Abstract
Description
1)組換えて発現させたSODは、耐酸性・耐アルカリ性が不十分である。
2)生成されたSODは、消化管内のペプシンやトリプシンによって分解され、失活し易い。
<1>高収率の突然変異型耐高温性SOD菌株の取得
好熱菌HB27(米国ATCC(American Type CultureCollection)細胞バンクから購入)のSODをコードする遺伝子を鋳型とし、以下の配列のプライマーで増幅させ、目標遺伝子を得た。
フォワードプライマー:5’−AAGAATTCATGCCGTACCCGTTCAAGCT−3’(配列番号1);
リバースプライマー:5’−CTGTCGACTCAGGCTTTGTTGAAGAAC−3’(配列番号2)。
回収キット(生工生物工程上海(股ふん)有限公司から購入)を用いて増幅後の産物を回収した。それを2種類の酵素(EcoRIとSalI)で切断し(Double Digestion)、同様の2種類の酵素で切断されたプラスミドベクターpET28a(+)(生工生物工程上海(股ふん)有限公司から購入)に連結させた。該組換えプラスミドをコンピテント化大腸菌BL21(DE3)(生工生物工程上海(股ふん)有限公司から購入)に形質転換し、スクリーニング、配列解析を経た後、高収率の突然変異型耐高温性SOD(以下、M型−SODと略称する)の菌株を得た。配列決定された、M型−SODをコードする遺伝子の具体的にヌクレオチド配列を以下に示す。
発酵工程は、一次菌種培養、二次菌種培養、タンク発酵、発現誘導という4つのステップを経て、最終的に発酵産物としてM型−SODを得た。具体的な過程は以下の通りである。
<1>耐熱性の比較
W型−SOD(Jianguo Liuら,Extremophiles, 2011:15:221 226,以下は同様)、本発明で提供されるM型−SODおよび通常SOD(Sigma、MFCD00132404)について、その熱安定性を比較した(Jianguo Liuら,Extremophiles, 2011:15:221 226に記載の条件と完全に同様の条件下で、異なる温度で1時間処理し、残留酵素の活性を測定した)。結果を図1に示す。図1から分かるように、本発明で提供されるM型−SODは、より幅広い温度範囲において、極めて高い安定性が維持されている。
W型−SOD、M型−SODおよび通常SOD(Sigma、MFCD00132404)について、異なるpH値での安定性を比較した(Jianguo Liuら,Extremophiles, 2011:15:221 226に記載の条件と完全に同様の条件で)。結果を図2に示す。図2から分かるように、M型−SODは、pH12の場合であっても97%の活性を有するが、W型−SODは、pH11の時点で40%未満の活性しかなかった。つまり、M型−SODは極めて突出した耐酸性・耐アルカリ性を有することが分かる。
中国薬典2010年版の基準に沿って、人工疑似胃液を作製した。即ち、希塩酸(塩酸234mLに水を加え、1000mLになるまで希釈したもの。該希釈液は9.5%〜10.5%のHClを含む)16.4mLに、水約800mLとペプシン(SINOPHARM、20141209)10gを加え、均一に振とうした後、1000mLになるまで水を加えて希釈し、人工疑似胃液を得た。
10U/mL〜300U/mLのトリプシン溶液(上海瑞永生物科技公司、RM1021−100g)を用い、M型−SODと通常SODとを、37℃で3時間インキュベートしたところ、M型−SODは活性が損なわれないが、通常SODは活性を約60%失ったことが分かった。これは、M型−SODは、トリプシンによる分解に対する耐性が極めて強いことを意味する。具体的には図4に示す。
<実験動物>
ゼブラフィッシュモデルは、以下の要件を満たしているため、疾病研究や薬物スクリーニングにおいて幅広く使用されている。(1)サンプルの大量供与が可能であるため、薬物のハイスループットスクリーニングが可能となる。(2)免疫学やゲノム学等において、ヒトとは高度な保全性を有する。(3)良好な追跡性を有する。(4)炎症に共通する最も重要な病理的特徴が明白、明確に表れる。ゼブラフィッシュは、国際的に公認されている最も重要な、薬物スクリーニング用動物モデルの1つである。ゼブラフィッシュの透明な体は、非常に強い造影特性を有するため、研究者による遺伝子操作は扱い易くなる。そのため、ゼブラフィッシュは、様々な炎症に対する反応を研究するための理想的なモデルとなっている。ゼブラフィッシュの消化系は哺乳類動物と極めて類似し、同様に肝臓、膵臓、胆嚢、および、吸収と分泌機能を有する線状分節型腸管を含む。また、腸上皮は、腸管の全域に亘って機能しており、且つ、吸収性細胞、杯状細胞、内分泌性細胞等、哺乳類動物にも見つけられる上皮細胞を含んでいる。したがって、近年、ゼブラフィッシュは、粘膜炎症、特に消化管の粘膜炎症を含む様々な炎症を研究するための典型的な動物モデルとなっている。中でも、最も代表的なものは炎症性腸疾患(IBD)に関するゼブラフィッシュ(動物)モデルである。
