別個の軽鎖及び重鎖の哺乳動物発現プラスミドを、等モル濃度比で合わせ、HEK−293又はCHO−S細胞をトランスフェクトするために使用した。小規模発現研究のために、リポフェクタミン及びHEK−293細胞を使用し、一方で、IgGのより大きなバッチの生成のためには、CHO−Sへのエレクトロポレーションが好ましかった。
精製MabによるTcdA活性のインビトロ中和
全ての中和スクリーニングアッセイを、96ウェルポリスチレンプレート内で実施した。アッセイは、成長したCACO−2細胞を使用し、MEM+20% FCS、2mM Q及びNEAAにおいてスクリーニングした。任意の抗体組み合わせは、そうでないと言及されない限り、等モル濃度比である。1日目:細胞を、1ウェルあたり50μl培地中3000個でプレート培養し、そして24時間にわたってインキュベートした;2日目:ヒト化Mabの精製サンプルを、96ウェル丸底ポリプロピレン滅菌プレートに添加した;PPプレートに、毒素Aを適切な致死用量、すなわちLD80以上を生じるために充分な濃度をスパイクし、そして1時間にわたって37℃でインキュベートした;50μlのこの混合物を、細胞プレートに添加し、そして96時間にわたってインキュベートした;5日目:メチレンブルー(0.5%メチレンブルー、50%エタノール)を添加した;室温で1時間インキュベートした;1% N−ラウリルサルコシンで細胞を溶解し、BIOTEK Synergy2プレートリーダーにおいて405nmにて読み取った。この結果を、図12〜24に示す。示したTcdA活性のEC50及び%最大中和は、選択された抗体は、単体の薬剤として非常に高い力価を有することを確認する。これらの2〜5の組み合わせは、最も良いEC50又は%最大中和において改善しなかった。CA922、923、995、997及び1000のMabを合わせた場合のいかなる相乗作用もの欠如は、重要な観察であり、各抗体単独が、特に高いレベルの親和性及び力価を有するという事実に起因し得る。例5における支持データは、Mabのいくつか(例えば、CA997)は、TcdAサブドメインに多数回結合可能であることを示す。従って、恐らく、これらの5つのMabは、TcdAのC末端細胞結合ドメインを標的した場合に達成可能である最大の親和性、力価及び結合価を示す。この抗体はまた、LD80からLD95を超えるまでの範囲に及ぶ非常に高い毒素濃度(LDmax)を中和する場合に有効であるが、EC50におけるいくつかの中程度の増大(すなわち、力価の下降)を、非常に高レベルの[TcdA]によって観察した。他のデータは、高いTcdA濃度を試験した場合、力価の実質的な低下を示した(20)ので、これらのデータはまた、非常に驚くべきものである。

これらのデータは、一定範囲の毒素濃度において、TcdB中和のためのMabの最も良い対は、CA1125及びCA1151であったことを示す。さらに、1125+1151組み合わせは、1125+1153と対照的に、相対的モル濃度比における変化によって大きく影響を受けなかった。

MabのTcdBサブドメインに対する結合価
抗C.ディフィシレTcdB抗体のTcdB1234に対する結合事象のモル数を、Biacore 3000(GE Healthcare)において、表面プラスモン共鳴(SPR)によって決定した。ストレプトアビジンを、CM5センサーチップ(GE Healthcare)上に約4000RUのレベルまでアミンカップリングを介して固定化し、ビオチン化TcdB1234を500〜600RUで結合させた。同じ抗TcdB抗体混合物(各抗体の最終濃度は500nMであった)の2回の20μl注入を、この表面にわたって10μl/分で注入し、飽和結合応答を記録した。表面を、HClを用いて毎周期後に再生した。全てのデータを、ストレプトアビジンのみの参照フローセルに対する応答を用いてバックグラウンド結合について修正した。
CA927は、TcdB1234に1回しか結合しないMabの代表と考えられるので、全ての応答は、CA927平均応答(表10の最終欄)の倍数に関して表現されている。
全ての抗体組み合わせは、付加物結合応答を有し、このことは、TcdB1234上には、これらの組み合わせによって結合される2以上の非競合的部位が存在することを示す。
