Abstract
Description
本発明は、シャペロンに融合する可溶性HTLV p24抗原、および診断適用における、たとえば単離した生体試料においてHTLV−IまたはHTLV−IIに対する抗体を検出するためのイムノアッセイにおける、それらの使用に関する。特に、本発明は、p24配列のN末端またはC末端のどちらかのドメインを含む可溶性HTLV−IまたはHTLV−II p24抗原フラグメントに関するものであり、その際、HTLV p24抗原フラグメントはシャペロンに融合していてもよい。さらに、本発明は、これらのHTLV−Iおよび−II融合抗原をコードする組換えDNA分子、ならびに発現ベクターを用いるそれらの組換え製造、ならびにそのような発現ベクターで形質転換した宿主細胞を含む。そのほか、本発明は、これらの数種類のHTLV p24抗原の組成物、および本発明の抗原を用いてHTLV抗体を検出するためのイムノアッセイ法に注目する。in vitro 診断アッセイにおけるHTLV p24抗原の使用、および抗HTLV抗体を検出するためのそれらのHTLV抗原を含む試薬キットも包含される。

本発明によるHTLV p24抗原は、組換えDNA技術により作製および産生できる。したがって本発明の他の観点は、前記にさらに定義したHTLV p24抗原およびそのバリアントをコードする組換えDNA分子である。
a)HTLV p24抗原をコードする遺伝子を含む前記の発現ベクターで形質転換した宿主細胞を培養する;
b)そのHTLV p24抗原をコードする遺伝子を発現させる;
c)そのHTLV p24抗原を精製する。
場合により、追加工程d)として、当技術分野で既知のリフォールディング法によりHTLV p24抗原を可溶性および免疫反応性のコンホメーションにするために、機能性可溶化を実施する必要がある。
a)体液試料を本発明によるHTLV p24抗原と混合することにより免疫反応混合物を調製する;
b)体液試料中に存在する、そのHTLV抗原またはHTLV抗原組成物に対する抗体が、そのHTLV抗原と免疫反応して免疫反応生成物を形成できるのに十分な期間、その免疫反応混合物を保持する;そして
c)そのいずれかの免疫反応生成物の存在および/または濃度を検出する。
抗体の検出のためのイムノアッセイは当技術分野で周知であり、したがってそのようなアッセイを実施するための方法ならびに実際の適用および操作も周知である。本発明によるHTLV p24抗原を用いて、使用する標識と無関係に、また検出様式(たとえば、放射性同位体アッセイ、エンザイムイムノアッセイ、電気化学発光アッセイなど)またはアッセイ原理(たとえば、試験片アッセイ、サンドイッチアッセイ、間接試験概念、または均一アッセイなど)と無関係に、抗HTLV抗体を検出するためのアッセイを改善することができる。当業者に知られているすべての生物学的液体を、抗HTLV抗体の検出のための単離した試料として使用できる。通常用いられる試料は、体液、たとえば全血、血清、血漿、尿または唾液である。
本発明のさらに他の対象は、イムノアッセイのための通常の検査添加剤に加えて、決定すべきHTLV抗体に特異的に結合するのに適した本発明によるHTLV p24抗原の少なくとも1つの抗原であっておそらく標識を保有するもの、および必要であれば他の通常の添加剤を含む、HTLVに対する抗体の検出のための試薬キットである。
さらに、前記に定めた試薬キットは、対照および標準溶液、ならびに平均的な当業者が用いる一般的な添加剤、緩衝剤、塩類、界面活性剤などを含む1以上の溶液中の試薬を、使用のための指示と一緒に含む。
実施例1
p24キャプシド融合ポリペプチドのクローニングおよび精製
発現カセットのクローニング
