Amaxaエレクトロポレーションシステム(溶液TでプログラムU23)を用いて、CHO−K1細胞をプラスミドpcDNA3.1_3xHA−Δ1−17−hCXCR2にトランスフェクトさせた。トランスフェクション細胞のプールを、トランスフェクション後の2日目から選択圧(1000αg/mL G418)の下に置いた。8日後に、ヒトCXCR2陽性集団を、FMAT Blue標識ヒトGRO−αを使って同定した。ヒトGro−α(Biosource、PHC1063)のFMAT Blue標識は、製造業者の指示(Applied Biosystems、4328408)に従ってFMAT Blue単官能反応性色素キットを用いて実行した。FACSaria(BD Biosciences)を用いて、単細胞を96ウェル細胞培養プレートに分配した。FMAT Blue標識ヒトGRO−αを使って、FACSarray(BD Biosciences)デバイスで、成長するクローンをΔ1−17ヒトCXCR2発現について試験した。最も高い発現を有するCHO−K1クローンを選択した(9000のMCF値)。
FuGene HDトランスフェクション試薬(Roche)を用いて、HEK293T細胞をプラスミドpcDNA3.1_cCXCR2にトランスフェクトさせた。トランスフェクションの2日後、50nMのFMAT Blue標識GRO−αを使って、FACSarray(BD Biosciences)デバイスで、細胞をcCXCR2発現について試験した。優れた発現(およそ12000のMCF値)を有する細胞をさらに用いた。
37℃/5%CO2で成長させ、1×非必須アミノ酸、0.15%炭酸水素ナトリウム、1mMピルビン酸ナトリウムおよび15%ウシ胎児血清(Invitrogen)を加えたMEMイーグル培地(Invitrogen)でルーチンに継代培養したラット好塩基球白血病細胞(RBL−2H3)を、製造業者のプロトコルに従ってエレクトロポレーション(Amaxa Biosystems)によるヌクレオフェクションにかけた。トランスフェクション細胞を37℃/5%CO2でインキュベートし、トランスフェクションの24時間後に、1mg/mLの最終濃度までジェネテシンを加えることによって、抗生物質選択を開始した。トランスフェクション細胞を選択培地で3〜5日の間成長させ、継代培養した後、連続希釈による96ウェルプレートへの単細胞選別にかけた。およそ2週後に、活発に成長するコロニーを増殖させ、cynoCXCR2転写産物発現についてその後分析した。分析のために、次に陽性クローンをさらに増殖させた。
10%の無テトラサイクリンウシ胎児血清(FBS)(Biosera)、1%ペニシリン/ストレプトマイシンと10μg/mLのブラストサイジンを加えた2mMのL−グルタミンを含有するハムのF12培地で、チャイニーズハムスター卵巣T−Rex(T−Rex(商標)−CHO、Invitrogen、#R718−07)を単層培養として37℃で維持した。このテトラサイクリンによって調節される発現(T−Rex(商標))細胞系は、テトラサイクリンリプレッサー(TetR)を安定して発現する。ヌクレオフェクション手法(細胞系NucleofectorキットT、Amaxa Biosystem、プログラムU−23)を用いて、次に両方のCXCR2構築物を発現する安定細胞系を生成した。トランスフェクション細胞を37℃/5%CO2でインキュベートし、トランスフェクションの48時間後に300μg/mLのZeocinで処理した。陽性形質転換体の選択を可能にするために、Zeocinの存在下で細胞を2週間培養し、その後、Mo−Flo FACSソーターを用いて単細胞選別を実行した。2週後に、300μg/mLのZeocin濃度のそれらの通常の培地で維持しながら、活発に成長するコロニーを増殖させた。
ヒトおよびカニクイザルCXCR2の異なるN末端および細胞外のループ(EL)のひと続きの配列を表すペプチドを、Bachemに注文した(表5)。「環状」と表されるペプチドでは、最初および最後のアミノ酸はシステイン残基で置き換えられ、野生型配列の天然に存在する内部システインはロイシン残基で置き換えられた。これらのペプチドは、隣接システイン残基を通して環化された。
