Description
実施例1:コンパウンドK成分の強化された紅参濃縮液の製造
本研究に用いられた薬用人参は、干し紅参(4年根)であり、原料の10倍(w/v)の水または酒精を入れ、70〜80℃で200rpmにて15時間ずつ3回還流抽出して紅参エキスを製造した後、これを常温または低温で湯煎・冷却してろ過した後、減圧濃縮して70brix以上の濃縮物を製造した。
コンパウンドK成分の強化された紅参濃縮液の非アルコール性脂肪肝の改善に対する機能を研究するために、雄性5週齢のC57BL/6マウスを用いて高脂肪食餌により誘起された動物モデルを構築して生体内の実験を行った。
11週間の飼育期間中に動物の体重を毎週1回の周期で測定し、統計学的な分析のためにスチューデントのT検定を行った。高脂肪食餌対照群(HFD)は正常食餌対照群(ND)と比較したのに対し、高脂肪食餌+シリマリン処理群(HFD+Sily)または高脂肪食餌+コンパウンドK成分の強化された紅参濃縮液処理群(HFD+Gx)は高脂肪食餌対照群(HFD)と比較した。実験群別の平均体重の変化の推移を下記表2に示す。
高脂肪食餌(HFD)の11週間の処理による脂肪肝を誘起した。目視したところ、正常食餌(ND)対照群に比べて高脂肪食餌(HFD)対照群の肝の色は相対的に黄色を帯びていた。これは、高脂肪食餌により肝組織にかなりの脂肪の蓄積が生じたものと認められる。
脂肪肝により肝機能の一部が損傷されれば、血液内の関連のある生化学的指標に変化が生じる。この実験では、脂肪肝の診断と関連して使用可能な主な肝機能指標を選定して群別にその違いを分析した。
肝組織内の中性脂質(Triglycerides、TG)の含量を分析し、その結果を図4に示す。
コンパウンドK成分の強化された紅参濃縮液が肝組織において脂質代謝を調節する機序を研究するために、まず、肝組織において脂質の生合成(Lipogenesis)および分解(Lipolysis)に与る主なタンパク質の遺伝子発現をリアルタイム逆転写酵素−ポリメラーゼ連鎖反応(RT−PCR)法を用いて分析し、その結果を図5に示す。

- コンパウンドK成分の強化された紅参濃縮液を有効成分として含有する非アルコール性脂肪肝症の予防および治療用薬剤学的組成物。
- 前記コンパウンドK成分の強化された紅参濃縮液は、薬剤学的組成物総重量に対して0.1〜99重量%で含有されることを特徴とする請求項1に記載の薬剤学的組成物。
- 前記コンパウンドKの含量は、0.5〜1.5mg/gの範囲に強化されたことを特徴とする請求項1に記載の薬剤学的組成物。
- コンパウンドK成分の強化された紅参濃縮液を有効成分として含有する健康機能食品。
- 前記コンパウンドK成分の強化された紅参濃縮液は、健康食品総重量に対して0.1〜99重量%で含有されることを特徴とする請求項4に記載の健康機能食品。
- 前記コンパウンドKの含量は、0.5〜1.5mg/gの範囲に強化されたことを特徴とする請求項1に記載の健康機能食品。
