体結合フラグメントを容易に生産することができる。特定の実施形態では、VH、及び/または、VLドメイン内のフレームワーク、及び/または、CDR残基はすべて、抗体が由来する元の生殖系列配列において認められる残基へと変異して戻される。その他の実施形態では、ある特定の残基だけが、元の生殖系列配列へと変異して戻され、例えば、変異した残基だけが、FR1の最初の8個のアミノ酸内に、または、FR4の最後の8個のアミノ酸内に認められ、または、変異した残基だけが、CDR1、CDR2、または、CDR3内に認められる。その他の実施形態では、フレームワーク、及び/または、CDR残基(複数可)の1つ以上は、異なる生殖系列配列(すなわち、抗体が元来由来している生殖系列配列とは異なる生殖系列配列)の対応する残基(複数可)へと変異する。さらに、本発明の抗原結合ドメインは、フレームワーク、及び/または、CDR領域内に2つ以上の生殖系列変異のあらゆる組み合わせを含有し得るものであり、例えば、特定の個々の残基は、特定の生殖系列配列の対応する残基へと変異するのに対し、元の生殖系列配列とは異なる特定のその他の残基は、維持され、または、異なる生殖系列配列の対応する残基へと変異する。一旦取得してしまえば、1つ以上の生殖系列変異を含有する抗原結合ドメインは、結合特異性の改善、結合親和性の増大、アンタゴニストまたはアゴニストの生物学的特性の改善または増強(場合によるが)、免疫原性の低下などの1つ以上の所望の特性について、容易に試験することができる。本発明の範囲には、この一般的な方法で得られた1つ以上の抗原結合ドメインを含む二重特異性抗原結合分子が含まれる。
本発明は、pH依存的な結合特性を有する、抗CD63抗体、及び、その抗原結合フラグメント(抗CD63抗体、または、その抗原結合フラグメントを含む、多重特異性抗原結合分子、及び、マルチドメイン治療タンパク質を含む)を含む。例えば、本発明の抗CD63抗体は、中性pHと比較して、酸性pHでCD63に対する結合の低下を示し得る。あるいは、本発明の抗CD63抗体は、中性pHと比較して、酸性pHでCD63に対する結合の増強を示し得る。表現「酸性pH」とは、約6.2未満、例えば、約6.0、5.95、5、9、5.85、5.8、5.75、5.7、5.65、5.6、5.55、5.5、5.45、5.4、5.35、5.3、5.25、5.2、5.15、5.1、5.05、5.0以下のpH値を含む。表現「中性pH」は、約7.0〜約7.4のpHを意味する。表現「中性pH」は、約7.0、7.05、7.1、7.15、7.2、7.25、7.3、7.35、及び、7.4のpH値を含む。
本発明の特定の実施形態によると、例えば、中性pHと比較した酸性pHで、FcRn受容体に結合する抗体を増強または低下させる1つ以上の突然変異を含むFcドメインを含む抗CD63抗体、及び、その抗原結合フラグメント(抗CD63抗体、または、その抗原結合フラグメントを含む、多重特異性抗原結合分子、及び、マルチドメイン治療タンパク質を含む)を提供する。例えば、本発明は、FcドメインのCH2またはCH3領域に変異を含む抗体を含み、変異(複数可)は、酸性環境において(例えば、pHが、約5.5〜約6.0の範囲のエンドソームにおいて)FcRnに対するFcドメインの親和性を高める。そのような突然変異は、動物に投与したときに、抗体の血清半減期の延長をもたらし得る。そのようなFc修飾の例として、例えば、250位(例えば、EまたはQ);250位及び428位(例えば、LまたはF);252位(例えば、L/Y/F/WまたはT)、254位(例えば、SまたはT)、及び、256位(例えば、S/R/Q/E/DまたはT)での修飾;または、428位、及び/または、433位(例えば、H/L/R/S/P/QまたはK)、及び/または、434位(例えば、H/FまたはY)での修飾;あるいは、250位及び/または428位の修飾;あるいは、307位または308位(例えば、308F、V308F)、及び、434位での修飾があるが、これらに限定されない。ある実施形態では、修飾は、428L(例えば、M428L)、及び、434S(例えば、N434S)の修飾;428L、259I(例えば、V259I)、及び、308F(例えば、V308F)の修飾;433K(例えば、H433K)及び、434(例えば、434Y)の修飾;252、254、及び、256(例えば、252Y、254T、及び256E)の修飾;250Q及び428Lの修飾(例えば、T250Q及びM428L);及び、307及び/または308の修飾(例えば、308Fまたは308P)を含む。