3頭のラマを、ヒトCXCR2を発現する哺乳動物細胞で7〜9回免疫化し、1頭のラマは、カニクイザルCXCR2を発現する哺乳動物細胞で6回免疫化した。このレジメンに、(不)完全フロイントアジュバントに混合したペプチド−キーホールリンペットヘモシアニン(KLH)コンジュゲートカクテルの4回の投与が続き、ペプチドは、ヒトとカニクイザルの両方のCXCR2の細胞外ループ番号2および3を表す(表5を参照)。他の8頭のラマを、ヒト完全長CXCR2またはpVAX1から発現されるΔ1−17CXCR2をコードするDNAで4〜5回免疫化し、その後、ヒト完全長CXCR2を発現する哺乳動物細胞の1回投与が続いた。3頭のさらなるラマを、pVAX1から発現されるカニクイザルCXCR2をコードするDNAで4回免疫化し、その後、カニクイザルCXCR2を発現する哺乳動物細胞の1回投与が続いた。免疫性の血液およびリンパ節試料を、抗原の各々の投与から4および8日後にとった。
cDNA試料は、免疫性血液およびリンパ節試料の全RNA調製物から作製した。Nanobodiesをコードするヌクレオチド配列は、プライマーABL051、ABL052およびABL003を用いて、一段階RT−PCR反応でヒトまたはカニクイザルのCXCR2で免疫化した全てのラマのcDNA試料から増幅した。プライマー配列は、表6に示す。試料中のIgG2およびIgG3cDNAから増幅させた700bpアンプリコンをゲルから単離し、その後、SfiI制限酵素部位を含有するABL050プライマーおよびABL003プライマーを用いるネステッドPCR反応で鋳型として用いた。PCR生成物をその後SfiIおよびBstEII(VHH遺伝子のFR4に天然に存在する)で消化し、ファージミドベクターpAX50の対応する制限部位にライゲーションし、大腸菌(Escherichia coli)TG−1でのエレクトロポレーションの後にライブラリーを得た。pAX50は、LacZプロモーター、大腸菌ファージpIIIタンパク質コード配列、アンピシリンまたはカルベニシリンの抵抗性遺伝子、マルチクローニング部位およびgen3リーダー配列を含有するpUC119から誘導された発現ベクターである。Nanobody(登録商標)コード配列とインフレームで、ベクターはC末端のc−mycタグおよび(His)6タグをコードした。ファージミドベクターは、geneIII生成物との融合タンパク質として個々のNanobodiesを発現する、ファージ粒子の生成を可能にする。
バクテリオファージの表面で発現された上述のpAX50 Nanobodyライブラリーは、CXCR2エピトープを提示するペプチド、膜抽出物および全細胞を用いて選択した。
溶出されたファージを指数的に成長するTG−1細胞に感染させ、次にそれらをカルベニシリン含有LB寒天平板の上で平板培養した。カルベニシリン抵抗性クローンを挿入断片の存在について分析し、陽性クローンの配列を検証した。カルベニシリンを加えたTB培地で対象のクローンを成長させ、発現のためにIPTGの添加によって誘導した。発現は37℃で4時間継続させ、続いて細胞を遠心沈降させた。大腸菌(E. coli)発現培養物からの一晩冷凍細胞ペレットをPBS(元の培養容量の1/10)に溶解し、穏やかな振盪条件下で4℃で1時間、インキュベートした。次に、細胞をもう一度遠心沈降させ、ペリプラズム間隙に分泌されたタンパク質を含有する上清を保存した。
ペリプラズム抽出物(上記)を、ヒトCXCR2への結合におけるGro−αとの競合についてFACSで分析した。2×105細胞を、4℃で30分間、FACS緩衝液(PBS+10%ウシ胎児血清(Sigma、F7524))中のペリプラズム抽出物の1/2希釈溶液とインキュベートした。次に、FACS緩衝液中の6nMのFMAT Blue標識ヒトGro−αの等量を加え、暗所において4℃でさらに30分間、インキュベーションを継続した。細胞を次にFACS緩衝液で3回洗浄し、最終的にFACS緩衝液に再懸濁した。死細胞は、ヨウ化プロピジウム(Sigma、P4170)で染色された。試料を次にFACSarray(BD Biosciences)で分析した。表7は、そのペリプラズム抽出物がヒトCXCR2に関してGro−αとの競合を示したNanobodiesを掲載する。
11.一価Nanobodiesの構築
