Description

本発明は、cGASおよびcGAMPに関する方法および組成物を、配合物(ワクチンアジュバントを含む)、薬物スクリーニング、治療法、および診断法でのその使用を含めて、提供する。
2'3'-cGAMPは、哺乳動物細胞によって産生される内因性二次メッセンジャーである; 2'3'-cGAMPはSTINGの高親和性リガンドである; 2'3'-cGAMPはI型インターフェロンの強力な誘導因子である; 2'3'-cGAMPの結合はSTINGの立体構造変化を誘導する。
外来遺伝要素に対する宿主防衛は、生きている生物の最も基本的な機能の1つである。細胞質における自己DNAまたは外来DNAの存在は、危険シグナルまたは外来からの侵入の兆候として真核生物により感知される(1)。DNAは、細菌もしくはウイルス感染、トランスフェクション、または狼瘡のような自己免疫疾患を引き起こすある病的な状態にある核もしくはミトコンドリアからの「漏出」により、細胞質へ導入されうる。哺乳動物細胞では、サイトゾルDNAは、小胞体タンパク質STING (MITA、MPYS、またはERISとしても知られる)を通じて、I型インターフェロン(IFN)および他のサイトカインの産生を誘発する(2)。STINGは、転写因子NF-κBおよびIRF3をそれぞれ活性化するサイトゾルキナーゼIKKおよびTBK1を動員しかつ活性化する。NF-κBおよびIRF3はその後、核に入り、ともに機能して、IFNおよび他のサイトカインを誘導する。DNA依存性RNAポリメラーゼIIIは、ポリ[dA:dT]のようなATに富むDNAを検出し、該DNAをRIG-I経路を刺激してIFNを誘導できるRNAリガンドへと転写するセンサーであることが示されている(3, 4)。しかしながら、大部分のDNA配列は、RNAポリメラーゼIII−RIG-I経路を活性化しない。代わりに、サイトゾルDNAがSTING依存性経路を配列依存的に活性化する。サイトゾルDNAがSTING経路をいかにして活性化するかは、依然として把握されていない。
DNAは、遺伝物質であることが示されるよりずっと以前に、免疫応答を刺激することが知られていたが、DNAが免疫刺激剤として機能する機構は、よく理解されていないままである(1)。DNAは、エンドソーム中のToll様受容体9 (TLR9)への結合を通じて樹状細胞におけるI型インターフェロンの産生を刺激しうるが、サイトゾル中のDNAがIFNをいかにして誘導するかは、不明なままである。特に、インターフェロン経路におけるサイトゾルDNAを検出するセンサーは、依然として把握されていない(2)。DAI、RNAポリメラーゼIII、IFI16、DDX41およびいくつかの他のDNAヘリカーゼを含む、いくつかのタンパク質は、IFNを誘導する潜在的なDNAセンサーとして機能することが示唆されているが、どれも広く受け入れられていない(3)。
本発明者らは、哺乳動物細胞またはサイトゾル抽出物へのDNAの送達が環状GMP-AMP (cGAMP)の産生を誘発し、環状GMP-AMPがSTINGに結合し、かつSTINGを活性化し、IRF3の活性化およびIFNβの誘導をもたらすことを示した(4)。cGAMPシンターゼ(cGAS)を同定するため、本発明者らは、cGAMP合成活性を含むマウス線維肉腫細胞株L929からサイトゾル抽出物(S100)を分画した。この活性は、ニシン精巣DNA (HT-DNA)の存在下においてATPおよびGTPとともにカラム画分をインキュベートすることによってアッセイされた。DNAをベンゾナーゼで消化し、95℃で加熱してタンパク質を変性させた後に、cGAMPを含有していた耐熱性上清を、ペルフリンゴリシンO (PFO)で透過処理したRaw264.7細胞(形質転換されたマウスマクロファージ)とともにインキュベートした。これらの細胞におけるcGAMP誘導性のIRF3二量体化は、ネイティブゲル電気泳動によって分析された(4)。このアッセイ法を用いて、本発明者らは、それぞれが4段階のクロマトグラフィーからなるが、カラムが異なるかまたは使われたカラムの順序が異なる、独立した3経路の精製を行った。特に、第3の経路には、ビオチン化DNAオリゴ(免疫刺激性DNAまたはISDとして知られる45 bpのDNA)を用いたアフィニティー精製段階を含めた。本発明者らは、これらの経路の分画から8000〜15,000倍の精製および2〜5%の活性回復の範囲を達成したものと推定した。しかしながら、これらの精製経路の各々の最終段階において、画分の銀染色から、cGAS活性で共精製された明瞭なタンパク質バンドは明らかにされず、推定上のcGASタンパク質の存在量はL929サイトゾル抽出物において非常に低い可能性があることが示唆された。