MabのTcdAサブドメインに対する結合価
抗C.ディフィシレTcdA抗体のTcdA123及びA456に対する結合事象のモル数を、Biacore 3000(GE Healthcare)において、表面プラスモン共鳴(SPR)によって決定した。ストレプトアビジンを、CM5センサーチップ(GE Healthcare)上に約4000RUのレベルまでアミンカップリングを介して固定化し、ビオチン化TcdA123を1つのフローセルに結合させ、そしてTcdA456を異なるフローセルに対し約500RUの応答で結合させた。同じ抗TcdA抗体1μMの2回の30μl注入を、両フローセルにわたって10μl/分で注入し、飽和結合応答を記録した。表面を、HClを用いて毎周期後に再生した。全てのデータを、ストレプトアビジンのみの参照フローセルに対する応答を用いてバックグラウンド結合について修正した。
抗体CA997及びCA1000は、TcdA123に、6つのCA997対1つのCA1000の比で結合し、一方で、これらは、TcdA456に、3つのCA997対1つのCA1000の比で結合する(表2)。
CA997全体は、A123に6回以上、そしてA456に6回以上結合し、一方で、CA1000は、A123に少なくとも1回、そしてA456に2回結合する。
TcdAによって起こったTEERの喪失のMab中和
C.ディフィシレ単層完全性アッセイを、Becton−Dickinson(BD)Caco−2 BioCoat HTSプレートシステムを用いて実施する。
1日目− Caco−2細胞を、500μlの基礎播種培地(BDによって供給される)中で、プレート挿入物の1ウェルあたり2×105/mlにて播種した。35mlの基礎播種培地を、フィーダートレイに添加した。細胞を、24時間にわたって37℃でインキュベートした。2日目−基礎播種培地を挿入物及びフィーダートレイから除去し、そしてEntero−STIM分化培地(BDによって供給される)と置き換えた。挿入物のウェルあたり500μl添加し、フィーダートレイに35ml添加した。細胞を、さらに72時間、37℃でインキュベートした。5日目−抗体を、ポリプロピレンプレート内及び毒素Aのアッセイウェルにおいて使用される濃度に対し2×濃度で調製した。毒素Aを、抗体に125ng/mlの濃度で加え、プレートを、1時間にわたって37℃でインキュベートした。1mlのCaco−2増殖培地(MEM+20% FCS、2mM Q、NEAA)を、標準24ウェルTCプレートの各ウェルに加えた。BioCoat挿入プレートを、24ウェルTCプレートに移した。Entero−STIM培地を、挿入物から除去し、400μlの毒素:Ab混合物と置き換えた。
抗C.ディフィシレ毒素抗体の、TcdA及びTcdBのサブドメイン:TcdA123、TcdA456及びTcdB1234に対する親和性。
抗C.ディフィシレTcdA及びTcdB抗体の相互作用についての動態定数を、CM5センサーチップを用いるBIAcore 3000において行われる表面プラスモン共鳴によって決定した。全ての実験を、25℃で実施した。Affinipure F(ab’)2断片ヤギ抗ヒトIgG,Fc断片特異的(Jackson ImmunoResearch)を、CM5センサーチップ(GE)上に、アミンカップリング化学を介して固定化し、≒7000応答単位(RU)の補足レベルとした。HBS−EP緩衝液(10mM HEPES pH7.4、0.15M NaCl、3mM EDTA、0.005%界面活性剤 P20、Biacore AB)を、10μL/分の流速でランニングバッファーとして使用した。各抗体の1μg/ml以下での10μL注入を、固定化した抗ヒトIgG、Fcを補足するために使用した。TcdA123、TcdA456又はTcdB1234を、12.5nMから二倍希釈にて30μL/分の流速にて補足した精製抗体にわたって滴定した。培養上清中に存在する抗体について、1濃度の12.5nMのTcdA123又はTcdA456及び50nMのTcdB1234を、30μl/分にて抗体の上を通過させた。動態を、n=2にて計算した。表面を、40mM HClの2回の10μL注入、及び5mM NaOH5μL注入により、10μL/分の流速にて再生した。