本発明は、治療状況、及び、所望の特定の標的化特性に応じて、高い、中位の、または、低い親和性でヒトCD63に結合する、抗体、及び、その抗原結合フラグメントを含む。例えば、一方のアームが、CD63に対して結合し、かつ、他方のアームが、標的抗原(例えば、腫瘍関連抗原)に結合する二重特異性抗原結合分子に関しては、標的抗原結合アームが、標的抗原には高い親和性で結合する一方で、抗CD63アームが、中位または低いの親和性でしかCD63に対して結合しない、ことが望ましい。このようにして、標的抗原を発現する細胞に対する抗原結合分子の優先的標的化は、一般的/非標的化CD63結合、及び、それに関連する結果として生じる有害な副作用を回避しながら達成し得る。
本発明の抗CD63抗体、及び、その抗原結合フラグメント(抗CD63抗体、または、その抗原結合フラグメントを含む、多重特異性抗原結合分子、及び、マルチドメイン治療タンパク質を含む)が結合するCD63のエピトープは、3つ以上(例えば、3つ、4つ、5つ、6つ、7つ、8つ、9つ、10個、11個、12個、13個、14個、15個、16個、17個、18個、19個、20個、または、それより多数)のCD63タンパク質のアミノ酸の単一連続配列からなり得る。あるいは、エピトープは、CD63の複数の非連続アミノ酸(または、アミノ酸配列)からなり得る。用語「エピトープ」とは、パラトープとして公知の抗体分子の可変領域における特異的抗原結合部位と相互作用する抗原決定基のことを指す。単一の抗原は、2つ以上のエピトープを有し得る。したがって、異なる抗体は、抗原での異なる領域に結合し得るものであり、かつ、異なる生物学的効果を有し得る。エピトープは、立体配座、または、線状のいずれにでもし得る。立体配座エピトープは、直鎖状ポリペプチド鎖の異なるセグメントから空間的に並置したアミノ酸によって生産される。直鎖状エピトープは、ポリペプチド鎖での隣接するアミノ酸残基によって生産される。特定の状況では、エピトープは、抗原上の糖類、ホスホリル基、または、スルホニル基の部分を含み得る。
特定の抗原に特異的な抗原結合ドメインは、当該技術分野で公知のあらゆる抗体生産技術で調製することができる。一旦取得してしまえば、2つの異なる抗原(例えば、CD63、及び、標的抗原)に特異的な2つの異なる抗原結合ドメインを、互いに適切に配置して、常法を使用して、本発明の二重特異性抗原結合分子を生産することができる。(本発明の二重特異性抗原結合分子を構築するために使用できる例示的な二重特異性抗体フォーマットの考察を、本明細書の別の箇所に提供している)。特定の実施形態では、本発明の多重特異性抗原結合分子の1つ以上の個々の構成要素(例えば、重鎖、及び、軽鎖)は、キメラ抗体、ヒト化抗体、または、完全ヒト型抗体に由来する。そのような抗体を作り出すための方法は、当該技術分野において周知である。例えば、本発明の二重特異性抗原結合分子の1つ以上の重鎖、及び/または、軽鎖は、VELOCIMMUNE(商標)技術を使用して調製することができる。VELOCIMMUNE(商標)技術(または、あらゆるその他のヒト抗体産生技術)を使用して、まず、特定の抗原(例えば、CD63)に対する高親和性のキメラ抗体が、ヒト可変領域及びマウス定常領域を有した状態で単離される。抗体を、親和性、選択性、エピトープなどの望ましい特徴について特徴決定して選択する。マウス定常領域を、所望のヒト定常領域と置換して、本発明の二重特異性抗原結合分子に組み込むことができる完全ヒト重鎖、及び/または、軽鎖を生産する。
本発明は、本明細書に開示した例示的な分子のものとは異なるが、CD63に結合する能力を保持するアミノ酸配列を有する抗原結合分子を含む。このような変異体分子は、親配列と比較してアミノ酸の1つ以上の付加、欠失、または、置換を含むが、説明した二重特異性抗原結合分子の生物学的活性とは本質的に等価である生物学的活性を呈する。

