Abstract
Description
中脳又は中頭は、視覚、聴覚、運動コントロール、睡眠/起床、覚醒状態(覚醒)及び温度レギュレーションに関連する中枢神経系の一部である。胚発生中、中脳は、神経管の中頭としても公知の2番目のベシクルから生じる。他の2つのベシクル(前脳及び後脳)とは異なり、中脳は、その後の神経発生では未分割のままである。
(1)中脳オルガノイドを作製するための方法であって、マトリックスを含む三次元細胞培養で培養された神経上皮幹細胞(のみ)と分化培地とを接触させることを含み、ここで、該培養が撹拌条件下で実施され、それにより、中脳オルガノイドを取得する、方法。
(2)前記神経上皮幹細胞がヒト神経上皮幹細胞であり、好ましくは、前記神経上皮細胞がSOX1、SOX2、PAX6及びネスチンを発現する、項目1に記載の方法。
(3)前記神経上皮幹細胞がhNESC−K7又はsmNPCである、項目2に記載の方法。

層流フードを使用して滅菌条件下で、細胞培養作業を実施した。37℃及び5%CO2一定のインキュベーター内で、全ての細胞を培養した。2D条件下で成長させた細胞の場合、冷ノックアウトDMEM(LIFE TECHNOLOGIES)に再懸濁したMATRIGEL(登録商標)(CORNING)で細胞培養プレート(NUNC(商標), THERMO SCIENTIFIC(商標))をコーティングした。希釈MATRIGEL 1.5mLを6ウェルプレートの各ウェルに追加した。プレートを室温(RT)で一晩保持した。コーティングプレートを4℃で最大1カ月間保存した。
2.脳オルガノイドの作製
Lancasterらによって確立されたプロトコール(Lancaster and Knoblich, 2014a; Lancaster et al., 2013)に若干の改変を加えて、脳オルガノイドを作製した。フィーダー依存性iPSCに代えて、開始集団としてフィーダー非依存性ヒトiPS細胞を使用した。第1段階として、胚様体(EB)を作製した。イントロダクションのセクション1.1に記載されている胚発生と同様に、これらの3D凝集体は、3つの生殖系列(内胚葉、中胚葉及び外胚葉)への分化を介して細胞特異化を受ける。次に、神経上皮組織の形成が始まり、細胞運命が神経系に限定された。神経上皮組織が発達すると、3D構造をMATRIGEL(これは構造支持を与え、組織が3D形状を維持するのを支援する)の小滴に移した。Engelbroth−Holm−Swarmマウス肉腫に由来するこのマトリックスは、ラミニン、コラーゲンIV、ニドゲン/エナクチン及びプロテオグリカンから構成されるので、細胞外マトリックス(ECM)に似ている。ビタミンAを含有するN2及びB27を補充したNeurobasal培地を使用することによって、神経分化を促す培地中で、MATRIGEL包埋組織を培養した。細胞成長及び生存率を増加させるために、非必須アミノ酸及びGlutaMax(商標)を追加した。還元剤2−メルカプトエタノールは、有毒な酸素ラジカルレベルの形成を防止した。組織を動的条件下に置いて、組織が底部に接着するのを防止した。それは栄養素交換をさらに増加させ、老廃物交換をコントロールする。この目的のために、Lancasterらは、スピニングバイオリアクターを使用した。このプロジェクトでは、約80rpmでスピニングしながら、オービタルシェーカー(IKA(登録商標))上の未処理10cm細胞培養ペトリ皿(GREINER)に組織を置いた。
2.1 ヒト人工多能性幹細胞の維持
CORIELLの野生型ヒト人工多能性幹細胞株を細胞培養6ウェルプレートに播種し、Essential8(商標)培地(LIFE TECHNOLOGIES)中で培養した。70〜80%コンフルエンスで、最後の播種後1週間以内に、細胞を継代した。表面から細胞を分離するために、培地を吸引し、0.5mM EDTA(INVITROGEN)を追加した。37℃及び5%CO2における4分間のインキュベーション時間後、EDTAを吸引した。PBSで細胞を洗浄し、Essential8(商標)培地に再懸濁した。細胞を新たなコーティング6ウェルプレートに1:3〜1:6の比で播種した。培地を毎日交換した。
