Abstract
Description
本発明は、特にワクチン製造のためのETEC CS6抗原の調製において有用な方法、ならびに、該方法によって得られる細胞およびワクチンに関連する。
大腸菌表面抗原6(CS6)は、開発途上国の乳幼児および小児、ならびにそのような地域への旅行者における下痢症の最も一般的な原因である毒素原性大腸菌(ETEC)の最もよく見られる非線毛性定着因子(CF)の1つである。
本開示の文脈においては、下記の用語は以下の意味を有する。
第一の局面において、ETEC CS6抗原の提示が少なくとも100%、好ましくは少なくとも200%、より好ましくは少なくとも300%増大するように、該抗原を発現する細胞を、0.6〜2.2重量%のフェノールを含む水溶液と接触させる段階を含む、細胞表面上におけるETEC CS6抗原の提示を増大させるための方法が開示される。
CS6抗原の提示を増大させるための方法
第一の局面において、本発明は、ETEC CS6抗原の提示が少なくとも20%増大するように、好適な温度で好適な時間、該抗原を発現する細胞を、0.6〜2.2重量%のフェノールを含む水溶液と接触させる段階を含むことを特徴とする、細胞表面上におけるETEC CS6抗原の提示を増大させるための方法を提供する。抗原提示の少なくとも20%の増大とは、細胞表面に存在し、かつ好適な方法(詳細については下記を参照のこと)によって検出可能な抗原の量が、本方法に供されていないが他の点では同等である細胞と比較して、少なくとも2倍になることを意味する。好ましくは、抗原提示の増大は、少なくとも30%、40%、50%、60%、70%、80%、または90%であり、より好ましくは少なくとも100%、125%、150%、175%、200%、250%、または300%である。最も好ましくは、増大は少なくとも100%である。
第二の局面において、本開示は、フェノール濃度、接触温度、および接触時間が、CS6抗原の提示の増大と同時に前記細胞の少なくとも107倍の不活化が生じるように選択される、第一の局面に従う方法を含む、CS6発現ETECに対する免疫化のための死滅全菌体ワクチンの製造のための方法を提供する。好ましくは不活化の程度は少なくとも108倍であり、より好ましくは109倍であり、さらにより好ましくは1010倍であり、最も好ましくは処理後に生存細胞が全く存在しない。細胞の不活化は、当技術分野において公知の任意の一般的な手段によって定量できる。好適な方法は本明細書において「材料および方法」という見出しのついた章において開示される。
第三の局面において、第一または第二の局面に従う方法によって得られる細胞が提供される。フェノール処理により、(インビトロでは例えば抗体によって、およびインビボでは免疫系によって、いずれにおいても)より多くの抗原がより検出可能となるように、明らかな変化が細胞壁および/または抗原の構造に生じる。言い換えれば、このように処理された細菌細胞は、構造におけるその変化を構造的用語で説明することはできないが、第一または第二の局面の方法によって新しい構造を獲得している。
図1Aおよび図1Bに表されるように、CS6の表面発現を有する野生型ETEC株から調製したCS6の構造遺伝子(cssA、cssB、cssC、cssD)を有するDNA断片をPCRで増幅し、クローニングし、発現ベクターpJT-CS6-ThyAを構築した。次いでこのプラスミドをチミン依存性の毒素非産生性大腸菌 C600-ΔthyA 株にエレクトロポレーションによって導入し、免疫ドットブロット法(図2A)で示されるように、増殖培地にIPTGを加えることによりCS6の表面発現を誘導した。誘導物質がないときにはCS6の発現は全く観察されなかった(データ非表示)。ドットブロット法を用いて組換えC600-CS6 株によるCS6の発現を調べた際に、本発明者らは、CF-CTB-ETECワクチンにおけるCS4+CS6ワクチン株として以前に使用されたCS6参照株 E11881/23と比較して、少なくとも8倍高いレベルのCS6をこの組換えC600-CS6 株が発現することを見出した(図2A)。同様に、阻害ELISA法を用いてCS6の表面発現を特異的に定量した場合にも、組換え株では参照株と比較しておよそ10倍多い量のCS6が見出された(図2B)。
細菌表面上のCS6抗原の特性を保ちながらCS6発現細菌を死滅させることを目的として、ホルムアルデヒドとフェノールの効果を比較した。予備研究により、0.3%または0.6%のホルムアルデヒドで細菌を処理すると、支障なく細菌を死滅させる一方で、検出可能なCS6抗原の完全な消失が生じることが示された(データ非表示)。対照的に、0.5%のフェノールで細菌を処理すると、供試細菌が死滅するだけでなく、CS6抗原が保たれてもいた(データ非表示)。一方、フェノールのより低い試験濃度0.25%では、細菌の完全な死滅が生じなかった。これらの結果により、CS6抗原を保ちながら細菌を不活化するためにフェノール処理が有用であり得ることが示された。したがって、最適な不活化法を見出すために、組換えC600-CS6株および比較のための他のCS6過剰発現株(TOP10-CS6-Amp)の双方の不活化について、種々の濃度のフェノールを試験した。