DNA小分子(Dbait)の導入は、損傷した染色体のDNA修復を阻害し、腫瘍、特に耐性腫瘍における放射線療法又は化学療法の効率を高めるための効率的方法を提供する。しかしながら、Dbait分子の増感活性は、腫瘍細胞内におけるそれらの送達効率に依存する。
1)coDbaitとクロロキンの組み合わせは、もしあったとしても、低い毒性をin vivoで示す。それにより、治療指数(有効用量/毒性用量の比率)を、ほぼ1(Dbait/PEIの場合)から20超(coDbaitの場合)に改善できる;毒性がないことが、マウス、ラット、ウサギ及びサルにおける静脈内注射、皮下注射後に、そして脳内注射後にさえ認められた;
2)coDbaitとクロロキンの組み合わせは、DNA−PK(Dbaitの主なターゲット)の持続的な遅延活性化をもたらし、治療効果を延長させる。より具体的には、活性又は効果の増加は、この期間を通じて認められる;
3)驚くべきことに、coDbaitは、Dbait/PEIと比較して、腫瘍/組織においてよく拡散される。
4)本発明者らは、クロロキンがcoDbaitの細胞取り込みを増加させるのに対して、コレステロールをsiRNA分子にコンジュゲーションさせた場合、この効果はあまりはっきりしないことを初めて観察した。

本発明の複合調製物において使用される各組み合わせパートナーの有効投与量は、使用される特定の化合物又は医薬組成物、投与方法、処置中の病状、処置中の病状の重症度に応じて変わり得る。従って、本発明の複合調製物の投与量レジメンは、投与経路及び患者の状態を含む様々な要因に従って選択される。通常の技術を有する医師、臨床医又は獣医は、病状の進行を予防、対抗又は停止するのに必要な単一有効成分の有効量を容易に決定及び規定できる。毒性を伴わずに効果をもたらす範囲内の有効成分濃度を達成する最適精度は、ターゲット部位に対する有効成分の有効性の動態に基づくレジメンを必要とする。
ポリエチレンイミン(PEI)と複合体化したか、又はコレステロールに結合した短いDNA(Dbait)の分布及び活性のマルチスケール比較。
Dbait/ベクター複合体及び細胞取り込みの特性評価
PEIは、DNA、オリゴヌクレオチド及びRNAと非共有高分子電解質間複合体を形成できることが示された。長いPEI鎖は、遺伝子トランスフェクションにおいて非常に効果的であるが、それ以上に細胞毒性である。本発明者らは、Dbaitを有するいくつかのPEI粒子ポリプレックスを試験し、それらの活性を、コレステロールに共有結合した改変Dbait(coDbaitと称する)と比較した。coDbaitは、コレステロールの脂肪鎖に共有結合したcoDbait分子であり、これは、追加のベクターなしで使用した。試験した各ベクターに関して、本発明者らの主な目標は、最高Dbait濃度で最も均一な粒径分布を有する製剤を開発することであった。動的レーザー光散乱(DLS)によって、粒子の直径及び表面電荷を測定した。多モード分析を使用して、本発明者らは、25Kdの平均サイズを有する分岐状PEI(bPEI25K)、及び、22Kdサイズを有する直鎖状PEI(PEI22K)又は11Kdサイズを有する直鎖状PEI(PEI11K)が、類似特性を有するDbaitとの複合体を形成することを見出した(表1)。
細胞培養物のDbait取り込み及び活性を分析することによって、全生物における薬物拡散、細胞取り込み及び活性について結論を出すことはできない。本発明者らは、Dbait−cy3をネイキッドで又はアジュバントと共に、1000細胞期(1K期)のゼブラフィッシュ胚(Kimmel et al., 1995, Dev Dyn 203:253-310)の細胞間隙に注射することによって、この組織を評価した。このプロトコールによって、ゼブラフィッシュ初期胚の急速に分裂する細胞で、細胞及び細胞内レベルのDbait−cy3分布並びにその活性を、共焦点顕微鏡法によりin vivo観察できた。1K期胚の動物極に注射したネイキッドなDbait−cy3は、全胚盤葉の全体に急速に拡散し、注射後15分までにもはや検出されなかった。Lutrolの添加は、Dbaitを注射地点付近の細胞外空間に保持することを可能にしたが、細胞取り込みを促進しなかった。Superfect又はPEIの存在下において、多数の蛍光パッチが細胞内に観察されたが、これは、効率的な細胞取り込みを示している。coDbait cy3は、パッチ状の細胞内蛍光と共に、原形質膜が持続的かつ濃く染色されるという別の種類の挙動を示した。注射前にCQと共に胚をインキュベーションすると、大きなcoDbait蛍光パッチが、拡散した細胞内分布に変わった。