実施例1で作製したM型−SOD(白色の粉末;使用直前に、超純水を用いて濃度20mg/mLの母液(mother solution)に調製する、即ち、その都度調製する)
プレドニゾン(白色粉末;バッチ番号28778;上海晶純実業有限公司より入手;使用直前に、100%ジメチルスルホキシドを用いて濃度15mg/mLの母液に調製する)
トリニトロベンゼンスルホン酸(以下、TNBSと略称する;茶褐色の液体;バッチ番号D1326018;上海晶純実業有限公司により入手)。
解剖顕微鏡(SZX7;OLYMPUS社、日本)
電動ズーム式連続倍率変更型蛍光顕微鏡(AZ100;Nikon社)
6穴プレート(Nest Biotech社)
メチルセルロース(Aladdin社、Shanghai、中国)。
モデル1:受精後3日の野生型AB系ゼブラフィッシュを、最終濃度4.5μMのTNBSを含む養魚用水で48時間培養し、ゼブラフィッシュの結腸炎モデルを作製した(具体的なプロセスは、A. Jemal, R. Siegel, J. Xuら. Ward. Clinicians, 2010,277-300;および、Intestinal Upregulation of Melanin-Concentrating Hormone inTNBS-Induced Enterocolitis in Adult Zebrafish. Plos One, 2013,8(12):1524-1528を参照)。モデルを作製した結果、モデルコントロールグループの腸内腔面積(197660)は、正常コントロールグループ(130711)と比べ、p<0.001であった。これは、モデル構築の成功を意味する。当該モデルは後述する実験1に用いられる。
<実験方法>
(1)M型−SODの非致死最大濃度(MNLC)の特定
異なる複数の濃度に設定したM型−SODを用いてゼブラフィッシュを処理(48時間処理)した。
表1の濃度−致死データによると、M型−SODのMNLCは150μg/mLであった。15μg/mL、50μg/mLおよび150μg/mLの3つの濃度を選択し、薬効学的評価実験に供する。
モデルコントロールグループ:TNBSでゼブラフィッシュモデルの結腸炎を誘発した(モデル1)。
各実験グループは何れも30匹のゼブラフィッシュを処理した(処理時間は何れも48時間)。M型−SODでの処理が完了した後、グループごとに、ランダムに15匹のゼブラフィッシュを選び、顕微鏡で観察し、撮影した画像を保存した。画像分析ソフトで画像を分析し、ゼブラフィッシュの腸内腔面積を計算すると共に、定量分析を行った。統計学的に処理した結果を下記式で表す。
モデルコントロールグループの腸内腔面積(197660)は、正常コントロールグループ(130711)と比較すると、p<0.001であった。これは、モデル構築が大いに成功したことを意味する。プレドニゾン剤を用いた陽性コントロールの腸内腔面積(147876)は、モデルコントロールグループと比べ、p<0.001であった。また、ゼブラフィッシュの結腸炎に対する、プレドニゾンによる治療効率は74%であった。また、濃度15μg/mL、50μg/mLおよび150μg/mLの各M型−SODは、結腸炎に対する治療効率が、それぞれ66%、82%および84%であった。各グループは、モデルコントロールグループと比べ、p<0.001であった。これは、M型−SODが結腸炎に対して顕著な治療効果を有することを意味する。詳細を表2、図5、図6、図7および図8に示す。
<実験方法>
テストのためにM型−SODの濃度を3つ選択した(15μg/mL、50μg/mLおよび150μg/mLの最終濃度で養魚用水に入れた)。
