Abstract

Description
本発明は、生理活性ポリペプチド、二つ以上のアルデヒド官能基を含んでいる非ペプチド性重合体リンカーによって免疫グロブリン不変領域に生理活性ポリペプチドと共有結合した複合体を製造する方法に関する。より具体的には、本発明は、生理活性ポリペプチド複合体の製造の時、還元剤の使用を調節することにより、低い製造収率及び原料ポリペプチドの変形などの問題を改善することにより特徴づけられた、生理活性ポリペプチド複合体を効率的に製造する方法に関する。
一般に、生理活性ポリペプチドは安定性が低いため変性しやすく、血液内でタンパク質分解を受ける。その次に、腎臓や肝臓を通じて排除される。薬理成分として生理活性ポリペプチドを含むタンパク質医薬品の血中濃度及び力価を維持するためには、タンパク質薬物を患者に頻繁に投与する必要がある。しかし、大部分注射剤の形であり、そのように、頻繁な注射は患者に苦痛を引き起こし、また、高い治療費用になる。このような問題を解決するために、タンパク質薬物の血中安定性を改善させ、血中薬物の濃度を高レベルに持続させて薬効を極大化するための努力がなされてきた。持続性製剤として使用するための必要用件として、患者に免疫反応を誘発することなく、タンパク質薬物の安定性を高めると共に、薬物自体の力価が十分に高く維持されなければならない。
タンパク質を安定化させ、酵素的分解による防止及び腎臓クリアランスのための従来のアプローチは、ポリエチレングリコール(polyethylene glycol: 以下 「PEG」と略す)のように溶解度の高い高分子のタンパク質薬物の表面を化学的に改造することである。目的のタンパク質の特定部位又は多様な部位への結合により、PEGは、溶解度を高めることによりタンパク質を安定化させ、タンパク質の加水分解を防止する(非特許文献1)。
例えば、特許文献1は、cGMPの合成を促進させるために、NPR-Aに結合するPEGsとの結合により、B型ナトリウム利尿ペプチド(BNP)を持続的に活性化させる方法を記述している。それによって、動脈内の血圧が減少する。このような事実から鬱血性心不全症の治療剤として用いられる。特許文献2は、そのリジン残基によるペグ化によるエキセンディン-4が生体内持続時間を増進させる方法を記述している。しかし、このような方法は、PEGの分子量を増加させることにより、ペプチド薬物の血液循環時間を延ばすことができるにもかかわらず、ペグ化は、分子量が増加するほどペプチド薬物の力価が顕著に低くなり、また、ペプチドとの反応性が低くなって収率が減少するような問題がある。
特許文献3は、GLP-1及びエキセンディン-4 又はその類似体とヒト血清アルブミン又は組換え遺伝子技術を用いて製造された免疫グロブリン断片(Fc)との融合タンパク質について記述している。特許文献4は、副甲状線ホルモン(PTH)又はその類似体がFcに結合されている。これらの方法が、低い収率と非特異性のようなペグ化の問題の解決策として評価されるが、それらの目的ペプチドの体内半減期の増加が期待に反して著しくなく、そして、場合によっては、融合タンパク質が、低い力価を有する。血中半減期の増加効果を極大化するために、多様な種類のペプチドリンカーが用いられる一方、免疫反応を誘発する可能性がある。さらに、BNPのようにジスルフィド結合を有するペプチドは、ミスフォールディングの確率が高いため、実際的に適用が困難である。さらに、天然に存在しないアミノ酸残基を有するリンカーは、遺伝子組換えによる生産が不可能である。
インスリンは、ヒトの膵臓のベータ細胞から分泌されるペプチドであり、体内の血糖値を調節するのに 重要である。インスリンが適切に分泌されないか、又は分泌されたインスリンが体内で適切に機能しない場合、血糖値が調節できずに上昇し、その結果、糖尿病になる。後者の場合を2型糖尿病と言う。この場合、インスリンが膵臓から分泌できず、血糖値の上昇するので、その結果、1型糖尿病になる。2型糖尿病の患者は、化学物質を主成分とする経口用血糖値降下剤を用いて治療し、一部の患者にはインスリンを用いて治療する。一方、インスリン注射は、1型糖尿病の患者に必須として要求される。
一般に、インスリン治療法は、毎日食事の前、又は後に注射を通じて1日3回のインスリンの投与により行われた。しかし、インスリンの1日3回の持続的投与は、患者にとって苦痛と不便である。これらの問題を解決するために多様な試みがあった。ペプチドの薬物の生体膜の透過度を増加させることを目指した一つの戦略は、口腔又は鼻腔吸入によってそれらを運ぶことである。しかし、注射と比べた時に、伝達効率が顕著に低く、そして生体内活性に必要なレベルにペプチド薬物を維持するのに困難が多い。
代替戦略として、薬物の過度な量は、1日に1回の投与で持続的な血中濃度を維持するために皮下に注射し、吸収が遅延されるようにできる。その中のいくつかは、商用として承認され (例えば、Lantus、Sanofi-aventis)現在患者に投与されている。別途に、インスリンに脂肪酸を修飾してインスリン結合体の結合を強くし、投与部位又は血中のアルブミンとの結合を通じて持続時間を延長する研究がなされてきた。薬物のいくつかは、商用(例えば、Levemir、NovoNordisk)として承認された。しかし、このような薬物は、投与部位に痛みが生じ、一日一回注射しなければならず、依然として患者に大きな負担となっている。
