本発明の菌株は、好ましくは、嫌気性条件下で増殖しうる。さらに、当該菌株は、そのような嫌気性条件下で、水不溶性セルロース及びタンパク質を分解することができることが、好ましい。
種々の環境試料から枯草菌株をスクリーニングし、動物への直接給餌微生物/プロバイオティクスとして優れた菌株を獲得した。この菌株は、標的動物の腸内でその潜在能力を最大限に発揮することを目的とするため、種々の環境条件及び消化管関連条件に耐性である菌株を予めスクリーニングした。菌株胞子を生成させて(Nicholson及びSetlow 1990)、洗浄し、80℃で20分間インキュベート(低温殺菌)した後、仔牛浸出ブロス寒天培地(veal infusion; VI, Difco(商標), no. 234420, Becton Dickinson GmbH, Heidelberg, Germany)を用いて、10倍希釈液中対数/1で滴定した。非増殖前で希釈度が2番目に高い液を-80℃で保存し、胞子状態以降の全ての評価で標準出発点として用いた。Larsen et al.(2014)に基づき、酸暴露の生存率を評価して、胃の通過をシミュレートした(Argenzio 2004a; Trampel and Duke 2004)。栄養細胞の増殖を、胃/前胃及び砂嚢の条件下での増殖を示す低pH、及び4mMまでの胆汁(B8631、CAS 8008-63-8、Sigma-Aldrich)の存在下で、pH7で評価して、胃又は砂嚢のクリアランス直後の小腸近位部における菌株の増殖を確認した(Argenzio 2004b; Trampel and Duke 2004)。嫌気性の腸における菌株の適応度(Argenzio 2004b; Trampel and Duke 2004)を、嫌気性条件下、2.5mM KNO3添加VI培地(Glaser et al, 1995)に標準化胞子溶液(AnaeroPakTM, Thermo Fisher Scientific)を接種して、評価した。さらに、菌株の嫌気性タンパク質分解活性及び細胞分解活性を、1%スキムミルク粉末(70166、Sigma-Aldrich)又は0.1%水不溶性AZCL−HEセルロース(I-AZCEL, Megazyme International, Bray, Ireland)を添加したVI寒天平板培地で、評価した。
実施例1で選択した枯草菌株DSM 32540の競合能を評価するために、市販の枯草菌株DSM 17299、及び/又はバチルス・リヒェニホルミス菌株DSM 17236を用いて、ベンチマーク分析を行った。近位小腸で行う菌株の準備(Argenzio 2004b; Trampel and Duke 2004)として、胃通過直後の中性pHの胆汁の存在下、0.3質量%ブタ胆汁(Sigma-Aldrich)添加VIB培地での菌株培養により、測定した。候補菌株の細胞懸濁液50uL、及び10mLのVIB、を含む100mLの三角フラスコ中で37℃、200rpm、インキュベートした後、一晩培養物約50uLと、pH7の0.3質量%のブタ胆汁を含むVIBの1mLとを、100ウェルハニカムプレート(Oy Growth Curves Ab Ltd, former Thermo Labsystems, Helsinki, Finland)に移して、OD0.2/mLを得た。BioLinkソフトウェアパッケージ(Oy Growth Curves Ab Ltd)を用いたBioscrien C MBRを用いて、15分毎にODを測定し、37℃、200rpmで48時間、菌株特異的増殖を観察した。各菌株の定量的評価は、0〜5時間の曲線下面積(AUC5、OD×時間(h))、0〜10時間の曲線下面積(OD×時間(h)のAUC10)、及び菌株が最大光学濃度(Tmax(h))に達するまでの時間として、比較した。結果を表2.1に示す。