二重参照バックグラウンドを減算した結合曲線を、標準的手順に従い、BIAevaluationソフトウェア(バージョン3.2)を用いて分析した。動態パラメータを、適合アルゴリズムから決定した。

サンプルを、以下を混合することによって調製した:2mg/mlの30μlのタンパク質サンプル、0.35% メチルセルロース、4% pH3〜10両性電解質(Pharmalyte)、合成pIマーカー(4.65及び9.77)、1μlの各ストック溶液及び最終容量を200μlにするためのHPLC等級の水。次いで、この混合物を、iCE280 IEFアナライザー(1500Vにて1分間事前焦点合わせをし、その後、3000Vで6分間焦点合わせをする)を用いて分析した。次いで、キャリブレーションした電気泳動図を、Empowerソフトウェア(Waters製)を用いて組み込んだ。
この方法は、Sypro orange蛍光色素を用い、タンパク質ドメインの変性プロセスをモニタリングする。この色素は、変性の結果として露出された露出疎水性領域に結合し、その結果、放出スペクトルの変化をもたらす。
サンプル(1mg/mlで5μl)を、Sypro orangeの5μlのストック溶液(30倍)と混合し、容量を、PBS、pH7.40で50μlにする。
このプレートを、正確な温度制御のための加熱デバイスを含む7900HT fast実時間PCRシステム内においた。温度は、20℃から99℃にまで上がった(1.1℃/分の傾斜率)。CCDデバイスは、ウェル内で変化する蛍光を同時にモニタリングする。アルゴリズムは、強度データを処理して、複数の変遷を考慮に入れるために使用される。
抗体を製造する間、サンプルを、ポンピング及び濾過などのプロセスによって生じた機械的ストレスに供する。これは、タンパク質の気体−液体境界の曝露及び生物活性の最終的喪失をもたらす剪断力に起因して、変性及び結果としての凝集を引き起こし得る。ボルテックスによるストレスは、凝集安定性の予測についての抗体サンプルの頑強さをスクリーニングする方法である。
表14は、抗TcdA及び抗TcdB IgG1分子の両方について、測定したpI及びTmデータのまとめを提供する。
分子の測定したpIは、高く(CA164_01000.g1_P3を除く)、PBS、pH7.4及び50m酢酸ナトリウム/125mM塩化ナトリウム、pH5などの製剤化緩衝液のpHから離れていた。これは、2以上のMabの共製剤化のために好適なpHを有する緩衝液が、選択され得ることを意味し得る。
全ての分子がIgG1であるので、FcドメインのTm(Tm(CH2))は、同じである。分子間の熱安定性の相違は、Fab’ドメインのTm(Tm(Fab))によって決定され得る。
抗TcdA分子について、ランクの順番(最も安定なものが1番目)は、CA922は997以上であり、997>923>995>1000である。抗TcdB分子について(最も安定なものが1番目)は、CA1151.g4>1125.g1、g4>1134.g4>1134.g5、1134.g5は1134.g6以上であり、1134.g6>1102.g1=1102.g4である。
異なる抗体間の異なる凝集安定性を決定することができ、図67は、PBS、pH7.4中の種々の抗TcdA IgG1分子に対するボルテックスを介した撹拌の効果を示す。
ランクの順番を決定することが可能であった(最も凝集安定性であるものが1番目):CA922は997以上であり、997>923であり、923は995以上であり、995>1000である。
図68は、種々の抗TcdB分子に対するボルテックスを介した撹拌の効果を示す。
凝集安定性の順番をランク付けすることが可能であったが、CA1125グラフトは、CA1134分子よりも安定であると考えられ、このCA1134分子は、CA1102分子よりも安定であった。
さらなる研究を実施し、抗TcdB分子(CA1151.g4)の凝集安定性を、より安定な分子CA1125.g2(図2を参照されたい)及びより凝集安定性である抗TcdA分子(CA922.g1及びCA997.g1)と直接的に比較した。結果は、図69において示され得る。
これらの4つのMabについてのさらなる結果を、図67及び68に示す。