本明細書では、抗CD63抗体、または、その結合フラグメント(それを含む多重特異性結合分子、または、マルチドメイン治療タンパク質を含む)、または、抗CD63抗体を含む抗体薬物コンジュゲート(例えば、本明細書の表1に示したHCVR/LCVR、または、CDR配列のいずれかを含む抗CD63抗体またはADC)を含む治療用組成物を、それを必要とする対象に対して投与する、ことを含む方法も開示している。治療用組成物は、抗CD63抗体、その抗原結合フラグメント、または、本明細書に開示すADCのいずれかと、医薬として許容可能な担体または希釈剤とを含むことができる。
例示的なCD63抗体
抗ヒトCD63抗体の生成
抗ヒトCD63抗体は、マウス(例えば、ヒト免疫グロブリン重鎖、及び、ヒトカッパ軽鎖可変領域をコードするDNAを含む遺伝子操作したマウス)を、ヒトCD63で免疫処置して得た。
表1は、本明細書に開示したマルチドメイン治療用抗CD63タンパク質を生成するために使用した、選択した抗CD63抗体の重鎖または軽鎖可変領域(それぞれ、HCVRまたはLCVR)、または、重鎖または軽鎖CDR(それぞれ、HCDR及びLCDR)をコードする核酸(NA)配列、及び、括弧内にはアミノ酸(AA)配列の配列識別子を示す。

ヒトCD63発現細胞に対する抗CD63抗体の相対的な細胞表面結合を、ヒトCD63を内因的に発現するCD63ポジティブHEK293細胞(ATCC、カタログ番号CRL−1573)、及び、CD63ネガティブHEK293/CD63ノックアウト細胞を使用したフローサイトメトリーを介してアクセスした。アッセイでは、2%FBS(Saradigm カタログ番号1500−500)(染色緩衝剤)を含有し、カルシウムとマグネシウムを含まないPBS(VWR、カタログ番号45000−446)が入った96ウェルV底プレート(Axygen Scientific、カタログ番号P−96−450−V−C−S)に細胞を播いた。次に、細胞を、100nM〜1.7pMの範囲の濃度の抗CD63抗体またはアイソタイプコントロール抗体と共に、氷上で、30分間、インキュベートした。抗体を含まないウェルを、コントロールとして使用した。HEK293/CD63KO細胞を、最高濃度(100nM)の抗体だけで染色した。次に、細胞を、染色緩衝剤で1回洗浄し、そして、PEコンジュケート抗マウスFc二次抗体(Jackson ImmunoResearch、カタログ番号115−115−164)と共に、100nMで、4℃で、30分間、インキュベートした。次いで、細胞を、洗浄し、そして、PBSで希釈したCytofix(BD Biosciences、カタログ番号554655)の50%溶液を使用して固定した。試料を、Intellicyte Hypercytフローサイトメーターに適用し、結果を、ForeCytソフトウェア(Intellicyte)で分析して、平均蛍光強度(MFI)を計算した。GraphPad Prismを使用して、12ポイント応答曲線での4つのパラメーターのロジスティック方程式を使用して測定値を分析し、そして、得られたEC50値を報告する(表12)。シグナル対ノイズ比(S/N)は、抗体不含のウェルに対する抗CD63抗体またはコントロール抗体MFIの比を計算して決定した(表12)。

本明細書に開示した抗CD63抗体が、CD63発現細胞に結合して内在化する能力を評価した。アッセイでは、10%FBS(ATCC、カタログ番号30−2020)、ペニシリン/ストレプトマイシン/L−グルタミン(Gibco、カタログ番号10378−016)、100μMピルビン酸ナトリウム(Millipore、カタログ番号TM5−005−C)、1mM HEPES(ThermoFisher、カタログ番号15630080)、10μg/mL インスリンウシ(Gemini BioProducts、カタログ番号700−912P)(増殖培地)を含有するRPMI(Irvine Scientific、カタログ番号9160)を入れた、96ウェルのコラーゲンコーティングプレート(Greiner、カタログ番号655956)に、T47D細胞(ATCC、カタログ番号HTB−133)を播種し、そして、5%CO2にて、37℃で、一晩インキュベートした。染色するために、細胞の4枚組プレートを、2%FBSを含み、カルシウムとマグネシウムが含まないPBS(Irving、カタログ番号9240)(染色緩衝剤)で希釈した10μg/mLの抗CD63抗体と共に、4℃で、30分間、インキュベートした。