直接比較すると、DSM 32540株は、0.3質量%のブタ胆汁存在下で、基準株DSM 17299よりも16.75時間早く、基準株DSM 17236よりも27.5時間早く、最大ODに達した。さらに、DSM 32540株は、DSM 17236株の増殖と比較して、最初の5時間で2.7倍、最初の10時間で3.2倍、最初の30時間で2.1倍速く、増殖した。さらに、DSM 32540株は、0.3%のニワトリ胆汁の存在下で増殖することができる。
DSM 32540株とDSM 17299株の増殖を、短鎖脂肪酸の存在下で、大腸についてより重要となる消化管で観察して比較し、評価した(Argenzio 2004b; Trampel and Duke 2004)。試験は、実施例1に記載の標準化された胞子溶液を用いて開始し、37℃、pH6のVI培地で、好気性菌培養して試験した。増殖対非増殖をパラメータとして読み取った。この試験では、Mcllvaine緩衝液(Palop et al. 1996)を用いてVI培地をpH6に調製後、0,05%酢酸(HA, 537020, CAS 64-19-7, Sigma-Aldrich)、0,05%プロピオン酸(HP, P1386, CAS 79-09-4, Sigma-Aldrich)又は0,2%乳酸(HL, W261106, CAS 50-21-5, Sigma-Aldrich)を添加した。結果を表3.1に示す。

実施例2で行った試験と同様に、DSM 32540株、DSM 17299株及びDSM 17236株の好気性キシラン分解活性を評価して、比較した。キシラナーゼ活性はLarsen et al. (2014)に記載されているように測定した。分析は、3回の独立実験系で行い、その後、ミリ単位/マイクロリットル溶液として平均化した。
実施例2で行った試験と同様、DSM 32540株及び野生型株DSM 10株を比較し、各培地で発現した代謝産物及び増殖阻害された病原体の数を、各々評価した。代謝産物発現分析については、出発培養物を培養して、Scholz et al. (2011)に記載されているように試験を行った。100mLフラスコで、10mLのLuria Bertamiブロス(LB, Thermo Fisher Scientific)を37℃、160rpmで24時間培養した培養液100 uLを、主培養物に用いた。主培養物を、0.2mL/LのKellyT微量金属溶液(LBKelly, Scholz et al. 2011)を含む10mLのLB、又は0.6%酵母抽出物を含む10mLのトリプチケーゼソイブロス((Oxoid, Thermo Fisher Scientific)(Y1625, CAS 8013-1-2, Sigma-Aldrich; 得られる培養液は「TSBYE」と略す)のいずれかを用いて、ともに100mLフラスコ内で37℃、24時間、160rpmで培養した。主培養物の4mLを、15mL試験管で、2mLのn−ブタノールと混合し、3分間ボルテックスした後、15分間超音波処理した。5000rpm、1分間遠心後、有機相を取り出して、真空乾燥し、高速液体クロマトグラフィー−エレクトロスプレーイオン化質量分析(HPLC-ESI-MS; Chen et al. 2006)を用いて、分析した。全ての試料を、負及び正の2つの異なるモードで測定し、質量スペクトルを得た。Teo and Tan (2005)で同様に報告されたピークの結果を、Da(ダルトン)の分子量に換算し、その結果を表5.1に示す。

熱ストレス等のストレス過多条件が原因となる酸化ストレス(Lin et al., 2006)では、活性酸素種が存在すると、DNA、タンパク質又は脂質の損傷の原因となる。プロバイオティクスは、抗酸化酵素活性を高めて、宿主の酸化的防御システムを増強しうる(Aluwong et al., 2013, Mishra et al., 2015)。そこで、抗酸化酵素活性、特にスーパーオキシドジスムターゼ及びカタラーゼ活性について、DSM 32540株をスクリーニングした。
