- 【特許請求の範囲】 1. c−myb RNAを切断する酵素的核酸分子であって、前記核酸の結合 アームが表II、XII−XXIVに規定される配列に相補的な配列を含むことを特徴 とする核酸分子。 2. c−fos、oct−1、SRF、PDGFレセプター、bFGFレセプ ター、アンギオテンシンII、およびエンドセリン由来弛緩因子をコードるす遺伝 子から選択される遺伝子から生成するRNAを切断する酵素的核酸分子。 3. 前記核酸分子がハンマーヘッドモチーフのものである、請求項1または2 に記載の酵素的核酸分子。 4. 前記核酸分子がヘアピン、デルタ肝炎ウイルス、VS核酸、グループIイ ントロンまたはRNAseP核酸モチーフのものである、請求項1または2に記 載の酵素的核酸分子。 5. 前記核酸が前記mRNAに相補的な12から100塩基を含む、請求項3 または4に記載の酵素的核酸分子。 6. 前記核酸が前記mRNAに相補的な14から24塩基を含む、請求項5記 載の酵素的核酸分子。 7. 本質的に、表III、XII−XXIVに掲げられる配列の群より選択される任 意の配列からなる酵素的核酸分子。 8. 請求項1または2に記載の酵素的核酸分子を含む哺乳動物細胞。 9. 前記細胞がヒト細胞である請求項8記載の細胞。 10. 請求項1または2に記載の1つの酵素的核酸分子または多数の酵素的分 子をコードする核酸を、哺乳動物細胞中においてその酵素的RNA分子の発現が 可能な様式で含む発現ベクター。 11. 請求項10記載の発現ベクターを含む哺乳動物細胞。 12. 前記細胞がヒト細胞である請求項13記載の細胞。 13. 請求項1または2に記載の酵素的核酸分子またはc−myb遺伝子から 生成するRNAを切断する酵素的核酸分子を投与することにより狭窄状態を治療 する方法。 14. 患者に請求項10記載の発現ベクターを投与することにより狭窄状態を 治療する方法。 15. 前記患者がヒトである、請求項13または14に記載の方法。 16. 患者または患者の細胞に請求項1または2に記載の酵素的核酸分子を投 与することにより癌を治療する方法。 17. 患者または患者の細胞に請求項10記載の発現ベクターを投与すること により癌を治療する方法。 18. 前記患者がヒトである、請求項16または17に記載の方法。 19. 酵素的核酸を、クロロキン、塩化アンモニウム、カルボニルシアニドp −トリフルオロメトキシフェニルヒドラゾン(FCCP)、モネンシン、コルチ シン、両親媒性ペプチド、ウイルス蛋白質およびウイルス粒子からなる群より選 択される化学物質と混合することにより前記酵素的核酸を投与する方法。 20. 前記核酸が少なくとも5個のリボース残基を含み、前記核酸が6個の5 ’末端ヌクレオチドの少なくとも3個においてホスホロチオエート結合を含み、 前記核酸が前記核酸の4位において2’−C−アリル修飾を含み、前記核酸が少 なくとも10個の2’−O−メチル修飾を含み、かつ前記核酸が3’−末端修飾 を含む、請求項3記載の酵素的核酸。 21. 前記核酸が前記3’−末端において3’−3’結合反転リボース部分を 含む、請求項20記載の酵素的核酸。 22. 前記核酸が少なくとも5個のリボース残基を含み、前記核酸が6個の5 ’末端ヌクレオチドの少なくとも3個においてホスホロチオエート結合を含み、 前記核酸が前記核酸の4位および/または7位において2’−アミノ修飾を含み 、前記核酸が少なくとも10個の2’−O−メチル修飾を含み、かつ前記核酸が その3’−末端に3’−3’結合反転リボースまたはチミジン部分を含む、請求 項3記載の酵素的核酸。 23. 前記核酸が少なくとも5個のリボース残基を含み、前記核酸が6個の5 ’末端ヌクレオチドの少なくとも3個においてホスホロチオエート結合を含み、 前記核酸が前記核酸分子の4位および/または7位において非ヌクレオチド置換 を含み、前記核酸が少なくとも10個の2’−Oメチル修飾を含み、かつ前記核 酸がその3’末端において3’−3’結合反転リボースまたはチミジン部分を含 む、請求項3記載の酵素的核酸。 24. 前記核酸が少なくとも5個のリボース残基を含み、前記核酸が6個の5 ’末端ヌクレオチドの少なくとも3個においてホスホロチオエート結合を含み、 前記核酸が前記核酸分子の4位および/または7位において6−メチルウリジン 置換を含み、前記核酸が少なくとも10個の2’−O−メチル修飾を含み、かつ 前記核酸がその3’末端において3’−3’結合反転リボースまたはチミジン部 分を含む、請求項3記載の酵素的核酸。 25. 前記核酸が少なくとも5個のリボース残基を含み、前記核酸が6個の5 ’末端ヌクレオチドの少なくとも3個においてホスホロチオエート結合を含み、 前記核酸が前記核酸の4位において2’−C−アリル修飾を含み、前記核酸が少 なくとも10個の2’−O−メチル修飾を含み、かつ前記核酸がその3’末端に おいて2’−3’結合反転リボースまたはチミジン部分を含む、請求項3記載の 酵素的核酸。 26. 少なくとも5個の連続する塩基においてc−mybに対する相補性を有 し、5’−ホスフェートを有する2’−5’−結合アデニレート残基を含むオリ ゴヌクレオチド。 27. c=myb RNAに対して酵素的活性を有する請求項26記載のオリ ゴヌクレオチド。 28. c−myb RNAとハイブリッドを形成しうる少なくとも20塩基を 含む、請求項26記載のオリゴヌクレオチド。