抗TcdA分子について、CA922.g1及びCA977.g1、CA922が、上の分析に基づいて好ましく、CA1000は除外して、全ての分子が、治療的IgG1としての使用のための好適な候補と考えられ得る。
抗TcdB分子について、生物物理学的特徴が、凝集安定性及びTmに基づくグラフトのファミリー内にグループ化され得る、CA1125グラフとは、潜在的により安定であると証明される。CA1102グラフトは、最も貧しいTmデータを示し、また、撹拌によるストレスを介して、最も大きな凝集の傾向を示した。
CA1151.g4を用いる研究は、この分子が、CA11125.g2と比較して僅かに増加した凝集安定性を示し、TcdA分子と同等であるとみられることを示した(CA922.g1及びCA997.g1)。全ての4つの分子は、同等のTm値を示した。CA997、CA1125及びCA1151は、非常に高いレベルの熱安定性及び撹拌後の非常に低いレベルの凝集形成を示す。
抗C.ディフィシレ毒素Mabハムスター感染研究。
ハムスター感染研究が、Ricerca Biosciences LLC、Cleveland、Ohio、USAによって実施された。研究プロトコールは、Ricerca IACUC委員会によって承認された。活性構成要素及び対照構成要素(組成物及び用量)を、計画された28日間の研究期間の完了後まで、Ricercaスタッフに対して盲検とした。
ゴールデンシリアン雄ハムスター(82〜103g、54日齢)を、HEPAフィルターをした使い捨てケージ内に個別に入れ、Teklad Global Diet 2016を食餌し、水を適宜与えた。順化後、ハムスターを、Mab混合物又はPBS(ビヒクル対照)で4日間(−3日目、−2日目、−1日目及び0日目)のそれぞれ1日1回事前投薬した(i.p.)。2用量のMabを、調査した:高用量=抗TcdA及び抗TcdB構成要素それぞれ50mg/kg及び低用量は、抗TcdA及び抗TcdB構成要素それぞれ5mg/kg。
試験した薬物組み合わせは、注射されたタンパク質の50%を構成する1つの抗TcdA抗体(CA997.g1)及び共に注射されたタンパク質の50%を構成するが単独では注射されたタンパク質の25%を構成する2つの抗TcdB抗体(CA1125.g2及びCA1151.g4)から構成されていた。ハムスターを、(−1日目に)50mg/kgのPBS中リン酸クリンダマイシン(s.c.)で感作し、その1日後(0日目)に、3.4×106c.f.u.の株ATCC43596由来の栄養細胞でチャレンジした。バンコマイシンを、1日目、2日目、3日目、4日目、5日目に、5日間5mg/kg日に2回投薬した(p.o.)。
生存能力チェックを、動物に対し1日に2回行い、死にかけていると見出された動物を安楽死させ、死として数えた。体重を、各投薬日、次いで、2週に1度及び生存個体の安楽死の前に決定した。全体的剖検を、全ての動物に対して行った。生存曲線を、カプラン及びメイヤーの方法によって作成した。生存曲線を、対数順位検定からのP値を、ボンフェローニ補正した閾値のP=0.005と比較して用いて分析した。バンコマイシン処置群を、分析に含めなかった。全ての統計学的試験を、Prism v5.04によって行った。全ての群は、5匹の動物を含むバンコマイシン処置群を除き、11匹の動物を含んだ。
生存曲線は、図63において見ることができる。PBS(対照)を受けたハムスターは全て、+2日目及び+3日目に死亡したが、一方,バンコマイシン処置を5日間受けた個体は、+10日目及び+11日目に全て死んだ。高用量のUCB Mab混合物を受けたハムスターは、+11日目まで生存したが、その後、28日間の研究の最終日までに、2匹のみが死んだ。低用量のUCB Mab混合物を受けたハムスターは、全て、+3日目までは生存し、その後、動物は、全てが死んだ+16日目までに、公平に安定的に失われた。このデータは、ハムスターにおける抗毒素Mabの使用についての刊行されたデータ(18)と比較した場合の、防御の例外的なレベル及び持続期間を示す。これらのインビボデータは、中和及び安定性についての非常に高レベルの性能のインビトロ観察を支持する。