表2に見られるように、0.25%のフェノールでは不活化されなかったが、0.5%、0.8%、1%、および1.6%のフェノールを用いて試験した双方の株については細菌が不活化され、阻害ELISA法によって試験した結果、表面CS6が保たれてもいた。0.8%のフェノールを用いて細菌を不活化した場合に最大のCS6レベルが見出され、この処理によって実際に、CS6表面抗原の推定量が非処理細菌と比較して再現性よく増大した(表2)。したがってこれらの結果に基づき、C600-CS6株および参照株E11881/23の双方を不活化するために0.8%の濃度のフェノールを使用し、これにより、阻害ELISA法によって試験すると組換え株では参照株よりも6倍多い量のCS6を有する死滅細菌を生じた。次いで、これらの不活化細菌をマウスの経口免疫処置に使用した。

組換え株C600-CS6の免疫原性の最初の試験において、本発明者らはBalb/Cマウス群およびC57 BI/6マウス群の双方を、フェノールで死滅させた同数の細菌を用いて免疫化し、CS6に対する血清の抗体応答をELISA法によって測定して比較した。いずれの系統のマウスにおいても著しい抗CS6応答が誘導されたが、C57 BI/6マウスにおける抗体応答は実質的にBalb/Cマウスよりも高かった(データ非表示)。本発明者らはしたがって、経口投与した、C600-CS6株および参照株E11881/23のフェノール死滅化ワクチン調製物の免疫原性を比較したさらなる経口免疫処置試験のためには、C57 BI/6マウスを使用した。結果は、全ての免疫化マウスがCS6に対する血清IgG+IgM抗体の産生を伴う応答をしたこと、および、参照株 E11881/23によって免疫化したマウスのものよりもC600-CS6株によって免疫化したマウスではCS6に対する抗体価が平均して60倍超高いことを示した(図3)。糞便および腸のIgA抗体のCS6に対する応答も調べた(図3)。いずれの場合においても、対応する参照株によって免疫化したマウスにおけるレベルと比較して、組換えC600-CS6株では平均して75倍高いレベルの、CS6に対する糞便および腸のIgA抗体が有意に誘導された。参照株では、免疫化していない対照マウスで見られるものよりもわずかに高い抗CS6粘膜IgAのレベルが誘導されたにすぎなかった。
C600-CS6株をロータリーシェーカー(150 rpm)において37℃で一晩(16〜18時間)培養した。一晩経った培養物のアリコートを1/100希釈し、得られた培養物を上記と同様に2時間インキュベートし、その後IPTGを最終濃度1 mMになるまで加え、CS6の発現を誘導した。この培養物を次いで、同じ条件でさらに6時間インキュベートした。次いで細菌を回収し、(培地からのいかなる残留物も存在しないように)PBSによって2回洗浄し、OD600=16(およそ2×1010細菌/mlに相当)の密度になるようPBS中に再懸濁した。

数値は2回の定量の平均である。
CS6抗原を過剰発現する大腸菌株(ETEX 24)を500リットルの発酵槽に播種した。IPTGにより発現を誘導した後、発酵を8時間続けた。細菌を回収し、500 kDの限外ろ過器で洗浄し、最終的に20×109細菌/mlの濃度で分注した。最終濃度0.8%(w/v)となるまでフェノールを加え、この懸濁液を常に撹拌しながら20℃で40時間置いた。リン酸緩衝液中で500 kDの限外ろ過膜で懸濁液を洗浄し、4℃で保存した。
細菌株および培養
本研究において使用された細菌株は表1に挙げられている。チミンに対して栄養要求性である毒素非産生性C600-ΔthyA大腸菌株(N.I.A.Carlin and M. Lebens, unpublished)をワクチン候補株C600-CS6の構築のために使用した。CF-ETEC+CTBワクチンにおいてCS4+CS6発現株として以前に使用されたETEC株E11881/23を参照株として使用した。CS6の発現のために、CFA培地中で細菌を増殖させ(Evans DG, Evans DJ Jr., Clegg S, Pauley JA. Purification and characterization of the CFA/I antigen of enterotoxigenic Escherichia coli. Infect Immun 1979;25:738-48)、必要な場合はアンピシリン(100μg/ml)を添加した。
C600-CS6組換え株を構築するために、まずプラスミドpJT-CFA/I-ThyAを作製した。プラスミドpJT-CFA/I-Cm(Tobias J, Holmgren J, Hellman M, Nygren E, Lebens M, Svennerholm A-M. Over-expression of major colonization factors of enterotoxigenic Escherichia coli, alone or together, on non-toxigenic E.coli bacteria. Vaccine 2010;28:6977-84)をXhoIおよびAvrIIによって消化し、クロラムフェニコール耐性遺伝子(cat)を除去した。次いでプラスミドpNC-4(N.I.A. Carlin; unpublished)をPCR反応において使用し、thyA遺伝子を増幅させた(コレラ菌(Vibrio cholerae)由来)。