2.2 hiPSCの免疫細胞化学的特性評価
ヒトiPS細胞を24ウェルプレート中のカバースリップ上に播種し、セクション2.1に記載されている条件下で培養した。3日後、4%パラホルムアルデヒド(PFA)によって細胞を4℃で40分間固定し、PBSで5分間にわたって3回洗浄した。0.2%Triton−X−100によって細胞膜を室温で10分間透過処理した。0.05%Triton−X−100 PBS溶液による3回の洗浄工程後、2%NGS+2%BSAの0.05%Triton−X−100 PBS溶液によって細胞を室温で60分間ブロッキングした。ブロッキングバッファーで一次抗体を希釈した(表1を参照のこと)。ウェットチャンバー内のパラフィルム上に、一次抗体溶液 35μl滴を調製した。カバースリップをこの小滴の上に載せ、4℃で一晩インキュベーションした。
翌日、3回交換したPBSでカバースリップを5分間洗浄した。一次抗体に対する二次抗体(表2)をヘキスト色素(INVITROGEN、1:10000)と一緒に希釈し、前述のようにパラフィルム上に小滴を調製した。カバースリップを室温で1時間インキュベーションし、次いで、PBSで3回洗浄し、H2Oで1回洗浄し、蛍光封入剤を使用してガラススライド上にマウントした。共焦点レーザー走査顕微鏡(ZEISS LSM 710)を使用して、染色を分析した。
2.3 EBの作製
EBの作製では、PBSでコロニーを1回洗浄し、0.5mM EDTA(INVITROGEN)を使用して分離した。37℃における4分間のインキュベーション期間後、EDTAを予熱StemPro(登録商標)Accutase(登録商標)(LIFE TECHNOLOGIES) 1mlと交換し、細胞を37℃でさらに4分間インキュベーションした。Essential8(商標)培地(LIFE TECHNOLOGIES) 1mlを含む1mlピペットチップを使用して、ディッシュからコロニーを分離した。単一細胞懸濁液を作るために、細胞懸濁液をファルコンチューブに移してトリチュレートした。細胞計数のために、2回反復分の5μlを収集した。細胞を室温、270gで5分間遠心分離し、トリパンブルーを用いた自動細胞カウンター(Countess II FL LIFE TECHNOLOGIES)を使用して、細胞を計数した。遠心分離後、上清を吸引し、ROCK阻害剤を含有するlow−FGF2 hESC培地(表3) 1mlに細胞を再懸濁した。次に、150μl当たり生細胞 9000個を得るために、low−FGF2 hESC培地を追加した。最後に、150μlを丸底超低接着96ウェルプレート(CORNING)(これは、細胞を沈降させてEBの形成を促す)の各ウェルにプレーティングした。37℃及び5%CO2のインキュベーター内で、細胞を培養した。培地の半分を新鮮low−FGF2 hESC培地と交換することによって、培地を隔日で交換した。ROCK阻害剤及びlow−FGF2を4日間だけ追加した。
2.4 神経誘導
6日後、カットピペットチップを用いて、境界において明るい平滑縁を有するEBを、神経誘導培地(表4)を含有する超低接着24ウェルプレート(CORNING)に移し、4日間培養した。
2.5 MATRIGEL小滴への神経上皮組織の移動
MATRIGEL小滴を作製するために役立つ基材を調製するために、パラフィルムの角材を200μlピペットチップ用の空のチップトレイに置き、手袋をした指でパラフィルムを穴に押し込むことによって、小さい窪みを作った。窪んだパラフィルムにエタノールを再度噴霧し、層流フード下、ペトリ皿中で乾燥させた。パラフィルムが乾燥したら、神経上皮組織を各窪みに移し、培地を徐々に除去した。次に、MATRIGEL 30μlを慎重に追加し、組織を小滴の中央に置いた(図2.1)。MATRIGELを重合させるために、それを37℃で30分間インキュベーションした。インキュベーション時間後、ビタミンAを含まない脳オルガノイド分化培地 15ml中に小滴を収集した(表5)。パラフィルムからMATRIGEL小滴を除去するために、小滴が落ちるまでディッシュを穏やかに振盪しながら、滅菌鉗子を使用してシートを保持した。ディッシュを静止条件下のインキュベーター内に置き、48時間後に培地を交換した。静置培養で4日後、培地を、ビタミンAを含有する新鮮分化培地と再度交換した。最後に、約80rpmで振盪しながら、インキュベーター内に設置したオービタルシェーカー上にディッシュを置き、培地を3日ごと又は4日ごとに交換した。