先行技術と直面した問題を克服するために、本発明者らは、インスリンのような生理活性ポリペプチドの血中の半減期の増進及び生体内の持続時間を同時に増加させる戦略として、非ペプチド性重合体を用いることにより結合した生理活性ポリペプチドと免疫グロブリン不変領域を含む結合体を製造した。しかし、結合体の構成物質が非常に高いため、結合体を高収率及び高純度で製造する方法が必要である。このような心得で、本発明者らは、結合体の製造反応中、反応溶液に最適の濃度で適正還元剤の種類を用いることにより、コストを節減し、高収率及び高純度で生理活性ポリペプチド結合体を製造する方法を開発し本発明を完成した。

本発明の目的は、反応の中で反応溶液に適正還元剤の種類と濃度を用いて、低い製造収率及び原料ポリペプチドの変形などの問題を改善することにより特徴づけられ、生理活性ポリペプチド、2つ以上のアルデヒド官能基を含む非ペプチド性重合体リンカー及び免疫グロブリン不変領域が共有結合により連結された結合体を製造する効果的方法を提供することにある。
上記目的を達成するための一様態として、本発明は、(1)1〜20 mMの濃度の還元剤の存在下で、二つ以上のアルデヒド官能基を有する非ペプチド性重合体を生理活性ポリペプチド又は免疫グロブリン不変領域との反応;及び(2)1〜100 mMの還元剤の存在の下に、上記段階(1)の反応液を生理活性ポリペプチド又は免疫グロブリン不変領域の他と反応させる段階を含む生理活性ポリペプチド-非ペプチド性重合体-免疫グロブリン不変領域の結合体の製造方法を提供する。
反応液は、非ペプチド性重合体と生理活性ポリペプチドとの結合体又は非ペプチド性重合体と免疫グロブリン不変領域との結合体及び/又は反応しない反応物を含むことができる。従って、本発明の方法は、段階(1)の後、反応混合物から生理活性ポリペプチド-非ペプチド性重合体の結合体又は免疫グロブリン-非ペプチド性重合体の結合体の分離をさらに含むことができる。
ここで用いられた用語、「非ペプチド性重合体」とは、少なくてもペプチド結合ではない任意の共有結合によって互いに結合される2つの繰り返しユニットを含む生体適合性重合体を意味する。本発明で使用可能な非ペプチド性重合体の例は、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールとの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、デキストラン、ポリビニルエチルエーテル、ポリ乳酸(PLA)及びポリ乳酸-グリコール酸(PLGA)のような生分解性重合体、脂質重合体、キチン類、ヒアルロン酸及びこれらの組み合わせを含む。好ましくは、ポリエチレングリコール(PEG)である。当技術の中で、既知の方法を使用して容易に製造することができる技術及び誘導物において、有名なその誘導物は、さらに現在の発明の範囲内である。該分野において知られているこれらの誘導体及び当該分野の技術水準で容易に製造できる誘導体も本発明の範囲に含まれる。
上記したように、非ペプチド性重合体は、2つ以上のアルデヒド官能基を有することができる。従って、上記列挙した非ペプチド性重合体は、それ自体で二つ以上のアルデヒド基を含んだものでもよいが、又はそれの2つ以上のアルコール基がアルデヒド基を含む置換基であることが好ましい。上記アルデヒド基を含む置換基は、プロピオンアルデヒド、又はブチルアルデヒドなどのアルキルアルデヒドであってもよい。1つの好ましい様態として、非ペプチド性重合体は、その両末端にそれぞれプロピオンアルデヒド基の置換を有するPEGあってもよい。
インフレーム融合により設計された融合タンパク質として用いられた従来のペプチド性リンカーの短所は、生体内で蛋白質分解酵素(プロテイナーゼ)により容易に切断されて、キャリアによる血中半減期延長を保障できない。しかし、本発明では、蛋白質分解酵素に対して抵抗性のある重合体を用い、よって、ペプチドの血中半減期は、キャリアと同様のレベルで維持することができる。従って、生体内のプロテイナーゼに対して抵抗性である限り、どんな非ペプチド性重合体も、制限なく、本発明で用いてもよい。非ペプチド性重合体の分子量は1〜100 kDaの範囲、好ましくは1〜20 kDaの範囲である。さらに、上記生理活性ポリペプチドと結合する本発明の非ペプチド性重合体は、1種類の重合体又は異なる重合体の組み合わせであてもよい。また本発明で有用である非ペプチド性重合体は、生理活性ポリペプチド及び免疫グロブリン不変領域と結合できる両末端又は三末端の官能基を有することができる。好ましくは、官能基はアルデヒド基であってもよい。
タンパク質薬剤の持続型製剤を製造するために、一般に用いられるPEGをとの結合は、タンパク質の安全性は増加させる。一方、PEGの分子量が増加を示すほどタンパク質との反応性が低くなって、よって製造収率が減少する。製造収率は、製造単価と産業適応性と密接な関係があるため、製造収率を高めることは非常に重要である。アルデヒド官能基を含んでいるPEGは、生理活性ポリペプチド又は免疫グロブリン不変領域のN-末端アミン基又はリジン残基側鎖のアミン基に結合することができ、その点では、PEGとタンパク質に対するモル比、反応溶液の濃度、反応時間、pH、温度などを含む様々な要因に依存して異なってもよい。非特許文献2では、インスリンペグ化は、モル比、反応時間、pH などを含む多様な要因を調節することにより、90%を超える高収率の5kアルデヒドmPEGが行われたことを記述している。特許文献5では、反応液に有機溶媒の添加によりペプチドペグ化の収率をの増加させることを報告している。特許文献6は、炭水化物の存在下で、PEGとr-metHuG-CSFを反応させることによって、ペグ収率の改善を記述している。