Novagen(米国ワイオミング州マディソン)のpET24a発現プラスミドをベースにして、HTLV−IおよびHTLV−IIに由来するp24融合タンパク質をコードする発現カセットを本質的に記載に従って得た(Scholz, C. et al., J. Mol. Biol. (2005) 345, 1229-1241)。HTLV−IおよびHTLV−IIに由来するp24抗原の配列をSwissProtデータベース(それぞれ、SwissProt ID P10274およびP03353)から検索した。N末端にグリシンリッチリンカー領域がインフレーム融合したHTLV−I(成熟キャプシドタンパク質のN末端プロリンリッチ15アミノ酸を欠如する)由来のp24キャプシド抗原アミノ酸146−344(ナンバリングはGag−Pro ポリタンパク質前駆体を参照する)をコードする合成遺伝子を、Medigenomix(ドイツ、マーチンスリード)から購入した。望ましくない副反応、たとえば酸化または分子間ジスルフィド架橋を阻止するために、p24の位置193、311および332のシステイン残基をアラニン残基に交換した。BamHIおよびXhoI制限部位が、p24−コーディング領域のそれぞれ5’および3’末端にあった。2つのEcSlyDユニット(SwissProt寄託no.P0A9K9の残基1−165)がグリシンリッチリンカー領域に連結したものをコードする、C末端にさらなるリンカー領域の部分を含むさらなる合成遺伝子を、同様にMedigenomixから購入した。NdeIおよびBamHI制限部位がこのカセットのそれぞれ5’および3’末端にあった。これらの遺伝子および制限部位は、シャペロン部分EcSlyD−EcSlyDとp24抗原部分の簡単なライゲーションによるインフレーム融合が可能になるように設計された。意図しない組換えプロセスを避けるために、かつ大腸菌宿主における発現カセットの遺伝子安定性を高めるために、EcSlyDユニットをコードするヌクレオチド配列を変性させ、エクステンションしたリンカー領域をコードするヌクレオチド配列を同様に変性させた;すなわち、同一アミノ酸配列をコードする異なるコドン組合わせを用いた。
すべてのp24融合タンパク質バリアントを実質的に同一のプロトコルを用いて精製した。特定のpET24a発現プラスミドを宿した大腸菌BL21(DE3)細胞を、37℃でLB培地+カナマイシン(30μg/ml)中において1.5のOD600になるまで増殖させ、1mMのイソプロピル−β−D−チオガラクトシドを添加することにより細胞質ゾル過剰発現を誘導した。誘導の3時間後、細胞を遠心分離(5000gで20分間)により収穫し、凍結し、−20℃で保存した。細胞溶解のために、冷却した50mMリン酸ナトリウム pH8.0、7.0M GdmCl、5mMイミダゾールに凍結ペレットを再懸濁し、懸濁液を氷上で2時間撹拌して細胞溶解を完了させた。遠心分離および濾過(0.45μm/0.2μm)の後、粗製の細胞溶解物を5.0mMのTCEPを含有する細胞溶解緩衝液でNi−NTAカラムに適用した。その後の洗浄工程は各ターゲットタンパク質に合わせて調整され、5mMから15mMに及ぶイミダゾール(50mMリン酸ナトリウム pH8.0、7.0M GdmCl、5.0mM TCEP中)であった。少なくとも10〜15体積の洗浄緩衝液を適用した。次いで、GdmCl溶液を50mMリン酸カリウム pH8.0、100mM KCl、10mMイミダゾール、5.0mM TCEPにより交換して、マトリックス結合したタンパク質のコンホメーショナルリフォールディングを誘導した。同時精製されるプロテアーゼの再活性化を避けるために、プロテアーゼインヒビターカクテル(Complete(登録商標) EDTAフリー,Roche)をリフォールディング緩衝液に含有させた。合計15〜20カラム体積のリフォールディング緩衝液を一夜の反応で適用した。次いで、TCEPとComplete(登録商標) EDTAフリーインヒビターカクテルの両方を、3〜5カラム体積の50mMリン酸カリウム pH8.0、100mM KCl、10mMイミダゾールで洗浄することにより除去した。その後、非特異的に結合したタンパク質混在物を除去するために、イミダゾール濃度 −まだ50mMリン酸カリウム pH8.0、100mM KCl中にある− を20〜80mM(各ターゲットタンパク質に応じて)に高めた。天然タンパク質を次いで同じ緩衝液中の500mMイミダゾールで溶離した。タンパク質含有画分を純度についてトリシン(Tricine)−SDS−PAGEにより査定し、プールした。最後に、タンパク質にサイズ排除クロマトグラフィー(Superdex HiLoad,Amersham Pharmacia)を施し、タンパク質含有画分をプールし、Amiconセル(YM10)で10〜20mg/mlに濃縮した。
分光測定