3.中脳オルガノイドの作製
3.1 ヒト神経上皮幹細胞
中脳オルガノイドを作製するために、ヒト神経上皮幹細胞(hNESC)は開始集団として機能した。これらの神経前駆細胞は、ロバストな不死性のエクスパンションが可能であり、ニューロン、アストロサイト、オリゴデンドロサイトを含むCNSの細胞に分化し得、神経堤系統にも分化し得るので、幹細胞の特性を有する。自己複製及びエクスパンションのために、それらは小分子を専ら必要とする。非神経BMP及び背側TGF−βシグナリングの阻害を介して、神経誘導を開始した。不死自己複製状態を維持するために、WNT及びSHHシグナルをトリガーした。WNTシグナリングは、神経板の外側縁において細胞の形成を誘導し、そのアンタゴニストSHHは、腹側神経管運命を規定する。CHIR99021を使用してカノニカルWNTシグナリング経路を刺激し、パルモルファミン(PMA)を追加してSHH経路を刺激した。hNESCは、運動ニューロン及びmDAに効率的に分化し得るので、初期ヒト発生及び神経変性疾患を研究するための強力なツールになる。
3.2 ヒト神経上皮幹細胞の維持
ヒト人工多能性幹細胞から誘導した野生型ヒト神経上皮幹細胞株(hNESC−K7)は、開始集団として機能した(Reinhardt et al., 2013)。新鮮補充N2B27維持培地(表6及び表7)中で、細胞を培養した。最後の播種後1週間以内に、80〜90%コンフルエンスで細胞を継代した。表面から細胞を分離するために、培地を温StemPro(登録商標)Accutase(登録商標)(LIFE TECHNOLOGIES) 700μlと交換し、細胞を37℃及び5%CO2で4〜6分間インキュベーションした。細胞を温DMEM/F12(LIFE TECHNOLOGIES) 5mlに再懸濁し、200gで3分間遠心分離した。上清を吸引した後、ペレットを補充N2B27維持培地に再懸濁し、コンフルエンスに応じて、細胞を新たなプレートに1:10〜1:20の比で播種した。培地を隔日で交換した。
3.3 hNESCの免疫細胞化学的特性評価
hNESCを24ウェルプレート中のカバースリップ上に播種し、セクション3.2に記載されている条件下で培養した。3日後、4%PFAで細胞を4℃で40分間固定し、PBSで5分間かけて3回洗浄した。セクション2.2に記載されているプロトコールにしたがって、異なる一次抗体(表8)及び対応する二次抗体(表2)を用いて、ICCを実施した。
3.4 三次元hNESCコロニーの作製及びエクスパンション
単一3D hNESCコロニーを作製するために、セクション3.2に記載されているようにhNESCを継代した。遠心分離してN2B27維持培地 1mlに再懸濁した後、トリパンブルーを用いた自動細胞カウンター(Countess II FL LIFE TECHNOLOGIES)を使用して、細胞を計数した。150μl当たり生細胞 9000個を得るために、N2B27維持培地を追加した。150μlを丸底超低接着96ウェルプレート(CORNING)(これは、細胞が1ウェル当たり1つの単一コロニーを形成することを可能にする)の各ウェルにプレーティングした。培地の半分を新鮮N2B27維持培地と交換することによって、培地を隔日で交換した。6日後、副次的なコロニーを除去して組織をエクスパンションするために、コロニーを超低接着24ウェルプレート(CORNING)に移した。2日後、セクション2.5に記載されているのと同じ方法で、コロニーをMATRIGEL小滴に包埋した。10cmペトリ皿中、静止維持条件下で、MATRIGEL包埋コロニーを2日間以上培養してから、10日目に、PMAを含有するN2B27分化培地(表9)を用いて分化を開始した。Reinhardt et al. 2013から若干の改変を加えて分化プロトコールを適合した。FGF8は、mDAニューロンへの分化効率を減少させるので、最初の8日間はFGF8を追加しなかった。分化条件下で2日後、ディッシュをオービタルシェーカー上に置き、約80rpmの動的条件下に置いた。培地を3日ごと又は4日ごとに交換した。分化の最初の6日間のみ、PMAを追加した。