細胞培養、ゼブラフィッシュ胚及びマウスのデータの整合性を評価するために、本発明者らは、Dbait/PEI1K、Dbait/PEI22K、Dbait/bPEI25K及びcoDbaitの反復投与に対するヌードマウスの皮膚の耐性を分析した。3日間の毎日皮下(SC)注射後に、異なる製剤化Dbaitの毒性を分析した。すべてのDbait/PEIが、注射による高い毒性を示し、これは、3.75mg/kgでは許容されたが、5mg/kgから、局所壊死及び虚血に関連する局所炎症(これは、処置の停止により急速に消失した)をトリガーした。静脈内(IV)注射の毒性は、同様の結果を示した:Dbait/PEI静脈内注射は、3mg/kgで致死的であり、これは、おそらく血液の詰まりによって注射中に死亡するためであった。潅流(0.4μL/mn)によって徐々に注射すると、6mg/kg Dbait/PEI(6nmole/注射)まで許容値が上昇したが、これは、IV毒性のほとんどが、ボーラス注射部位の局所濃度に起因することを裏付けている。CQの有無にかかわらず、coDbaitは、使用した経路(SC、IVボーラス又はIV潅流)が何であれ、すべての試験用量(最大800mg/kg/注射;800nmole/注射)でいかなる毒性も示さなかった。
製剤化Dbaitの抗腫瘍効果を、放射線療法と組み合わせて、SK28異種移植ヒトメラノーマで試験した。各放射線照射の5時間前に、腫瘍内注射を使用して、Dbait/ベクター複合体を投与した。
本研究において、本発明者らは、投与プロトコール及び製剤の開発を導くためのアッセイのセットを使用した。これらのアッセイにより、マウスにおける前臨床アッセイを実施する前に、Dbaitの異なる製剤を比較できた。Dbait細胞取り込み(これは、抗腫瘍薬物効果の必要条件工程である)の効率を評価するため、並びに、動物における前臨床試験に最も適切なプロトコール及び製剤を選択するために、細胞及びゼブラフィッシュ胚のアッセイを使用した。ゼブラフィッシュ胚における全体的な毒性は、マウスの皮膚又は全身注射後の毒性に相関しなかった。特に、ゼブラフィッシュ胚におけるcoDbaitの高い毒性は、薬物と接触する細胞のほとんどがおそらく死亡したことを示したのに対して、マウスの皮膚は、coDbait高用量の注射に対していかなる反応も示さなかった。この違いは、ゼブラフィッシュ初期胚における毒性が、健常組織感受性よりもむしろ腫瘍感受性の指標であることを示唆している。実際、Dbait分子は、正常皮膚ではなく腫瘍において特異的に毒性であることが示されている(Quanz et al., 2009、前掲)。細胞培養において同等のDNA−PKcs活性化をトリガーしたDbait/PEI(5μM)及びcoDbait(50μM)+CQは、ゼブラフィッシュ胚に対する毒性効果が類似しており(図3D)、マウス腫瘍に対して有意な抗腫瘍活性を示した(表2、図5及び6)。この観察結果は、腫瘍細胞と特性(分裂指数並びに生化学的特性及び表現型形質を含む)が共通しているゼブラフィッシュ胚細胞の抗腫瘍活性に対する感受性と一致している。この仮説に一致して、本発明者らは、最近、1〜16細胞期のゼブラフィッシュの割球にネイキッドなDbaitを直接的に細胞内注射することによって、Dbaitの抗増殖活性を実証した。
Dbait及び粒子形成
前述のように(Quanz et al., 2009、前掲)、Eurogentec (Seraing, Belgium)又はAgilent Technologies Nucleic Acid Solution Division (Boulder, USA)の自動化固相オリゴヌクレオチド合成によって、Dbait及びcoDbait分子を得た。変性逆相HPLC及び/又はHPLC−IEXによって、それらを精製した。いくつかのDbait誘導体を、蛍光Cy3(λ励起=540nm;λ放出=560nm)又はCy5.5(λ励起=Xnm;λ放出=Xnm)でラベル化した。直鎖状PEI(11kDa及び22kDa)は、Polyplus-Transfection (Illkirch, France)製であり、300mM窒素濃度の即時使用可能な溶液として提供された。SIGMA-Aldrich (Saint Quentin, France)から、分岐状bPEI25kdを購入した。In Cell Art (Nantes, France)から、Lutrolを購入した。Dbait及びPEI溶液(ストックPEI)を、10%スクロース又は150mM NaCl(in vitroトランスフェクション実験用)中で希釈して、様々な比率のベクター/Dbaitを得た。