STING発現を欠くHEK293Tにおけるm-cGASの過剰発現では、IFNβは誘導されなかったが、HEK293T細胞におけるSTINGの安定発現によって、これらの細胞はm-cGASによるIFNβの誘導において高度にコンピテントとなった。重要なことには、推定上の触媒残基G198およびS199のアラニンへの点突然変異によって、m-cGASがIFNβを誘導する能力が消失した。これらの変異、ならびに他の推定上の触媒残基E211およびD213のアラニンへの変異によっても、m-cGASがHEK293T-STING細胞においてIRF3の二量体化を誘導する能力が消失した。c-GASによるIFNβ誘導の大きさは、MAVS (RNAセンサーRIG-Iの下流で機能するアダプタータンパク質)によって誘導されるものに匹敵し、DAI、IFI16、およびDDX41を含む、他の推定上のDNAセンサーによって誘導されるものよりも数桁高かった。cGASおよび他の推定上のDNAセンサーの過剰発現が細胞におけるcGAMPの産生をもたらしたかどうか判定するため、加熱処理した細胞抽出物由来の上清を、PFO透過処理したRaw264.7細胞とともにインキュベートし、その後にIRF3二量体化の測定を行った。HEK293T-STING細胞において発現された全てのタンパク質のなかで、cGASだけが細胞においてcGAMP活性を生成することができた。
本発明者らは、異なる2組のsiRNAを用いて、L929細胞においてm-cGASをノックダウンし、両方のsiRNAオリゴがHT-DNAによるIFNβの誘導を顕著に阻害したこと、および阻害度がm-cGAS RNAのノックダウンの効率と相関していたことを見出した。本発明者らはまた、異なるm-cGAS領域を標的化するshRNA配列を安定的に発現する2つのL929細胞株を樹立した。これらの細胞がHT-DNAに応答してIFNβを誘導する能力は、対照shRNA (GFP)を発現している別の細胞株と比べて大幅に損なわれていた。重要なことに、L929-sh-cGAS細胞においてcGASを発現させることによって、IFNβの誘導が回復した。また、L929-sh-cGAS細胞においてSTINGもしくはMAVSを発現させることにより、またはこれらの細胞へcGAMPを送達することにより、IFNβが誘導された。対照的に、cGASの発現またはcGAMPの送達は、L929-shSTING細胞においてIFNβを誘導できなかったが、STINGまたはMAVSの発現はこれらの細胞におけるIFNβの誘導を回復した。定量的RT-PCR分析から、対応するshRNAを安定的に発現するL929細胞株においてcGASおよびSTINGをノックダウンすることの特異性および効率性が確認された。これらの結果から、cGASがSTINGの上流で機能し、サイトゾルDNAによるIFNβの誘導に必要とされることが示唆される。
cGASがcGAMP合成を触媒するのに十分であるかどうか試験するため、本発明者らは、HEK293T細胞においてFlagタグ付のh-cGASを発現させ、見掛け上均質になるまでそれを精製した。HT-DNAの存在下で、精製したc-GASタンパク質はcGAMP活性の産生を触媒し、それによって、透過処理したRaw264.7細胞におけるIRF3の二量体化を刺激した。DNase-I処理によってこの活性は消失した。cGAS活性は、RNAポリ(I:C)ではなく、ポリ(dA:dT)、ポリ(dG:dC) 、およびISDを含む他のDNAによっても刺激された。cGASによるcGAMPの合成には、CTPまたはUTPではなく、ATPおよびGTPの両方が必要とされた。これらの結果から、精製cGASタンパク質のシクラーゼ活性が、RNAではなく、DNAにより刺激されたことが示唆される。