4.ヒト大脳オルガノイド及び中脳オルガノイドの免疫組織化学的特性評価
脳発生中に、細胞はそれら自身を再編成し、異なる機能領域及び相互依存領域を形成する。同時に、神経上皮細胞は、CNSの様々なニューロン及び支持グリア細胞に分化する。脳オルガノイド及び中脳オルガノイドが、発生中のヒト脳に特徴的な異なる細胞型及び脳領域を発達させるかを分析するために、免疫組織化学的(IHC)染色を実施した。神経前駆細胞、幼若ニューロン及び成熟ニューロン、アストロサイト、オリゴデンドロサイト、ドーパミン作動性ニューロンの存在を分析した。また、免疫蛍光染色を実施して、脳オルガノイド及び中脳オルガノイドが中脳同一性を確立したかを検証した。
4.1 切片化
6日目(n=3)、7日目(n=3)、16日目(n=3)、30日目(n=3)及び44日目(n=8)に、シェーカー上で、脳オルガノイド及び中脳オルガノイドを4%PFAによって室温で一晩固定した。IHCの前に、900μm超の直径を有するより大きいオルガノイドを切片化した。PBSで固定組織を15分間かけて3回洗浄し、温3%低融点アガロースに包埋し、固体になるまで冷却した。次いで、アガロースのブロックをトリミングし、金属ブロックホルダー上に接着し、0.5mm/秒の速度及び70Hzの周波数でビブラトーム(LEICA, VT100 S)を使用して厚さ150μmに切片化した。
4.2 IHC
TBS+++(0.5%Triton−X−100、0.1%アジ化ナトリウム、0.1%クエン酸ナトリウム及び5%ウシ胎児血清又は正常ヤギ血清を含む1×Tris緩衝生理食塩水)を含有する24ウェルプレートに浮遊アガロース切片を収集し、シェーカー上、室温で少なくとも1時間ブロッキング/透過処理した。TBS+++で一次抗体(表10を参照のこと)を希釈し、抗体溶液 300μlを各ウェルに追加した。シェーカー上で切片を4℃で48時間インキュベーションし、その後、3回交換したTBSで15分間洗浄した。シェーカー上、TBS+++で希釈した二次抗体(表11を参照のこと)で切片を室温で2時間インキュベーションし、続いて、TBSによる3回の洗浄工程を15分間行った。最後に、H2Oで切片をリンスし、封入剤を用いてガラススライド上にマウントした。共焦点レーザー走査顕微鏡(ZEISS LSM 710)を使用して、染色を分析した。
II.結果
中脳オルガノイドの作製
1.1 hNESCの免疫細胞化学的特性評価
ヒト神経上皮幹細胞のICCにより、維持条件下における神経前駆体マーカーSOX2及びネスチンの安定発現が明らかになった。hNESCでは、別の神経前駆体マーカーSOX1も発現していたが、蛍光強度が変動した。hNESCでは、FOXA2(腹側神経管のマーカー)がわずかに発現していた。初期分化時には、少数のhNESCが、ニューロン特異的クラスIIIβ−チューブリン(TUJ1)及びダブルコルチン(DCX)(これらは、初期ニューロンの指標である)を発現するニューロンに分化する(図3)。
1.2 中脳オルガノイドの発生
中脳オルガノイドの作製のために、開始集団として野生型hNESC株K7を使用した。丸底超低接着プレート上に播種したヒトNESCは高密度の球状コロニーを形成し、周辺の死細胞はごくわずかであった。いくつかのウェルでは、小さい副次的なコロニーが発達し、数日後、これは主要コロニーと一体化した。最初の3日以内に、コロニーは明るくなり始め、維持条件下で平滑縁を示した(これは、組織が健康であることを示す)。しかしながら、8日目の初期オルガノイドは、中央において暗いコアを発達させた(これは、細胞死を示す)(図4a、b)。2日間の分化後、オルガノイドは、組織の表面に沿って小さい突起を発達させた(図4c)。3週間以内に、突起は伸びて約1mmの長さに達し、コロニーの外側の細胞体はごくわずかであった。MATRIGEL支持がないコロニーでは、これは観察されなかった(図4d〜f)。