しかし、2つ以上の反応基を有するPEGを含む非ペプチド性重合体が2つの異なるポリペプチド間のリンカーとして用いられる場合、2回以上の反応段階によって、それによって全収率を低下させる。特に、2次反応段階(二つ以上の官能基を有する非ペプチド性重合体が結合された生理活性ポリペプチド又は免疫グロブリン不変領域がそれぞれ免疫グロブリン不変領域又は生理活性ポリペプチドと反応する、以下 「カップリング反応」という)。生理活性ポリペプチド又は免疫グロブリン不変領域が2つ以上の反応基を有する非ペプチド性重合体と反応させる1次反応段階と比べて顕著に低い収率で行われることが観察された。
本発明において、1次反応の中で還元剤の濃度は、2次カップリング反応の収率と相関性を有することを証明する。1次反応の中で低い濃度の還元剤、より短い反応時間、低い反応温度によりカップリング反応による高収率が観察された。
ここで用いられる用語として、「還元剤」は非ペプチド性重合体アルデヒド基とポリペプチド(生理活性ポリペプチド、免疫グロブリン不変領域)のアミン基との反応から形成される可逆的イミン二重結合を還元するために機能する混合物を意味する。それによって、共有結合を生成させる当該分野に知られたあらゆる合成物を意味し、本発明の目的上、還元剤は、非ペプチド性重合体が生理活性ポリペプチド又は免疫グロブリン不変領域と共有結合を形成する反応溶液に含まれてもよい。この技術は、一般的に使用される限り、どんな還元剤も本発明で使用されてもよい。還元剤の例として、これには制限されるものではないが、シアノ水素化ホウ素ナトリウム、ボラン-ピリジン複合体、水素化ホウ素ナトリウム、ボラン-ジメチルアミン コンプレックス、ボラン-トリメチルアミン コンプレックス又はトリアセトキシ水素化ほう素 ナトリウムを含んでもよい。適切な還元剤は、生理活性ポリペプチド又は免疫グロブリン不変領域の種類と反応溶媒に依存して、選択することができる。
還元剤は、生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体の間の結合のために用いられた。反応液は、生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体の間の結合反応の間の反応のために、1〜20 mMの濃度を含んでもよい。カップリング反応のために、1〜100 mMの濃度を含んでもよい。より好ましくは、還元剤は、生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体の間の結合反応のために、1〜20 mMの濃度で用いられてもよい。カップリング反応の時、1〜40 mMの濃度を含んでもよい。
生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体の間の結合反応(段階(1)の反応)時間は、1〜16時間で、0〜25℃の温度で行ってもよい。さらに、カップリング反応(段階 (2)の反応)の時間は1〜48 時間であってもよい。
好ましくは、段階(1)の反応は、1〜20 mMの濃度の還元剤の存在下で、0〜25℃で1〜16時間行い、一方、段階(2)の反応は、1〜40 mMの濃度の還元剤の存在下で 、1〜48時間行ってもよい。
本発明の様態として、インスリン、2つ以上のアルデヒド基を官能基として含むPEGリンカー及び免疫グロブリン不変領域がお互い結合された結合体の収率を増加させるために、還元剤としてシアノ水素化ホウ素ナトリウムを多様な条件で用いられた。生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体を連結する1次反応の時に低い濃度の還元剤の使用、短い反応時間、低い反応温度により、2次反応段階であるカップリング反応収率が向上することを確認した (表1)。
本発明の様態として、反応は、還元剤のボラン-ピリジン複合体を様々な濃度別で行われ、生理活性ポリペプチド又は免疫グロブリン不変領域と非ペプチド性重合体との1次反応の中で低い濃度の還元剤を用いるほど2次反応段階であるカップリング反応収率が向上することを確認した。還元剤として使われたシアノ水素化ホウ素ナトリウムはボラン-ピリジン複合体の使用の時に比べ高収率を示した(表2)。
また、本発明者は、2次カップリング反応の収率は、還元剤の濃度が高濃度であるほど向上することを確認した。
本発明の様態として、カップル反応は還元剤のシアノ水素化ホウ素ナトリウムの様々な濃度で行われ、高濃度の還元剤の存在下で、カップリング反応の改善された収率が得られた。しかし、非常に高濃度の還元剤は、免疫グロブリン不変領域の変性を引き起こす。これを避けるために、20 mMのシアノ水素化ホウ素ナトリウム存在下で、13時間行われ、これらの収率は、維持され、免疫グロブリン不変領域の変性が結果的に最小化されることが観察された(表3及び表4)。
