Abstract
Description
(a) オレイン酸塩を低級アルコール溶媒に約35℃から65℃の温度で溶解するステップ;
(b) ステップ(a)の溶液に非極性溶媒を添加するステップ;
(c) ステップ(b)の溶液に、低級アルコールに溶解した鉄塩を添加するステップ;
(d) ステップ(c)の溶液を、約50℃で少なくとも5分間撹拌するステップ;
(e) ステップ(d)の反応混合物を約15℃から30℃へ冷却するステップ;
(f) 場合によりステップ(e)の反応混合物を濾過するステップ;
(g) 前記低級アルコール相から前記非極性溶媒相を分離するステップ;
(h) 前記非極性溶媒相を洗浄し乾燥するステップ;
(i) ステップ(h)の非極性溶媒から揮発物を蒸発させて除去するステップ;及び
(j) ステップ(i)の生成物を極性溶媒と混合して、固体鉄オレイン酸錯体を得る、ステップを含む。
(k) 前記固体鉄オレイン酸錯体を濾過で単離するステップ;
(l) ステップ(j)又は(k)の固体鉄オレイン酸錯体を極性溶媒で洗浄するステップ;
(m) ステップ(l)の固体鉄オレイン酸錯体を非極性溶媒に溶解するステップ;
(n) ステップ(m)の固体鉄オレイン酸錯体を濾過するステップ;
(o) ステップ(n)の固体鉄オレイン酸錯体に過剰量の極性溶媒を添加するステップ;
(p) ステップ(o)の懸濁液を約1から10時間撹拌するステップ;
(q) 前記鉄オレイン酸錯体を濾過するステップ;
(r) ステップ(q)の鉄オレイン酸錯体を極性溶媒で洗浄するステップ;及び
(s) ステップ(r)のオレイン酸錯体を乾燥して、粉末固体鉄オレイン酸錯体を得るステップ、を実施する。
(a) オレイン酸及び前記定義された鉄オレイン酸錯体又は前記本発明の方法により得られる鉄オレイン酸錯体、及び場合によりオレイルアミンを第1の有機溶媒中に懸濁させるステップ;
(b) 前記懸濁物を既定の速度で最大340℃から500℃へ温度を上げるステップ;
(c) ステップ(b)の最高温度で前記懸濁物を約0.5から6時間熟成するステップ;
(d) 前記懸濁物を冷却するステップ;
(e) 第2の有機溶媒を添加するステップ;
(f) 非溶媒を添加してナノ粒子を沈殿させ、過剰の溶媒を除去するステップ;
(g) 前記ナノ粒子を前記第2の有機溶媒中に分散させるステップ;
(h) ステップ(g)の分散物をポリマーの溶液と混合するステップ;及び
(i) 場合により、前記第2の有機溶媒を除去する、ステップを含む。
(j) ステップ(i)で得られるナノ粒子又はナノ粒子溶液を精製するステップ;
(k) ステップ(i)又は(j)で得られるナノ粒子又はナノ粒子溶液を酸化剤又は還元剤で処理するステップ;
(l) ステップ(i)又は(j)で得られるナノ粒子の表面を、前記コーティングを除去、置換又は変更することで変性するステップ;
(m) ステップ(i)から(l)で得られるナノ粒子を、ミセル、リポソーム、ポリマーソーム、血液細胞、ポリマーカプセル、デンドリマー、ポリマー又はヒドロゲルで、カプセル化又はクラスタ化するステップ;及び
(n) ステップ(i)から(m)で得られるナノ粒子をターゲットリガンドで修飾するステップ、を含む。

(j) ステップ(i)の生成物を極性溶媒と混合して、固体鉄オレイン酸錯体を得る、ステップを含む。
(k) 固体鉄オレイン酸錯体を濾過して単離するステップ;
(l) ステップ(j)又は(k)の固体鉄オレイン酸錯体を極性溶媒で洗浄するステップ;
(m) ステップ(l)の固体鉄オレイン酸錯体を非極性溶媒に溶解するステップ;
(n) ステップ(m)の固体鉄オレイン酸錯体を濾過するステップ;
(o) ステップ(n)の固体鉄オレイン酸錯体に過剰量の極性溶媒を添加するステップ;
(p) ステップ(o)の懸濁物を約1から10時間撹拌するステップ;
(q) 鉄オレイン酸錯体を濾過するステップ;