それらの機能性及びニューロンネットワーク活性をさらに確認するために、本発明者らは、オルガノイド全体に対してFluo-4AMカルシウムイメージングを実施した。本発明者らは、活動電位によって誘発されたカルシウム移行に基づいて自発ニューロン活性を測定した(図12A、B)。注目すべきことに、蛍光トレースのいくつかは規則的な発火パターン(これは、緊張性電気生理学的活性の指標である)を示し、ドーパミン作動性ニューロンのペースメーカー活性に似ていた(Moreno et al. (2015), “Differentiation of neuroepithelial stem cells into functional dopaminergic neurons in 3D microfluidic cell culture.” Lab on a chip 15, 2419-2428; Cummings et al. (2014)” Alzheimer’s disease drug-development pipeline: few candidates, frequent failures.” Alzheimer’s research & therapy 6, 37)。カルシウムイメージングに加えて、多電極アレイ(MEA)システムを使用して、電気生理学的活性を調べた。この方法論は、活動電位によって生成される細胞外電場電位の非侵襲的記録を可能にする。60〜70日目(50〜60の分化)に、48ウェル組織培養プレート中で、中脳オルガノイドを16電極のグリッド上に置いた(図12C)。単相スパイク及び二相スパイクの形態の個々の電極によって、自発活性を数日間にわたって検出した(26.12±5.1スパイク/活性電極(n≧3)、図12D、E)。さらに、スパイクは、近いタイミングで複数の電極上で発生したが、これは、ニューロンネットワーク同期性を表す(図12F)。これらの知見は、中脳オルガノイドが機能的シナプス結合を発達させ、自発ニューロン活性を示すことを示している。

(b)幼若ニューロン;及び/又は
(c)幼若ドーパミン作動性ニューロン
を含む、請求項30、31又は33のいずれか一項に記載の方法。 - 後期中脳オルガノイドが、
(a)神経前駆細胞;
(b)幼若ニューロン;
(c)幼若ドーパミン作動性ニューロン;
(d)成熟ニューロン;
(e)成熟ドーパミン作動性ニューロン;
(f)中脳オルガノイドの非対称組織化;
(g)オリゴデンドロサイト;
(h)オリゴデンドロサイト前駆体;
(i)アストロサイト;
(j)中脳オルガノイド内のドーパミン作動性ニューロンのクラスタ化;及び/又は
(k)中脳オルガノイドからマトリックスを通して伸びる突起
を含む、請求項30、32又は33のいずれか一項に記載の方法。 - 前記神経前駆細胞がマーカーSOX2及び/若しくはネスチンの発現を特徴とし、
前記幼若ニューロンがマーカーTUJ1の発現を特徴とし、
前記幼若ドーパミン作動性ニューロンがマーカーTUJ1及びチロシンヒドロキシラーゼ(TH)の発現を特徴とし、
前記成熟ニューロンがマーカーMAP2の発現を特徴とし、
前記成熟ドーパミン作動性ニューロンがマーカーMAP2及びTHの発現を特徴とし、
前記オリゴデンドロサイトがマーカーO4の発現を特徴とし、
前記オリゴデンドロサイト前駆体がマーカーNG2の発現を特徴とし、並びに/又は
前記アストロサイトがマーカーGFAP及び/若しくはS100bの発現を特徴とする、請求項33〜35のいずれか一項に記載の方法。 - 中脳オルガノイドの非対称組織化が、ドーパミン作動性ニューロンの非対称極性組織化及び/又は中脳オルガノイド内の神経前駆細胞の非対称組織化である、請求項33〜36のいずれか一項に記載の方法。
- 前記中脳オルガノイド内のドーパミン作動性ニューロンの非対称極性組織化が、
(a)成熟ドーパミン作動性ニューロンが中脳オルガノイドの最外部にあり;及び/又は