Fwd−EVQL−MfeIおよびRev−TVSS−BstEIIプライマー(表1)を有する機能的ファージミドクローンでのPCRによって得られた、Nanobody含有DNA断片を、MfeIおよびBstEIIで消化し、pAX100ベクターにライゲーションし、大腸菌(E. coli)TG−1コンピテント細胞に形質転換させた。pAX100は、LacZプロモーター、カナマイシン抵抗性遺伝子、マルチクローニング部位およびOmpAリーダー配列を含有するpUC119から誘導された発現ベクターである。Nanobodyコード配列とインフレームで、ベクターはC末端のc−mycタグおよびHis6タグをコードした。カナマイシン抵抗性クローンを挿入断片の存在について分析し、陽性クローンの配列を検証した。
対象のNanobodiesをコードする発現ベクターを含有するTG−1細胞を、TB培地プラス100μg/mlカナマイシンを含有するバッフル振盪フラスコで成長させ、発現のために1mM IPTGの添加によって誘導した。発現は37℃で4時間継続させた。細胞を回収した後に、ペリプラズム抽出物を調製し、固定化金属親和性クロマトグラフィー(HisTrap FF Crude、GE Healthcare)と、続く脱塩(HiPrep26/10、GE Healthcare)またはPBSでのゲル濾過クロマトグラフィー(Superdex75 HR16/10、GE Healthcare)によって、His6標識Nanobodiesを精製した。
ヒトおよびカニクイザルのCXCR2でのFACSリガンド競合アッセイで、精製した一価の抗CXCR2 Nanobodiesを3nMのFMAT Blue標識Gro−αに対して滴定した(表10)。ヒトCXCR2では、ブロッキング効力の範囲は二桁のnMからnM未満であるが、カニクイザルCXCR2では、それらは一桁から二桁のnMの間である。
(1)細胞内カルシウムのアゴニストによって誘導される放出の測定(FLIPR)
ヒトまたはカニクイザルのCXCR2受容体を発現するRBL細胞を96ウェルプレートに播種し、37℃で一晩インキュベートした。実験当日、Fluo−4色素を37℃で30分の間細胞に加え、続いて精製した一価の抗CXCR2 Nanobodiesと30分の間インキュベーションした。最後に、蛍光定量的画像化プレートリーダー(FLIPR)を用いてGRO−αの添加を実施し、続いて細胞内カルシウムの放出に対応する蛍光性シグナルを検出した。ヒトCXCR1を発現するL2071細胞を用いて、選択アッセイを実施した。アッセイプロトコルはCXCR2についての記載と同じであったが、アゴニストとしてIL−8が用いられた。平均IC50値の要約を表11に示し、さらに、試験したNanobodiesのいずれも、CXCR1受容体での細胞内カルシウムのアゴニストによって誘導される放出の阻害を試験濃度(1μM最大濃度)で示さなかった。
精製した一価の抗CXCR2Nanobodiesを、ヒトCXCR2受容体を発現するCHO細胞から調製したGRO−α、GDP、SPAビーズおよびCHO−CXC2膜と一緒に、96ウェルプレートで60分の間インキュベートした。これに、[35S]GTPγSの添加、およびさらなる60分間のインキュベーションが続いた。最後に、プレートを遠心分離した後、Topcountで読み取った。平均IC50値の要約を表11に示す。
(1)ヒト好中球全血形状変化アッセイ(hWBSC)
ドナーは、全身薬物療法を受けていない健康な正常ボランティアであった(Novartis Horshamドナーパネル)。52mMのEDTA(滅菌)で血液凝固を阻止した全血を、1mL EDTA対9mL血液の比で回収した。血液を室温で回収し、使用前に37℃に予熱した。ケモカインによる刺激の前に、80μLの全血を、室温で10分の間CXCR2 Nanobodiesとプレインキュベートした(用量応答につき10ポイント(0.03〜1.144×10−7μM);10μLの形状変化アッセイ緩衝液を加えた0化合物以外の全てのウェルに、10μLのrhGROα(2nMの近似EC70濃度)を加えた。試料を静かに振盪し、37℃でさらなる5分間インキュベートした。管を次に氷上に置き、250μLの氷冷最適化CellFix(商標)溶液を加え、管を静かに振盪し、さらなる5分間インキュベートし、その後、1.4mLの1×塩化アンモニウム溶解溶液を全ての管に加え、さらなる20分間氷上に放置した。赤血球溶血の後、FACSCaliburフローサイトメーター(Becton Dickinson)で試料を分析した。細胞集団は、前方散乱/側方散乱(FSC/SSC)ゲーティングと、続く第1のプロットからのゲート制御顆粒球を用いたFSC/FL−2プロットによって同定した。好中球はFL−2プロットの上で好酸球と区別されたが、その理由は後者がより高い自己蛍光性を有するからである。1試料につき5000個のイベントを計数した。
