CRM197トキソイドに融合したN末端シグナル配列をコードする配列を含むプラスミドを、標準的な分子生物学技術を用いて作製した。13の異なるシグナル配列を用い(表2)、これらをGenEMBLデータベースから選択した。
本実験において、実施例1において作製されたものと同様だが、ペリプラズムシグナル配列を欠失するプラスミドを作製した。この設計プロセスを図2に要約する。まず、CRM197配列(配列番号31)をMDSCRM1(配列番号27)およびMDSCRM2(配列番号28)と命名された2つのプライマーをテンプレートとして用いて、標準的なPCR技術によって増幅した(使用した温度周期は、(94℃2分―55℃2分―72℃2分30秒)×25回―72℃10分―終止4℃であった)。
シグナル配列FlgIを、改良されたコンストラクトを作製するための使用のために選択した。以下のクローン化プロセスを図3に要約する。
本実験において、実施例1において作製したDsbAシグナル配列コンストラクトで形質転換した細胞を培養し、発現を誘導した。B834(DE3)、HMS174(DE3)、およびBLR(DE3)組換え株の前培養物(3mL)を、カナマイシン(50μg/mL)の存在下で1%グルコースを補充したLBT培地において37℃で一晩、攪拌の下で増殖した。翌日、培地の光学密度OD620が0.1に到達した後、この前培養物の試料を、50μg/mLのカナマイシンを有する20mLのLBT培地へと転化した。これらの細胞を撹拌の下で30℃で増殖しておいた。このプラスミドにおいて、CRM197発現は、lacオペレーターの制御下にあり、それゆえ、コードされたCRM197の発現は、イソプロピル−ベータ−Dチオガラクトピラノシド(IPTG)の添加の際に誘導されることができる。光学密度が0.6に到達すると、IPTG(1mMの終濃度になるまで添加)を用いた誘導を実施し、培養物をさらに30℃で3時間増殖させておいた。
実施例4の工程を反復したが、この場合、100mMのK2HPO4/KH2PO4緩衝液を誘導前に培養物に添加し、それにより培地をpH7.5に緩衝した。発現を1mMのIPTGの添加によって誘導し、培養物を23℃または30℃のいずれかで一晩増殖させておいた。上清およびペレットにおいて発現したタンパク質のレベルを、実施例4において説明されるように、クーマシー染色およびウェスタンブロット技術を用いて評価した。
DE3細胞を実施例2において製造したプラスミドで形質転換し、培養し、CRM197の発現を実施例4において説明される通り誘導した。実施例4に説明される通り、CRM197発現レベルを、SDS−PAGEゲルにおおける全細菌細胞の産物を泳動し、クーマシーブルー染色およびウェスタンブロット技術を用いることによって評価した。細菌抽出を標準的なPBS緩衝液におけるワンショット技術(Constantの系)によって実施した。遠心分離後、得られた可溶性画分および不溶性画分をゲルに負荷した。
BLR(DE3)大腸菌細胞を実施例3において製造したコンストラクトで形質転換し、培養し、実施例5の最適化されたプロトコールにおいて説明された通り、発現を誘導した。加えて、いくつかの試料細胞を、100mMのK2HPO4/KH2PO4緩衝液の添加なしで増殖した。同様に、発現したタンパク質のレベルを、実施例4に説明される通り、クーマシー染色およびウェスタンブロット技術を用いて評価した。誘導時に、温度を23℃または30℃のいずれかで維持し、試料を誘導時、誘導2時間後、誘導4時間後、および一晩の誘導後に採取した。
前培養物を、大腸菌株B2355の凍結種培養を用いて調製した。この株は、大腸菌由来のFlgIのシグナルペプチドとCRM197の成熟部分の間の融合タンパク質をコードする配列を含むpET26b誘導体(これは、図3および実施例3において説明されたプラスミドpRIT16681である)を用いて形質転換されたB834(DE3)株である。シード培養能は、1mLあたりおよそ1×1010コロニー形成単位として決定された。
方法
