Description
(1)鋳型となるRNA、逆転写酵素、PCR増幅酵素、少なくとも2種のプライマー、及び二本鎖DNA分解酵素を含む溶液を調製する工程と、
前記溶液をインキュベーションする工程と、

実施例1において、エキソヌクレアーゼIII(1U)を添加しなかった以外は、実施例1と同様に1step−RT−PCRを行い、その後に、融解曲線解析を行った。
Claims (8)
- 鋳型となるRNA、逆転写酵素、PCR増幅酵素、少なくとも2種のプライマー、及び二本鎖DNA分解酵素を含む溶液を調製する工程と、
前記溶液をインキュベーションする工程と、
を含む1ステップRT−PCR法。 - 前記二本鎖DNA分解酵素が、直鎖状二本鎖DNA分解酵素である、請求項1に記載の1ステップRT−PCR法。
- 前記直鎖状二本鎖DNA分解酵素が、エキソヌクレアーゼである、請求項2に記載の1ステップRT−PCR法。
- 前記直鎖状二本鎖DNA分解酵素が、エキソヌクレアーゼIII、T7エキソヌクレアーゼ、及びランダムエキソヌクレアーゼからなる群から選ばれる、請求項2又は3に記載の1ステップRT−PCR法。
- 逆転写酵素が、Avian Myeloblastosis Virus由来の逆転写酵素、Moloney Murine Leukemia Virus由来の逆転写酵素、及びそれらの遺伝子改変型逆転写酵素からなる群から選ばれる、請求項1〜4のうちのいずれかに記載の1ステップRT−PCR法。
- 逆転写酵素、PCR増幅酵素、及び二本鎖DNA分解酵素を含む1ステップRT−PCR反応用キット。
- 逆転写酵素、PCR増幅酵素、及び二本鎖DNA分解酵素を含む1ステップRT−PCR反応用組成物。
- 鋳型となるRNA、逆転写酵素、PCR増幅酵素、少なくとも2種のプライマー、及び二本鎖DNA分解酵素を含む溶液を調製する工程と、
前記溶液をインキュベーションする工程と、
を含む1ステップRT−PCR法におけるプライマーダイマーの抑制方法。