ここで用いられた用語「生理活性ポリペプチド」とは、一般的概念として生体内である生理機能を有するポリペプチドを意味し、ポリペプチド構造を有するという共通点を有し、多様な生理活性を示す。特定の機能に関与する物質の欠乏や過度な結果で体が生物学的に異常である時、上記生理活性ポリペプチドは、遺伝子発現と生理機能を調整し、それによって、異常を修正する。一般的な例は、タンパク質薬物である。

本発明の様態で用いられたインスリンは、体内の血糖値が高い時、膵臓から分泌され、血液から肝、筋肉、脂肪組織から糖を吸収してグリコーゲンとして貯蔵することにより、脂肪を分解を抑制することによって、エネルギー源として用いられることを抑制して血糖値を調節する機能を有する生理活性ペプチドの一種である。生理活性ペプチドは、インスリンアゴニスト、前駆物質、誘導体、断片、変異体などを含む。天然型インスリン、速効性インスリン、持続型インスリンが好ましい。
天然型インスリンは、膵臓から分泌するホルモンであり、細胞内グルコースの吸収を促進し、脂肪の分解を抑制して体内の血糖値を調節する重要な役割をする。血糖値調節機能を有するインスリンは、血糖値調節機能がないプロインスリン前駆体から多様に生成される。インスリンのアミノ酸配列は下記の通りである。

インスリンアゴニストは、インスリンの構造と関係なく、生体内でインスリン受容体に結合でき、インスリン同様の生物学的活性を示す物質を意味する。
インスリン誘導体は、天然型インスリンと少なくとも80%以上がアミノ酸配列で相同性を示し、それらは、体内で血糖値を調節する機能を有するペプチドを意味する。アミノ酸残基の一部のグループが化学的に置換(例; alpha-methylation、alpha-hydroxylation)、除去(例; deamination)又は修飾(例; N-methylation、glycosylation、fatty acid)されたでものであってもよい。
インスリン断片は、インスリンのアミノ末端又はカルボキシ末端から少なくとも1つ又はそれ以上のアミノ酸が追加又は削除されることにより製造された体内で血糖値調節機能を有するペプチドを意味する。追加されたアミノ酸は、天然に存在しないアミノ酸(例; D型アミノ酸)でもよい。
インスリン変異体は、天然型インスリンとアミノ酸残基が一つ以上異なるアミノ酸配列を有するペプチドであり、しかし、体内で血糖値調節機能を有するペプチドを意味する。
インスリンアゴニスト、断片及び変異体の製造のために、それぞれ用いられた方法は、独立的にも、組み合わせで用いられてもよい。例えば、本発明でインスリンペプチドは、天然型インスリンとアミノ酸残基が1つ以上異なり、N-末端アミノ酸残基が脱アミノ化された、アミノ酸配列を有し、体内で血糖値調節機能を有するペプチドを含んでもよい。
ここで用いられる用語、「免疫グロブリン不変領域」とは、軽鎖及び重鎖可変領域、重鎖そして不変領域1(CH1)と軽鎖不変領域(CL)を除いた、免疫グロブリンの断片を意味し、これらのFc領域は、重鎖不変領域2(CH2)及び重鎖不変領域3(CH3)(又は重鎖不変領域4(CH4)を含む)の部分を含む。重鎖不変領域にヒンジ部分を含むこともある。必要によって、免疫グロブリンFc領域は、ヒンジ領域をさらに含む。また、本発明の免疫グロブリン不変領域は、天然型の免疫グロブリン不変領域が、実質的に同等又は向上した効果を有する限り、免疫グロブリンの重鎖と軽鎖可変領域のみを除いて、一部又は全体重鎖不変領域1(CH1)及び/又は軽鎖不変領域(CL)を含む拡張されてもよい。さらに、本発明の免疫グロブリン不変領域は、CH2及び/又はCH3に該当する重要な部分のアミノ酸配列が除去された領域であってもよい。従って、本発明の免疫グロブリン不変領域は、(2)CH1ドメイン、CH2ドメイン、CH3ドメイン及びCH4ドメイン、(2)CH1ドメイン及びCH2ドメイン、(3)CH1ドメイン及びCH3ドメイン、(4)CH2ドメイン及びCH3ドメイン、(5)1つ又は2つ以上の不変領域ドメインと免疫グロブリンヒンジ領域(又はヒンジ領域の一部)との組み合わせ、(6)重鎖不変領域各ドメインと軽鎖不変領域の二量体であり得る。
Fc領域を含む免疫グロブリン不変領域は、生体内で代謝される生分解性のポリペプチドである。そのため、薬物のキャリアとして安定に用いることができる。さらに、免疫グロブリンFc領域は、免疫グロブリン全体分子に比べ比較的に低い分子量であるため、結合体の製造、精製及び収率の面でより有利である。さらに、アミノ酸配列が抗体ごとに異って、高い非均質性を示すFab部分が除去されるため、同質性が著しく増加され、血中抗原性の誘発可能性を低下させた結合体を提供する。
免疫グロブリン不変領域は、ヒト又は牛、山羊、豚、マウス、ラビット、ハムスター、ラット、ギニアピッグなどの動物起源であり得、好ましくは、ヒト起源である。また、免疫グロブリン不変領域は、IgG、IgA、IgD、IgE、IgM来由又はこれらの組み合わせ又はこれらのハイブリッドによる不変領域からなる群から選択されることができる。好ましくは、ヒトの血液に最も豊かなIgG又はIgM来由であり、最も好ましくは、リガンド結合タンパク質の血液の半減期を改善させると公知となったIgG来由であり得る。
本発明において、「組み合わせ」とは、二量体又は多量体を形成するために、同一起源の短鎖免疫グロブリン不変領域(好ましくが、Fc領域)をコードするポリペプチドが相違する起源の短鎖ポリペプチドと結合を形成することを意味する。すなわち、IgG Fc、IgA Fc、IgM Fc、IgD Fc及びIgEのFc断片からなる群から選択された2つ以上の組み合わせにより製造された二量体又は多量体であってもよい。