モデルコントロールグループのゼブラフィッシュの腸管における好中球細胞の個数(9.4)は、正常コントロールグループ(1.6)と比べ、p<0.001であった。これは、モデル構築が大いに成功したことを意味する。プレドニゾン剤を用いた陽性コントロールにおける好中球細胞の個数(3.8)は、モデルコントロールグループと比べ、p<0.001であった。これは、プレドニゾンが、結腸炎の炎症解消に対して促進作用を有することを意味する。その炎症解消の促進率は71%であった。また、濃度15μg/mL、50μg/mLおよび150μg/mLの各M型−SODは、結腸炎の炎症解消に対する促進率がそれぞれ34%、49%よび74%であった。各グループは、モデルコントロールグループと比べ、それぞれp<0.05、p<0.01、およびp<0.001であった。これは、M型−SODが結腸炎の炎症解消に対して顕著な促進作用を有することを意味する。詳細を表3、図9、図10および図11に示す。

M型−SODで処理するグループ:処理方法、グループ別での濃度設定は、実験1と同様である。
正常コントロールグループのゼブラフィッシュは、腸粘膜が滑らかで無傷であり、腸のヒダが明確に見える。モデルコントロールグループのゼブラフィッシュは、腸内が拡張し、腸粘膜が薄くなり、腸のヒダが消え、粘膜の糜爛や潰瘍が見られた。また、プレドニゾン剤で治療した陽性コントロールは、組織学の観点から、腸粘膜が明らかに改善された。また、濃度15μg/mL、50μg/mLおよび150μg/mLのM型−SODで処理したグループは、腸内腔面積や腸のヒダがほぼ回復し、腸粘膜の炎症が顕著に改善され、炎症による目立った潰瘍が見られなかった。詳細を図12に示す。
炎症性腸疾患(Inflammatory bowel disease;IBD)は、よく見られる疾患であり、通常、クローン病(Crohn's disease;CD)および潰瘍性結腸炎(ulcerative colitis;UC)と総称される。クローン病は、局所性腸炎、分節性腸炎または肉芽腫性小腸結腸炎とも呼ばれる。この疾患は、原因不明な胃腸の慢性肉芽腫性疾患であり、回腸末端および近位結腸によく見られ、段階的または跳躍的な病変分布を呈し、腸壁全層に亘る炎症である。潰瘍性結腸炎は、非特異性潰瘍性結腸炎とも呼ばれ、原因不明な直腸および結腸の慢性炎症性疾患である。その病変は、主に大腸粘膜および粘膜下組織に及び、最も一般的な患部は直腸および遠位結腸である。但し、重度な病変の場合、結腸全体および回腸末端にまで及ぶことがある。上記疾患は、通常、下痢、腹痛、ないしは血便といった人体症状が現れる。現在、IBDの発病メカニズムであろうものは次のように考えられている。何らかの遺伝的な決定要素によって罹患し易くなっている敏感性個体は、感染因子または腸内腔の抗原の作用により、粘膜関連リンパ組織が刺激され、アップレギュレーション化したT細胞反応が引き起こされる。これにより、様々なサイトカインによるネットワークが活性化し、局所組織の炎症発生、炎症拡大および炎症持続が誘発され、腸壁のダメージおよび対応する臨床症状が引き起こされる。その治療には、糖質ステロイド(GCS)系薬物(プレドニゾン、副腎皮質ホルモン)および抗生物質等がよく使用されるが、治療効果が薄く、副作用が強く、さらに薬物耐性が発生し易い。本発明者らは、本発明における錠剤(1錠あたり、10000UのM型−SODが含まれる)を作製し、本発明による治療効果を検証した。錠剤の作製方法は以下の通りである。
(1)80メッシュ篩で篩分けしたM型−SOD粉末を用意した。
UCでは、特異性に関する診断基準が不足しており、CDでは、診断を確定することが可能な病理組織学的結果(非乾酪性肉芽腫)が得られ難い。そのため、現在、IBDの病因および疾患類型を判断することは比較的困難である。しかし、IBD患者によく見られる臨床症状として、長年繰り返される腹痛、下痢等が現れるため、これらの症状の改善または緩和の有無を観察、記録することは可能である。
毎晩1錠、10日間の連続服用を1クールとした。
有効:10日間の使用において、下痢や腹痛がなく、固形便が排便される。
<実験動物>
遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュ(杭州環特生物科技股ふん有限公司)を自然交尾、繁殖させた。受精後3日の合計810匹を実験グループごとに30匹ずつ振り分けた。なお、実験動物は、28℃の養魚用水(水質:逆浸透水1Lあたり200mgの快速溶解性海塩を添加したもの;導電率480〜510μS/cm、pH6.9〜7.2、硬度53.7〜71.6mg/L CaCO3)中で飼育していた(実験動物の使用許可証番号は、SYXK(浙)2012−0171)。また、飼育の管理は国際AAALACの認可基準を満たしていた。
実施例1で作製したM型−SOD(白色の粉末;使用直前に、超純水を用いて濃度20mg/mLの母液に調製する、即ち、その都度調製する)
インドメタシン(白色粉末;バッチ番号1108939;上海晶純実業有限公司より入手;100%ジメチルスルホキシドを用いて濃度60mMの母液に調製する)