フォワードプライマーP1(配列番号:1)はthyAの上流98 bpから開始する配列に対して相同であり、Eco31IおよびAvrIIのための制限部位を有しており、リバースプライマーP2(配列番号:2)はthyAの下流75 bpで終結する配列に対して相同であり、5'末端にEco31IおよびXhoIのための制限部位を有していた(表1)。PCR条件は以下の通りであった:95℃で5分間、次いで94℃で15秒間、58℃で30秒間、および72℃で50秒間を31サイクル、最終伸長を72℃で7分間。次いで、得られたthyAを含む1065 bpの断片をゲル抽出し、XhoIおよびAvrIIによって切断した。増幅され消化されたthyAを、消化されたpJT-CFA/I-Cmとライゲーションすることによって、pJT-CFA/I-ThyAが得られた(図1A)。次いでこのプラスミドを、エレクトロポレーションによって大腸菌C600-ΔthyAに導入し、組換え株(C600-ΔthyA/pJT-CFA/I-ThyA)を単離した。
CS6発現株をCFA培地中でロータリーシェーカー(150 rpm)において37℃で一晩(16〜18時間)培養した。一晩経った培養物のアリコートをCFA培地中に1/100希釈し、得られた培養物を上記と同様に2時間インキュベートした。組換え株の培養物に、最終濃度1 mMになるまでIPTGを加え、CS6の発現を誘導し、次いで同じ条件で培養物をさらに6時間インキュベートした。次いで細菌を回収し、OD600=0.8(およそ109 細菌/mlに相当)の密度になるようPBS中に再懸濁した。
記載されているように、ドットブロット法および阻害ELISA法を用いて、組換えC600-CS6株およびETEC参照E11881/23株によるCS6の発現レベルを定量化するために、CS6に対して特異的なモノクローナル抗体(MAb 2a:14)(Helander A, Grewal HM, Gaastra W, Svennerholm A-M. Detection and characterization of the coli surface antigen 6 of enterotoxigenic Escherichia coli strains by using monoclonal antibodies. J Clin Microbiol 1997;35:867-72)を使用した(Tobias J, Holmgren J, Hellman M, Nygren E, Lebens M, Svennerholm A-M. Over-expression of major colonization factors of enterotoxigenic Escherichia coli, alone or together, on non-toxigenic E.coli bacteria. Vaccine 2010;28:6977-84 および Tobias J, Lebens M, Bolin I, Wiklund G, Svennerholm A-M. Construction of non-toxic Escherichia coli and Vibrio Cholerae strains expressing high and immunogenic levels of enterotoxigenic E.coli colonization factor I fimbriae. Vaccine 2008;26:743-52)。
CS6発現株の不活化についてホルムアルデヒドとフェノールを試験し、比較した。最終濃度0.3%(w/v;0.1 M)または0.6%(0.2 M)のホルムアルデヒド、および最終濃度0.25%〜1.6%(w/v;0.026〜0.17 M)のフェノールをPBS中1010細菌/mlの密度の細菌培養物に加えた。懸濁液を60 rpmで振とうしながら37℃で2時間インキュベートし、次いで撹拌せずに4℃で3日間置いた。この細菌懸濁液を次いで遠心分離し、洗浄し、同容量のPBS中に再懸濁し、使用するまで4℃で保存した。双方の不活化法において、0.1 mlの各懸濁液を血液寒天培地上に拡げ、37℃で1週間までインキュベートし、増殖の欠如を調べた。経口免疫処置に使用する前に、阻害ELISA法によって不活化細菌上のCS6のレベルを調べた。
経口(胃内)免疫処置のために雌のBalb/CおよびC57 BI/6マウスの群(Charles River; 6〜8週齢;5個体/群)を使用した。全てのマウスに、0.3 mlの3%炭酸水素ナトリウム溶液中3×108個のC600-CS6株または参照株のいずれかのフェノール死滅化細菌(0.8%濃度のフェノールを使用して不活化)および7.5μgのCTの用量を、2日間隔で2回、ベビーフィーディングカテーテルによって胃内に投与し(初回の免疫処置)、その2週間後の第2回目の免疫処置においては同じ2回の免疫処置を2日間隔で行った。