本発明において、「ハイブリッド」とは、単鎖の免疫グロブリン不変領域(好ましくは、Fc領域)内に存在する2個以上の相違する起源の免疫グロブリン不変領域コードする配列を意味する用語である。本発明で、種々の形態のハイブリッドが可能である。例えば、IgG Fcの CH1、CH2、CH3及びCH4、IgM Fc、IgA Fc、IgE Fc及びIgD Fcからなるグループから選択される1つ〜4つのドメインによるハイブリッドドメインで構成され、ヒンジを含むことができる。
IgGはIgG1、IgG2、IgG3及びIgG4のサーブクラスに分けられ、本発明では、これらの組み合わせ又はこれらのハイブリッドもを含んでもよい。好ましくは、IgG2及びIgG4サーブクラスであり、最も好ましくは、補体依存的毒性(CDC)のようなエフェクター機能がほとんどないIgG4のFc領域であってもよい。
免疫グロブリン不変領域は、天然型糖鎖、天然型に比べて増加された糖鎖、天然型に比べて減少した糖鎖又は糖鎖が除去された形態であり得る。免疫グロブリン領域の糖鎖付加又は除去の増減又は減少は、例えば、化学的方法、酵素学的方法及び微生物を用いた遺伝工学的方法の利用による通常の方法により取得された。ここで、脱グリコシル化の時に、補体(C1q)の免疫グロブリン不変領域の結合力が顕著に低下され、抗体-依存性細胞毒性又は補体-依存性細胞毒性が減少又は除去されるため、それによって、生体内の不必要な免疫反応は誘発しない。このような点で、糖鎖が除去されているか又は非糖鎖化された免疫グロブリン不変領域は、薬物のキャリアとしての本来の目的により符合する。従って、本発明の薬物キャリアとして最も適切な免疫グロブリンFc領域は、、ヒトIgG4来由の非-糖鎖化されたFc領域である。ヒト来由のFc領域は、ヒト生体で抗原として作用し、抗原に対する新たな抗体を生成するなどの好ましくない免疫反応を起こす非-ヒト来由のFc領域に比べて好ましい。
さらに、本発明において、天然型アミノ酸配列を有する免疫グロブリン不変領域だけではなく、そのアミノ酸配列変異体は、免疫グロブリン不変領域の範囲内に含まれてもよい。ここで用いられる用語「アミノ酸配列誘導体」とは、天然アミノ酸配列中の一つ以上のアミノ酸残基が欠失、挿入、非保全的又は保全的置換又はこれらの組み合わせの結果、野生型と相違したアミノ酸配列を有するポリペプチドを意味する。例えば、IgG Fcの場合、結合に重要であると知られた214〜238、297〜299、318〜322 又は327〜331番のアミノ酸残基が変形のために適当な部位として用いられてもよい。二硫化結合を形成可能な部位が除去されたり、天然型 FcからN-末端のいくつかのアミノ酸が除去されたり、又は天然型FcのN-末端にメチオニン残基が付加するなど、多様な種類の誘導体が可能である。さらに、エフェクター機能を無くすために、補体結合部位、例えば、C1q結合部位又はADCC部位が天然型Fc領域から除去されるでもよい。免疫グロブリン不変領域のアミノ酸配列変異体を製造する技術は、特許文献7,8などに開示されている。
分子の活性を変更させないペプチド分子又はタンパク質のアミノ酸置換は、該当分野に公知となっている(H.Neurath、R.L.Hill、The Proteins、Academic Press、New York,197 9)。最も一般的な置換は、アミノ酸残基 Ala/Ser、Val/Ile、Asp/Glu、Thr/Ser、Ala/Gly、Ala/Thr、Ser/Asn、Ala/Val、Ser/Gly、Thr/Phe、Ala/Pro、Lys/Arg、Asp/Asn、Leu/Ile、Leu/Val、Ala/Glu、Asp/Gly間で起こる。場合によっては、アミノ酸がリン酸化、硫化、アクリル化、糖化、メチル化、ファルネシル化、アセチル化、アミル化などにより修飾されてもよい。
上記記述した免疫グロブリン不変領域誘導体は、本発明の免疫グロブリン不変領域と同一な生物学的活性を示すが、しかし、熱、pH などに対する構造的安全性を改善した。
この免疫グロブリン不変領域は、ヒト及び牛、山羊、豚、マウス、ラビット、ハムスター、ラット、ギニアピッグなどの動物又はヒトから単離した天然型から得られることもでき、又は形質転換された動物細胞微生物から得られた組換え型又はこの誘導体であってもよい。天然型不変領域は、 ヒト又は動物の生体から分離した全体免疫グロブリンをタンパク質分解酵素を処理することにより得ることができる。免疫グロブリンは、パパインによりFab及びFCに切断され、続けて、ペプシンによりpF'c及びF(ab)2に切断される。これをサイズ排除クロマトグラフィーなどによりFc又はpF'cを分離することができる。
この免疫グロブリン不変領域は、ヒト及び牛、山羊、豚、マウス、ラビット、ハムスター、ラット、ギニアピッグなどの動物又はヒトから単離した天然型から得られることもでき、又は形質転換された動物細胞微生物から得られた組換え型又はこの誘導体であってもよい。天然型不変領域は、 ヒト又は動物の生体から分離した全体免疫グロブリンをタンパク質分解酵素を処理することにより得ることができる。免疫グロブリンは、パパインによりFab及びFCに切断され、続けて、ペプシンによりpF'c及びF(ab)2に切断される。これをサイズ排除クロマトグラフィーなどによりFc又はpF'cを分離することができる。