DNAによるcGAS活性の刺激から、c-GASがDNAセンサーであることが示唆される。実際に、GST-RIG-I N末端[RIG-I(N)]ではなく、GST-m-cGASおよびGST-h-cGASの両方がビオチン化ISDによって沈降された。対照的に、ビオチン化RNAはcGASに結合しなかった。欠失分析から、C末端断片の残基213〜522番ではなく、残基1〜212番を含むh-cGAS N末端断片が、ISDに結合することが示された。残基161〜522番を含むさらに長いC末端断片がISDに結合したことから、配列161〜212番がDNA結合にとって重要である可能性が示唆される。しかしながら、h-cGAS由来の残基161〜212番の欠失では、ISD結合があまり損なわれなかったことから、cGASがN末端の位置に別のDNA結合ドメインを含むことが示唆された。実際に、残基1〜160番を含むN末端断片もISDに結合した。このように、cGASはN末端の位置に2つの別々のDNA結合ドメインを含みうる。それにもかかわらず、残基1〜212番を含むh-cGASのN末端は、DNAを結合するのに必要かつ十分の両方であることが明らかである。
cGASがサイトゾルDNAセンサーであるかどうか判定するため、本発明者らは、THP1細胞からサイトゾルおよび核の抽出物を調製し、免疫ブロッティングにより内因性h-cGASの局在について分析した。h-cGASはサイトゾル抽出物において検出されたが、核抽出物においてはほとんど検出できなかった。THP1抽出物を分画遠心法にさらに供して、細胞内小器官を互いにおよびサイトゾルから分離した。ほぼ同じ量のh-cGASがS100およびP100 (100,000×gの遠心分離後のペレット)において検出され、このタンパク質が細胞質中で可溶性であるものの、かなりの割合のタンパク質が軽小胞(light vesicle)または細胞小器官と結び付いていることが示唆された。ミトコンドリアおよびERを含有することが、それぞれVDACおよびSTINGの存在から裏付けられたP5においては、cGASタンパク質は検出されなかった。cGASはまた、主にERおよび重小胞(heavy vesicle)を含有していたP20おいて検出できなかった。
本実施例において、本発明者らは、定量的質量分析を従来のタンパク質精製と組み合わせて、未精製の細胞抽出物から部分的に精製された生物学的に活性なタンパク質を同定するという戦略を開発した。この戦略は一般に、非常に存在量が少ないか、活性が不安定であるか、または出発材料が不十分であることに起因して、均質になるまで精製することが難しいタンパク質に適用可能である。原理の証明として、本発明者らはこの戦略を用いて、マウスタンパク質E330016A19を、cGAMPを合成する酵素として同定した。この発見は、魚類からヒトまで保存されている大きなcGASファミリーの同定につながり、細菌、古細菌、および原生動物でしか以前に見出されていなかった、環状ジヌクレオチドを合成する進化的に保存された酵素を、脊椎動物が含むということを正式に実証した(11〜13)。コレラ菌は、NTaseドメインを含むが哺乳動物cGASに対する有意な一次配列相同性を欠くコレラ菌のシクラーゼDncV (VC0179)を通じて、cGAMPを合成することができる(12)。
微生物感染の先天性免疫感知は、Toll様受容体(TLR)のような膜タンパク質ならびにNOD様受容体(NLR)およびRIG-I様受容体(RLR)のようなサイトゾルタンパク質を含む生殖細胞系にコードされたパターン認識受容体によって媒介される(Iwasaki and Medzhitov, 2010; Ronald and Beutler, 2010; Takeuchi and Akira, 2010)。実質的に全ての感染性微生物が、その生活環において核酸を含み、かつ必要とするため、先天性免疫系は宿主防衛の中心戦略として微生物のDNAおよびRNAを認識するように進化した。具体的には、いくつかのTLRがエンドソーム内腔中のRNAまたはDNAを検出するためにエンドソーム膜上に局在するのに対して、RLRは、細胞質中のウイルスおよび細菌のRNAの検出に関与している。