Uvikon XLダブルビーム分光光度計でタンパク質濃度測定を実施した。Pace (1995), Protein Sci. 4, 2411-2423が記載した方法を用いてモル吸光係数(ε280)を決定した。個別の融合ポリペプチドに用いたモル吸光係数(εM280)を表1に明示する。
実施例3
融合タンパク質へのビオチン部分およびルテニウム部分のカップリング
融合ポリペプチドのリジン ε−アミノ基を、タンパク質濃度10〜30mg/mlでN−ヒドロキシ−スクシンイミド活性化ビオチンおよびルテニウム標識分子によりそれぞれ修飾した。標識/タンパク質比をそれぞれの融合タンパク質に応じて2:1から5:1(mol:mol)まで変更した。反応緩衝液は150mMリン酸カリウム pH8.0、100mM KCl、0.5mM EDTAであった。反応を室温で15分間実施し、緩衝化L−リジンを10mMの最終濃度になるまで添加することにより停止した。標識の加水分解による不活性化を避けるために、それぞれの原液を乾燥DMSO(seccosolv品質,Merck,ドイツ)中に調製した。反応緩衝液中25%までのDMSO濃度が、試験したすべての融合タンパク質によって良好に耐容された。カップリング反応の後、粗製タンパク質コンジュゲートをゲル濾過カラム(Superdex 200 HiLoad)に通すことにより、未反応の遊離標識を除去した。
HTLVイムノアッセイにおける種々のp24キャプシド抗原バリアントの免疫反応性(すなわち、抗原性)
HTLV p24キャプシド抗原のポリペプチド融合バリアントの免疫反応性(抗原性)を、自動Elecsys(登録商標) 2010およびcobas e 411分析計(Roche Diagnostics GmbH)で査定した。Elecsys(登録商標)はRocheグループの登録商標である。測定は二重抗原サンドイッチフォーマットで実施された。
非対称二重抗原サンドイッチフォーマットにおけるオリゴマー型シャペロンキャリヤータンパク質の組合わせ
イムノアッセイを、本質的に実施例4に記載したように実施した。オリゴマー型シャペロン、たとえばFkpAおよびSkpは、それらの各クライアント抗原の機能的オリゴマー化を達成するための融合パートナーとして有利に使用できる。ターゲットタンパク質(すなわち、それらのクライアント抗原またはゲスト抗原)へのFkpAまたはSkpの融合により、イムノアッセイにおいてIgM分子を検出するのに適切な明確に規定されたオリゴマー型融合ポリペプチドを得ることができる。ここでは、本発明者らは、DAGS(二重抗原サンドイッチ)フォーマットに用いる場合、Skp−XおよびFkpA−X融合ポリペプチドの特定の好ましい組合わせがあるかどうかという疑問に対処した(注釈:“X”は一般にいずれかのターゲットタンパク質または抗原を表わす)。言い換えると、本発明者らはFkpA−X融合ポリペプチドを(信号発生)ルテニウム側よりもむしろ(捕捉)ビオチン側に用いるのが得策であるかどうかという疑問に対処した。逆に、本発明者らはSkp−X融合ポリペプチドを(捕捉)ビオチン側よりもむしろ(信号発生)ルテニウム側に用いるのが得策であるかどうかという疑問をもっていた。
CD検出したSkp−p24/CTD(267−350)およびFkpA−p24/CTD(267−350)の熱誘導アンフォールディング
近UV CDスペクトルをサーモスタット付きセルホルダーを備えたJasco−720分光偏光計で記録し、平均残基楕円率(mean residue ellipticity)に換算した。緩衝液は150mMリン酸カリウム pH8.0、100mM KCl、0.5mM EDTAであった。光路長は0.2cm、タンパク質濃度は218μΜ(Skp−p24モノマーについて)または147.5μΜ(FkpA−p24モノマーについて)であった。測定範囲は250〜330nmであり、バンド幅は1.0nmであり、0.5nmの解像度でスキャン速度は20nm/分であり、応答は1秒であった。信号−対−ノイズ比を改善するために、スペクトルを9回測定して平均した。
Claims (13)
- SEQ ID NO.2またはSEQ ID NO.6に特定されたHTLV p24のN末端ドメイン(NTD)を含み、SEQ ID NO.3およびSEQ ID NO.7に特定されたC末端ドメイン(CTD)を欠如する、可溶性HTLV p24抗原。