ドナーは、全身薬物療法を受けていない健康な正常ボランティアであった(Novartis Horshamドナーパネル)。52mMのEDTA(滅菌)で血液凝固を阻止した全血を、1mL EDTA対9mL血液の比で回収した。白血球は、標準プロトコルを用いて単離した:4%デキストランを20mLの血液凝固を阻止した血液に加え、静かに混合し、次に30分間氷上でインキュベートして赤血球を沈降させた。末梢血単核細胞(PMN)を含有する上清をFicoll−Paque(登録商標)密度勾配の上に次に重層し、18℃で25分間、300×gで遠心分離した。PMNに富む分画を500μLの1XPBSに再懸濁し、低張性ショックを使用して赤血球溶血を実行した。20mLの氷冷滅菌の無エンドトキシン蒸留水をペレットに加え、溶解を30〜40秒間起こさせた後に、20mLの2×PBSを加えた。試料を静かに混合し、18℃で10分間300×gで遠心分離し、顆粒球を得た。顆粒球ペレットを500μLの1×PBSに再懸濁し、50mLの×1 PBSで2回洗浄した。顆粒球ペレットをRPMI1640、pH7.4、プラス2.5%FBSに再懸濁し、計数し、2e6/mLの最終濃度まで希釈した。Becton Dickinsonからの3μm PET膜とトランスウェルプレートを用いて、移動を測定した。簡潔には、6nMのGROα(EC80−EC100)をプレートの底ウェル(1000μL/ウェル)に加えた後に、多重ウェル挿入断片を適正位置に下げ、RTで30分間様々な濃度のNanobody(一価については0.13〜1000nM、二パラトープについては0.6pM〜30nM)とプレインキュベートしたPMNを次に挿入断片に加えた(500μL/ウェル)。プレートを次に37℃で90分間インキュベートし、底チャンバーに移動した細胞は、FACSCaliburフローサイトメーターを用いて計数された。フローサイトメーターは、1試料につき20秒の設定時間、FSC/FL−2プロット上のR2ゲートの中でのイベント数を計数するように設定された。
前腕または脚のいずれかからとられた静脈血を、1mLクエン酸ナトリウム対9mL血液の比の、3.8%クエン酸ナトリウム(滅菌)で血液凝固を阻止した。血液を室温で回収し、使用前に37℃に予熱した。ケモカインによる刺激の前に、80μLの全血を、室温で10分の間CXCR2 Nanobodiesとプレインキュベートした(用量応答につき10ポイント(0.03〜1.144×10−7μM);10μLの形状変化アッセイ緩衝液を加えた0化合物以外の全てのウェルに、10μLのrhGROα(30nMの近似EC70〜90濃度)を加えた。試料を静かに振盪し、37℃でさらなる5分間インキュベートした。管を次に氷上に置き、250μLの氷冷最適化CellFix(商標)溶液を加え、管を静かに振盪し、さらなる5分間インキュベートし、その後、2mLの溶解緩衝液(Sigma Aldrich #R7757)を全ての管に加え、さらなる40〜60分間氷上に放置した。赤血球溶血の後、FACSCaliburフローサイトメーター(Becton Dickinson)で試料を分析した。細胞集団は、前方散乱/側方散乱(FSC/SSC)ゲーティングと、続く第1のプロットからのゲート制御顆粒球を用いたFSC/FL−2プロットによって同定した。好中球はFL−2プロットの上で好酸球と区別されたが、その理由は後者がより高い自己蛍光性を有するからである。1試料につき5000個のイベントを計数した。
16.二価Nanobodiesの構築
二価Nanobodiesを構築するために2つのアプローチを用いた。