20リットルの発酵槽(Biolafitte)を用いた。9リットルのバッチ相培地を発酵槽へと無菌的に転移させた(Zabriskie et al. (J. Ind. Microbiol. 2:87-95 (1987))から応用)。培地のpHを塩基の添加により、6.8に再調整した。次に、3mLの希釈していない照射済み消泡剤(SAG471)も発酵槽に添加した。温度(28℃)、頭部圧力(0.5バール)、曝気速度(1分間当たり20リットルの撒き散らした空気)、および初期撹拌速度(300rpm)を設定した後に接種した。これらの条件下の溶存酸素レベルは100%であった。頭部圧力および曝気速度を、発酵の間定常レベルで維持した。
本実験において、応答−表面方法論(J.ind. Microbiol. Biotechnol. 37:195-204 (2010))を用いて、組換えタンパク質のペリプラズム産生を最大化するために、3つのパラメータについての最適な値を決定した。研究した3つの発酵パラメータは、増殖相の間のpH、誘導中のpH、および誘導中の供給速度であった。これら3つのパラメータについての値を、Doehlertの均一なシェル設計(Doehlert (Applied Statistics 19:231-239 (1970)))に従って選択した。表5に示される値を用いて、15の発酵を実施した。

c.配列番号24の少なくとも10のアミノ酸の断片であって、前記細菌宿主細胞のペリプラズムへの前記細菌毒素ポリペプチドの輸送を配向させることができる断片、
をコードする、請求項1〜3のいずれか一項に記載のポリヌクレオチド。 - 前記3’毒素部分は、配列番号32のアミノ酸配列を有するポリペプチドをコードする、請求項1〜5のいずれか一項に記載のポリヌクレオチド。
- 前記3’毒素部分は、配列番号31を含んでなる、請求項1〜5のいずれか一項に記載のポリヌクレオチド。
- 前記5’シグナル配列部分は、前記3’毒素部分の直ちに5’である、請求項1〜7のいずれか一項に記載のポリヌクレオチド。
- 誘導性プロモーターに連結された請求項1〜8のいずれか一項に記載のポリヌクレオチドを含んでなるベクター。
- 前記誘導性プロモーターは、十分な量のイソプロピル−ベータ−D−チオガラクトピラノシド(IPTG)の添加によって活性化される、請求項9に記載のベクター。
- 請求項9または10に記載のベクターを含んでなる宿主細胞。
- 前記宿主細胞は、グラム陰性菌である、請求項11に記載の宿主細胞。
- 前記宿主細胞は、大腸菌細胞である、請求項12に記載の宿主細胞。
- a)請求項11〜13のいずれか一項に記載の細菌宿主細胞の培養物を増殖する工程と、
b)細菌トキソイドがペリプラズムに発現するよう、前記ポリペプチドの発現を誘導する工程と
を含んでなる、細菌毒素を作製するための方法。 - A.グラム陰性宿主細胞の培養物を増殖すること、および
B.タンパク質がペリプラズムに発現するよう、ポリペプチドの発現を誘導すること
による、組換えポリペプチドのペリプラズム発現のための方法であって、この中で、以下の工程のうちの1つ以上は発現中に作用する:
i.工程a)のpHは、工程b)のpHよりも低く、
ii.工程a)の温度は、工程b)の温度よりも高く、または

- 工程a)のpHは、工程b)のpHよりも低い、請求項14〜17のいずれか一項に記載の方法。
- 工程a)において、pHは6.0〜7.0のまたは6.5〜7.0の範囲である、請求項14〜19のいずれか一項に記載の方法。
- 工程b)において、pHは、6.5〜10.0、7.0〜8.0、6.5〜9.0、または7.0〜8.5の範囲である、請求項14〜20のいずれか一項に記載の方法。
- 工程b)の間、pHは維持される、請求項21に記載の方法。
- 前記pHは、リン酸緩衝液、トリス緩衝液、およびヒスチジン緩衝液からなる群に由来する緩衝液を用いて維持される、請求項21または22のいずれか一項に記載の方法。
- 前記緩衝液は、10〜200mM、50〜100mM、10〜50mM、100〜200mM、または50〜150mMの濃度である、請求項23に記載の方法。