in vitroの研究によれば、病原体複製は、低分子量物質が介在すると阻害されることが示されている(Oelschlager 2010)。このリストの上位には、乳酸等の短鎖脂肪酸があげられる(Oelschlager 2010)。病原体が阻害される理由として考えられるのは、乳酸産生によるpHの低下である(Fuller 1992)。乳酸菌は、動物飼料のプロバイオティクスとしても用いられており、炭水化物の発酵では、乳酸は主要最終産物として生成されることが知られている(Halasz 2009)。枯草菌株DSM 32540は、in vitroで乳酸を産生しうることが示されている。
実験は、168頭の25日間離乳した子ブタ(ランドレース(Landrace)×ヨークシャ(Yorkshire))を対象とし、無作為に3つの処理群に割り付け(表8.1)、これを8区(反復)にし、各区に平均6.5±0.5kgの7頭をあてがった。3種類の処置は、主にトウモロコシ−大豆粕に基づき(表8.2):1.基本制御(対照);2.対照+30gのバージニアマイシン/飼料MT(AGP);3.対照+2.0×109cfu/g(DSM 32540株)含有250g/MTの枯草菌株DSM 32540株である。実験的処置として、1〜42日齢から、マッシュ型で自由摂取させた。
表8.1:実験的処置
試験では、離乳後21日目の子ブタ64頭(平均体重7.3+0.35kg)を2つの投与群に無作為に割り付け(表9.1)、各4匹の6区(反復)に分けた。3種類の処置は、主にトウモロコシ−大豆粕に基づき(表9.2)、1.基本制御(対照);2.対照+2.0×109cfu/g(DSM 32540株)含有250g/MTの枯草菌株DSM 32540株である。実験的処置として、1〜42日齢から、マッシュ型で自由摂取させた。
Claims (19)
- 以下の群:
a)DSMZに寄託番号DSM 32540として寄託された枯草菌(Bacillus subtilis)株;
b)前記DSM 32540株の全ての同定特性を有する、前記DSM 32540として寄託された枯草菌株の変異体;
c)前記a)又はb)の調製物;
d)前記a)、b)又はc)に含まれる化合物の有効な混合物を含有する調製物;
から選択される、枯草菌株又はその調製物。 - 請求項1のb)前記DSM 32540として寄託された枯草菌株の変異体であって、前記変異体のDNA配列は、DSM 32540株のDNA配列と少なくとも95%同一である、DSM 32540として寄託された枯草菌株の変異体。
- 枯草菌株又はその調製物であって、前記枯草菌株が以下の:
a)配列番号2で表されるポリヌクレオチド配列と、少なくとも99.5%、好ましくは少なくとも99.8%、とりわけ100%の配列同一性を有する、yqfD配列;
b)配列番号3で表されるポリヌクレオチド配列と、少なくとも99.5%、好ましくは少なくとも99.8%、とりわけ100%の配列同一性を有する、gyrB配列;
c)配列番号4で表されるポリヌクレオチド配列と、少なくとも99.5%、好ましくは少なくとも99.8%、とりわけ100%の配列同一性を有する、rpoB配列;
d)配列番号5で表されるポリヌクレオチド配列と、少なくとも99.5%、好ましくは少なくとも99.8%、とりわけ100%の配列同一性を有する、groEL配列;
の少なくとも1つ、好ましくは全てを示す、枯草菌株又はその調製物。 - 枯草菌株が配列番号1で表されるポリヌクレオチド配列と、少なくとも99.5%、好ましくは少なくとも99.8%、とりわけ100%の配列同一性を有する、16S rDNA配列を表す、請求項1〜4のいずれか一項に記載の枯草菌株又はその調製物。
- 嫌気的に増殖ができ、特に、嫌気性条件下で、水不溶性のセルロース及びタンパク質を分解することができる、請求項1〜4のいずれか一項に記載の枯草菌株。