ヌクレオチド間の2'-5'および3'-5'の両方のホスホジエステル結合は、天然に存在することが知られているが、2'-5'結合はあまり多く見られない。cGASにより産生された天然cGAMPの内部ホスホジエステル結合は、まだ決定されていない。本発明者らはそれゆえ、全4つの可能なホスホジエステル結合を含むcGAMP分子を化学的に合成した(表S1)。公表されている方法を改変した手順を用いて、cGAMPアイソフォームの化学合成を行った(Gaffney et al., 2010; Zhang et al., 2006)。簡単にするために、本発明者らは、ホスホジエステル結合を形成するGMPのOH位置とそれに続くAMPのOH位置にしたがって、これらのcGAMP分子を命名する; 例えば、2'3'-cGAMPは、GMPの2'-OHとAMPの5'-リン酸との間にホスホジエステル結合を、およびAMPの3'-OHとGMPの5'-リン酸との間に別のホスホジエステル結合を含む。本発明者らはまた、精製cGASタンパク質を用いて、DNAの存在下でATPおよびGTPから天然cGAMPを酵素的に合成した(Sun et al., 2013)。精製されたcGAS生成物および合成cGAMP異性体を、核磁気共鳴(NMR)分光学によって分析した。際立っていたのは、cGAS生成物の1H NMRスペクトルが、合成2'3'-cGAMPのものとは同一であったが、他のcGAMP異性体のものとは異なっていたことである。特に、アノマープロトン(H1')は、3'-リン酸による一重線および2'-リン酸による二重線であった。一貫して、2',3'-cGAMPのリン酸だけが、天然cGAMPのものと同じ31P NMR化学シフトを有していた。本発明者らはまた、高い分解能および質量精度を有する機器Q-Exactiveを用いて、天然cGAMPおよび合成cGAMPの質量分析を行った。これらの一価分子([M+H]+)の各々の全質量は675.107であり、cGAMPの理論的質量と厳密に一致していた。高エネルギー衝突解離(HCD)を用いて断片化されたcGAS生成物のタンデム質量(MS/MS)スペクトルは、合成2'3'-cGAMPのものと同一であり、2'2'-cGAMPおよび3'3'-cGAMPのものとほぼ同じであったが、同一ではなかった。3'2'-cGAMPのMS/MSスペクトルは、2'3'-cGAMPおよびcGAS生成物のものと最も異なるように思われた。逆相HPLC分析により、天然cGAMPが2'3'-cGAMPと同時に溶出するが、他のcGAMP分子とは同時に溶出されないことが示された。本発明者らはまた、円偏光二色性(CD)によりcGAS生成物の立体配置を判定し、それがD-リボースに由来することを確認した。天然cGAMPのCDスペクトルは、2'3'-cGAMPのものとよく重なった。近紫外線(UV) CDスペクトルから、4種のcGAMPが有意に異なる高次構造をとり、2'3'および2'2'-cGAMPが、3'2'-および3'3'-cGAMPのものとは異なるCDバンドパターンを形成することが示唆される。まとめると、これらの結果は、cGASがインビトロにおいて2'3'-cGAMPを合成するという決定的証拠を提供する。
哺乳動物細胞が、混合ホスホジエステル結合を含む内因性cGAMPを産生しうるかどうか試験するため、本発明者らは、マウス細胞株L929およびヒト単球THP1にニシン精巣DNA (HT-DNA)をトランスフェクションし、その後、細胞溶解物を95℃で加熱してタンパク質を変性させ、上清を質量分析による内因性cGAMPの分析のために調製した(Wu et al., 2013)。両細胞株由来の内因性分子のMS/MSスペクトルは、cGAS生成物および2'3'-cGAMPのものと同一であったことから、内因性二次メッセンジャーは2'3'-cGAMPであることが示唆された。