- 前記緩衝液は、80〜100mMのリン酸緩衝液である、請求項14〜24のいずれか一項に記載の方法。
- 前記方法は、発酵槽において実施される、請求項14〜25のいずれか一項に記載の方法。
- 工程b)のpHの上昇は、塩基、例えば水酸化ナトリウムまたはアンモニウムの添加によって達成される、請求項19〜26のいずれか一項に記載の方法。
- 前記pHは、工程b)の間、水酸化ナトリウムまたはアンモニウムの添加によって、6.5〜10.0、7.0〜8.0、6.5〜9.0、または7.0〜8.5の間に維持される、請求項18〜27のいずれか一項に記載の方法。
- 工程a)における第一の基質供給速度と、工程b)における第二の基質供給速度とを含んでなり、かつ、該第二の基質供給速度は、該第一の基質供給速度よりも低い、請求項14〜28のいずれか一項に記載の方法。
- 前記第二の基質供給速度は、前記第一の基質供給速度の5%〜90%、20%〜80%、または20%〜30%の間に維持される、請求項29に記載の方法。
- 前記細菌毒素は、ジフテリア毒素またはその点突然変異したバリアントである、請求項14〜30のいずれか一項に記載の方法。
- 前記細菌毒素は、CRM197である、請求項31に記載の方法。
- 前記細菌毒素は破傷風毒素である、請求項14〜30のいずれか一項に記載の方法。
- 工程a)の温度は、工程b)の温度よりも高い、請求項14〜33のいずれか一項に記載の方法。
- 工程a)は、20〜40℃の温度で実施される、請求項14〜34のいずれか一項に記載の方法。
- 工程b)は、20〜28℃、21〜27℃、21〜24℃、または22〜23℃の温度で実施される、請求項14〜34のいずれか一項に記載の方法。
- 発現は、十分な量のIPTGの添加によって、工程b)において誘導される、請求項14〜36のいずれか一項に記載の方法。
- IPTGは、0.1〜10mMの間の終濃度になるよう添加される、請求項37に記載の方法。
- 前記宿主細胞は、誘導時に0〜2.5の間の、または0.4〜1.5の間の光学密度(OD650)を有する、請求項14〜38のいずれか一項に記載の方法。
- 前記発酵槽は、10〜2000リットルの培養物を含む、請求項26〜39のいずれか一項に記載の方法。
- 前記発酵槽は、100〜200Lの間のまたは約150Lの培養物を含む、請求項26〜40のいずれか一項に記載の方法。
- 溶存酸素レベルは、5%超である、請求項26〜41のいずれか一項に記載の方法。
- 前記溶存酸素は、15〜25%である、請求項42に記載の方法。
- シグナルペプチドは、前記細菌宿主細胞内の前記細菌毒素から除去されて、成熟した細菌毒素を得る、請求項14〜43のいずれか一項に記載の方法。
- 前記細胞ペーストを前記培養物から収穫するさらなる工程c)を含んでなる、請求項14〜44のいずれか一項に記載の方法。
- 前記細菌毒素を例えば、成熟した細菌毒素として精製するさらなる工程を含んでなる、請求項14〜45のいずれか一項に記載の方法。
- i)請求項14〜41のいずれか一項に記載の方法を用いて細菌毒素を作製する工程と、ii)工程a)の前記細菌毒素を抗原に抱合させる工程とを含んでなる、抱合体を作製するための方法。
- 前記抗原は、細菌糖質である、請求項47に記載の方法。
- 前記細菌糖質は、ストレプトコッカス・ニューモニアエ、インフルエンザ菌、ナイセリア・メニンジティディス、またはS.アウレウスに由来する莢膜糖質である、請求項48に記載の方法。
- A.請求項14〜49に記載の方法を用いて細菌毒素または抱合体を作製する工程と、 B.該細菌毒素またはその抱合体を医薬として許容し得る賦形剤と混合する工程と、
を含んでなる、ワクチンを製造するための方法。 - 前記細菌毒素は、工程b)においてさらなる抗原と混合される、請求項50に記載の方法。
- 前記細菌毒素は、工程b)においてアジュバントと混合される、請求項50または51に記載の方法。
