3.局部脳組織中のAMP/ADP/ATPの測定を、紫外線検出を用いる高速液体クロマトグラフィー(HPLC)によって測定した。酵素を不活性化させて死後変化を防ぐために、マウスをマイクロウェーブ固定により屠殺することを要した。
4.局部脳組織中の脳アセチルコリン(Ach)の測定を、電気化学検出を用いるHPLCによって測定した。酵素を不活性化させて死後変化を防ぐために、マウスをマイクロウェーブ固定により屠殺することを要した。
5.脳内ドーパミン、セロトニン、及びノルエピネフリン局部脳組織を、電気化学検出を用いるHPLCによって測定した。
6.血漿及び脳組織の完全定量的アミノ酸プロファイリングを、紫外線検出を用いるHPLCによって実施した。GABA及びその他の興奮性及び阻害性アミノ酸を脳組織で測定した。
7.尿有機酸と血液アシル−カルニチンをガスクロマトグラフ質量分析によって測定した。

Claims (30)
- 神経疾患又は神経変性疾患に対する治療を必要とする成人患者を特定するステップと、
前記成人患者に生理学的に効果的な量の製剤を投与するステップとを含み、前記製剤が一般式:
- R1、R2、及びR3炭素鎖が、ペンタノイン、ペンタノイルカルニチン、n−ペンタデカン酸、5炭素脂肪酸前駆体、及びその誘導体から選択される5炭素長である、請求項1に記載の方法。
- R1、R2、及びR3炭素鎖が7炭素長である、請求項1に記載の方法。
- 奇数鎖トリグリセリドがトリヘプタノインである、請求項1に記載の方法。
- 神経変性疾患がアルツハイマー病(AD)又は加齢である、請求項1に記載の方法。
- アルツハイマー病(AD)に対する治療を必要とする成人患者を特定するステップと、前記患者にADの症状を治療又は軽減するために十分な量でトリヘプタノインの製剤を経腸的又は非経口的に投与するステップとを含む、ADを患っている前記成人患者を治療する方法。
- 製剤が、香味剤、ビタミン、ミネラル補給剤、タンパク質補給剤、着色剤、及び防腐剤からなる群から選択される、1又は2以上の任意の添加物を含む、請求項6に記載の方法。
- 組成物を投与すると、ヒト対象の血液中の1又は2以上の循環ケトン体のレベルが上昇する、請求項6に記載の方法。
- 高脂質低炭水化物食をヒト対象に提供する食餌組成物であって、一般式:
- R1、R2、及びR3炭素鎖が、ペンタノイン、ペンタノイルカルニチン、n−ペンタデカン酸、5炭素脂肪酸前駆体、及びその誘導体から選択される5炭素長である、請求項9に記載の組成物。
- R1、R2、及びR3炭素鎖が7炭素長である、請求項9に記載の組成物。
- 奇数鎖脂肪酸がトリヘプタノインである、請求項9に記載の組成物。
- ヒト対象が、健康なヒト対象、又はアルツハイマー病(AD)、老年認知症、血管性認知症、ピック病、クロイツフェルト・ヤコブ病、パーキンソン病、及び加齢からなる群から選択される1又は2以上の神経学的状態を患っているヒト対象である、請求項9に記載の組成物。
- ADを患っていることが疑われるヒト対象への投与に適合された、請求項9に記載の組成物。
- 投与すると、ヒト対象の血液中の1又は2以上の循環ケトン体のレベルが上昇する、請求項9に記載の組成物。
- 中鎖トリグリセリド、5、7、及び15個の炭素脂肪酸からなる群から選択される奇数炭素鎖脂肪酸及びそのトリグリセリド、又はその両方を含む、食用に適した食餌製剤。
- 脂肪酸がペンタン酸である、請求項16に記載の製剤。
- 脂肪酸がヘプタン酸である、請求項16に記載の製剤。
- 奇数鎖トリグリセリドがトリヘプタノインである、請求項16に記載の製剤。
- 組成物が、神経変性疾患、アルツハイマー病(AD)、老年認知症、血管性認知症、ピック病、クロイツフェルト・ヤコブ病、パーキンソン病、及び加齢に関連する症状を治療又は軽減するために用いられる、請求項16に記載の製剤。
- ADを患う患者への送達に適合された、請求項16に記載の製剤。
- 経腸投与に適合された、請求項16に記載の製剤。
- 高脂質低炭水化物食をヒト対象に提供する食餌組成物であって、官能担体中のペンタノイン、ペンタノイルカルニチン、n−ペンタデカン酸、5炭素脂肪酸前駆体、並びにその誘導体から選択される、単離及び精製された短鎖脂肪酸、及び香味剤、ビタミン、ミネラル補給剤、タンパク質補給剤、着色剤、並びに防腐剤からなる群から選択される、1又は2以上の任意の添加物とを含む、食餌組成物。
- アルツハイマー病(AD)、老年認知症、血管性認知症、ピック病、クロイツフェルト・ヤコブ病、パーキンソン病、及び加齢からなる群から選択される、1又は2以上の神経学的状態を患っていることが疑われるヒト対象への投与に適合された、請求項23に記載の組成物。
- 投与すると、ヒト対象の血液中の1又は2以上の循環ケトン体のレベルが上昇する、請求項23に記載の組成物。
- 投与すると、アセチル−CoAの血清レベルが上昇する、請求項23に記載の組成物。
- アルツハイマー病(AD)に対する治療を必要とする成人患者を特定するステップと、C5、C7、C9、C11、C13、C15、又はそのトリグリセリドの少なくとも1つを含む奇数鎖脂肪酸の製剤をADの症状を治療又は軽減するために十分な量で患者に投与するステップとを含む、ADを患っている前記成人患者を治療する方法。
- 製剤が、香味剤、ビタミン、ミネラル補給剤、タンパク質補給剤、着色剤、及び防腐剤からなる群から選択される、1又は2以上の任意の添加物を含む、請求項27に記載の方法。