本発明者らは、等温滴定熱量計実験を行って、天然cGAMPおよび合成cGAMPへのSTINGの結合の親和性(Kd)を測定した。細菌の二次メッセンジャー環状di-GMPへの結合を媒介することが以前に示されたヒトSTINGの残基139〜379番を包含するC末端ドメイン(CTD) (Burdette et al., 2011; Huang et al., 2012; Ouyang et al., 2012; Shang et al., 2012; Shu et al., 2012; Yin et al., 2012)を大腸菌において発現させ、ITC実験のために見掛け上均質になるまで精製した。以前の報告と一致して、本発明者らは、c-di-GMPが1.21 uMのKdでSTINGに結合することを見出した。興味深いことに、天然cGAMPも合成2'3'-cGAMPもともに、曲線適合が困難であるような高い親和性でSTINGに結合した。さらに、発熱過程であるc-di-GMPの結合とは異なり、STINGへの天然cGAMPおよび2'3'-cGAMPの結合が吸熱性であったことから、そのエネルギーがSTINGの立体構造変化のために用いられうることが示唆された(下記参照)。STINGに対する天然2'3'-cGAMPおよび合成2'3'-cGAMPのKdを得るため、本発明者らは、さまざまな量のこれらの化合物を競合体として、STING-c-di-GMP複合体へ滴定した。これらの測定により、cGAS生成物に対して4.59 nMおよび2'3'-cGAMPに対して3.79 nMのKdが得られた。競合実験は、3'2'-cGAMPについても行った。というのは、STINGへのその結合がほとんど熱変化を生じなかったからである。この化合物は1.61 uMのKdでSTINGに結合する。2'2'-および3'3'-cGAMPをSTINGに対して直接滴定し、Kd値はそれぞれ、287 nMおよび1.04 uMであると算出された。したがって、2'3'-cGAMPのKdはc-di-GMP、3'2'-cGAMPおよび3'3'-cGAMPのKdよりもおよそ300倍低く、2'2'-cGAMPのKdよりもおよそ75倍低かった。
本発明者らは、さまざまな量のcGAMP異性体およびc-di-GMPをL929細胞へ送達し、q-RT-PCRによってIFNβの誘導を測定した。cGAMP分子は、15 nMから42 nMに及ぶEC50でIFNβを誘導したが、c-di-GMPは500 nM超のEC50を有していた。したがって、さまざまな環状ジヌクレオチドの結合親和性は、細胞に基づくアッセイ法においてそのEC50とあまり相関していないように思われた。この理由は明らかではないが、異なる化合物は細胞中で異なる安定性または分布を有する可能性がある。それにもかかわらず、これらの実験は、cGAS生成物2'3'-cGAMPがSTINGに対する高親和性リガンド(Kd: およそ4 nM)および細胞における強力なIFNβ誘導因子(EC50: およそ20 nM)であるという直接的な証拠を提供する。
本発明者らは、精製cGAS生成物とともにSTING C末端ドメイン(CTD) (残基139〜379番)をC2空間群で同時に結晶化した。この複合体の構造を、探索モデルとしてアポSTING構造(PDBコード: 4F9E)を用いた分子置換によって解析し、1.88Åの分解能にまで精緻化した(表M1)。結晶学的な非対称単位の中に1つのSTINGプロトマーが存在し、これは、結晶学的な二回対称性でつなげられた、別のプロトマーと一緒に蝶形の二量体を形成する。結合したcGAMP分子は二回軸に位置する(詳細は下記を参照のこと)。STINGの秩序だった領域(Asn152からGlu336まで)は、4本のαヘリックスによって囲まれた中央のねじれβシートにより特徴付けられる、アポSTINGと類似の全体構造をとる。しかしながら、cGAMPとの複合体中のSTINGは、単量体の構造および二量体の配置の両方でアポSTINGとはいくつかの著しい違いを示す。アポ二量体と比べて、複合体構造の二量体中の2つのプロトマーは、cGAMP結合部位に対してかなり内側に回転する。