(b)幼若ドーパミン作動性ニューロンが中脳オルガノイドの内部にある
という局在性を特徴とする、請求項37に記載の方法。 - 幼若ドーパミン作動性ニューロンが、成熟すると中脳オルガノイドの最外部に向かって遊走する、請求項37又は38に記載の方法。
- 中脳オルガノイド内の神経前駆細胞の非対称極性組織化が、神経前駆体が中脳オルガノイドの内部コア周辺の環状構造にあるという局在性を特徴とする、請求項37に記載の方法。
- 前記中脳オルガノイド内のドーパミン作動性ニューロンのクラスタ化が、中脳オルガノイドの特定領域における2個超、3個、4個、5個、6個、7個、8個、9個、10個又はそれ以上のドーパミン作動性ニューロンの蓄積を特徴とする、請求項33〜40のいずれか一項に記載の方法。
- 前記中脳オルガノイドからマトリックスを通して伸びる突起が、0.1mm、0.2mm、0.3mm、0.4mm、0.5mm、0.6mm、0.7mm、0.8mm、0.9mm、1mm、1.1mm、1.2mm又はそれ以上の長さを有する、請求項33〜41のいずれか一項に記載の方法。
- 請求項1〜42のいずれか一項に記載の方法によって取得可能な、中脳オルガノイドユニット。
- 細胞応答を誘発するその能力について目的の化合物を試験するための方法であって、
(a)請求項43に記載の中脳オルガノイドと前記目的の化合物とを接触させること;及び
(b)前記目的の化合物が細胞応答を誘発するかを決定すること
を含む、方法。 - 請求項43で定義される中脳オルガノイドにおけるドーパミン作動性ニューロンの分化及び/又はドーパミン作動性ニューロンの死を促進又は阻害する分子を同定するための方法であって、該中脳オルガノイドと目的の分子とを接触させることを含み、
コントロールと比較したドーパミン作動性ニューロンへの分化の増加が、該目的の分子がドーパミン作動性ニューロンの分化を促進し、及び/又はドーパミン作動性ニューロンの死を阻害することを示し、コントロールと比較したドーパミン作動性ニューロンへの分化の減少が、該目的の分子がドーパミン作動性ニューロンの分化を阻害し、及び/又はドーパミン作動性ニューロンの死を誘導することを示す、方法。 - 前記目的の化合物が、薬物、小分子、ホルモン、成長因子、結合タンパク質、核酸分子、ペプチド、タンパク質又は(共培養)細胞である、請求項44又は45に記載の方法。
- 前記細胞の種類がドーパミン作動性ニューロンである、請求項44に記載の方法。
- 神経突起伸長を比較することによって、ドーパミン作動性ニューロンへの分化を測定する、請求項45に記載の方法。
- THの発現を比較することによって、ドーパミン作動性ニューロンへの分化を測定する、請求項45又は48に記載の方法。
- コントロールが、目的の分子と接触されていない中脳オルガノイドである、請求項45、48又は49のいずれか一項に記載の方法。
- 請求項43に記載の中脳オルガノイドユニットを含む、組成物。
- 中脳オルガノイドユニットを作製するための、撹拌条件下、マトリックスを含む三次元細胞培養で培養された神経上皮幹細胞の使用。
- a)場合により、人工多能性幹細胞(iPSC)を取得/提供すること;
b)
(i)アクチビン/トランスフォーミング成長因子−β(TGF−β)シグナリング阻害剤;
(ii)カノニカルWNTシグナリング活性化剤;
(iii)骨形成タンパク質(BMP)シグナリング阻害剤;及び
(iv)SHH経路活性化剤
を含む培地中で、前記iPSCを培養すること;並びに
c)
(i)アクチビン/TGF−βシグナリング阻害剤;
(ii)カノニカルWNTシグナリング活性化剤;
(iii)BMPシグナリング阻害剤;及び
(iv)SHH経路活性化剤
を含む培地中で、b)において取得した細胞を培養すること;並びに
d)
(i)カノニカルWNTシグナリング活性化剤;
(ii)SHH経路活性化剤;及び
(iii)抗酸化剤
を含む培地中で、c)において取得した細胞をさらに培養すること
を含み、それにより、神経上皮幹細胞を取得する方法によって、神経上皮幹細胞を取得する、請求項1〜42のいずれか一項に記載の方法。