- ウェルシュ菌(C.perfringens)ATCC 13124株;ウェルシュ菌BB−081_Cpe株;ウェルシュ菌BB_031_Cpe株;ディフィシル菌(C.difficile)DSM 1296株;黄色ブドウ球菌亜種アウレウス菌(S.aureus subsp.aureus)DSM 20231株;ガリナセウス菌(S.gallinaceus)DSM 15349株;豚レンサ球菌(S.suis)DSM 9682株;カンピロバクター・コリ菌(C.coli)DSM 4689株;及びエンテロコッカス・セコラム菌(E.cecorum)DSM 20683株から選択される菌株の少なくとも1つ、好ましくは、全て菌株の増殖を阻害することができる、請求項1〜5のいずれか一項に記載の枯草菌株。
- 0.05質量%の酢酸、0.05質量%のプロピオン酸、及び/又は0.2質量%の乳酸の存在下で増殖することができる、請求項1〜6のいずれか一項に記載の枯草菌株。
- セルラーゼ活性、キシラナーゼ活性、カタラーゼ活性、スーパーオキシドジスムターゼ活性の少なくとも1つ、好ましくは、全ての酵素活性を呈する、請求項1〜7のいずれか一項に記載の枯草菌株。
- 2mM、好ましくは、4mMの胆汁の存在下で増殖することができる、請求項1〜8のいずれか一項に記載の枯草菌株。
- 飼料又は食品中のプロバイオティック成分(DFM)としての、請求項1〜9のいずれか一項に記載の枯草菌株又はその調製物の使用。
- 請求項1〜9のいずれか一項に記載の枯草菌株又はその調製物と、及び好ましくは、タンパク質、炭水化物、脂肪、さらなるプロバイオティクス、プレバイオティクス、酵素、ビタミン、免疫調節剤、代用乳、ミネラル、アミノ酸、抗コクシジウム剤、酸性製品、医薬品、及びそれらの組み合わせから選択される、少なくとも1つのさらなる飼料又は食品成分と、を含む、飼料又は食品の組成物。
- 請求項1〜9のいずれか一項に記載の枯草菌株又はその調製物、及び薬学的に許容される担体を含む、医薬組成物。
- 動物又はヒトの健康状態、特に腸の健康状態を改善する、請求項11又は12に記載の組成物。
- 動物、特にブタ又は鳥類に、請求項1〜9のいずれか一項に記載の枯草菌株又はその調製物、及び/又は請求項11に記載の飼料組成物、及び/又は請求項12に記載の医薬組成物を、給餌する方法。
- 請求項1〜9のいずれか一項に記載の枯草菌又はその調製物、又は請求項11〜13のいずれか一項に記載の組成物を、動物又はヒトに投与して、前記動物又はヒトの健康状態、特に腸の健康状態を改善する方法。
- 請求項1〜9のいずれか一項に記載の少なくとも1の枯草菌及び/若しくは少なくとも1のその調製物、又は請求項11〜13のいずれか一項に記載の少なくとも1の組成物を動物に投与することにより、前記動物の健康を改善する、及び/又は、前記動物の一般的な体調を改善する、及び/又は、飼料転換率を改善する、及び/又は、前記動物の死亡率を低下させる、及び/又は、動物の生存率を高める、及び/又は、動物の体重を増加させる方法、及び/又は、動物の病気に対する抵抗力を高める、及び/又は、動物の免疫応答を高める、及び/又は、動物の健全な腸内微生物叢を確立又は維持する、及び/又は、動物の糞便を介して排泄される病原体を低下させる、方法。
- 堆肥又は汚染された液体の有害な環境影響を制御、及び/又は回避する方法であって、堆肥、汚染された液体、落葉落枝、ピット、又は堆肥池に、請求項1〜9のいずれか一項に記載の少なくとも1の枯草菌、及び/又は少なくとも1のその調製物、及び/又は請求項11〜13のいずれか一項に記載の少なくとも1の組成物、を適用する工程を含む、方法。
- 水又は水溶液、特に飲料水又は飼育水、の水質を制御、及び/又は改善する方法であって、請求項1〜9のいずれか一項に記載の少なくとも1の枯草菌、及び/又は少なくとも1のその調製物、及び/又は請求項11〜13のいずれか一項に記載の少なくとも1の組成物、を、水又は水溶液に適用する工程を含む、方法。
- 栽培植物の微生物病を処置、及び/又は予防する方法であって、請求項1〜9のいずれか一項に記載の少なくとも1の枯草菌、及び/又は少なくとも1のその調製物、及び/又は請求項11〜13のいずれか一項に記載の少なくとも1の組成物、を、栽培植物に適用する工程を含む、方法。