このさらに接近した配置によって、2つのプロトマーの間にcGAMPを包囲するさらに深いポケットが作り出される。さらに、cGAMP結合部位は、2つのプロトマーの各々の残基219〜249番によって形成される連結ループおよび四本鎖逆平行βシートの蓋によって覆われる。対照的に、アポ構造中のこのセグメントはほとんどが無秩序である(Ouyang et al., 2012; Yin et al., 2012)。βシートの形成は結晶充填によるものではない。蓋内のドメイン間相互作用は、Tyr245の側鎖とGly234の主鎖カルボニル酸素原子との間の、Ser243の側鎖とLys236の主鎖アミド窒素原子との間の、ならびにAsp237およびLys224の側鎖の間の、いくつかの有極接点の対を伴う。
結晶学的な二回軸は非対称性の2'3'-cGAMP分子を貫通するので、cGAMPは二回対称性でつなげられた2つの幾何学的配置をとるはずである。これは、STING二量体中の2つのプロトマーが、グアニジン部分またはアデノシン部分のいずれかと相互作用する確率が等しいと予想されるという事実と一致している。本発明者らはそれゆえ、cGAMPおよびいくつかの周囲アミノ酸残基に関して2つの代替的な立体構造を0.5の占有率で割り当てた。精緻化された構造のシミュレーテッドアニーリングオミットマップ(simulated annealing omit map)から、cGAMPに対しての適切な密度が示される。2'3'-cGAMPは、電子密度図によく適合するが、他のアイソフォームはそうでない。STINGに結合したc-di-GMPと比べて、cGAMPは、STING二量体界面間の隙間においておよそ2.5Åさらに深くに位置する。さらに、蝶形の2つの翼は、2つのSTINGプロトマーのさらに接近した配置により、STING:cGAMP構造中で互いにおよそ20Åさらに近くに位置する。cGAMP結合ポケットのさらなる分析から、cGAMPが、広範な極性および疎水性相互作用によって十分に配位されることを示す。cGAMPプリン塩基群の環は、周囲の4つの芳香族残基、すなわち2つのプロトマーの各々に由来するTyr240およびTyr167に積み重なる。特に、cGAMPの2つのαリン酸基は、2つのプロトマーに由来する両方のArg238および1つのプロトマーに由来するArg232と接する。GMPの遊離3'-OHは、ポケットの下方部分由来の2つのSer162残基の方に向いている。グアニン塩基は、Glu260およびThr263の側鎖、ならびにVal239の主鎖カルボニル酸素と直接相互作用する。これらの独特な有極接点は、2'3'-cGAMPがSTINGに対する特異的かつ高親和性リガンドである理由を説明する。加えて、cGAMP結合に関与するβシート由来の残基(Arg232、Arg238、Val239)は、蓋の形成およびSTINGのさらなる活性化を制御する可能性が高い。
環状di-GMPが結合したSTINGの結晶構造に関する3つの過去の報告では、Arg232をヒスチジンと置換された希少なヒト変種を用いていた(Ouyang et al., 2012; Shu et al., 2012; Yin et al., 2012)。ヒト集団由来DNAの広範な配列決定により、Arg232対立遺伝子はよく見られるため、野生型STINGと見なされるべきであると示されている(Jin et al., 2011)。STINGのH232変種の使用は、これらの研究においてc-di-GMPがSTINGの有意な立体構造変化を誘導しなかった理由を説明しうる(Ouyang et al., 2012; Shu et al., 2012; Yin et al., 2012)。マウスSTINGのArg231 (ヒトSTINGにおけるArg232に等しい)のアラニンへの変異によって、DNAではなく、環状di-GMPによるIFNβの誘導が消失することが過去の報告によって示された(Burdette et al., 2011)。