一価の構成単位をコードするプラスミドDNAで、PCR増幅を実行した。Fwd−EVQL−MfeIおよびGlySerリンカーの一部をコードするリバースプライマーを用いてN末端構成単位を増幅したのに対して、GlySerリンカーの残りの部分をコードするフォワードプライマーおよびRev−TVSS−BstEIIを用いてC末端構成単位を増幅した(表6)。N末端断片をMfeIおよびBamHIで消化し、C末端断片をBamHIおよびBstIIで消化した。次にこれらを同時にpAX100ベクターにライゲーションし、大腸菌(E. coli)TG−1コンピテント細胞に形質転換させた。

精製した多価の抗CXCR2 Nanobodiesを、ヒトCXCR2受容体を発現するCHO細胞から調製したアゴニスト(GRO−α、IL−8またはENA−78)GDP、SPAビーズおよびCHO−CXC2膜と一緒に、96ウェルプレートで60分の間インキュベートした。これに、[35S]GTPγSの添加、およびさらなる60分間のインキュベーションが続いた。最後に、プレートを遠心分離した後、Topcountで読み取った。平均IC50値の要約を、表15に示す。
Schild分析は、IL−8およびGRO−αによって刺激された[35S]GTPγS蓄積アッセイを用いて実行した。このアッセイフォーマットは、[35S]GTPγSの添加の前にアゴニストおよびNanobodyの平衡を可能にし、結果として、機構の誤解釈につながるかもしれない半平衡のいかなる人為産物も避けられるはずである。これを実行するために、Nanobodyの漸増濃度の存在下でアゴニストの濃度反応曲線を求めた。2つの一価Nanobodies、54B12および163E3と、生じた多価Nanobodyのデータが例として与えられ、図1に示される。データはGRO−αの濃度反応曲線を示すが、IL−8を用いたときにも類似のデータが観察された。
セクション15に記載されるのと同じ方法
熱シフトアッセイ(TSA):5μlの精製した一価Nanobody(80mg/ml)を、10μlの緩衝液(100mMリン酸塩、100mMホウ酸塩、100mMクエン酸塩、115mM NaCl、3.5から9の範囲の異なるpHで緩衝した)中の5μlの蛍光性プローブSypro Orange(Invitrogen、Carlsbad、CA、カタログ#S6551)(最終濃度10×)と混合した。次に試料を37から90℃まで4.4℃/sでLightCycler 480II機器(Roche、Basel、Switzerland)で加熱し、その後、それらを37℃まで2.2℃/sで冷却した。熱誘導アンフォールディングの結果、タンパク質の疎水性パッチが露出し、それにSypro Orangeが結合し、蛍光強度の増加をもたらす。蛍光強度曲線の一次導関数の屈折点は、融点(Tm)の尺度の役目をする。(Ericssonら2006(Annals of Biochemistry、357:289〜298頁)。

ショットガン突然変異誘発は、真核生物細胞中の大きなライブラリーの突然変異標的タンパク質の発現および分析を可能にする、所有権のあるハイスループット細胞発現技術を用いる。機能の変化をアッセイするために、タンパク質中のあらゆる残基は、通常複数の他のアミノ酸に個々に突然変異させられる。タンパク質は標準の哺乳動物細胞系の中で発現されるので、真核生物での翻訳または翻訳後のプロセシングを必要とする困難なタンパク質さえもマッピングすることができる。

ショットガン突然変異誘発マッピングによって同定された重要なアミノ酸は、3つのCXCR2 MAbの結合部位を規定する。MAb RD HBC792は、CXCR2のN末端領域をマッピングし、重要な残基の近接性はエピトープが本来は線状であることを示唆する。MAb RD HBC793およびRD HBC794は、CXCR2のECL1およびECL3によって主に形成される高次構造が複雑なエピトープに結合するようである。ケモカイン受容体で細胞外ループを所定の位置に保つ2つのジスルフィド架橋を形成することが公知である細胞外Cys残基の突然変異は、MAb 793および794の結合も排除するので、エピトープ相互作用に直接に関与するとは考えられていない。793および794のエピトープはかなり重複するが、2つの間の僅差が明白である。
細胞内カルシウムのアゴニストによって誘導される放出(FLIPR)
CXCR2選択性をチェックするために、異なる一価の抗CXCR2ナノボディを、個々のナノボディ構成単位が35GSリンカーによって分離されている、一価、二価または二パラトープ性の構築物として合わせた。
