Abstract
Description
1. 技術分野
本発明は、有効成分としてヤブラン(Liriope platyphylla)の抽出物を含む、ニコチン中毒の予防又は治療用の組成物に関する。
保健福祉部の報告によると、韓国の成人人口の22.6%(男性39.3%、女性5.5%)が慢性的に喫煙することが知られている。喫煙により引き起こされる有害物質への曝露により、平均余命は、約8〜10年短縮され、現在、喫煙は、がんの主要な原因として挙げられている。統計的に、がんによる死亡者の3人に1人が喫煙者である。特に、韓国の男性の肺がんの90%が喫煙に起因していることが知られている。さらに、喫煙は、種々のがん、例えば膀胱がん、膵臓がん、咽頭がん、子宮頸がん、及び食道がんの発症に直接的又は間接的に関与することが知られている。
1)抽出溶媒を添加することによってヤブランを抽出する工程、
2)工程2)の抽出物を、ろ過する工程、及び
3)工程3)のろ過された抽出物を減圧下で濃縮し、そしてそれを乾燥させることによってヤブランの抽出物を調製する工程
を含む調製方法によって調製されるが、必ずしもそれに限定されない。
ヤブラン温水抽出物の調製
ヤブラン根を乾燥させ、2.1Lの水を300gの乾燥ヤブラン根に添加し、それを、還流装置中、80℃で4時間にて、2回抽出した。還流及びろ過の後、抽出物を減圧下で乾燥凍結させた。その結果、65%の収率で、ヤブランの温水抽出物197gを得た。
ヤブランエタノール抽出物の調製
<2-1>ヤブラン30%エタノール抽出物の調製
ヤブラン根を乾燥させ、2.1Lの30%エタノール(v/v)を300gの乾燥ヤブラン根に添加し、それを、還流装置中、70℃で4時間にて、2回抽出した。還流及びろ過の後、抽出物を減圧下で乾燥凍結させた。その結果、63%の収率で、ヤブランの30%エタノール抽出物191gを得た。
ヤブラン根を乾燥させ、2.1Lの50%エタノール(v/v)を300gの乾燥ヤブラン根に添加し、それを、還流装置中、60℃で4時間にて、2回抽出した。還流及びろ過の後、抽出物を減圧下で乾燥凍結させた。その結果、57%の収率で、ヤブランの50%エタノール抽出物171gを得た。
ニコチン及びヤブラン抽出物の処置濃度に応じた神経細胞株(PC-12細胞株)の細胞生存率を、MTTアッセイによって測定した。
PC-12細胞におけるニコチンにより制御されたチロシンヒドロキシラーゼ(TH)タンパク質及びドーパミントランスポーター(DAT)タンパク質の発現に対するヤブラン抽出物の効果を確認するために、ウェスタンブロッティングを行った。
実験動物の準備
体重230〜260gのSprague-Dawley雄ラットを購入した(Orient Bio)。韓国毒性学会(Korean Institute of Toxicology)の動物飼育室内の管理条件(温度23±3℃、湿度30〜70%、明/暗サイクル12/12時間)でラットを飼育した。実験開始まで、食物及び水を自由に摂取できるように十分な食物及び水をラットに与えた。さらに、測定装置箱への曝露に対するストレスなどの環境要因に関する可変要素を最小化するために、実験開始前の少なくとも3日間、1日当たり30分間、Sprague-Dawley雄ラットを自発運動活性測定装置箱内で馴らした。
ヤブラン抽出物が、ニコチンによって自発運動活性を増加させるニコチン中毒現象を抑制できるかどうかを調べた。
Claims (9)
- 有効成分としてヤブラン(Liriope platyphylla)の抽出物を含む、ニコチン中毒の予防又は治療用の医薬組成物。
- 前記抽出物が水、C1〜C2低級アルコール、又はその混合物を溶媒として使用して抽出されている、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 前記抽出物がヤブランの花、葉、茎、実、及び根からなる群から選択されるいずれか1種から抽出されている、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 前記中毒が、耐性、欲求及び離脱症状からなる群から選択されるいずれか1つである、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 離脱症状が、抑うつ、不安、不眠症及び体重増加からなる群から選択されるいずれか1つである、請求項4に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 前記抽出物がチロシンヒドロキシラーゼ(TH)タンパク質の発現を低減する、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 前記抽出物がドーパミントランスポーター(DAT)タンパク質の発現を増加させる、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 前記抽出物が、ニコチンによって増加した自発運動活性を抑制する、請求項1に記載のニコチン中毒の予防又は治療用の医薬組成物。
- 有効成分としてヤブランの抽出物を含む、ニコチン中毒の予防又は改善用の健康機能性食品。