(r) ステップ(q)の鉄オレイン酸錯体を極性溶媒で洗浄するステップ;及び
(s) ステップ(r)の鉄オレイン酸錯体を乾燥して、分かつ固体鉄オレイン酸錯体とするステップ、を含む。
好ましくは、前記オレイン酸イオンは図1に示す構造を持つ。
(a) オレイン酸と、前記の鉄オレイン酸錯体、又は本発明の方法で得られ得る又は得られた鉄オレイン酸錯体、場合によりオレイルアミンとを第1の有機溶媒中に懸濁させるステップ;
(b) 前記懸濁物の温度を所定の速度で、最大340℃から500℃まで温度を上げるステップ;
(c) ステップ(b)の最大温度で前記懸濁物を約0.5から6時間熟成させるステップ;
(d) 懸濁物を冷却するステップ;
(e) 第2の有機溶媒を添加するステップ;
(f) 非溶媒を添加してナノ粒子を沈殿させ過剰の溶媒を除去するステップ;
(g) 前記第2の有機溶媒中に前記ナノ粒子を分散させるステップ;
(h) ステップ(g)の分散物をポリマーの溶液と混合するステップ;及び
(i) 場合により前記第2の有機溶媒を除去するステップ、を含む。
(j) ステップ(i)で得られたナノ粒子又はナノ粒子溶液を精製するステップ;
(k) ステップ(i)又はステップ(j)で得られたナノ粒子又はナノ粒子溶液を酸化剤又は還元剤で処理するステップ;
(l) ステップ(i)、(j)又は(k)で得られたナノ粒子の表面を、前記コーティングを除去、交換又は変更することで変性するステップ;
(m) ステップ(i)から(l)で得られたナノ粒子を、ミセル、リポソーム、ポリマーソーム、血液細胞、ポリマーカプセル、デンドリマー、ポリマー又はヒドロゲルで、カプセル化又はクラスタ化するステップ;及び
(n) ステップ(i)から(m)で得られたナノ粒子をターゲットリガンドで修飾するステップ、により、処理、変性又は変更され得る。
実施例1−オレイン酸鉄の合成(サンプル1)
オレイン酸ナトリウム(20.27g、66.6mmol)をMeOH(180ml)中に50℃で窒素雰囲気下で溶解した。ヘキサン(720ml)を前記透明無色のメタノール溶液に、激しく撹拌しながら小分けして添加した。FeCl3・6H2O(6.0g、22.2mmol)をMeOH(15ml)中に溶解し、これを、窒素雰囲気下、ゆっくりと前記暖められたオレイン酸塩に滴下漏斗から添加した。反応混合物の色は黄色に変化し、後オレンジ色になり、一方塩化ナトリウムの白色沈澱が生成した。FeCl3・6H2O溶液の添加が終了後、反応混合物は50℃で約1時間撹拌された。混合物を室温に冷却した後、濾過し、分液漏斗に移した。オレンジ−赤色MeOH相を、黄−緑色のヘキサン相と分離した。生成物をその後MeOHで、MeOH相が無色になるまで洗浄した(4x100ml)。黄−緑色ヘキサン溶液は、MgSO4で乾燥し、濾過し、溶媒を蒸発させて、暗赤色粘調オイルが残った。このオイルをアセトン中(500ml)で一夜撹拌して固化させた。得られた赤茶色固体生成物をオレンジ色溶液から濾過し、アセトンで洗浄した(50ml)。この固体をヘキサン(60ml)に溶解させ、得られた溶液をシリンジフィルタ(0.45μmポアサイズ)に通して濾過し、滴下漏斗に移した。得られた暗赤色溶液を、激しい撹拌下でアセトン(500ml)中に滴下した。小さい赤灰色塊沈澱が形成され、この凝集物は一夜撹拌することで脱凝集化した。得られた粉末生成物を、オレンジ赤色アセトン溶液から濾過し、残った大きい生成物凝集物はスパチュラで細かくした。赤茶色粉末を合わせてアセトンで洗浄した(3x50ml)。生成物を、最後に、オイル・ポンプで真空乾燥し、シリカ上で貯蔵した。