好ましくは、組換えヒト免疫グロブリン不変領域は、微生物から得た。
上述のように、生理活性ポリペプチド-非ペプチド性重合体-免疫グロブリン不変領域結合体は、本発明の製造方法により高純度と高収率だけでなく、低コストで製造することができる。従って、本発明の方法は、産業的に有用である。さらに、薬の順応度を改善した生理活性ポリペプチドの持続型剤形の開発に有用に用いられることができる。
本発明のさらなる理解は、例示するための次の例により得ることができる。しかし、本発明をに限定するものとして行われるわけではない。
実施例1: シアノ水素化ホウ素ナトリウム還元剤を用いたインスリンのペグ化及びモノペグ化インスリンの精製
インスリン粉末を10 mM HClに溶解させた後、3.4k propion-ALD2 PEG (プロピオンアルデヒド基を2個有しているPEG、IDB、韓国)をベータチェーンのN-末端をペグ化させた。5mg/mlのインスリンは、PEGとのモル比を1 : 2で、4℃〜常温で約2時間反応させた。反応は50 mMのクエン酸ナトリウムpH 6.0、45%のイソプロパノールからなり、2〜20 mMの濃度のシアノ水素化ホウ素ナトリウム還元剤の存在下で反応させた。反応液は、クエン酸ナトリウム(pH 3.0)、45%のEtOHを含むバッファーの溶出とKCl濃度勾配を用い、SP-HP(GE Healthcare)カラム上でロードしてモノペグ化インスリンを精製した。
下記表1は、インスリンと免疫グロブリンFc断片を含む結合体の製造の時、還元剤のシアノ水素化ホウ素ナトリウム条件によるインスリンのペグ化収率を示す。
実施例2: ペグ化を用いた、シアノ水素化ホウ素ナトリウム還元剤の条件によるモノペグ化インスリン-免疫グロブリンFc領域結合体の製造収率変化。
インスリン-PEG-免疫グロブリンFc領域結合体の製造の収率を検査するために、実施例1で製造されたモノペグ化したインスリンは、免疫グロブリンFcとのモル比が1:1で、全体タンパク質の濃度を20 mg/mlで、25℃で13時間反応させた。このカップリング反応は、還元剤として20 mMのシアノ水素化ホウ素ナトリウムの存在下で、22 mMのリン酸カリウム、 10%のエチルアルコール pH 8.2を含む100 mMのHEPESバッファーの中で行われた。
反応液は、反応しないインスリン、反応しない免疫グロブリンFc領域、インスリン-PEG-免疫グロブリンFc領域複合体、モノ−ペグ化インスリン(インスリン-PEG)が2個以上結合した免疫グロブリンFc領域を、Tris-HCl(pH 7.5)バッファーとNaCl濃度勾配を用いて、Source 15Q(GE Healthcare)カラムにロードして分離精製した。インスリン-PEG-免疫グロブリンFc領域複合体の製造の収率は、クロマトグラフィーによる精製の後、280nmのUV吸光度で決定された。
下記表1に、インスリンのペグにおける免疫グロブリンFc領域を含むカップリング反応の収率が、還元剤として用いたシアノ水素化ホウ素ナトリウム条件により要約される。
実施例3: ボラン-ピリジン複合体還元剤を用いたインスリンのペグ化及びモノペグ化インスリンの精製
インスリン粉末を10 mM HClに溶解させ、3.4k propion-ALD2 PEG (プロピオンアルデヒド基を2個有しているPEG、IDB、韓国)をインスリンベータチェーンのN-末端にペグ化した。このような点で、5mg/mlのインスリンは、PEGとのモル比を1 : 2で、4℃で約2時間反応させた。この反応は、還元剤として3〜20 mMのボラン-ピリジン複合体還元剤の存在下で、45%のイソプロパノール中の50 mMのクエン酸ナトリウムバッファーpH 6.0の中で行われた。反応液は、クエン酸ナトリウム (pH 3.0)、45%のEtOHを含むバッファーの溶出とKCl濃度勾配を用い、SP-HP (GE Healthcare)カラム上でロードしてモノペグ化インスリンを精製した。
下記表2に、インスリンと免疫グロブリンFc領域を含む結合体の製造間、還元剤ボラン-ピリジン複合体条件によるインスリンのペグ化が下記の表2に要約される。
実施例4: ペグ化を用いたボラン-ピリジン複合体還元剤の条件によるモノペグ化インスリン-免疫グロブリンFc領域複合体の製造収率の変化。
還元剤の濃度によるインスリン-PEG-免疫グロブリンFc領域結合体の製造の収率を検査するために、実施例3の方法で製造されたモノペグ化したインスリンが、免疫グロブリンFc断片とのモル比が1:1にで、全体タンパク質の濃度を20 mg/mlにして25℃で13時間反応させた。このカップリング反応は、20 mMのシアノ水素化ホウ素ナトリウムの存在下で、22 mMのリン酸カリウム、pH 8.2の10 %のエチルアルコールを含む100 mMのHEPESの中で行われた。
反応液は、反応しないインスリン、反応しない免疫グロブリンFc断片、インスリン-PEG-免疫グロブリンFc断片結合体、モノペグ化インスリン(インスリン-PEG)が2個以上結合した免疫グロブリンFc断片結合体を、Tris-HCl(pH 7.5)バッファーの溶出とNaCl濃度勾配を用い、Source 15Q (GE Healthcare)カラム上でロードし分離精製した。インスリン-PEG-免疫グロブリンFc領域複合体の製造の収率は、クロマトグラフィーによる精製の後、280nmのUV吸光計で決定された。