感染の急性期(1〜5日目)の間、死と体重との間は明らかにリンクしていなかった。図64〜65。従って、ハムスターは、圧倒的なTcdA及びTcdBの直接的及び間接的な影響により死んだと考えることができる。中和Mabの部分的防御(UCB低用量)に起因して急性期を生き延びたハムスターは、恐らく、胃の損傷及び栄養状態の変化に起因して、体重を落した。研究の28日間を生き残ったハムスターの多くは、UCB高用量Mabの防御効果に起因し、体重喪失から回復し、実際には体重を増やしたことは、注目すべき点である。このことは、UCB Mabの優れた防御効果が、胃を正常に機能させることが可能であるという証拠として採用され得る。
結果を、図63〜66に示す。
図66中の写真は、TcdA及びTcdBによって起こる盲嚢の膨潤及び血液滲出についての代表的な全体的病理学(左画像、PBS対照、2日目の動物死)及びUCB高用量Mabによる防御後の、正常な大便に満たされた盲嚢(右画像、UCB高用量、28日目まで生存した動物)を示す。これらのデータは、高用量のUCB Mabによる防御後、大腸は、正常な形態及び機能に戻り得ることを示す。
精製Mabによる、種々のリボタイプの株由来のTcdAの中和。
臨床的感染を、種々の異なる株によって引き起こす。株相違を、リボタイプが鍵となる多くの種々の方法を用いて性質決定する。異なるリボタイプ株を種々の病原性、感染及び胞子形成特性を有することを観察する。上で示されるTcdA中和の全ては、VPI10463として公知の株から精製されたTcdAを用いた。しかし、発生に関連する優勢且つ積極的に病原性の株は、リボタイプ027と呼ばれる。他の鍵となるリボタイプとしては、078、001、106が挙げられる。アミノ酸配列相違は、異なるリボタイプによって生成された毒素間で観察されていて、従って、Mabが広範なセットの臨床的単離物からの毒素を中和可能であることが重要である。CA922、997及び1000を、株027及び078由来のTcdAを中和する能力について試験し、その能力を、VPI10463由来のTcdAに対して比較した。Mabを、4の[TcdA]において試験し、有意差なしでLD80、LD90及びLD95において、全ての毒素の中和が可能であることを見出した。

健康なハムスターにおけるヒトIgG1(20mg/kg)のPK研究。ハムスターPKは、マウス又はラットにおけるMabと類似の半減期を有することが見出された(t1/2 6〜8日間)。i.p.及びs.c.投薬は、本質的に同じであった。薬物動態学及びhIgG1 Mabの胃への分布を、「正常」(非感染)ゴールデンシリアンハムスターにおいて研究した。精製Mabを、CARE Research LLC,Fort Collins,Colorado、USAにより雄のハムスター(120〜135g)に投与し、サンプルを、UCB Pharma.によりアッセイした。この研究は、CARE IACUC委員会によって承認された。8匹の動物に20mg/kgのIgG1の単回用量を与え、4匹に、i.p.で投薬し、4匹にs.c.で投薬した。血液を、投薬の1、3、8、24、48、72、103及び168時間後に収集し、血清を、−80℃での保存の前に分離した。また、血液を、アッセイ対照を提供するために、2匹の未処理ハムスターからも採取した。安楽死後、2cmの長さの結腸を、各ハムスター由来の盲嚢接合部以降から切り出した。結腸切片を、洗浄緩衝液(50%(v/v)Sigmaプロテアーゼインヒビターカクテル(P2714)含有50%(v/v)PBS)で洗い流した後、切開し、下にある筋肉からの粘膜の分離及び除去を行った。粘膜サンプルを、0.5mlの洗浄緩衝液中に入れ、視覚的に均一になるまでホモジナイズし、輸送の直前までウェットアイス上で4℃にて保存した。抗ヒトIgG1 ELISA Nunc maxisorp 96ウェルプレートを、Goat F(ab’)2抗ヒトIgG−Fcγ断片(Jackson 109−006−098)を含む0.1M NaHCO3 pH8.3中で一晩コートし、プレートを、PBS−Tween(PBS/0.1%(v/v)Tween 20)で洗浄し、次いで、1.0%(w/v)BSA及びPBS中0.1%(v/v)Tweenでブロックした。血清サンプルを、サンプルコンジュゲート緩衝液(1%(w/v)BSA、PBS中0.2% Tween)中に希釈し、洗浄後、サンプルコンジュゲート緩衝液及び2.5M H2SO4停止溶液を含むTMB中のヤギ抗ヒトκ−HRP(Cambridge Bioscience 2060−05)によって明らかにした。
