Abstract
Description
(i)第一分離ステップにおいて、溶離剤として水性有機溶媒を含む複数の連結したクロマトグラフィーカラムを有する擬似または実移動床式クロマトグラフィー装置で供給混合液を精製して、中間生成物を得るステップと、
(ii)第二分離ステップにおいて、溶離剤として水性有機溶媒を含む複数の連結したクロマトグラフィーカラムを有する擬似または実移動床式クロマトグラフィー装置を用いて、(i)で得られた中間生成物を精製して、PUFA生成物を得るステップと
を含み、
(a)第一および第二分離ステップが同じクロマトグラフィー装置で順次行われ、中間生成物が第一と第二の分離ステップの間で回収され、クロマトグラフィー装置のプロセス条件が、各分離ステップで供給混合液の異なる成分からPUFA生成物が分離されるように、第一と第二の分離ステップの間で調整される、または
(b)第一および第二分離ステップが、別々の第一および第二クロマトグラフィー装置でそれぞれ行われ、第一分離ステップから得られた中間生成物が第二クロマトグラフィー装置に導入され、各分離ステップで供給混合液の異なる成分からPUFA生成物が分離される、方法を提供する。
(b)第一分離ステップで使用した装置からの抽残液流の一部は、第一分離ステップで使用した装置に再循環され、かつ/または
(c)第二分離ステップで使用した装置からの抽出液流の一部は、第二分離ステップで使用した装置に再循環され、かつ/または
(d)第二分離ステップで使用した装置からの抽残液流の一部は、第二分離ステップで使用した装置に再循環される。
(b)第一分離ステップで使用した装置からの抽残液流の一部は、第一分離ステップで使用した装置に再循環され、
(c)第二分離ステップで使用した装置からの抽出液流の一部は、第二分離ステップで使用した装置に再循環され、
(d)第二分離ステップで使用した装置からの抽残液流の一部は、第二分離ステップで使用した装置に再循環される。
(2)より極性が低い成分とともにPUFA生成物を含む中間生成物は、第一分離ステップで抽出液流として収集され、PUFA生成物は、第二分離ステップで抽残液流として収集される。
実施例
SMBによって生成された2種のPUFA生成物に存在する環境汚染物質の量を蒸留によって調製された同様の油と比較することを目的として、実験を行った。これらの油の汚染物質プロファイルを以下の表1に示す。
蒸留によって調製された同等の油と比較して、SMBによって調製された油に存在する異性体不純物の量を決定することを目的として、実験を行った。
SMBによって生成された、EPAが豊富な2種の生成物を、蒸留によって生成されたEPAが豊富な油と比較した。それらの構成成分であるPUFAの重量%分析を以下に示す。
SMBによって生成された、EPA/DHAが豊富な生成物を、蒸留によって生成されたEPA/DHAが豊富な油と比較した。それらの構成成分であるPUFAの重量%分析を以下に示す。

Claims (17)
- 供給混合液から多価不飽和脂肪酸(PUFA)生成物を回収するためのクロマトグラフィー分離方法であって、
(i)第一分離ステップにおいて、溶離剤として水性有機溶媒を含む複数の連結したクロマトグラフィーカラムを有する擬似または実移動床式クロマトグラフィー装置で供給混合液を精製して、中間生成物を得るステップと、
(ii)第二分離ステップにおいて、溶離剤として水性有機溶媒を含む複数の連結したクロマトグラフィーカラムを有する擬似または実移動床式クロマトグラフィー装置を用いて、(i)で得られた中間生成物を精製して、PUFA生成物を得るステップと
を含み、
(a)第一および第二分離ステップが同じクロマトグラフィー装置で順次行われ、中間生成物が第一と第二の分離ステップの間で回収され、クロマトグラフィー装置のプロセス条件が、各分離ステップで供給混合液の異なる成分からPUFA生成物が分離されるように、第一と第二の分離ステップの間で調整される、または
(b)第一および第二分離ステップが、別々の第一および第二クロマトグラフィー装置でそれぞれ行われ、第一分離ステップから得られた中間生成物が第二クロマトグラフィー装置に導入され、各分離ステップで供給混合液の異なる成分からPUFA生成物が分離される、方法。 - 各装置が前記複数の連結したクロマトグラフィーカラムから液体が収集され得る抽出液流および抽残液流を有する、請求項1に記載の方法。
- 第一分離ステップで得られた中間生成物が、供給混合液と比較してPUFA生成物が豊富である、請求項1または2に記載の方法。
- (a)中間生成物が第一分離ステップで抽残液流として収集され、PUFA生成物が第二分離ステップで抽出液流として収集される、または
(b)中間生成物が第一分離ステップで抽出液流として収集され、PUFA生成物が第二分離ステップで抽残液流として収集される、前記請求項のいずれか一項に記載の方法。 - 第一分離ステップで、より極性が低い供給混合液成分からPUFA生成物が分離され、第二分離ステップで、より極性が高い供給混合液成分からPUFA生成物が分離される、前記請求項のいずれか一項に記載の方法。
- PUFA生成物が少なくとも1つのω−3PUFAを含む、前記請求項のいずれか一項に記載の方法。
- PUFA生成物がEPAおよび/またはDHAを含む、請求項6に記載の方法。
- 溶離剤が、水とアルコール、エーテル、エステル、ケトンまたはニトリルとの混合液である、前記請求項のいずれか一項に記載の方法。
- 溶離剤が水とメタノールの混合液である、請求項8に記載の方法。
- 供給混合液が魚油または魚油由来の供給原料であり、PUFA生成物がEPAまたはEPAエチルエステルであり、純度90%を超える、好ましくは純度95%を超える、より好ましくは純度97%を超える純度でPUFA生成物が生成される、前記請求項のいずれか一項に記載の方法。
- (a)第一分離ステップで使用した装置からの抽出液流の一部が、第一分離ステップで使用した装置に再循環され、かつ/または
(b)第一分離ステップで使用した装置からの抽残液流の一部が、第一分離ステップで使用した装置に再循環され、かつ/または、
(c)第二分離ステップで使用した装置からの抽出液流の一部が、第二分離ステップで使用した装置に再循環され、かつ/または
(d)第二分離ステップで使用した装置からの抽残液流の一部が、第二分離ステップで使用した装置に再循環される、前記請求項のいずれか一項に記載の方法。 - 各分離ステップで使用する水性有機溶媒の溶離剤が、異なる水:有機溶媒比率を有する、前記請求項のいずれか一項に記載の方法。
- 各分離ステップで使用する水:有機溶媒比率が、各分離ステップで供給混合液の異なる成分からPUFA生成物を分離できるように調整される、前記請求項のいずれか一項に記載の方法。
- 第一分離ステップの溶離剤の水:有機溶媒比率が、第二分離ステップの溶離剤の水:有機溶媒比率より低い、前記請求項のいずれか一項に記載の方法。
- 第一分離ステップの溶離剤の水:有機溶媒比率が0.5:99.5から1.5:98.5容積部であり、第二分離ステップの溶離剤の水:有機溶媒比率が4.5:95:5から5.5:94.5容積部である、前記請求項のいずれか一項に記載の方法。
- 第一および第二分離ステップの溶離剤の水:有機溶媒比率が、第一および第二分離ステップで使用する装置の1個または複数のカラムに水および/または有機溶媒を導入することによって制御される、前記請求項のいずれか一項に記載の方法。
- 前記請求項のいずれか一項に定義されたクロマトグラフィー装置を制御するためのコンピュータープログラムであって、実行時に、前記請求項のいずれか一項に記載の方法を実施するように装置に指示するコード手段を含む、コンピュータープログラム。