表2において、免疫グロブリンFc領域とカップリング反応の収率がをインスリンのペグ化における還元剤ボラン-ピリジン複合体の条件により要約される。
実施例5: シアノ水素化ホウ素ナトリウム還元剤の濃度と反応時間によるカップリング反応収率及び免疫グロブリンFc変形体形成。
カップリング反応において、還元剤の濃度と反応時間による免疫グロブリンFc変形体の形成を検査するために、モノペグ化したインスリンと免疫グロブリンFcのモル比が 1:1になるようにし、全体タンパク質の濃度を20 mg/mlにして25℃で13〜43時間反応させた。このカップリング反応は、 還元剤として5〜40 mMのシアノ水素化ホウ素ナトリウムの存在下で、22 mMのリン酸カリウム、10%のエチルアルコール pH 8.2を含む100 mMのHEPESの中で行われた。
反応液は、 反応しないインスリン、反応しない免疫グロブリンFc領域、インスリン-PEG-免疫グロブリンFc領域複合体、モノペグ化インスリン(インスリン-PEG)が2個以上結合した免疫グロブリンFc領域複合体を、Tris-HCl(pH 7.5)バッファーの溶出とNaCl濃度勾配を用い、Source 15Q(GE Healthcare)カラム上でロードし分離精製した。インスリン-PEG-免疫グロブリンFc領域複合体の製造の収率は、クロマトグラフィーによる精製の後、280nmのUV吸光度で決定された。
表3に、インスリンと免疫グロブリンFc断片を含む結合体の製造のためのカップリング反応の収率が、還元剤のシアノ水素化ホウ素ナトリウム濃度と反応時間により要約された。
カップリング反応において、還元剤の濃度による免疫グロブリンFc変形体の形成は、NaCl濃度勾配を用い、そしてTris-HCl(pH 8.0)バッファーに溶出したPropac SAX-10(DIONEX)カラム上のLCにより観察した。
表4は、インスリンと免疫グロブリンFc領域を含む結合体の製造のためのカップリング反応におけるシアノ水素化ホウ素ナトリウム濃度とカップリング反応時間による免疫グロブリンFc変形体の製造の収率を示す。
実施例6: シアノ水素化ホウ素ナトリウム還元剤を用いた免疫グロブリンFcのペグ化及びモノペグ化した免疫グロブリンFcの精製
免疫グロブリンFcのN-末端は、5K propion-ALD2 PEG(プロピオンアルデヒド基を3つ有しているPEG、NOF、日本)とペグ化された。このような点で、10 mg/mlの免疫グロブリンFcは、PEGとのモル比を1 : 2で、4℃〜常温で約4.5時間反応させた。この反応は、還元剤として20 mMの濃度のシアノ水素化ホウ素ナトリウムの存在下で、100 mMのリン酸カリウム pH 6.0の中で行われた。反応液は、Tris-HCL(pH 7.5)バッファーの溶出とNaCl濃度勾配を用い、Source 15Qカラム上でロードし、モノペグ化した免疫インスリンを精製した。
実施例7: シアノ水素化ホウ素ナトリウム還元剤を用いたモノペグ化した免疫グロブリンFc領域-インスリン結合体の製造
インスリン-PEG-免疫グロブリンFc領域複合体の製造のために、実施例6のモノペグ化した免疫グロブリンFcは、インスリンのモル比が 1 : 4で、全体タンパク質の濃度を20 mg/mlにして 4℃で約13時間反応させた。このカップリング反応は、還元剤として20 mMのシアノ水素化ホウ素ナトリウムの存在下で、100 mMのリン酸カリウムバッファーpH 6.0の中で行われた。
反応液は、Source 15Qカラム上にロードされ1次精製した。2次生成は、さらにSource 15ISO(GE Healthcare)カラムで行い、インスリン-PEG-免疫グロブリンFc領域複合体を得た。
次に、本発明の好ましい態様を示す。
1. (1)1〜20mMの還元剤の存在下で、二つ以上のアルデヒド官能基を有する非ペプチド性重合体と生理活性ポリペプチド又は免疫グロブリン不変領域のいずれか一つと反応させる段階;及び
(2)1〜100mMの還元剤の存在下で、段階(1)の反応混合物を生理活性ポリペプチド又は免疫グロブリン不変領域の他の反応段階を含む生理活性ポリペプチド-非ペプチド性重合体-免疫グロブリン不変領域複合体の製造方法。
2. さらに、段階(1)の後、反応混合物から生理活性ポリペプチド-非ペプチド性重合体の結合体又は免疫グロブリン不変領域-非ペプチド性重合体の結合体の分離をさらに含む上記1に記載の方法。
3. 前記還元剤は、非ペプチド性重合体のアルデヒド基と生理活性ポリペプチド又は免疫グロブリン不変領域のアミン基が結合して生成される可逆的イミン二重結合を還元させることにより、共有結合を形成する作用をする上記1に記載の方法。
4. 前記還元剤は、シアノ水素化ホウ素ナトリウム、ボラン-ピリジン複合体、水素化ホウ素ナトリウム、ボラン-ジメチルアミン複合体、ボラン-トリメチルアミン複合体又はトリアセトキシ水素化ほう素 ナトリウムからなる群から選択される上記1に記載の方法。
5. 前記還元剤は、段階(2)において、1〜40 mMの濃度で用いられる上記1に記載の方法。
6. 段階(1)における反応は、1〜16時間で行われる上記1に記載の方法。
7. 段階(2)における反応は、1〜48時間で行われる上記1に記載の方法。