M型−SODを用いてゼブラフィッシュを処理した(処理時間は5時間)。M型−SODのテスト濃度は、15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLの5つとした。各実験グループは何れも30匹のゼブラフィッシュを処理した。実験期間中は、毎日、死亡したゼブラフィッシュの数をカウントし、死亡したゼブラフィッシュを取り除いた。M型−SODでの処理が完了した後、各実験グループのゼブラフィッシュ死亡数を統計することで、MNLCを特定した。結果、M型−SODの濃度が2000μg/mLの場合、5時間作用した後でも、ゼブラフィッシュに対する毒害作用が現れないことが分かった。したがって、2000μg/mLを、SODによる抗炎症効果を評価する最高濃度とした。
<グループ>
実験グループ1:正常対照グループ、具体的には何ら処理されていない、遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュ;

濃度を模索するテストの結果、SODの濃度が2000μg/mLまでの場合、ゼブラフィッシュに対する毒害作用が現れなかった。したがって、2000μg/mLを、SODによる抗炎症効果を評価する最高濃度とした。抗炎症効果を評価するための濃度をそれぞれ、15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLに設定した。
10μMの硫酸銅五水和物を水に溶解させた溶液を用いて、遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュを処理し、ゼブラフィッシュ炎症モデルを構築した(具体的な方法は、d'Alenconら, BMC Biology. 2010,8:151,A high-throughputchemically inducedinflammation assay in zebrafish.を参照)。モデルコントロールグループでは、炎症部位(聴覚細胞領域)における好中球細胞の個数(15.5)は、正常コントロールグループ(1.3)と比べ、p<0.001であった。これは、モデル構築の成功を意味する。
遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュをランダムに選び、6穴プレートの1穴ごとに30匹を入れ、硫酸銅を用いてゼブラフィッシュ炎症モデルに誘発した。M型−SODを水溶液とし、それぞれ15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLの濃度にした。また、陽性コントロールのインドメタシン濃度を60μMにした。穴ごとに3mLの薬液を投入すると共に、正常コントロールグループおよびモデルコントロールグループを設けた。M型−SODによる処理の時間は5時間であった。処理が完了した後、グループごとに、ランダムに10匹のゼブラフィッシュを選び、蛍光顕微鏡で観察し、撮影した画像を保存した。NIKON製NIS−ElementsD 3.10(高級画像処理ソフト)で画像を分析し、炎症化ゼブラフィッシュにおける好中球細胞の個数(N)を計算し、定量分析を行った。試料の炎症解消率の計算式は以下の通りである。
モデルコントロールグループでは、炎症部位の好中球細胞の個数(15.5)は、正常コントロールグループ(1.3)と比べ、p<0.001であった。これは、モデル構築の成功を意味する。また、インドメタシンを用いたグループでは、炎症部位における好中球細胞の個数(1.9)は、モデルコントロールグループと比べてp<0.001であり、その炎症解消率は88%であった。これは、インドメタシンが抗炎症効果を有することを意味する。15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLのM型−SODを用いたグループでは、炎症部位における好中球細胞の個数は、それぞれ14.9、11.8、8.3、3.6、1.4および1.2であり、その炎症解消率は、それぞれ4%、24%、47%、77%、91%および92%であった。モデルコントロールグループと比べ、M型−SODを用いたグループは、15μg/mLおよび50μg/mLのグループを除き、残りのグループが、p<0.05、p<0.01、およびp<0.001であった。これは、本実験の濃度条件下のM型−SODが、硫酸銅で誘発されたゼブラフィッシュ炎症に対して顕著な抗炎症効果を有することを意味する。また、効果と投与量との相関性を表している。詳細を表4、図13、図14および図15に示す。