鉄オレイン酸(0.100g、0.12mmol、0.24mmol(Fe))、オレイン酸(0.435g、1.54mmol)及びイコサン(0.60g)を、リフラックスコンデンサーと、反応混合物に漬けた温度センサーを持つ三つ首フラスコに混合した。混合物を、3.3℃/分で温度360℃まで加熱し、その温度で2時間保持した。50℃に冷却後、ヘキサン(10ml)を添加し、均一溶液とした。次に、アセトン(20ml)を加え、沈澱させて固体を形成させ、これを遠心分離器(4900rpm、30分)で集め、溶媒を捨てた。洗浄のため、集めた固体物質をヘキサン(5ml)に再懸濁させ、上記のようにアセトン(10ml)を添加して沈殿させ、遠心分離し集めた。洗浄手順をもう一度繰り返し、集めた固体をヘキサンに懸濁させて鉄酸化物ナノ粒子の安定な黒色良い駅を生成した。続いて、実施例2で得られた鉄酸化物ナノ粒子を磁気粒子スペクトル(MPS)で特徴付けた。
鉄オレイン酸(0.100g、0.12mmol、0.24mmol (Fe))、オレイン酸(0.684g、2.42mmol)及びイコサン(0.60g)を混合し、実施例2で説明したように処理した。鉄酸化物ナノ粒子が実施例2で記載したように得られ、磁気粒子スペクトル(MPS)により特徴付けられた。
鉄オレイン酸(0.100g、0.12mmol、0.24mmol(Fe))、オレイン酸(0.560g、1.98mmol)及びイコサン(0.60g)を混合し、実施例2と同様に処理した。実施例2と同様に鉄酸化物が得られ、磁気粒子スペクトル(MPS)により特徴付けられた。
Claims (15)
- 鉄オレイン酸錯体を形成する方法であり、前記方法は次のステップ:
(a) オレイン酸塩を低級アルコール溶媒に約35℃から65℃の温度で溶解するステップ;
(b) 前記ステップ(a)の溶液に非極性溶媒を加えるステップ;
(c) ステップ(b)の溶液に低級アルコールに溶解した鉄塩を加えるステップ;
(d) ステップ(c)の溶液を約50℃の温度で少なくとも5分間撹拌するステップ;
(e) ステップ(d)の反応混合物を約15℃から30℃に冷却するステップ;
(f) 場合により、ステップ(e)の反応混合物を濾過するステップ;
(g) 前記低級アルコール相から前記非極性溶媒相を分離するステップ;
(h) 前記非極性溶媒相を洗浄して乾燥するステップ;
(i) ステップ(h)の前記非極性溶媒相から揮発性物を蒸発させて除去するステップ;及び
(j) ステップ(i)の生成物を極性溶媒と混合して個体鉄オレイン酸錯体を得るステップを、含む方法。 - 請求項に記載の方法であり、ステップ(a)の温度が約50℃である、方法。
- 請求項1又は2のいずれか一項に記載の方法であり、前記オレイン酸塩がオレイン酸ナトリウムであり、及び/又は前記低級アルコール溶媒がメタノールであり、及び/又は前記非極性溶媒がヘキサンであり、及び/又は前記極性溶媒がアセトンであり、及び/又は前記鉄塩が鉄塩化物、好ましくは鉄(III)塩化物(FeCl3)である、方法。
- 請求項3に記載の方法であり、過剰量のオレイン酸ナトリウムが使用される、方法。
- 請求項4に記載の方法であり、オレイン酸ナトリウム:FeCl3のモル比は3:1である、方法。
- 請求項1乃至5のいずれか一項に記載の方法であり、混合ステップ(j)が約1から10時間実施される、方法。
- 請求項1乃至6のいずれか一項に記載の方法であり、追加のステップ、
(k) 前記固体鉄オレイン酸錯体を濾過して単離するステップ;
(l) ステップ(j)又は(k)の固体鉄オレイン酸錯体を極性溶媒で洗浄するステップ;
(m) ステップ(l)の固体鉄オレイン酸錯体を非極性溶媒中に溶解するステップ;
(n) ステップ(m)の固体鉄オレイン酸錯体を濾過するステップ;
(o) ステップ(n)の固体鉄オレイン酸錯体に過剰量の極性溶媒を添加するステップ;
(p) ステップ(o)の懸濁物を約1から10時間撹拌するステップ;
(q) 前記鉄オレイン酸錯体を濾過するステップ;
(r) ステップ(q)の鉄オレイン酸錯体を極性溶媒で洗浄するステップ;及び