8. 段階(1)における反応は、0〜25℃の温度で行われる上記1に記載の方法。
9. 段階(1)における反応は、1〜20 mMの濃度の還元剤の存在下で、1〜16時間、0〜25℃で行われ、段階(2)における反応は、1〜40mMの濃度の還元剤の存在下で、1〜48 時間行われる上記1に記載の方法。
10. 非ペプチド性重合体が、その二つ以上のアルデヒド官能基を通じて生理活性ポリペプチド及び免疫グロブリン不変領域とそれぞれ共有結合している上記1に記載の方法。
11. 非ペプチド性重合体の官能基が生理活性ポリペプチド及び免疫グロブリン不変領域とそれぞれ結合され、ここでアミノ基はN-末端又はリジン残基に存在する上記1に記載の方法。

(2)1〜100mMの還元剤の存在下で、段階(1)の反応混合物を生理活性ポリペプチド又は免疫グロブリン不変領域の他の反応段階を含む生理活性ポリペプチド-非ペプチド性重合体-免疫グロブリン不変領域複合体の製造方法。
2. さらに、段階(1)の後、反応混合物から生理活性ポリペプチド-非ペプチド性重合体の結合体又は免疫グロブリン不変領域-非ペプチド性重合体の結合体の分離をさらに含む上記1に記載の方法。
3. 前記還元剤は、非ペプチド性重合体のアルデヒド基と生理活性ポリペプチド又は免疫グロブリン不変領域のアミン基が結合して生成される可逆的イミン二重結合を還元させることにより、共有結合を形成する作用をする上記1に記載の方法。
4. 前記還元剤は、シアノ水素化ホウ素ナトリウム、ボラン-ピリジン複合体、水素化ホウ素ナトリウム、ボラン-ジメチルアミン複合体、ボラン-トリメチルアミン複合体又はトリアセトキシ水素化ほう素 ナトリウムからなる群から選択される上記1に記載の方法。

- (1)1〜20mM未満の還元剤の存在下で、二つ以上のアルデヒド官能基を有する非ペプチド性重合体と生理活性ポリペプチド又は免疫グロブリン不変領域のいずれか一つと反応させる段階;及び
(2)1〜100mMの還元剤の存在下で、段階(1)の反応混合物を生理活性ポリペプチド又は免疫グロブリン不変領域の他方と反応させる段階を含む生理活性ポリペプチド-非ペプチド性重合体-免疫グロブリン不変領域複合体の製造方法。 - さらに、段階(1)の後、反応混合物から生理活性ポリペプチド-非ペプチド性重合体の結合体又は免疫グロブリン不変領域-非ペプチド性重合体の結合体の分離をさらに含む請求項1に記載の方法。
- 前記還元剤は、非ペプチド性重合体のアルデヒド基と生理活性ポリペプチド又は免疫グロブリン不変領域のアミン基が結合して生成される可逆的イミン二重結合を還元させることにより、共有結合を形成する作用をする請求項1に記載の方法。
- 前記還元剤は、シアノ水素化ホウ素ナトリウム、ボラン-ピリジン複合体、水素化ホウ素ナトリウム、ボラン-ジメチルアミン複合体、ボラン-トリメチルアミン複合体及びトリアセトキシ水素化ほう素ナトリウムからなる群から選択される請求項1に記載の方法。
- 前記還元剤は、段階(2)において、1〜40mMの濃度で用いられる請求項1に記載の方法。
- 段階(1)における反応は、1〜16時間で行われる請求項1に記載の方法。
- 段階(2)における反応は、1〜48時間で行われる請求項1に記載の方法。
- 段階(1)における反応は、0〜25℃の温度で行われる請求項1に記載の方法。
- 段階(1)における反応は、1〜20mM未満の濃度の還元剤の存在下で、1〜16時間、0〜25℃で行われ、段階(2)における反応は、1〜40mMの濃度の還元剤の存在下で、1〜48時間行われる請求項1に記載の方法。
- 非ペプチド性重合体が、その二つ以上のアルデヒド官能基を通じて生理活性ポリペプチド及び免疫グロブリン不変領域とそれぞれ共有結合している請求項1に記載の方法。
- 非ペプチド性重合体の官能基が、生理活性ポリペプチドのN-末端又はリジン残基側鎖及び免疫グロブリン不変領域のN-末端又はリジン残基側鎖とそれぞれ結合される請求項1に記載の方法。
- 非ペプチド性重合体が、ポリエチレングリコール、ポリプロピレングリコール、エチレングリコールとプロピレングリコールとの共重合体、ポリオキシエチル化ポリオール、ポリビニルアルコール、ポリサッカライド、デキストラン、ポリビニルエチルエーテル、ポリ乳酸(PLA)及びポリ乳酸-グリコール酸(PLGA)、脂質重合体、キチン類、ヒアルロン酸及びこれらの組み合わせからなる群から選択される請求項1に記載の方法。
- 非ペプチド性重合体が、ポリエチレングリコールである請求項1に記載の方法。
- 非ペプチド性重合体の分子量が、1〜100kDaの範囲である請求項1に記載の方法。
- 免疫グロブリン不変領域が、非糖鎖化されることを特徴とする請求項1に記載の方法。
- 免疫グロブリン不変領域が、CH1、CH2、CH3及びCH4ドメインからなる群から選択された1つ〜4つのドメインからなる請求項1に記載の方法。
- 免疫グロブリン不変領域が、ヒンジ領域をさらに含む請求項1に記載の方法。
- 免疫グロブリン不変領域が、IgG、IgA、IgD、IgE、IgM又はこれらの組み合わせ又はこれらのハイブリッドから由来する不変領域からなる群から選択される請求項1に記載の方法。
- 免疫グロブリン不変領域が、IgG1、IgG2、IgG3、IgG4、これらの組み合わせ及びこれらのハイブリッドの不変領域からなる群から選択される請求項1に記載の方法。