168時間の時点で採取した血液から収集した血清サンプル及び結腸サンプルを、この後、除去した。

hIgG1は、胃の「掻き取り」において見出され得る、すなわち、hIgG1は、健康な胃の血管構造に入り、「予防的投薬」において防御的であり得ることもまた、示される。この効果は、ヒトは、同族のhFcRnを有するので、ヒトにおいてなおより深遠である。
C.ディフィシレ感染を有するハムスターにおける血清レベル
この研究は、ゴールデンシリアンハムスターにおけるi.p.投与(種々の用量は、以下に詳説する)後に、CA725.0、CA726.0、CA997.g1、CA1125.g2及びCA01151.g4の血清濃度を決定することであった。
ヒト化Mabを、トリプシン消化後の液体クロマトグラフィータンデム質量(LC−MS/MS)分析を用いて定量した。定量化を、スパイクしたウマミオグロビンを内部標準として使用し、公知の濃度でブランクのマトリクスにスパイクした信頼のおける標準的材料に対する比較によって達成した。
調査した全てのヒト化Mabに対して共通する独特の(「プロテオティピックな」)ペプチドを選択し(DTLMISR、CH2領域ペプチド)、両サンプル及びキャリブレーションサンプルを、概説するようにトリプシン消化した。5μl血清サンプルのトリプシン消化を、変性/アセトニトリルによる還元/Tris(2−カルボキシエチル)ホスフィン及びヨードアセトアミドによるカルバミド−メチル化(Sigma−Aldrich、Poole、UK)の後に、配列決定グレード修飾トリプシン(Promega、Southampton、UK)を用いて一晩実施した。
LC−MS/MSシステムは、CTC HTS−x Autosampler(CTC Analytics、Zwingen、Switzerland)、Agilent 1290 LCシステム(Agilent Technologies、Stockport、UK)及びSciex 5500 QTrap MSシステム(AB Sciex、Warrington、UK)からなり、エレクトロスプレーモードで操作されるTurbo Vイオン供給源を備える。分析物を、2〜95%(v/v)水/アセトニトリル(0.1%ギ酸)の勾配を用いたOnyx Monolithic C18カラム(100×4.6mm、Phenomenex, Macclesfield, UK)を用いて分離し、1.5mL/分で6分間にわたって送った。注入容量は、10μLであった:溶出物の全ては、質量分析供給源に導入した。質量分析の供給源温度を、600℃に維持し、他の供給源パラメータ(例えば、衝突エネルギー、デクラスタリング電位、カーテンガス圧など)を最適化し、目的のペプチドについて最大感度を達成した。目的のプロテオティピックペプチド各々の選択的遷移を、モニタリングした。
独特の(「プロテオティピック」)ペプチドを、目的の分析物の全てについて選択した;サンプルを、トリプシン消化後に分析した。
モニタリングしたペプチドに基づいて計算した血漿濃度を、以下で概説する。
CA164_00997及びCA164_01151について、干渉ピークを、MRMトレースにおいて観察した。この理由のため、これら2つの分析物は、サンプル中で定量できなかった。
総h−IgGを、目的の全ての分析物に共通するペプチドを用い、全てのサンプルにおいて定量化した。これは、5つ全ての分析物を合わせた標準曲線を用いて行われた。このアプローチの妥当性は、CA164_00725及びCA164_00726について観察された濃度の合計が、総h−IgGについて観察された濃度と、(実験誤差範囲内で)一致しているという事実によって実証される。
このアプローチを用い、CA164_00997、CA164_01125及びCA164_01151を投薬された動物のサンプル中のh−IgGの総濃度を決定した。
得られたデータ全体は、目的の全ての5つの分析物の曝露が、所定の用量と類似したことを示す。

Claims (41)
