Disclosed herein is a dietary composition, which contains both red ginseng extract and Curcuma longa L. extract as active ingredients, and thus has an excellent effect on fatigue recovery through the synergistic effect of red ginseng extract and Curcuma longa L. extract. The present invention relates to a dietary composition, which contains a combination of red ginseng extract and Curcuma longa L. extract so as to show the synergistic effect of red ginseng and Curcuma longa L. , and thus has an excellent effect on recovery from mental fatigue (stress) and physical fatigue (exercise load) and the effect of increasing physical exercise duration ability.
[Background Art] Generally, fatigue refers to a state in which the ability to perform physical and mental activities is reduced. When a sign of fatigue appears, the time for the body to take a rest and recover from fatigue is required, but it is difficult in a busy modern society to correctly maintain the cycle of such fatigue and recovery from fatigue. The accumulation of fatigue caused by overwork can cause chronic fatigue and also cause many diseases, including digestive ulcer, hypertension, diabetes and the like. Also, cancer, stroke and heart disease are the three major causes of death of modern persons and are mainly attributable to overwork. With the recent well- being trend, the importance of exercise has been highlighted in order to enjoy life and to prevent and treat various adult diseases. Due to such social concerns, there has been an increasing need for the ingestion of scientific food for increasing exercise performance ability and recovering from physical fatigue, not only in exercise players, but also general persons. Products associated with fatigue recovery, known in Korea and foreign countries, are mostly products, containing vitamin B or various Chinese medicinal materials, including ginseng or red ginseng, evening primrose seed oil, Eleutherococcus Senticosus extract, etc. Also, the effects thereof include comprehensive and slightly abstract effects, such as tonic, beauty, hangover removal, blood circulation, immune enhancement and the like.
Panax ginseng CA. The effects of ginseng, known to date, include action on the central nervous system, anti-carcinogenic action and anticancer activity, immune function regulatory action, anti-diabetic action, liver function-improving action, cardiovascular disorder- improving and anti-atherogenic actions, blood pressure- controlling action, improvement of menopausal disorder and osteoporosis, anti-stress and anti-fatigue actions, antioxidant activity and aging inhibitory effects (Korean Ginseng, "components and effects", the Korea Ginseng Research Institute, 56-112, 1996) . Meanwhile, red ginseng is prepared by steaming raw ginseng (fresh ginseng) with water vapor or other means and drying the steamed ginseng and has a color ranging from light yellowish brown to reddish brown due to a browning reaction, which occurs during the drying process after steaming the fresh ginseng. Among various kinds of ginseng, red ginseng is known to exhibit excellent effects compared to white ginseng. Curcuma longa L. is a perennial plant belonging to the family Zingiberaceae and has been used for a long time as a Chinese medicinal material, spice and edible food in South Asia and Southeast Asia. The root and stem of Curcuma longa L. perform antioxidant and cell protective roles, and the main components thereof are known to be curcumin and curcumin derivatives (Chatterjee, S. et al, Food Research International, 32, 487-490, 1999). Due to the curcumin components, Curcuma longa L. was reported to have antioxidant effects, anticancer effects, antiinflammatory effects, antiviral effects, blood lipid- lowering effects, the effect of reducing cholesterol accumulated in the liver, and the like (An. B. et al, Korean J. Medicinal Crop Sci. 14(3) 168-172. 2006).
With respect to the synergistic effect of red ginseng or ginseng and Curcuma longa L. , there was a study on an effect on the induction of cancer cell death (Volate, S. et al, Carcinogenesis. 26(8): 1450-6., 2005). However, it is considered that there is yet no report on the red ginseng or ginseng and Curcuma longa L. on anti-fatigue, that is, on the use thereof for recovery from stress and exercise load. [Technical Problem]
Accordingly, the present inventors have conducted studies to provide a composition having a composite effect allowing recovery from mental fatigue and physical fatigue and, as a result, have found that a combination of red ginseng extract and Curcuma longa L. extract has an excellent effect on fatigue recovery, thereby completing the present invention.
Therefore, it is an object of the present invention to provide a dietary composition capable of showing the effect of allowing recovery from mental and physical fatigue through the synergistic effect of red ginseng extract and Curcuma longa L. extract.
[Technical Solution] To achieve the above object, in one aspect, the present invention provides a dietary composition containing a combination of red ginseng extract and Curcuma longa L. extract as active ingredients.
In another aspect, the present invention provides the use of the dietary composition for recovery from physical fatigue or for recovery from mental stress.
In still another aspect, the present invention provides the use of the dietary composition for increasing exercise duration ability. Hereinafter, the present invention will be described in further detail.
The present invention provides a dietary composition containing a combination of red ginseng extract and Curcuma longa L. extract as active ingredients. Also, the present invention provides the use of the dietary composition for recovery from physical fatigue or for recovery from mental stress .
In the present invention, the red ginseng extract (liquid) and the Curcuma longa L. extract (powder) are mixed with each other at a weight ratio of 1:7-3:1. Particularly, in the dietary composition for recovery from physical fatigue or mental stress, the red ginseng extract and the Curcuma longa L. extract are preferably mixed with each other at a weight ratio of 75:172. This is because the dietary composition of the present invention is easily formulated in the above-specified weight range, the composition containing the extracts within the above- specified weight ratio is suitable for ingestion, and the effects of red ginseng and Curcuma longa L. are complementarily bound within the above-specified range to show an excellent effect on recovery from physical fatigue and mental stress.