しかしながら、STING-cGAMP複合体に関する本発明者らの結晶構造に基づけば、Arg232のヒスチジンへの変異は、cGAMPの結合およびSTINGによる下流のシグナル伝達をかなり弱めるものと予想され、Arg232のアラニンへの変異はさらにいっそう有害なはずである。本発明者らはそれゆえ、2組の実験においてSTINGのArg232の機能を調べた。第一に、本発明者らは、L929細胞においてRNAiにより内因性STINGをノックダウンし、それをヒトSTINGのWT、R232A、またはR232Hと置き換えた。これらの安定細胞株にHT-DNAをトランスフェクションし、または安定細胞株を2'3'-cGAMPで処理し、その後q-RT-PCRによってIFNβの測定を行った。WT STINGを発現する細胞は、DNAまたはcGAMP刺激に応答してIFNβを誘導することができたが、R232AまたはR232Hのいずれかを発現するものには欠陥があった。対照として、二本鎖RNA類似体ポリ[I:C]は、これらの細胞株の全てでIFNβの発現を刺激した。第二に、本発明者らは、内因性STINGおよびcGASの検出不能な発現を有するHEK293T細胞においてWTまたは変異体STINGを安定的に発現させた(Sun et al., 2013)。細胞に次いで、ヒトcGAS発現プラスミドをトランスフェクションし、その後にIFNβ RNAの測定を行った。R232A変異体ではなく、WT STINGは、cGASによるIFNβの誘導を支持することができた。R232H変異体は部分的に欠陥があった。これはおそらく、正電荷を持つヒスチジンが、弱くともArg232の機能のいくつかの代わりになりうるからである。RIG-I経路の不可欠なアダプタータンパク質MAVS (Seth et al., 2005)は、これらの細胞株の全てにおいてIFNβを誘導することができた。まとめると、本発明者らの構造的および機能的データから、STINGの機能におけるArg232の重要な役割が強く示唆され、不可欠なサイトゾルDNAセンサーとしてのcGASの役割がさらに強調される。
本発明者らの以前の研究から、cGASは、サイトゾルDNAセンサーと同定され、かつ基質としてATPおよびGTPを用いてcGAMPを合成するシクラーゼと同定された(Sun et al., 2013; Wu et al., 2013)。cGAMPはその後、STINGに結合しかつSTINGを活性化する二次メッセンジャーとして機能する。ここで本発明者らは、化学合成およびいくつかの生物物理学的手法を利用して、cGAS生成物の内部ホスホジエステル結合をさらに特徴付け、それが2'3'-cGAMPであることを決定付けた。その後、Gaoらが、cGASの構造をそのアポ結合型およびDNA結合型として報告したが、それによって、cGASが実際に、ATPおよびGTPからのcGAMP合成を触媒するDNA活性化環状GMP-AMPシンターゼであることが確認された(Gao et al., 2013)。このエレガントな研究はまた、DNA結合がcGASの活性化につながる構造機構を解明した。異なる手法を用いて、Gaoらはまた、切断型cGASタンパク質がインビトロにおいて2'3'-cGAMPを合成することを見出した。しかしながら、Gaoらは、2'3'-cGAMPが何らかの生物学的または生化学的活性を有するかどうか試験しなかった。またGaoらは、内因性2'3-cGAMPが哺乳動物細胞において産生されるかどうか示していなかった。本報告において、本発明者らは、DNAによるマウスおよびヒト細胞の刺激が、内因性2'3'-cGAMPの産生につながることを示す。さらに、本発明者らは、2'3'-cGAMPが他のcGAMP異性体およびc-di-GMPよりもはるかに高い親和性でSTINGに結合することを実証する。本発明者らはさらに、2'3'-cGAMPおよび他のcGAMP異性体が、細胞においてIFNβを誘導する際にc-di-GMPよりもはるかに強力であることを示す。
2'3'-cGAMP結合ヒトSTING CTD構造の座標は、RCSBタンパク質データバンクに寄託されている(PDB: 4KSY)。