- 抗原TcdA又はTcdBに対して特異的なモノクローナル抗体であって、前記抗体が、標的抗原に対して高い親和性を有し、クロストリジウム・ディフィシレ(clostridium difficile)感染又は前記感染の危険にある患者において下痢の持続期間及び/又は重症度、罹患率及び/又は死亡率を低下するために好適である、上記抗体。
- 前記抗体が、高い力価、例えば、毒素がLD80以上で使用された場合に、200ng/ml以下、例えば、150ng/ml以下、詳細には100ng/ml以下のEC50を有する、請求項1に記載のモノクローナル抗体。
- 毒素がLD80以上で使用された場合に、前記抗体のEC50が0.1〜10ng/mlの間である、請求項2に記載のモノクローナル抗体。
- 毒素がLD80以上で使用された場合に、毒素の最大阻害が50〜100%の間である、請求項2又は請求項3に記載のモノクローナル抗体。
- 前記抗体が、標的抗原に複数回結合する、請求項1から4までのいずれか一項に記載のモノクローナル抗体。
- 前記抗体が、標的抗原に2、3、4、5、6、7、8、9、10、11、12、13、14又は15回以上結合する、請求項5に記載のモノクローナル抗体。
- 前記抗体が、TcdAに特異的である、請求項1から6までのいずれか一項に記載のモノクローナル抗体。
- 前記抗体が、TcdBに特異的である、請求項1から6までのいずれか一項に記載のモノクローナル抗体。
- 前記抗体が、1nM以下、例えば600pM、例えば50〜600pMの親和性を有する、請求項1から8までのいずれか一項に記載のモノクローナル抗体。
- 前記抗体が、中和抗体であり、高濃度の毒素、特に、リボタイプ003、012、027及び078に対して有効なものを含む、請求項1から9までのいずれか一項に記載のモノクローナル抗体。
- 前記抗体が、TEERアッセイにおいて、前記アッセイの開始の4時間後に測定した場合、60〜80ng/mlの範囲内のEC50を有する、請求項1から10までのいずれか一項に記載のモノクローナル抗体。
- TcdAに特異的に結合し、重鎖を含み、重鎖の可変ドメインは、CDR-H1用の配列番号44に示される配列を有するCDR、CDR-H2用の配列番号45に示される配列を有するCDR、及びCDR-H3用の配列番号46に示される配列を有するCDRのうち少なくとも1つを含む、請求項1に記載のモノクローナル抗体。
- さらに軽鎖を含み、軽鎖の可変ドメインは、CDR-L1用の配列番号41に示される配列を有するCDR、CDR-L2用の配列番号42に示される配列を有するCDR、及びCDR-L3用の配列番号43に示される配列を有するCDRのうち少なくとも1つを含む、請求項12に記載のモノクローナル抗体。
- 配列番号49に示される配列を含む重鎖及び配列番号47に示される配列を含む軽鎖を有する、請求項13に記載のモノクローナル抗体。
- TcdAに特異的に結合し、重鎖を含み、重鎖の可変ドメインは、CDR-H1用の配列番号54に示される配列を有するCDR、CDR-H2用の配列番号55に示される配列を有するCDR、及びCDR-H3用の配列番号56に示される配列を有するCDRのうち少なくとも1つを含む、請求項1に記載のモノクローナル抗体。
- さらに軽鎖を含み、軽鎖の可変ドメインは、CDR-L1用の配列番号51に示される配列を有するCDR、CDR-L2用の配列番号52に示される配列を有するCDR、及びCDR-L3用の配列番号53に示される配列を有するCDRのうち少なくとも1つを含む、請求項15に記載のモノクローナル抗体。
- 配列番号59に示される配列を含む重鎖及び配列番号57に示される配列を含む軽鎖を有する、請求項16に記載のモノクローナル抗体。
- TcdBに特異的に結合し、重鎖を含み、重鎖の可変ドメインは、CDR-H1用の配列番号124に示される配列を有するCDR、CDR-H2用の配列番号125に示される配列を有するCDR、及びCDR-H3用の配列番号126に示される配列を有するCDRのうち少なくとも1つを含む、請求項1に記載のモノクローナル抗体。