PEIに関してはアミン窒素の数、及び、Dbaitに関してはリン酸塩の数によって、PEI/Dbaitの比率(又はN/Pの比率)を決定した。典型的には、0.6mg/mL及びN/P6の複合体300μLに関して、Dbait(180μg、0.54μmolのリン酸塩)及び所望量のポリマー溶液(11,4μLのPEIストック溶液は、0.3μmolのアミン窒素を含む)を、150μL(10%スクロース)にそれぞれ希釈した。製造業者(Qiagen, Courtaboeuf, France)に従って、10μl Superfect/μgDNAの比率でSuperfect/Dbait粒子を調製した。アガロースゲル電気泳動法によって、ベクター/Dbaitの錯体を分析した。サンプル(18μL)をブロモフェノールブルー色素(1μL)と混合し、次いで、1,5%アガロースゲル上で、TAEバッファー1X(40mMTris−酢酸塩、pH8.3、1mMEDTA)を含む電気泳動チャンバーにロードした。ゲルを、100ボルトで30分間ランした。次いで、ゲルをエチジウムブロマイド(EtBr)で15分間染色し、UV光の下でバンドを観察した。
自動化固相オリゴヌクレオチド合成によって、Dbait分子を作製した。配列は、5’−GCTGTGCCCACAACCCAGCAAACAAGCCTAGA−(H)−TCTAGGCTTGTTTGCTGGGTTGTGGGCACAGC(配列番号4)(式中、Hは、ヘキサエチレングリコールリンカーである)である。SV40でトランスフォーメーションした線維芽細胞MRC−5を使用して、培養細胞での研究を実施した。100%湿度、95%空気及び5%CO2の条件下、10%FCS及び抗生物質(100μg/mLストレプトマイシン及び100μg/mLペニシリン)を含む完全DMEM(Gibco, Cergy Pontoise, France)の単層培養で、細胞を37℃で増殖させた。特に明記しない限り、60mm直径プレートにおける1.2mLの血清不含MEM培地中でトランスフェクションを実施した。製造業者の説明書に従って、jetPEI(Polyplus-transfection, Illkirch, France)によるトランスフェクションを、6のN/P比率で実施した。つまり、Dbaitを150mM NaCl中で希釈し、150mM NaCl中で等量のPEIと穏やかに混合し、血清不含DMEM培地に添加した。coDbaitを、血清不含DMEM培地に直接添加した。Superfect試薬を用いて、Dbait分子のトランスフェクションを(60mm直径プレートにおける)1.2mLの血清含有DMEM培地中で5時間実施し、次いで、特に示さない場合は、細胞を1時間放置して回収した。エレクトロポレーションの場合、Gene Pulser II (Bio-Rad, Marnes-la-Coquette, France)を使用して、1.2×106個の細胞を、2μgのDbaitでトランスフェクションした。5時間のトランスフェクションの終了時に、培地を完全培地に交換し、分析する前に、細胞を、指示された時間増殖させた。トランスフェクション前に、クロロキン(50μM)を30分間添加した。
細胞を、Dbait−cy3を有する異なる複合体で5時間トランスフェクションし、5時間又は24時間増殖させ、PBSで速やかに洗浄した。フローサイトメトリーによって、細胞を直接分析した。フローサイトメトリーによる免疫蛍光検出の場合、免疫検出の前に、細胞を2%パラホルムアルデヒド中で10分間固定した。透過処理により、同じ細胞における免疫蛍光検出及びDbait検出を害するDbaitのほとんどが排除されたことに留意すべきである。細胞を4%ホルムアルデヒド中で15分間固定し、0.2%Triton X−100中で1時間透過処理し、2%BSAでブロッキングし、氷上で一次抗体、マウスモノクローナル抗体抗γ−H2AX(Upstate Biotechnology, Temecula, CA, USA)と共に2時間インキュベーションし、1/200の希釈、30分間、室温(RT)で、Alexa-488 (Molecular Probes, Eugene, OR, USA)、Texas Red (Rockland, Gilbertsville, PA, USA)とコンジュゲーションした二次抗体により可視化した。細胞をPBSで洗浄し、50μg/mLヨウ化プロピジウム、25U/ml RNaseAを含むPBS中で再懸濁した。FACScaliburフローサイトメーター(BD Biosciences, Franklin Lakes, NJ, USA)によって細胞を分析し、BD CellQuest Pro (BD Biosciences)及びフリーのWinMDI 2.8 (Scripps Research Institute, La Jolla, CA, USA)ソフトウェアを使用してデータを分析した。