Also, in the inventive dietary composition for increasing exercise duration ability, the red ginseng extract (liquid) and the Curcuma longa L. extract (powder) may be mixed with each other at a weight ratio of 1:7-3:1, and preferably 10:19. More preferably, the extracts are mixed such that the weight ratio of saponin to curcumin is 1 : 2 as calculated in terms of the total saponin content of the red ginseng extract and the curcumin content of the Curcuma longa L. extract. This is because the dietary- composition of the present invention is easily formulated in the above-specified weight range, the composition containing the extracts within the above-specified weight ratio is suitable for ingestion, and the effects of red ginseng and Curcuma longa L. are complementarily bound within the above-specified range so as to show an excellent effect on physical exercise duration ability. The sum of the amounts of the red ginseng extract and the Curcuma longa L. extract may be 10~5-100 wt% (dry weight) based on the total amount of the composition.
Also, the red ginseng extract and the Curcuma longa L. extract, which are used in the present invention, may be obtained by heating and extracting each of the plants in water or alcohol, filtering the extract and concentrating the filtrate under reduced pressure.
Moreover, in the present invention, the red ginseng extract and the Curcuma longa L. extract may be concentrated using a process such as spray drying or freeze drying, in addition to a vacuum concentration process.
In addition, the red ginseng extract and the Curcuma longa L. extract, which are used in the present invention, may be commercial extract materials prepared using water or alcohol .
In addition to the above-described active ingredients, the inventive composition may, if necessary, contain one or more selected from excipients, saccharides, pigments, oil and fat, proteins and the like. The formulation of the inventive composition is not specifically limited and can be selected depending on the intended use thereof. For example, the inventive composition can be provided in the form of powders, granules, capsules, pastes, beverages and the like. [Advantageous Effects]
As described above, the inventive composition containing the red ginseng extract in combination with Curcuma longa L. extract allows recovery from mental stress by inhibiting the increase in stress hormone Cortisol caused by mental stress and inhibiting the increases in serum creatine phosphokinase activity, blood glucose and blood lactic acid. Also, the inventive composition can show the effect of recovery from physical fatigue by increasing exercise duration ability against physical fatigue (exercise load) and inhibiting the increases in blood glucose, lactic acid and lactate dehydrogenase levels. In addition, the inventive composition can provide the effect of increasing exercise duration ability. [Description of Drawings] FIG. 1 is a graphic diagram showing the swimming time of mice to exhaustion in a stress control group and stress groups (test groups 1-3) .
FIG. 2 shows the results of staining and microscopic observation for the storage of glycogen in mouse liver tissue before and after exercise in a control group (general diet) , a group fed with ginseng extract and a group fed with a combination of red ginseng extract and Curcuma longa L. extract. [Mode for Invention] Hereinafter, the present invention will be described in further detail with reference to examples and test examples. It is to be understood, however, that these examples and test examples are illustrative only, and the scope of the present invention is not limited thereto.
Reference Example 1: Preparation of red ginseng extract
1 kg of red ginseng was added to about 10-fold volume of water or alcohol and extracted 2-3 times at 90-100 °C for 12-24 hours for each extraction. The extract was filtered and concentrated under reduced pressure, thus obtaining red ginseng extract (ginseng saponin content of more than 70 mg/g) .
Reference Example 2: Preparation of Curcuma longa L. extract
1 kg of Curcuma longa L. was added to about 10-fold volume of water or alcohol and extracted 2-3 times at 90- 100 °C for 12-24 hours for each extraction. The extract was filtered and concentrated under reduced pressure, thus obtaining Curcuma longa L. extract (curcumin content of more than 60 mg/g) .
Example 1: Preparation of health functional food The red ginseng extract, prepared in Reference Example 1, and the Curcuma longa L. extract, prepared in Reference Example 2, were uniformly mixed at a weight ratio of 75:172. The mixture was used in the following tests about its effects of recovering from fatigue and increasing exercise duration ability.
Test Example 1: Test of effect on recovery from mental stress
1. Samples and test animals
As test samples, the composition prepared in Example
1, the red ginseng extract of Reference Example 1 and the Curcuma longa L. extract of Reference Example 2 were used.
As test animals, 5-week-old male Wistar rats and 6-week-old male hairless mice were used.
In the test animals, control groups were fed with AIN- 93G as basal diet, and test groups were fed with a mixture of each of test samples (red ginseng and Curcuma longa L.) with AIN-93G.
2. Test method
The test animals were grouped into a non-stress control group, a stress control group and stress groups, in which the stress groups were divided into test group 1 fed with red ginseng extract alone, test group 2 fed with Curcuma longa L. extract alone, and test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract. Each group consists of 10 mice. In the test, a restraint cage was used as restraint stress. As test diets, modified diets based on AIN- 93G were used, and the animals were bred for 4 weeks using a free feeding method.
To induce restraint stress, the animals were restricted in a restraint case for 3 hours before sacrifice after breeding with the test diets. Then, the animals were anesthetized and sacrificed, and samples (serum or plasma) were collected from the animals.
As biomarkers for evaluating fatigue recovery effects, blood Cortisol, glucose, lactic acid and creatine phosphokinase levels were measured.
3. Results
<Coraparison of blood Cortisol level>
The blood Cortisol levels of the non-stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 1 below. [Table l]