- さらに軽鎖を含み、軽鎖の可変ドメインは、CDR-L1用の配列番号121に示される配列を有するCDR、CDR-L2用の配列番号122に示される配列を有するCDR、及びCDR-L3用の配列番号123に示される配列を有するCDRのうち少なくとも1つを含む、請求項18に記載のモノクローナル抗体。
- 配列番号129に示される配列を含む重鎖及び配列番号127に示される配列を含む軽鎖を有する、請求項19に記載のモノクローナル抗体。
- TcdBに特異的に結合し、重鎖を含み、重鎖の可変ドメインは、CDR-H1用の配列番号154に示される配列を有するCDR、CDR-H2用の配列番号155に示される配列を有するCDR、及びCDR-H3用の配列番号156に示される配列を有するCDRのうち少なくとも1つを含む、請求項1に記載のモノクローナル抗体。
- さらに軽鎖を含み、軽鎖の可変ドメインは、CDR-L1用の配列番号151に示される配列を有するCDR、CDR-L2用の配列番号152に示される配列を有するCDR、及びCDR-L3用の配列番号153に示される配列を有するCDRのうち少なくとも1つを含む、請求項21に記載のモノクローナル抗体。
- 配列番号159に示される配列を含む重鎖及び配列番号157に示される配列を含む軽鎖を有する、請求項22に記載のモノクローナル抗体。
- TcdAに特異的に結合し、重鎖及び軽鎖を有し、重鎖可変領域は、配列番号9、配列番号19、配列番号29及び配列番号39からなる群より選択される配列を含み、軽鎖可変領域は、配列番号7、配列番号17、配列番号27及び配列番号37からなる群より選択される配列を含む、請求項1に記載のモノクローナル抗体。
- TcdBに特異的に結合し、重鎖及び軽鎖を有し、重鎖可変領域は、配列番号69、配列番号79、配列番号89、配列番号99、配列番号109、配列番号119、配列番号139、配列番号149及び配列番号159からなる群より選択される配列を含み、軽鎖可変領域は、配列番号67、配列番号77、配列番号87、配列番号97、配列番号107、配列番号117、配列番号137、配列番号147及び配列番号157からなる群より選択される配列を含む、請求項1に記載のモノクローナル抗体。
- 1つ又は複数の請求項1から25までのいずれか一項に記載の抗体を含む、医薬組成物。
- TcdBに特異的な2つ以上の抗体を含む、請求項26に記載の医薬組成物。
- TcdAに特異的な2つ以上の抗体を含む、請求項26に記載の医薬組成物。
- 前記組成物中の少なくとも1つの抗体が、TcdAに対し特異的であり、前記組成物中の少なくとも1つの抗体が、TcdBに対し特異的である、請求項26に記載の医薬組成物。
- 前記組成物が、TcdBに特異的な少なくとも第2の抗体をさらに含む、請求項29に記載の医薬組成物。
- 前記組成物が、標的抗原(単数又は複数)に対する2、3、4、5、6、7、8、9、10、11、12、13、14、15種の異なる抗体、例えば2、3、4、又は5種の抗体を含む、請求項26から30までに記載の医薬組成物。

- クロストリジウム・ディフィシレ(clostridium difficile)感染を有するか又はその危険にある患者を処置する方法であって、治療有効量の請求項1から25までのいずれか一項に記載のモノクローナル抗体又は請求項26から33までのいずれか一項に記載の医薬組成物を投与することを含む、上記方法。
- 前記処置が、クロストリジウム・ディフィシレ(Cclostridium difficile)処置のためのさらなる処置、例えば、メトロニダゾール、バンコマイシン、クリンダマイシン、フィダキソマイシン及びこれらの組み合わせからなる群より選択される処置と組み合わせて使用される、請求項35に記載の処置の方法。

- 毒素中和の測定、TEER測定、熱安定性測定(Tm)、振盪凝集測定及び共製剤化のために好適な等電点(pI)を組み合わせることによる、クロストリジウム・ディフィシレ(clostridium difficile)感染の処置のための抗体混合物を選択する方法。
- 例えば、クロストリジウム・ディフィシレ(clostridium difficile)感染又はその合併症の処置又は予防などのワクチン接種における使用のための、トキソイド又はこれを含む医薬組成物と組み合わせた、請求項1から26までのいずれか一項に記載のモノクローナル抗体。