野生型又はトランスジェニック(βアクチン:egfp−ras)フィッシュ系統の自然産卵から、ゼブラフィッシュの卵を得た。落射蛍光照明(Leica MZ16F)及び空気注入器(Eppendorf FemtoJet)を備える解剖顕微鏡上に固定したNarishige (MN 153)マイクロマニピュレーターを、1K細胞期にDbait注射を実施するのに使用した。KopF垂直ピペットプラー(KopF 720)でガラスキャピラリー(Harvard Apparatus GC100-10)を引き伸ばして、注射針を作製した。2〜5nlのDbait溶液を10細胞周期の胚の動物極に注射し、40x/0.8NA水浸漬対物レンズを備える共焦点レーザー走査顕微鏡イメージング(upright Leica SP2)のためにすぐに加工した。480nm(eGFP)及び561nm(cy3)の同時励起によって、イメージングを実施した。24hpfまで、胚を28.5℃でさらに成長させた。解剖ストップ(dissecting stop)の下で1日齢の幼生を観察し、図3に記載するように、表現型を分類した。注射前のクロロキン処置は、50μMクロロキン含有胚培地(The Zebrafish Book)における2時間のインキュベーションであった。Dbait(coDbait)−cy3 50μMを、単独で又はPEI25K(N/P=9の比率)、PEI11K(N/P=6の比率)、Superfect(10μl/1μgDbait)と組み合わせて注射した。
成体雌性ヌードマウス(Charles River strain; L’arbresle, France)の側腹に106個の腫瘍細胞を注射することによって、SK28又はU87G異種移植腫瘍を得た。実験を開始する前に、動物を研究室で少なくとも1週間飼育した。明暗周期(12時間:12時間)、相対湿度(55%)及び温度(21℃)のコントロール条件下で、動物5〜6匹/ケージであった。餌及び水道水は、自由に利用可能であった。約12日後に、測定した皮下腫瘍が150〜200mm3である場合、多くても12匹ずつの均一なグループにマウスを分けて、異なる処置を施した。動物の身体の約3分の2を保護するためのシールドデザインを用いて、137Cs単位(0.5Gy/分)で放射線照射を実施した。熱ミネセンス線量測定法によって、線量をコントロールした。1週間あたり5Gyのセッションを3回の間隔で、6回のセッションによって、30Gyの総線量を2週間で送達した。PEIとの混合をNaCl(Polyplus Transfection, Strasbourg, France)なしで実施した点を除いて、in vitro研究に関して前述したように、100μLの10%スクロース中でDbait分子を調製した。注射前に、Dbait混合物を室温で15分間インキュベーションした。coDbaitを10%スクロース中で必要濃度に希釈した。各放射線療法セッションの5時間前に、指示量のDbaitの腫瘍内注射を実施した。関連アッセイのプロトコールに従って、100μLの10%グルコースをモック処置動物に注射した。キャリパー測定によって3日ごとに腫瘍サイズを評価し、定式(2x長さx幅2)によってサイズを算出した。マウスの体重を量り、腫瘍の写真を毎週撮影した。腫瘍が2000mm3に達した場合、倫理的理由により、動物を屠殺した。いくつかの分析において使用したこの評価項目は、死亡日とした。Local Committee on Ethics of Animal Experimentation (Orsay, France)がすべての実験を承認した。
各処置及び各腫瘍型に関して、腫瘍反応の記述的分析を実施した。1日目を、最初の処置セッションの日とした。すべての動物を少なくとも150日間追跡した。Kaplan-Meier法に従って、平均寿命を評価した。各処置グループにおける個々のマウスの腫瘍容積の4倍から、コントロールグループの平均腫瘍容積の4倍を引くことによって、腫瘍成長遅延(TGD)を算出した。個別測定を使用して、各処置グループの平均TDGを算出した。Kaplan-Meier評価によって全生存曲線を評価し、データが正規分布に従わないので、ノンパラメトリックLogRank検定を使用して比較した。statELソフトウェア(ad Science, Paris, France)を使用して、分析を実施した。最初に、同じ腫瘍型を有する各グループに関して、グローバルなLogRankを実施した。次いで、Dbaitによる処置をモック処置コントロールと比較した。動物の数(n)、相対リスク(RR)及びp値を表2に報告する。有意水準0.05で、すべての試験を有意であると判断した。