好中球は、化学誘引を媒介する高レベルのCXCR2受容体を発現する、炎症での主な炎症誘発性細胞の1つである。CXCR2リガンド(GROα)または対照刺激(FMLP)による刺激の後の好中球の形状変化は、フローサイトメトリーによって数量化することができ、活性化マーカーとして用いることができる。
ヒト血清中のCXCR20079−35GS−CXCR20076(79−76)、CXCR20079−35GS−CXCR20076−ALB8(79−76−Alb8)CXCR20079−35GS−CXCR20086(79−86)およびCXCR20079−35GS−CXCR20086−ALB8(79−86−Alb8)に結合するIgG抗体の形成を、健康なドナーの血清(44人の男性および44人の女性)をサンドイッチELISAでスクリーニングすることによって評価した。簡潔には、ナノボディCXCR20079−35GS−CXCR20076−ALB8を、マイクロタイタープレートの上に直接に固定化する。抗ナノボディ抗体を固定化ナノボディによって捕捉し、西洋ワサビペルオキシダーゼに結合している抗ヒトIgG(Fc特異)抗体によって検出する。基質TMBとのインキュベーションの後、酵素反応の有色生成物の光学密度(OD)を450nmで測定する。用いる陽性対照抗体は、ヒト抗CXCR2抗体NOV0205(1M8)(ファージディスプレイによって内部に生成される)、バッチACE00277である。アッセイは検証されないので、予備的カットオフポイントを推定する、平均の標準化OD(OD試料/OD陰性対照)。

- ケモカイン受容体CXCR2に対するものであるか、またはそれに結合する、2つの免疫グロブリン抗原結合ドメインを含むポリペプチドであって、前記第1の抗原結合ドメインは、ラクダ科動物に由来する重鎖抗体の単一重鎖からの第1のVHHドメインもしくはその断片に含まれるか、またはヒト化を含めて配列最適化されたその変異体であり、前記第2の抗原結合ドメインは、ラクダ科動物に由来する重鎖抗体の単一重鎖からの第2のVHHドメインもしくはその断片に含まれるか、またはヒト化を含めて配列最適化されたその変異体であり、ポリペプチドのC末端は、抗原結合ドメインの配列への、少なくとも1つのさらなるアミノ酸残基の伸長部を含み;第1の抗原結合ドメインはCXCR2上の第1のエピトープを認識し、第2の抗原結合ドメインはCXCR2上の第2のエピトープを認識するポリペプチド。

- 100nM未満のIC50で、ヒトCXCR2を発現するRBL細胞からのGro−α誘導カルシウム放出を阻害することができる、請求項1〜38のいずれか一項に記載のポリペプチド。

- アテローム硬化症、糸球体腎炎、炎症性腸疾患(クローン病)、血管形成ならびに黄斑変性症、糖尿病性網膜症および糖尿病性神経障害を含む新しい血管発達を特徴とする疾患、多発性硬化症、乾癬、壊疽性膿皮症、加齢性黄斑変性疾患、目のベーチェット病、ブドウ膜炎、特発性のPAH、家族性PAHおよび関連PAHを含む肺動脈高血圧(PAH)、慢性炎症性疾患、慢性関節リウマチ、骨関節炎、非小細胞がん腫、結腸がん、膵がん、食道がん、卵巣がん、乳がん、固形腫瘍および転移、黒色腫、肝細胞がん、虚血再かん流傷害、鎌状赤血球症での溶血性輸血誘発血管閉塞危機、虚血/再かん流傷害、急性の脳卒中/心筋梗塞、非開放性頭部損傷、外傷後炎症またはインスリン抵抗性糖尿病の処置において使用するための、請求項1〜42のいずれか一項に記載のポリペプチド。
- 請求項1〜42のいずれか一項に記載のポリペプチドの有効量を対象に投与することを含む、慢性閉塞性肺疾患(COPD)または(COPD)の増悪を処置する方法。
- 請求項1〜42のいずれか一項に記載のポリペプチドの有効量を対象に投与することを含む、嚢胞性線維症、喘息、特に重度の喘息および喘息の増悪、急性の肺損傷、急性の呼吸窮迫症候群、特発性の肺線維症、気道リモデリング、閉塞性細気管支炎症候群または気管支肺異形成からなる群から選択される状態を処置する方法。

- 喘息、特に重度の喘息および喘息の増悪、急性の肺損傷、急性の呼吸窮迫症候群、特発性肺線維症、気道リモデリング、閉塞性細気管支炎症候群または気管支肺異形成の処置のための医薬の製造における、請求項1〜42のいずれか一項に記載のポリペプチドの使用。