細胞を、染色緩衝剤で2回洗浄した後に、Alexa−Flour 488コンジュゲート二次抗体(Jackson Immunoresearch、カタログ番号115−547−003、または、Jackson Immunoresearch、カタログ番号109−547−003)の10μg/mLと共に、4℃で、30分間、インキュベートを行い、続いて、染色緩衝液で、さらに2回洗浄した。2枚のプレートを直ちに固定し、そして、4%パラホルムアルデヒド(PFA;ThermoFisher、カタログ番号28908)+5uM DRAQ5(ThermoFisher、カタログ番号62251)を含むPBSで、20分間、染色した(非内在化プレート)。残りの2枚のプレートを、増殖培地と共に、37℃で、2時間、インキュベートした後に固定を行い、そして、PBSで希釈した4%PFA+5μM DRAQ5の溶液を使用して、20分間、染色した(内在化プレート)。固定した後に、すべてのプレートを、PBSで1回洗浄した。1枚の非内在化プレートと1枚の内在化プレートを、50μg/mLの抗Alexa Fluor 488抗体(Regeneron)を含むPBSと共に、4℃で、一晩、インキュベートして、表面のAlexa Fluor 488蛍光を反応停止させた。残りのプレートは、PBSだけでインキュベートした。共焦点画像を、Opera Phenix(Perkin Elmer)にて、40倍の倍率で得た。ハーモニー分析ソフトウェア(PerkinElmer)を使用して、DRAQ5で標識した細胞を同定し、そして、総Alexa−Fluor 488相対蛍光単位(RFU)/細胞を決定した。4℃での総結合(反応停止させていない4℃のウェルでのRFU値)、37℃での総結合(反応停止させていない37℃のウェルのRFU値)、総内在化RFU、及び、%内在化を、表1に示したように、各抗体について決定した。すべての計算について、第2のAbだけのコントロールウェルでのバックグラウンド蛍光を、それぞれのウェルから控除した。総内部化RFUは、次のようにして計算した:反応停止させていない37℃の試料での総RFU−37℃での表面RFU。表面RFUとは、反応停止していない37℃でのRFU−反応停止させた37℃でのRFU)/QEとして定義する。QE(反応停止効率)は:1−(反応停止した4℃での試料の総RFU/反応停止していない4℃での試料の総RFU)として定義する。%内在化は、次の式:(37℃での総内在化RFU/37℃での総RFU)*100で決定した。

精製した抗CD63モノクローナル抗体に結合する異なるCD63(EC2)ループ試薬の平衡解離定数(KD)を、リアルタイム表面プラズモン共鳴をベースとしたBiacore T200バイオセンサー、または、Biacore 2000バイオセンサーを使用して決定した。すべての結合研究を、10mM HEPES、150mM NaCl、3mM EDTA、及び、0.05%v/v界面活性剤Tween−20、pH7.4(HBS−ET)ランニング緩衝剤にて、25℃と37℃で実施した。BiacoreCM5センサーチップ表面を、まずは、ウサギ抗マウスFc特異的ポリクローナル抗体(GE Healthcare、カタログ番号BR100838)、または、抗ヒトFabキット(GE Healthcare、カタログ番号28958325)のいずれかとのアミンカップリングで誘導体化して、抗CD63モノクローナル抗体を捕捉した。C末端Myc−Myc−ヘキサヒスチジン(hCD63 ECループ2−MMH;配列番号338)で発現した組換えヒトCD63細胞外ループ2、または、C末端ヒトFcタグ(hCD63 ECループ2−hFc;配列番号339)で発現した組換えヒトCD63細胞外ループ2のいずれかに関して、結合研究を行った。異なる濃度のhCD63 ECループ2−MMH、または、hCD63 ECループ2−hFc(4倍希釈で50nM〜12.5nM、または、3倍段階希釈で90nM〜0.37nMのいずれかで試験した)を、まず、HBS−ETランニング緩衝剤で調製し、そして、それらを、捕捉した抗CD63モノクローナル抗体表面に、35μL/分、または、50μL/分の流速で、4分間かけて注入し、一方で、モノクローナル抗体に対して結合したCD63試薬の解離を、HBS−ETランニング緩衝剤にて、8分間または10分間モニターした。結合率(ka)と解離率(kd)は、Scrubber 2.0c曲線適合ソフトウェアを使用して、リアルタイム結合センサーグラムを、物質移動制限のある1:1結合モデルに適合させて決定した。結合解離平衡定数(KD)、及び、解離半減期(t1/2)は、反応速度から、次のように計算した:

抗CD63モノクローナル抗体のパネルの間での結合競合を、Octet HTXバイオセンサープラットフォーム(Pall ForteBio Corp.)でのリアルタイムの無標識バイオレイヤー干渉測定アッセイを使用して決定した。実験全体を、25℃で、10mM HEPES、150mM NaCl、3mM EDTA、及び、0.05%v/v界面活性剤Tween−20、1mg/mL BSA、pH7.4(HBS−EBT)緩衝剤にて、1000rpmの速度でプレートを震盪しながら実施した。2つの抗体が、C末端myc−myc−ヘキサヒスチジンタグ(hCD63 ECループ2−MMH;配列番号xx)で発現した組換えヒトCD63 ECループ2でのそれぞれのエピトープに対する結合について、互いに競合できるか否かを評価するために、約0.61nmのhCD63 ECループ2−MMHを、まず、hCD63 ECループ2−MMHの20μg/mL溶液を含むウェルに、バイオセンサーチップを3分間沈めて、抗Penta−His抗体でコーティングしたOctetバイオセンサーチップ(FortebioInc、#18−5122)に捕捉させた。次に、抗原で捕捉したバイオセンサーチップを、第1の抗CD63モノクローナル抗体(以下、mAb−1と称する)を、mAb−1の50μg/mL溶液を含むウェルに、210秒間浸漬させたて飽和させた。次いで、バイオセンサーチップを、第2の抗CD63モノクローナル抗体(以下、mAb−2と称する)の50μg/mL溶液を含むウェルに、120秒間浸漬させた。バイオセンサーチップは、実験のそれぞれのステップの間に、HBS−EBT緩衝剤で洗浄した。実験の全過程でリアルタイムの結合応答をモニターし、そして、それぞれのステップの終わりに結合応答を記録した。mAb−1と事前に複合体化したhCD63 ECループ2−MMHに対するmAb−2結合の応答を比較し、そして、表10に示したように、異なる抗CD63モノクローナル抗体の競合/非競合挙動を決定した。

本実施例で説明したようにして生成した抗CD63抗体が、二重特異性抗原結合分子の一部として内在化する能力を評価するために、抗体を、二重特異性フォーマットに再構築し、そして、一方の結合アームを、抗CD63抗体VH/VLペア(表1の親抗体を参照されたい)とし、また、他方を、無関係な結合アームとした。二重特異性抗体を作り出す標準的な方法を使用しており、例示的な方法として、例えば、本明細書の一部を構成するものとして、それぞれの内容を援用する、米国特許出願公開第2010/0331527号、及び、米国特許第US5731168号に記載されたものがある。二重特異性抗体を、ヒトCD63発現細胞を使用して、内在化する能力について試験した。アッセイでは、10%FBS、及び、ペニシリン−ストレプトマイシン/L−グルタミン(Gibco、カタログ番号10378016)を含有するDMEMを入れた、透明底の黒色ポリ−D−リジンでコーティングした96ウェルプレート(Greiner、カタログ番号655946)に、10,000細胞/ウェルの密度で、ヒトCD63を内因的に発現するHEK293細胞を播種した。2日後に、培地を、抗CD63二重特異性抗体と、10μg/mLから始まって、0.157μg/mLに至る2倍希釈系列のネガティブコントロール抗体とを含む新たな培地との交換を行い、併せて、培地だけのコントロールも交換した。次に、細胞を、37℃で、3時間インキュベートして、抗体の内在化を可能にした。インキュベートの後に、細胞を、PBSで洗浄し、4%パラホルムアルデヒド(Thermo Scientific、カタログ番号28908)で、室温で、20分間固定し、続いて、0.2%Triton X−100(Spectrum Chemical、カタログ番号TR135)で、5%正常ヤギ血清(NGS)(Gibco、カタログ番号PCN5000)にて、室温で、20分間、透過処理した。次いで、細胞を、2ug/mLのロバ抗マウスIgG Alexa Fluor−647 Fab(Manufacture、カタログ番号115−606−006)、または、2μg/mLのヤギ抗ヒトIgG Alexa Fluor−647 Fab(Jackson ImmunoResearch、カタログ番号115−606−006)のいずれかと、5%NGSにて、室温で、1時間、インキュベートした。