初回の免疫処置の前と最終免疫処置の2週後に採血を行い、その時に糞塊(FP)も採取し、以前に説明されているように抽出物を調製した(Nygren E, Holmgren J, Attridge SR. Murine antibody responses following systemic or mucosal immunization with viable or inactivated Vibrio cholerae. Vaccine 2008;26:6784-90)。さらに、マウスを屠殺した後の時点で、ヘパリン-PBS溶液で灌流して組織から血液を除去し、小腸組織を採取し、以前に説明されているように(Villavedra M, Carol H, Hjulstrom M, Holmgren J, Czerkinsky C. "PERFEXT": a direct method for quantitative assessment of cytokine production in vivo at the local level. Res Immunol 1997;148:257-66)、2%(w/v)サポニン-PBS溶液で抽出した(Perfext法)。
以前に説明されているようにELISA法によって、CS6に対するIgG+IgMおよびIgAの抗体価を、血清、糞便および腸の抽出物において定量した(Rudin A, Svennerholm A-M. Colonization factor antigens(CFAs) of enterotoxigenic Escherichia coli can prime and boost immune responses against heterologous CFAs. Microb Pathog 1994;16:131-9)。ELISA法においてコーティング抗原として使用するためのCS6(最終濃度0.7μg/ml)は、以前に説明されているTOP10-CS6過剰発現株から(Tobias J, Lebens M, Kallgard S, Nicklasson M, Svennerholm A-M. Vaccine 2008;26:5373-80)、一連の硫酸アンモニウム沈殿とゲルろ過によって精製した。個々のマウスからの血清は低結合性マイクロタイタープレート(Greiner)を用いて試験し、サンプルは最初に1/100希釈してから3倍段階希釈した。糞塊抽出物および小腸組織の抽出物は、高結合性マイクロタイタープレート(Greiner)において、開始希釈率1/3から3倍段階希釈で試験した。バックグラウンド値よりも0.4高いA450吸光度を生じたサンプルの希釈率の逆数として抗体価を算出した。説明されているように、ELISA法によって糞便および腸の抽出物サンプルにおける総IgAも測定し(Nygren E, Holmgren J, Attridge SR. Vaccine 2008;26:6784-90)、抗原特異的IgA抗体の値を総IgAのμg当たりのIgA力価単位として表した。
ELISA法による全ての実験は別々の機会で少なくとも2回行われた。統計解析はスチューデントt検定によって行い、P<0.05(両側検定)を有意差として見なした。
Claims (15)
- ETEC CS6抗原の提示が少なくとも100%増大するように、該抗原を発現する細胞を、0.6〜2.2重量%のフェノールを含む水溶液と接触させる段階を含む、細胞表面上におけるETEC CS6抗原の提示を増大させるための方法。
- 前記抗原の提示が少なくとも200%、好ましくは少なくとも300%増大する、請求項1記載の方法。
- 前記接触させる段階の継続時間が1〜72時間である、前述の請求項のいずれか一項記載の方法。
- 前記接触させる段階の継続時間が1.5〜42時間である、請求項3記載の方法。
- 前記接触させる段階中の温度が18〜42℃である、前述の請求項のいずれか一項記載の方法。
- 前記接触させる段階中の温度が18〜38℃、好ましくは18〜22℃または36〜38℃である、請求項5記載の方法。
- フェノール濃度が0.8〜2.0重量%、好ましくは1.0〜2.0重量%である、前述の請求項のいずれか一項記載の方法。
- 前記細胞が大腸菌(Escherichia coli)細胞を含む、前述の請求項のいずれか一項記載の方法。
- 前記抗原が、組換えにより前記細胞によって過剰発現される、前述の請求項のいずれか一項記載の方法。
- 阻害ELISA法またはドットブロット法によって、好ましくは阻害ELISA法によって、前記細胞による前記抗原の提示を、処理されていないが他の点では同等な細胞によるCS6抗原の提示と比較する段階をさらに含む、前述の請求項のいずれか一項記載の方法。
- 前記細胞の少なくとも107倍の不活化が、CS6抗原の提示の増大と同時に生じるように、フェノール濃度、接触温度、および接触時間が選択される、前述の請求項のいずれか一項記載の方法を含む、CS6発現ETECに対する免疫化のための死滅全菌体ワクチンの製造のための方法。
- フェノール濃度が0.6〜2.0重量%、接触時間が6〜72時間、および接触温度が18〜22℃である、請求項11記載の方法。
- フェノール濃度が0.75〜0.85重量%、接触時間が40±2時間、および接触温度が20±1℃である、請求項12記載の方法。
- 前述の請求項のいずれか一項記載の方法によって得られる細胞。
- 請求項14記載の細胞を含む、CS6発現ETECに対する免疫化のためのワクチン。