<濃度グループ>
実験グループ1:正常コントロールグループ、具体的には何ら処理されていない、遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュ;

LPSを注射する方式で、遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュを処理し、ゼブラフィッシュ炎症モデルを構築した。モデルコントロールグループでは、炎症部位(卵黄嚢領域)における好中球細胞の個数(17.8)は、正常コントロールグループ(2.4)と比べ、p<0.001であった。これは、モデル構築の成功を意味する(具体的なプロセスは、Li-LingYangら,Molecules2014,19,2390-2409.Endotoxin MoleculeLipopolysaccharide-Induced Zebrafish Inflammation Model:A Novel ScreeningMethod for Anti-Inflammatory Drugs.を参照)。
遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュをランダムに選び、6穴プレートの1穴ごとに30匹を入れ、LPSを用いてゼブラフィッシュ炎症モデルに誘発した。M型−SODを水溶液とし、それぞれ15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLの濃度にした。また、陽性コントロールのインドメタシン濃度を60μMにした。穴ごとに3mLの薬液を投入すると共に、正常コントロールグループおよびモデルコントロールグループを設けた。5時間処理した後、グループごとに、ランダムに10匹のゼブラフィッシュを選び、蛍光顕微鏡で観察し、撮影した画像を保存した。NIKON社製のNIS−Elements D 3.10(高級画像処理ソフト)で画像を分析し、炎症化ゼブラフィッシュにおける好中球細胞の個数(N)を計算し、定量分析を行った。試料の炎症解消率の計算式は以下の通りである。
モデルコントロールグループでは、炎症部位における好中球細胞の個数(17.8)は、正常コントロールグループ(2.4)と比べ、p<0.001であった。これはモデル構築の成功を意味する。また、インドメタシンを用いたグループでは、炎症部位における好中球細胞の個数(4.9)は、モデルコントロールグループと比べ、p<0.001であり、その炎症解消率は72%であった。これは、インドメタシンが抗炎症効果を有することを意味する。15μg/mL、50μg/mL、150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLのM型−SODを用いたグループでは、炎症部位における好中球細胞の個数は、それぞれ17.7、14.9、12.3、11.7、7.4および6.3であり、その炎症解消率は、それぞれ0%、16%、31%、34%、58%および65%であった。モデルコントロールグループと比べ、M型−SODを用いたグループは、15μg/mLおよび50μg/mLのグループを除き、残りのグループが、p<0.05、p<0.01、およびp<0.001であった。これは、本実験の濃度条件下のM型−SODが、LPSで誘発されたゼブラフィッシュ炎症に対して顕著な抗炎症効果を有することを意味する。また、効果と投与量との相関性を表している。詳細を表5、図16、図17および図18に示す。
<グループ>
実験グループ1:正常コントロールグループ、具体的には何ら処理されていない、遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュ;
実験グループ2:モデルコントロールグループ;
実験グループ3:60μMのインドメタシン剤を用いる陽性コントロール;
実験グループ4:150μg/mLのM型−SODを用いる;
実験グループ5:450μg/mLのM型−SODを用いる;
実験グループ6:1350μg/mLのM型−SODを用いる;
実験グループ7:2000μg/mLのM型−SODを用いる。
遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュの尾ビレを器械で切断し、ゼブラフィッシュ炎症モデルを構築した。モデルコントロールグループでは、炎症部位における好中球細胞の個数(20.2)は、正常コントロールグループ(4.8)と比べ、p<0.001であった。これはモデル構築の成功を意味する。なお、稚魚の尾ビレの一部を切断した稚魚尾創傷モデルは、最も知られているゼブラフィッシュダメージモデルである。本発明者は、このような遺伝子導入ゼブラフィッシュの損傷モデルについて、好中球細胞の特有なペルオキシターゼ触媒の転写を制御しつつ発現させた緑色蛍光タンパク(EGFP)を利用し、損傷部位への好中球細胞の走化性に関する最新メカニズムを研究した(具体的には、J.Yuan, S.Shaham,S.Ledoux,et al. TheC.elegans cell death gene ced-3 encodes a protein similar to mammalianinterleukin-1β-converting enzyme[J].Cell, 1993,641-652.を参照)。
遺伝子導入により好中球細胞を蛍光化したゼブラフィッシュの尾ビレを器械で切断した後、6穴プレートの1穴ごとに30匹をランダムに入れた。M型−SODを水溶液とし、それぞれ150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLの濃度にした。また、陽性コントロールのインドメタシン剤の濃度を60μMにした。穴ごとに3mLの薬液を投入すると共に、正常コントロールグループおよびモデルコントロールグループを設けた。5時間処理した後、グループごとに、ランダムに10匹のゼブラフィッシュを選び、蛍光顕微鏡で観察し、撮影した画像を保存した。NIKON社製のNIS−Elements D 3.10(高級画像処理ソフト)で画像を分析し、炎症化ゼブラフィッシュにおける好中球細胞の個数(N)を計算し、定量分析を行った。試料の炎症解消率の計算式は以下の通りである。
モデルコントロールグループでは、炎症部位における好中球細胞の個数(20.2)は、正常コントロールグループ(4.8)と比べ、p<0.001であった。これは、モデル構築の成功を意味する。また、インドメタシンを用いたグループでは、炎症部位における好中球細胞の個数(7.6)は、モデルコントロールグループと比べ、p<0.001であり、その炎症解消率は62%であった。これは、インドメタシンが抗炎症効果を有することを意味する。150μg/mL、450μg/mL、1350μg/mLおよび2000μg/mLのM型−SODを用いたグループでは、炎症部位における好中球細胞の個数は、それぞれ14.3、9.9、8.4および8.3であり、その炎症解消率は、それぞれ29%、51%、58%および59%であった。モデルコントロールグループと比べ、M型−SODを用いたグループは、p<0.01、およびp<0.001であった。これは、本実験の濃度条件下のM型−SODが、ゼブラフィッシュの尾切断損傷による炎症に対して顕著な抗炎症効果を有することを意味する。また、効果と分量との相関性を表している。詳細を表6、図19、図20および図21に示す。
(実験1 皮膚炎性湿疹に対する本発明による治療の臨床観察)
合計51名の被験者に、本製品(M型−SODをグリセリンに溶解、均一混合し、最終濃度が4000〜6000U/mgになるように調製したもの)を試験的に投与した。実験の内訳は、男性24名および女性27名のうち、年齢5〜15歳の被験者が14名、20〜30歳の被験者が26名、40〜50歳の被験者が6名、50歳以上の被験者が5名であった。
皮膚炎性湿疹の症状として、皮膚が赤色に変わり、重度な場合には、少々腫れ、または丘疹が発生して頂部に水疱が生じ、かゆくなる。掻き破った後、かさぶたが形成される(図22中、上側の写真を参照)。
1日3回、本発明の製品を患者の皮膚表面に塗布し、7日間の使用を1クールとした。
有効:1週間使用した後、皮膚が正常に回復した。
合計47名の被験者に、本製品(M型−SODをグリセリンに溶解、均一混合し、最終濃度が4000〜6000U/mgになるように調製したもの)を試験的に投与した。実験の内訳は、男性22名および女性25名のうち、年齢15〜30歳の被験者が18名、30〜50歳の被験者が23名、50歳以上の被験者が6名であった。
手・足の白癬の症状として、足の指(手の指)の間の皮膚に角化、水疱、丘疹、糜爛、発赤性腫れが発生し、局所的発赤性腫れや、強烈な痒みが常に伴う(図23における上側の2つの写真を参照)。
1日3回、本発明の製品を患者の皮膚表面に塗布し、7日間の使用を1クールとした。
有効:1週間使用した後、皮膚が正常に回復した(図23中、下側の2つの写真を参照)。
