Abstract
Description
− 通常、発酵槽である生体内変換部分であって、微生物は、バニリン及びほとんどの場合に基質からの副生成物を合成する、生体内変換部分、
− バイオマス濾過部分であって、細胞は、合成された生成物と残留基質とを含む濾液から物理的に分離され、その後、発酵槽中でリサイクルされる、バイオマス濾過部分、及び
− 選択的な分離部分であって、バニリンなどの目的生成物は、発酵槽へリサイクルされる他のものから抽出される、選択的な分離部分。
− 基質の生物変換によってバニリンを形成することが可能な少なくとも1種の微生物を発酵槽中で培養すること、
− 前記微生物によって変換される少なくとも1種の基質を前記発酵槽に供給すること、及び
− 前記発酵槽の発酵ブロスからそのように製造されたバニリンを回収すること
からなる工程を含み、
− 前記基質が少なくとも部分的に発酵槽の発酵ブロスに連続的に添加され、及び
− 前記基質の連続添加中、発酵ブロスの流れが分離装置によって発酵槽から連続的に抽出され、それにより、(i)前記微生物を含まない発酵ブロス組成物(a)と、(ii)前記微生物を含む発酵ブロス組成物(b)とが与えられ、
前記バニリンが組成物(a)の全て又は一部から回収され、及び組成物(b)が全て又は少なくとも部分的に発酵槽でリサイクルされる、方法に関する。
前述したように、本発明の方法は、基質をバニリンに変換できる少なくとも1種の微生物を発酵槽中で培養する予備工程を含む。
既に前述したように、複数の種類の炭素基質を検討することができる。
式中、trは、時間(h)単位での滞留時間であり、
Vは、L単位での発酵槽の有効体積であり、
Q=Q(a)=Qfeedは、L/h単位での流量である。
既に本明細書の上で提示したように、これは、一部の発酵ブロスの連続的な抽出のための、並びにその結果の(i)前記微生物を含まない発酵ブロス組成物(a)と、(ii)発酵ブロス組成物(b)との連続的な形成のための分離装置を介して進行し、後者は、少なくとも部分的に発酵槽中でリサイクルされる。
0.1gのKH2PO4、0.4gのNa2HPO4、12H2O、1gの酵母エキス、0.02gのMgSO4,7H2O、38gの水中に2gのグルコースを含む溶液10gが入っている500mLの振とうフラスコに1mLのストレプトマイセス・セトニィ(Streptomyces setonii)ATCC39116株を接種し、37℃、150rpmで24時間培養する。
ストレプトマイセス・セトニィ(Streptomyces setonii)を実施例1に従って培養した。300mLの溶液を発酵槽から取り出す。pHを8.3に上げて一定に保ち、曝気を0.47vvmに下げ、撹拌速度を変化させて溶存酸素を30%で一定に維持する。温度及び圧力は変更しない。
ストレプトマイセス・セトニィ(Streptomyces setonii)を実施例1に従って培養した。300mLの溶液を発酵槽から取り出す。pHを8.3に上げて一定に保ち、曝気を0.47vvmに下げ、撹拌速度を変化させて溶存酸素を30%で一定に維持する。温度及び圧力は変更しない。
Claims (17)
- 基質からの生物変換によってバニリンを製造する方法であって、少なくとも、
− 基質の生物変換によってバニリンを形成することが可能な少なくとも1種の微生物を発酵槽中で培養すること、
− 前記微生物によって変換される少なくとも1種の基質を前記発酵槽に供給すること、及び
− 前記発酵槽の発酵ブロスからそのように製造されたバニリンを回収すること
からなる工程を含み、
− 前記基質が少なくとも部分的に発酵槽の発酵ブロスに連続的に添加され、及び
− 前記基質の連続添加中、発酵ブロスの流れが分離装置によって発酵槽から連続的に抽出され、それにより、(i)前記微生物を含まない発酵ブロス組成物(a)と、(ii)前記微生物を含む発酵ブロス組成物(b)とが与えられ、
前記バニリンが組成物(a)の全て又は一部から回収され、及び組成物(b)が全て又は少なくとも部分的に発酵槽でリサイクルされる、方法。 - 基質の最初の投入量が発酵槽に添加され、及び引き続き又は同時に、一部の基質が発酵槽中の発酵ブロスに連続的に添加される、請求項1に記載の方法。
- 発酵槽の滞留時間が1〜24時間、好ましくは3〜10時間の範囲である、請求項1又は2に記載の方法。
- 基質の供給の流量Qfeed及び組成物(a)の流れの流量Q(a)が、前記発酵槽中の発酵ブロスの一定の体積を維持するように調整される、請求項1〜3のいずれか一項に記載の方法。
- 前記基質がフェルラ酸である、請求項1〜4のいずれか一項に記載の方法。
- 基質が、7〜9の範囲のpHを有する溶液の形態で連続的に供給される、請求項1〜5のいずれか一項に記載の方法。
- 前記溶液が前記発酵槽の発酵ブロスのための栄養成分も含む、請求項6に記載の方法。
- 発酵ブロス中において基質の濃度が5〜60g/Lの範囲である、請求項1〜7のいずれか一項に記載の方法。
- 微生物が、バシラス(Bacillus)、ストレプトマイセス(Streptomyces)、シュードモナス(Pseudomonas)、及びセラチア(Serratia)からなる群において選択され、好ましくはストレプトマイセス・セトニィ(Streptomyces setonii)である、請求項1〜8のいずれか一項に記載の方法。
- 分離装置がバニリンの選択的な同時抽出も可能にする、請求項1〜9のいずれか一項に記載の方法。
- 前記分離装置が、膜溶媒抽出(MBSE)に基づく濾過膜装置である、請求項10に記載の方法。
- 分離装置が、未変換の基質と生物変換生成物とを含む組成物(a)を形成する、請求項1〜9のいずれか一項に記載の方法。
- 組成物(a)からのバニリンの選択的な回収を可能にする後続の且つ補完的な分離技術を含む、請求項12に記載の方法。
- 補完的な分離技術が、吸着カラムの樹脂へのバニリンの選択的な吸着に基づく、請求項13に記載の方法。
- 補完的な分離技術が液−液抽出に基づく、請求項13に記載の方法。
- 補完的な分離技術が、バニリンを含まず且つ未変換の基質を含む組成物の回収を可能にし、前記バニリンを含まず且つ未変換の基質を含む組成物が少なくとも部分的に発酵槽中でリサイクルされる、請求項13〜15のいずれか一項に記載の方法。
- 前記回収されたバニリンの精製工程又は精製工程の組み合わせを更に含む、請求項1〜16のいずれか一項に記載の方法。