- SEQ ID NO.3またはSEQ ID NO.7に特定されたHTLV p24のC末端ドメインを含み、SEQ ID NO.2およびSEQ ID NO.6に特定されたN末端ドメインを欠如する、可溶性HTLV p24抗原。
- 可溶性HTLV p24抗原がシャペロンに融合する、請求項1または2に記載の可溶性HTLV p24抗原。
- シャペロンがオリゴマー型シャペロンである、請求項3に記載の可溶性HTLV p24抗原。
- オリゴマー型シャペロンがSkpおよびFkpAからなる群から選択されるシャペロンである、請求項4に記載の可溶性HTLV p24抗原。
- 可溶性かつ免疫反応性のHTLV p24抗原を製造する方法であって、以下の工程を含む前記方法:
−請求項1〜5のいずれか1項に記載のHTLV p24抗原をコードする組換えDNA分子が利用可能な状態で連結したものを含む発現ベクターで形質転換された宿主細胞を培養すること;
−前記HTLV p24抗原を発現させること;および、
−前記HTLV p24抗原を精製すること
。 - 請求項1〜5のいずれか1項に記載のHTLV p24抗原およびSEQ ID NO.25によるアミノ酸配列を含むHTLV gp21抗原を含むHTLV抗原組成物であって、p24およびgp21抗原が別個のポリペプチドとして発現された、前記組成物。
- 単離した試料においてHTLVに対して特異的な抗体を検出するための方法であって、請求項1〜5のいずれか1項に記載のHTLV p24抗原または請求項7に記載のHTLV抗原組成物をHTLV抗体の捕捉試薬および/または結合パートナーとして使用する、前記方法。
- 単離した試料においてHTLVに対して特異的な抗体を検出するための方法であって、以下を含む前記方法:
a)体液試料を請求項1〜5のいずれか1項に記載のHTLV p24抗原または請求項7に記載のHTLV抗原組成物と混合することにより免疫反応混合物を調製すること;
b)体液試料中に存在する、前記HTLV抗原またはHTLV抗原組成物に対する抗体が、前記HTLV抗原またはHTLV抗原組成物と免疫反応して免疫反応生成物を形成できるのに十分な期間、前記免疫反応混合物を保持すること;および、
c)前記いずれかの免疫反応生成物の存在および/または濃度を検出すること
。 - 単離した試料においてHTLVに対して特異的な抗体を検出するための請求項9に記載の方法であって、ここで前記免疫反応が下記を含む非対称二重抗原サンドイッチフォーマットで実施される、前記方法:
a)直接または間接的に固相に結合でき、生体親和性結合対の一部であるエフェクター基を保有する第1のHTLV p24抗原、および検出可能な標識を保有する第2のHTLV p24抗原をその試料に添加すること、ここで前記第1および第2のHTLV p24抗原は抗HTLV抗体に特異的に結合すること;
b)第1抗原、試料抗体および第2抗原を含む免疫反応混合物を調製すること、ここで免疫反応混合物を調製する前、途中または後に、生体親和性結合対の対応するエフェクター基を保有する固相を添加すること;
c)体液試料においてそれらのHTLV p24抗原に対する抗HTLV抗体がそれらのHTLV p24抗原と免疫反応して免疫反応生成物を形成できるのに十分な期間、その免疫反応混合物を保持すること;
d)液相を固相から分離すること;
e)固相もしくは液相または両方におけるそのいずれかの免疫反応生成物の存在を検出すること。 - HTLVに対して特異的な抗体を検出するための請求項10に記載の方法であって、第1抗原はFkpAに融合したHTLV p24抗原であってビオチン部分を保有し、かつ第2抗原はSkpに融合したHTLV p24抗原であって電気化学発光性ルテニウム錯体で標識されているか、または、
第1抗原はSkpに融合したHTLV p24抗原であってビオチン部分を保有し、かつ第2抗原はFkpAに融合したHTLV p24抗原であって電気化学発光性ルテニウム錯体で標識されている、前記方法。
- 抗HTLV抗体の検出のための in vitro 診断検査における、請求項1〜5のいずれか1項に記載のHTLV p24抗原または請求項7に記載のHTLV抗原組成物の使用。
- 少なくとも、請求項1〜5のいずれか1項に記載のHTLV p24抗原または請求項7に記載のHTLV抗原組成物を含む、抗HTLV抗体の検出のための試薬キット。