合計74名の被験者に、本製品(M型−SODをグリセリンに溶解、均一混合し、最終濃度が4000〜6000U/mgになるように調製したもの)を試験的に投与した。実験の内訳は、男性38名および女性36名のうち、年齢15〜30歳の被験者が34名、30〜50歳の被験者が31名、50歳以上の被験者が9名であった。
抗原に起因する皮膚病である過敏性皮膚炎の主な症状として、人体が何らかの抗原に晒された場合に生じた皮膚の発赤性腫れ、痒み、膨疹、皮の剥け等(図24中、上側の写真を参照)。
1日3回、本発明の製品を患部の皮膚表面に塗布し、7日間の使用を1クールとした。
有効:1週間使用した後、皮膚が正常に回復した(図24中、下側の写真を参照)。
炎症は、感染源、外傷または組織虚血に対する生体の防御反応である。炎症に共通する最も重要な病理学的特徴は、白血球(好中球細胞および大食細胞)の遷移、集結および浸潤である。中でも、好中球細胞の走化、遷移および集結による効果は、急性ダメージおよび炎症反応に対する効果的防御のための生体免疫反応メカニズムである。特に、生体自身の組織が損傷された場合や、体外の物質に侵された場合には、このような防御効果はさらに重要になる。
Claims (13)
- アミノ酸配列が配列表中の配列番号4に示されることを特徴とするスーパーオキシドジスムターゼ。
- 請求項1に記載のスーパーオキシドジスムターゼをコードする遺伝子であって、
好ましくは、上記遺伝子の配列が、配列表中の配列番号3に示されるDNA配列である、遺伝子。 - 請求項2に記載の遺伝子を含む発現ベクター、遺伝子導入細胞系、および宿主菌。
- 請求項1に記載のスーパーオキシドジスムターゼを有効治療量で含むことを特徴とする薬物組成物。
- 抗菌剤、および/または薬学的に許容可能な担体、および/または賦形剤をさらに含むことを特徴とする請求項4に記載の薬物組成物。
- 調剤の形態が内服型または外用型であり、
上記内服型は、好ましくは経口液剤、錠剤、丸剤、カプセル剤、顆粒剤、経口散剤であり、
上記外用型は、好ましくは外用散剤、軟膏剤、貼布剤、外用液剤、坐剤、スプレー剤、エアゾール剤、吸入剤であることを特徴とする請求項4または5に記載の薬物組成物。 - 請求項1に記載のスーパーオキシドジスムターゼを有効量で含むことを特徴とする化粧品組成物。
- 請求項1に記載のスーパーオキシドジスムターゼを含むことを特徴とする食品添加剤。
- 請求項1に記載のスーパーオキシドジスムターゼを作製する方法であって、
スーパーオキシドジスムターゼをコードする請求項2に記載の遺伝子を発現させることによるか、または化学的合成方法により、請求項1に記載のスーパーオキシドジスムターゼを得ることを特徴とする方法。 - 請求項1に記載のスーパーオキシドジスムターゼ、または請求項2に記載の遺伝子、または請求項3に記載の発現ベクター、遺伝子導入細胞系、および宿主菌、または請求項4〜6のいずれか1項に記載の薬物組成物の、抗炎症薬物の作製における応用。
- 上記炎症は、粘膜炎症、外因性炎症因子による炎症、または皮膚炎症を含む請求項10に記載の応用。
- 上記粘膜炎症は、呼吸器系、消化管、生殖器官、聴覚器官、視覚器官の粘膜炎症を含み、好ましくは口腔潰瘍、歯齦炎、腸炎、胃炎、膣炎、骨盤内炎症、膀胱炎、子宮頸管炎、鼻炎、咽頭炎、中耳炎、外耳道炎、結膜炎、角膜炎であり、
上記外因性炎症因子による炎症は、生物的因子、物理的因子、外因性化学的因子またはこれらの組み合わせによる炎症を含み、
生物的因子による炎症は、好ましくは細菌、ウイルス、リケッチア、病原虫、真菌、スピロヘータおよび寄生虫が生体に作用することにより引き起こされる炎症を含み、
物理的因子による炎症は、好ましくは物理的力、高温、低温、電離放射線、紫外線が生体に作用することにより引き起こされる炎症を含み、さらに好ましくはやけど、熱傷、凍傷、日焼け、切り傷、放射線ダメージにより引き起こされる炎症、または、外科手術後もしくは放射線治療後に発生する炎症であり、
外因性化学的因子による炎症は、好ましくは放射性物質、強酸、強アルカリ、強酸化剤、アルコール、化学品が生体に作用することにより引き起こされる炎症を含み、さらに好ましくはアルコール性もしくは薬物誘発性肝炎、化学療法後に発生する炎症であり、
上記皮膚炎症は、手・足の白癬、毛嚢炎、接触性皮膚炎、湿疹、蕁麻疹、神経性皮膚炎、脂漏性皮膚炎を含み、好ましくは乳幼児湿疹であることを特徴とする請求項11に記載の応用。 - 請求項1に記載のスーパーオキシドジスムターゼ、または請求項2に記載の遺伝子、または請求項3に記載の発現ベクター、遺伝子導入細胞系、および宿主菌の、化粧品の作製における用途であって、
上記化粧品は、好ましくは老化や皮膚の色素沈着を防止する化粧品である、用途。