The blood glucose levels of the non-stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 3 below. [Table 3]

As shown in Table 4 above, the blood lactic acid levels were increased due to mental stress, and in all the test groups 1, 2 and 3, the increase in blood lactic acid level caused by mental stress was inhibited. Particularly, the test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract showed the most excellent effect (p<0.05).
Test Example 2: Test of effect on recovery from physical fatigue
1. Samples and test animals
As test samples, the composition prepared in Example 1, the red ginseng extract of Reference Example 1 and the
Curcuma longa L. extract of Reference Example 2 were used. As test animals, 5 -week-old male Wistar rats and 6 -week-old male hairless mice were used.
In the test animals, control groups were fed with AIN-93G as basal diet, and test groups were fed with a mixture of each of test samples (red ginseng and Curcuma longa L.) with AIN-93G. 2. Test method
The test animals were grouped into a non-stress group, a stress control group and stress groups, in which the stress groups were divided into test group 1 fed with red ginseng extract alone, test group 2 fed with Curcuma longa L. extract alone, and test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract. Each group consisted of 10 mice. In the test, swimming load was used, and as test diets, modified diets based on AIN-93G were used. The animals were bred for 4 weeks using a free feeding method.
The test groups were fed freely with the respective feeds containing the test samples for 4 weeks . In the exercise load test, the mice were forced to swim in a constant-temperature water pool, and the total swimming time immediately after exhaustion (death) was recorded. At 2 weeks, 3 weeks and 4 weeks after feeding with the test materials, the mice were subjected to adaptation training by forcing them to swim once a week for 5 min, 5 min and 10 min, respectively. After 4 weeks of feeding with the test materials, the test animals were forced to swim in a water tank and stimulated with a rod such that the limbs were all moved. Immediately after the mice were exhausted, the total swimming time were measured. When the mice did not rise to the surface of water for 7-10 seconds after the mice were went down below the surface, the mice were judged to be exhausted. Immediately after exhaustion, the animals were sacrificed, and samples (serum or plasma) were collected. In order to evaluate the effect of the test materials on recovery from fatigue, exercise duration time and blood glucose, lactic acid, creatine phosphokinase and lactate dehydrogenase (LDH) levels as biomarkers were measured. Also, from the control groups (general diet) , the group fed with red ginseng extract and the group fed with a combination of red ginseng extract and Curcuma longa L. extract, the liver tissue was sampled before and after the swimming test (exercise) . Then, the liver tissue samples were subjected to haematoxylin and eosin staining and microscopically compared with respect to the storage of glycogen.
3. Results
<Comparison of swimming time of mice> The swimming time of mice to exhaustion in the stress control group and the stress groups (test groups 1, 2 and 3) is shown in FIG. 1.
As shown in FIG. 1, the test groups 1, 2 and 3 all showed an increase in the exercise duration time by physical fatigue, compared to the stress control group. Particularly, the test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract showed the greatest increase in exercise duration time (p<0.05). In the test groups, because the exercise duration time by physical fatigue was increase, it can be seen that the exercise duration ability was increased. <Comparison of blood glucose level>
The blood glucose levels of the non-stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 5 below. [Table 5]