二次抗体溶液を除去した後に、細胞を、PBSで洗浄し、続いて、2滴/mLのNucBlue(Invitrogen、カタログ番号R37605)を含む新たなPBSを添加して、生細胞の核を染色した。抗体の内在化と核を、ImageXpress High−Content Imaging System(Molecular Devices)にて、40倍の倍率で、画像化を行い、そして、抗体の内在化を、MetaXpress Software Transfluor Application Module(Molecular Devices)を使用して定量した。抗体内在化を、ピット積分強度/細胞±標準偏差(SD)として報告する。

抗hCD63 ScFv::GAAポリヌクレオチド、及び、遺伝子療法ベクターの構築
ヒトGAA(hGAA;配列番号369)、または、ヒトGAAに対して、そのC末端で融合した抗ヒトCD63一本鎖可変フラグメント(ScFv)(抗hCD63 ScFv−hGAA)の発現をコードするAAV2/8ウイルスを、標準的な三重トランスフェクションプロトコールを使用して生成する(Gray et al. 2011;“Production of recombinant adeno−associated viral vectors and use in vitro and in vivo administration”, Current Protocols in Neuroscience, John Wiley & Sons, New York (1999), pp.4.17.1−4.17.25, Vol 1も参照されたい)。生産のために、1×107個のHEK293細胞を、15cmプレートに播く。翌日に、(A)肝臓特異的セルピナ1エンハンサー(配列番号367)を含み、TTRプロモーター(配列番号368)駆動性ヒトGAAをコードするコントロールpAAVベクター、または、肝臓特異的セルピナ1のエンハンサー(配列番号367)を含み、TTRプロモーター(配列番号368)駆動性hCD63 ScFv−hGAA(図2を参照されたい)をコードする試験pAAVのいずれかの8μg、及び、(B)pAAV RC2/8−由来ベクター(Gao、2002)、及び、16μgのpHelper(Agilent、カタログ番号240074)を、PEIpro(Polyplus transfection,New York,NY,カタログ番号115−100)媒介性トランスフェクションを、1:1の比(1ul PEIpro:1μg DNA)で用いて、トランスフェクションする。トランスフェクションの72時間後に、細胞を回収し、そして、標準的な凍結融解法を用いて、20mM Tris−HCl、1mM MgCl2、2.5mM KCl、100mM NaClからなる緩衝剤に溶解する。次に、ベンゾナーゼ(Sigma、カタログ番号E1014−25KU)を、最終濃度0.5U/μLで試料に添加し、次いで、これを、37℃で、60分間、インキュベートする。続いて、ウイルスを、(Zolotukhin et al.,1999,Gene Ther 1999;6:973−985)に記載されているようにして、イオジキサノール勾配超遠心分離を用いて精製し、続いて、qPCRで滴定する。
AAV後のマウスポンペモデルのグリコーゲン含量
関連のグリコーゲン蓄積インビボモデルにおける、AAVが送達した抗hCD63 ScFv−GAA融合体(配列番号364)と、AAVが送達したGAAとの効果を決定するために、両方の療法をポンペ病マウスモデルに対して送達することとし、同マウスは、マウスGAA遺伝子の欠失についてホモ接合性であり、そして、マウスCD63の代わりに、ヒトCD63の発現についてホモ接合性であり、系統の背景は、75%のC57BL/6;25%の129SvJである。本明細書では、これらのマウスを、CD63 HumIn GAA KOマウス、または、CD63hu/hu;GAA−/−マウスとも称する。

GAAに対する免疫応答
抗ヒトGAA抗体の血清レベルを測定するために、製造業者の仕様書に従って、血清分離チューブ(BD Biosciences、カタログ番号365967)を使用して、すべての処置群から末期採血の間に回収した血液から血清を分離する。これとは別に、96ウェル高タンパク質結合プレート(ThermoFisher、カタログ番号15041)を、PBSで希釈した20μgのhGAA(R&D Systems、カタログ番号8329−GH−025)で、一晩、コーティングする。プレートを、PBS+0.05% Tween(PBS−T)で、3回洗浄する。プレートを、0.5%BSAを含むPBS−Tでブロッキングし、そして、マウス血清の1:300〜1:5.1e7の範囲の希釈系列を、一晩、プレートに加える。HRPコンジュゲートヤギ抗マウスIgG抗体(Jackson Immuno Research、カタログ番号115−035−164)、及び、BD Opt EIA基質キットを使用して、総抗マウスIgG(サブクラス1+2a+2b+3)を測定する。1Nリン酸を使用して、比色反応を停止する。次いで、Spectramax i3プレートリーダー(Molecular Devices)で、450nmでの吸光度を読み取る。希釈曲線を、シグモイド曲線に適合させ、そして、この曲線から力価を計算する。
血清GAA
実験の過程でヒトGAA血清レベルを測定するために、尾部出血を介して、試料を、毎月回収する。製造業者の仕様書に従って、血清セパレーターチューブ(BD Biosciences、カタログ番号365967)を使用して、血液から血清を分離する。次いで、1μLの単離した血清を、4%〜20%のNovex wedgewellプレキャストゲルにロードし、220Vで、45分間泳動させ、標準手順書を使用して、200mAで、1時間、ニトロセルロース膜に移送する。次いで、ニトロセルロース膜を、1:2000希釈で使用する抗GAA一次抗体(Abcam、番号ab137068)、及び、1:1000希釈で使用する抗GAPDH抗体(Abcam、番号AB9484)の12mLを使用してプロービングし、そして、4℃で、一晩、インキュベートする。一次抗体によるインキュベートの後に、膜を、1×TBST/洗浄で、5分間、3回洗浄する。次いで、抗ウサギIgG(LiCor、926−32211)、及び、抗マウスIgG(LiCor、925−68070)(Licor,Lincoln,NE)二次抗体の1:15000希釈物の12mLを、膜に添加し、そして、室温で、1時間、インキュベートする。二次抗体によるインキュベートの後に、膜を、1×TBST/洗浄で、5分間、2回、そして、1×TBSで、5分間、1回洗浄する。次いで、LiCor Odyssey機器(LI−COR Biotechnology)を使用して、膜を画像化して定量する。
グリコーゲンの組織測定、及び、筋肉組織の組織学的特徴決定
グリコーゲンの組織測定:個々の組織のグリコーゲン含量を測定するために、心臓、四頭筋、腓腹筋、横隔膜、ヒラメ筋、及び、EDLの組織を、CO2で窒息させた直後のすべての群由来のマウスから摘出し、次いで、液体窒素にて急速凍結し、及び、−80℃で保存する。グリコーゲン測定のために、各組織の約50mgを、蒸留水中で、1mg対25μLの水の比率のステンレス鋼ビーズを用いて、ベンチトップ用ホモジナイザーで溶解する。グリコーゲン分析溶解物を、105°で、15分間加熱し、そして、21000×gで遠心分離して破片を除去する。グリコーゲン測定は、蛍光アッセイに関する製造業者の指示書に従って、グリコーゲンアッセイキット(Sigma−Aldrich、番号MAK016)を使用して実施する。各試料の蛍光を、535nmの励起、及び、587nmの発光で、蛍光プレートリーダー(Molecular Devices、Spectramax i3)にて測定する。グリコーゲンの計算量は、製造業者が提供した以下の式を使用して計算する。
Claims (25)
- 抗CD63抗体、または、その抗原結合フラグメントであって、前記抗体、または、その抗原結合フラグメントは、表に記載のHCVR/LCVRアミノ酸配列ペアを含み、または、ヒトCD63に対する結合について、表1に示したHCVR/LCVRを含むリファレンス抗体と競合する、前記抗体、または、その抗原結合フラグメント。
- 前記リファレンス抗体が、配列番号2/10、配列番号18/26、配列番号34/42、配列番号50/58、配列番号66/74、配列番号82/90、配列番号98/106、配列番号114/122、配列番号130/138、配列番号146/154、配列番号162/170、配列番号178/186、配列番号194/202、配列番号210/218、配列番号226/234、配列番号242/250、配列番号258/266、配列番号274/282、配列番号290/282、配列番号298/282、配列番号306/282、配列番号314/282、配列番号322/282、及び、配列番号330/282に記載したものからなる群から選択するHCVR/LCVRアミノ酸配列ペアを含む、請求項1に記載の抗CD63抗体、または、その抗原結合フラグメント。
- 前記抗体、または、その抗原結合フラグメントが、表1に示したHCVR/LCVRアミノ酸配列ペアを含むリファレンス抗体と同じヒトCD63でのエピトープに対して結合する、請求項1または請求項2に記載の抗CD63抗体、または、その抗原結合フラグメント。
- 前記抗体、または、その抗原結合フラグメントが、配列番号2/10、配列番号18/26、配列番号34/42、配列番号50/58、配列番号66/74、配列番号82/90、配列番号98/106、配列番号114/122、配列番号130/138、配列番号146/154、配列番号162/170、配列番号178/186、配列番号194/202、配列番号210/218、配列番号226/234、配列番号242/250、配列番号258/266、配列番号274/282、配列番号290/282、配列番号298/282、配列番号306/282、配列番号314/282、配列番号322/282、及び、配列番号330/282からなる群から選択するHCVR/LCVRアミノ酸配列ペアを含むリファレンス抗体と同じヒトCD63でのエピトープに対して結合する、請求項3に記載の抗体、または、その抗原結合フラグメント。
- 請求項1〜4のいずれか1項に記載の単離した抗CD63抗体、または、その抗原結合フラグメントと、ヒトCD63に対する結合について競合する第1の抗原結合ドメインと、標的抗原に結合する第2の抗原結合ドメインとを含む二重特異性抗原結合分子。
- 前記標的抗原が、腫瘍関連抗原である、請求項5に記載の二重特異性抗原結合分子。
- 前記第1の抗原結合ドメインが、CD63を発現するが、標的抗原を発現しないヒト細胞に結合せず、例えば、CD63及び標的抗原の両方を発現するヒト細胞だけに結合する、請求項5または請求項6に記載の二重特異性抗原結合分子。
- 前記第1の抗原結合ドメイン、及び、前記第2の抗原結合ドメインのそれぞれが、完全にヒトである、請求項5〜7のいずれか1項に記載の二重特異性抗原結合分子。
- 前記抗原結合分子が、細胞で発現するヒトCD63、及び、ヒト標的抗原の両方に対して結合し、かつ、その細胞でのヒト標的抗原のCD63内在化、及び/または、分解を誘導する、請求項5〜8のいずれか1項に記載の二重特異性抗原結合分子。
- 前記抗体が、ヒトCD63を発現するが、ヒト標的抗原を発現しない細胞では内在化されない、請求項5〜9のいずれか1項に記載の二重特異性抗原結合分子。
- 前記抗体が、完全にヒトである、請求項5〜10のいずれか1項に記載の二重特異性抗原結合分子。
- 前記第1の抗原結合ドメインが、ヒトCD63だけを発現する細胞には結合しないが、ヒトCD63と前記標的抗原の両方を発現する細胞に対して結合する前記抗体の低い結合親和性で、ヒトCD63に結合する、請求項5〜11のいずれか1項に記載の二重特異性抗原結合分子。
- 請求項1〜4のいずれか1項に記載の抗CD63抗体、または、その抗原結合フラグメント、及び、酵素ドメインを含むマルチドメイン治療用タンパク質。
- 前記酵素ドメインが、GAA、または、その生物学的に活性な部分を含む、請求項13に記載のマルチドメイン治療用タンパク質。
- 配列番号364に記載のアミノ酸配列を含む、請求項13または請求項14に記載のマルチドメイン治療用タンパク質。
- 請求項1〜4のいずれか1項に記載の抗CD63抗体、または、その抗原結合フラグメント、請求項5〜12のいずれか1項に記載の二重特異性抗原結合分子、または、請求項13〜15のいずれか1項に記載のマルチドメイン治療用タンパク質、をコードする配列を含むポリヌクレオチド。
- 前記ヌクレオチド配列が、請求項13〜15のいずれか1項に記載のマルチドメイン治療用タンパク質をコードするAAVベクターである、請求項16に記載のポリヌクレオチド。