HIVを含むレトロウイルスは、先天性免疫応答を活性化しうるが、レトロウイルスに対する宿主センサーは、ほとんど知られていない。ここで本発明者らは、HIV感染が環状GMP-AMP (cGAMP)シンターゼ(cGAS)を活性化してcGAMPを産生し、これがアダプタータンパク質STINGに結合し、かつそれを活性化して、I型インターフェロンおよび他のサイトカインを誘導することを示す。インテグラーゼではないHIVの逆転写酵素の阻害剤が、ウイルスによるインターフェロン-β誘導を抑止したことから、逆転写されたHIV DNAが先天性免疫応答を誘発することが示唆された。マウスまたはヒト細胞株におけるcGASのノックアウトまたはノックダウンは、HIV、マウス白血病ウイルス(MLV) 、およびサル免疫不全ウイルス(SIV)によるサイトカインの誘導を遮断した。これらの結果から、cGASがレトロウイルスDNAを検出すること、ならびにcGASがHIVおよび他のレトロウイルスの先天性免疫センサーであることが示唆される。
21.クローン#18は、3つの染色体全てにおいて、フレームシフト変異を有している。フレームシフトに加えて、クローン#36は、1つの染色体において、触媒ドメイン内の3アミノ酸(215〜217)を除去する9-bpの欠失を含んでいた一方、クローン#94は、1つの染色体において12-bpの欠失を有し、かつ別の染色体において18-bpの欠失を有し、これによりそれぞれ触媒ドメイン内の4アミノ酸(214〜217)および6アミノ酸(212〜217)が除去された。
動物細胞の細胞質への微生物DNAの侵入は、感染を除去するのに役立つ宿主免疫反応のカスケードを誘発する; しかしながら、細胞質中の自己DNAが自己免疫疾患を引き起こすことがある。生化学的手法により、先天性免疫応答を誘発するサイトゾルDNAセンサーとしての環状GMP-AMP (cGAMP)シンターゼ(cGAS)の同定につながった。ここで本発明者らは、線維芽細胞、マクロファージ、および樹状細胞を含む、cGAS欠損(cGas-/-)マウス由来の細胞が、DNAトランスフェクションまたはDNAウイルス感染に応答してI型インターフェロンおよび他のサイトカインを産生できなかったことを示す。cGas-/-マウスは、野生型マウスよりも単純ヘルペスウイルス-1 (HSV1)による致命的感染に感受性が高かった。本発明者らはまた、cGAMPが、抗原特異的T細胞活性化および抗体産生を後押しするアジュバントであることを示す。
Claims (11)
- 所定量の環状GMP-AMP (cGAMP)を含む、薬学的組成物であって、該cGAMPがGMPの2'-OHとAMPの5'-リン酸との間のホスホジエステル結合、および、AMPの3'-OHとGMPの5'-リン酸との間に別のホスホジエステル結合を含む、該薬学的組成物。
- 所定量の2'3'-cGAMP、2'2'-cGAMP、または3'2'-cGAMPを含む、薬学的組成物。
- cGAMPが2'3'-cGAMPである、請求項1記載の薬学的組成物。
- 他の環状ジヌクレオチドを本質的に含まない、請求項1記載の薬学的組成物。
- 請求項1記載の薬学的組成物を含む、アジュバント。
- 所定の免疫原および請求項1記載の薬学的組成物を含む、ワクチン。
- 免疫原が標的病原体に選択的である、請求項6記載のワクチン。
- 免疫応答を誘導または促進する方法に用いるための請求項1記載の薬学的組成物であって、該方法がそれを必要としている哺乳動物に該組成物の有効量を投与する段階を含む、薬学的組成物。
- 粘膜(舌下もしくは鼻腔内)に、筋肉内に、または皮下に投与される、請求項8記載の薬学的組成物。
- 環状GMP-AMPシンターゼ(cGAS)、ATPおよびGTPを含む混合物を、シンターゼがATPおよびGTPをcGAMPに触媒転換する条件の下で、形成する段階であって、シンターゼ、ATPおよびGTPが所定の量で存在する、段階
を含む、または結果として生じるcGAMPを単離もしくは検出する段階をさらに含む、請求項1記載の環状GMP-AMP (cGAMP)を生成する方法。 - 前記混合物がDNAをさらに含み、かつ転換がDNA依存的である、請求項10記載の方法。