Particularly, the test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract showed the most excellent effect (p<0.05).
<Comparison of blood lactic acid level> The blood lactic acid levels of the non-stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 6 below. [Table 6]

As shown in Table 6 above, the blood lactic acid levels were increased due to the lack of blood oxygen supply caused by physical stress (exercise load) , and in all the test groups 1, 2 and 3, the increase in blood lactic acid level caused by physical stress was inhibited. Particularly, the test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract showed the most excellent effect (p<0.05).
<Comparison of blood creatine phosphokinase level>
The blood creatine phosphokinase levels of the non- stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 7 below. [Table 7]

<Comparison of blood lactate dehydrogenase (LDH) level>
The blood lactate dehydrogenase (LDH) levels of the non-stress control group, the stress control group and the stress groups (test groups 1, 2 and 3) are shown in Table 8 below. [Table 8]

As shown in Table 8 above, the activity of lactate dehydrogenase (LDH) catalyzing the formation of lactic acid in an oxygen-free condition was increased due to physical fatigue (exercise load) , but in all the test groups 1, 2 and 3, the increase in the activity of lactate dehydrogenase (LDH) was inhibited. Particularly, the test group 3 fed with a combination of red ginseng extract and Curcuma longa L. extract showed the most excellent effect (p<0.05) .
<Comparison of glycogen in liver tissue>
The storage of glycogen in liver tissue before and after exercise was directly observed with a microscope after staining. Based on vacuolization (white portion) , the glycogen content in liver tissue could be compared between the test groups, and the observation results are shown in FIG. 2. As can be seen in FIG. 2, the group fed with a combination of red ginseng extract and Curcuma longa L. extract had increased glycogen content before and after exercise. Because the difference in glycogen content is considered to be a direct basis that indicates exercise duration ability, it can be seen that the administration of a combination of red ginseng extract and Curcuma longa L. extract leads to a further increase in exercise duration ability, [industrial Applicability] As described above, a dietary composition containing a combination of red ginseng extract and Curcuma longa L. extract has an excellent effect on recovery from mental stress and physical stress. Thus, the dietary composition containing red ginseng extract and Curcuma longa L. extract as active ingredients can assist in recovering from stresses. Also, the dietary composition increases exercise duration ability through recovery from physical stress, and thus can be used as a dietary composition for increasing exercise duration ability.

The dietary composition of Claim 1, wherein the red ginseng extract and the Curcuma longa L. extract are mixed with each other at a ratio of 1:7 - 3:1.

The dietary composition of Claim 1, wherein the red ginseng extract and the Curcuma longa L. extract are contained in an amount of 10"5 - 100 wt% (dry weight) based on the total weight of the composition.
[Claim 4]
The dietary composition of Claim 1, wherein each of the red ginseng extract and the Curcuma longa L. extract is obtained by extracting each of the plants in water or alcohol, filtering the extract and concentrating the filtrate under reduced pressure.
[Claim 5]
Use of a dietary composition, containing a combination of red ginseng extract and Curcuma longa L. extract as active ingredients, for recovery from mental stress.
[Claim 6]
Use of a dietary composition, containing a combination of red ginseng extract and Curcuma longa L. extract as active ingredients, for recovery from physical stress. [Claim 7]
The use of Claim 6, wherein the dietary composition increases exercise duration ability.