(s) ステップ(r)の鉄オレイン酸錯体を乾燥して、粉末状固体鉄オレイン酸錯体を得るステップ、を実施する方法。 - 請求項7に記載の方法であり、ステップ(o)で少なくとも4:1の過剰量のアセトンが添加される、方法。
- 請求項1乃至8のいずれか一項に記載の方法で得られ得る鉄オレイン酸錯体。
- 請求項10に記載の方法であり、前記補助リガンドL1、L2、L3及びL4は、お互いに独立して、アセトン、メタノール、エタノール、水、テトラヒドロフラン、イミダゾール、メチルイミダゾール、ピリジン、ホルムアミド、ジメチルホルムアミド、ピロリドン、1−メチル−2−ピロリドン、ヒドロキシド、フロリド、クロリド、ブロミド、ヨード、スルフェート、ビスルフェート、ホスフェート、ビホスフェート、ナイトレート、スルフィド、ビスルフィド、オキザレート、ラクテート、シアニド、シアネート、イソシアネート、チオシアネート、イソチオシアネート、アセチルアセトネート、カルボネート、ビカルボネート、アジド、ベンゾエート、アルキレート、メタクリレート、スルファイト、ビスルファイト、メトキシド、エトキシド、シクロヘキサンスルフォネート、メタンスルフォネート、エタンスルフォネート、プロパンスルフォネート、ペンタンスルフォネート、ヘキサンスルフォネート、オクタンスルフォネート、デカンスルフォネート、ドデカンスルフォネート、オクタデカンスルフォネート、シトレート、タータレート、ボレート、水素化ボレート、 二水素化ボレート、ナイトライト、パーボレート、パーオキシド、チオスルフェート、メタチオネート、アセテート、プロピオネート、ブチレート、ペンタノエート、ヘキサノエート、ヘプタノエート、オクタノエート、デカノエート、ドデカノエート、ペンタデカノエート、ヘキサデカノエート、オクタデカノエート又はオレートである、方法。
- 請求項10又は11に記載の鉄オレイン酸錯体であり、前記錯体が分子式Fe2O(oa)2(OH)2(OC(CH3)2)2を持つ、鉄オレイン酸錯体。
- 請求項9乃至12のいずれか一項に記載の鉄オレイン酸錯体、又は請求項1乃至8のいずれか一項に記載の方法により得られ得る鉄オレイン酸錯体を、ナノ粒子の製造のための前駆体としての使用。
- 鉄オレイン酸ナノ粒子を形成する方法であり、前記方法は次のステップ:
(a) オレイン酸と、請求項9乃至12のいずれか一項に記載の鉄オレイン酸錯体又は請求項1乃至8のいずれか一項に記載の方法で得られ得る鉄オレイン酸錯体と、及び場合によりオレイルアミンを、第1の有機溶媒中に懸濁させるステップ;
(b) 前記懸濁物を、所定の速度で、最大340℃から500℃まで温度を上げるステップ;
(c) ステップ(b)の前記懸濁物を前記最大温度で約0.5から6時間熟成させるステップ;
(d) 前記懸濁物を冷却するステップ;
(e) 第2の有機溶媒を添加するステップ;
(f) 非溶媒を添加することでナノ粒子を沈殿させ過剰溶媒を除去するステップ;
(g) 前記第2の有機溶媒中に前記ナノ粒子を分散させるステップ;
(h) ステップ(g)の分散物をポリマー溶液と混合するステップ;及び
(i) 場合により、前記第2の有機溶媒を除去するステップ、を含む方法。 - 請求項14に記載の方法であり、1又はそれ以上の次の追加のステップ、
(j) ステップ(i)で得られ得るナノ粒子又はナノ粒子溶液を精製するステップ;
(k) ステップ(i)又は(j)で得られ得るナノ粒子又はナノ粒子溶液を酸化剤又は還元剤で処理するステップ;
(l) ステップ(i)又は(j)で得られ得るナノ粒子の表面を、前記コーティングを除去、置換又は変更することで変性するステップ;
(m) ステップ(i)から(l)で得られ得るナノ粒子を、ミセル、リポソーム、ポリマーソーム、血液細胞、ポリマーカプセル、デンドリマー、ポリマー又はヒドロゲルなどのキャリアでカプセル化又はクラスタ化するステップ;及び
(n) ステップ(i)から(m)で得られ得るナノ粒子をターゲットリガンドで修飾するステップ、を含む方法。