これらの仕様:中間粘度:1.150cP、屈折率:1.45、散乱角:90°、温度:25°CによるZetasizer nanoシリーズ(Malvern instruments, Paris, France)での動的光散乱(DLS)によって、ベクター/Dbaitの粒子サイズを測定した。データは、1サンプルあたり3〜5回の測定の平均であり、各測定は、10〜15回のサブラン(sub−run)のデータを平均している。機器に付属の多モード数分布ソフトウェアを使用して、データを分析した。ゼータ電位測定に関して、粒子を10%スクロース/10mM NaCl中で希釈して、0.1mg/mLの最終Dbait濃度とし、以下の仕様:3回の測定、中間粘度:1.054cP、中間誘電率:79、温度:25°Cで測定した。
酢酸ウラニルでネガティブ染色することによって、透過電子顕微鏡法用のサンプルを調製した。サンプル1滴(10μL)をグリッド(formvar/carbon on 200 mesh copper, AGAR scientific)上に付着させ、余分な液体を吸い取り紙で除去する前に、3分間放置した。次いで、複合体を、10μLの水性酢酸ウラニル(2%)で2分間染色し、余剰分を吸い取り紙で除去した。Jeol JEM-100S電子顕微鏡で観察を行った。
別のコンジュゲートDbait分子を調製し、以下に記載する:
本発明者らは、フローサイトメトリー分析によって、コレステロールにコンジュゲーションしたDbait、特にCoDbaitの細胞取り込みを、Dbaitと比較して測定した。
Claims (14)
- 少なくとも1つの自由末端と、ヒトゲノムの遺伝子と80%未満の配列同一性を有する20〜200bpのDNA二本鎖部分とを有するコンジュゲート核酸分子及びキノリンエンドソーム溶解剤を含む、医薬組成物であって、脂溶性分子又は細胞レセプターをターゲティングしてレセプター介在性エンドサイトーシスを可能にするリガンドから選択されるエンドサイトーシスを促進する分子に前記核酸分子が共有結合している、医薬組成物。
- 少なくとも1つの自由末端と、ヒトゲノムの遺伝子と80%未満の配列同一性を有する20〜200bpのDNA二本鎖部分とを有するコンジュゲート核酸分子及びキノリンエンドソーム溶解剤を含む、同時、個別又は逐次使用のための複合調製物としての製品であって、脂溶性分子又は細胞レセプターをターゲティングしてレセプター介在性エンドサイトーシスを可能にするリガンドから選択されるエンドサイトーシスを促進する分子に前記核酸分子が共有結合している、製品。
- キノリンエンドソーム溶解剤が、コンジュゲート核酸分子の前及び/又は同時に投与される、詰求項2記載の製品。
- キノリンエンドソーム溶解剤が、少なくとも1週間の前処置として経口経路により投与され、次いで、コンジュゲート核酸分子及びキノリンエンドソーム溶解剤が、同時、個別又は逐次使用のための複合調製物として投与される、請求項2又は3記載の製品。
- DNA損傷抗腫瘍剤をさらに含む、請求項1〜4のいずれか一項に記載の組成物又は製品。
- ガンの処置における使用のための、請求項1〜5のいずれか一項に記載の組成物又は製品。
- 処置が、放射線療法又は、好ましくはDNA損傷抗腫瘍剤による化学療法をさらに含む、請求項6記載の組成物又は製品。
- DNA損傷抗腫瘍剤が、トポイソメラーゼI又はIIの阻害剤、DNA架橋剤、DNAアルキル化剤、代謝拮抗剤及び有糸分裂紡錘体の阻害剤からなる群より選択される、請求項7記載の組成物又は製品。
- キノリンエンドソーム溶解剤が、クロロキン又はヒドロキシクロロキン、好ましくはクロロキンである、詰求項1〜8のいずれか一項に記載の組成物又は製品。
- エンドサイトーシスを促進する分子が、単鎖又は二本鎖脂肪酸、例えばオクトデシル及びジオレオイル、トコフェロール、葉酸塩又は葉酸、コレステロール、糖、例えばガラクトース及びマンノース並びにそれらのオリゴ糖、ペプチド、例えばRGD及びボンベシン、及びタンパク質、例えばインテグリンからなる群より選択され、好ましくは、ジオレオイル、オクタデシル、葉酸及びコレステロールからなる群より選択され、より好ましくは、コレステロールである、請求項1〜9のいずれか一項に記載の組成物又は製品。
- コンジュゲート核酸分子が、下記式:
