Abstract
下記式(I):(式中、R1、R2およびR3は、それぞれ飽和脂肪酸残基であって、その少なくとも1つはペンタデカン酸残基である。)で表されるトリグリセリドを有効成分として含むインスリン分泌促進剤である。ペンタデカン酸トリグリセリドの有する、血糖値上昇時におけるインスリン分泌促進作用を見出し、このトリグリセリドを用いて血糖値改善や糖尿病改善するための医薬品および健康食品を提供する。
Description
本出願は、日本国において、2019年11月27日に出願された特願2019-213850号に基づく優先権を主張するものであり、当該出願に記載された内容は全て、参照によりそのまま本明細書に援用される。また、本明細書において引用した全ての特許、特許出願及び文献に記載された内容は全て、参照によりそのまま本明細書に援用される。
本発明は、インスリン分泌促進剤並びにこれを用いた血糖値改善剤、糖尿病改善剤及び健康食品、機能性表示食品、特定保健用食品等の食品に関する。
糖尿病はインスリンの分泌不全あるいはインスリン感受性の低下により、血糖値が恒常的に上昇し、目や腎臓など様々臓器の機能が障害される慢性疾患である。日本国内において糖尿病の罹患者数は、糖尿病の疑いのある者を合わせて約1,000万人に上り、また、全世界では糖尿病の患者数は約4億人に上り、その数は現在も増え続けている。糖尿病患者に対しては、従来、スルフォニル尿素(SU)やDDP4阻害剤などに代表されるインスリン分泌促進薬が多く使用されている。しかし、SU剤についてはインスリンの過剰分泌による低血糖の危険性、DDP4阻害剤についてはインスリン分泌促進効果がやや低いといった問題点があるため、インスリン分泌促進効果を有する新規治療剤の開発に向けてなおも熾烈な開発競争が行われている。しかしながら、現状、これらに代わるインスリン分泌作用を促進する有効な成分は見出されていなかった。
ところで、オーランチオキトリウム(Aurantiochytrium)属藻類を利用して、奇数鎖脂肪酸を主要成分として含有するトリグリセリドおよび高度不飽和脂肪酸を含有するトリグリセリドとの混合物を製造する方法が知られている(例えば、特許文献1参照)。哺乳類や鳥類の体内の脂肪酸は、炭素数が偶数の脂肪酸がほとんどであり、奇数鎖脂肪酸は、ヒトおよび家畜と家禽の体脂肪、筋肉、臓器、乳および卵には、0.1~数%程度しか含まれていない。奇数鎖脂肪酸は偶数鎖脂肪酸と同様にβ-酸化によりエネルギー源となるが、β-酸化系で最後に生成されたプロピオニル-CoAがスクシニル-CoAに変換されてTCAサイクルに補充されることにより、TCAサイクルを正常に維持する機能も担っている。TCAサイクルの機能を維持することにより、運動時や運動後の筋肉の損傷により起こる筋肉痛の軽減や(非特許文献1参照)、ミトコンドリアのエネルギー代謝を改善して代謝性疾患や老化を改善することが期待されている(非特許文献2参照)。
また、奇数鎖トリグリセリドを含むケトン産生食の使用により、認知を改善し、循環ケトン体を増加させ、アルツハイマー病(AD)患者のアミロイド-β(Aβ)沈着を減少させる、ADと加齢に対する治療方法についても報告(特許文献2参照)があるが、糖尿病分野における作用については未だ知られていない。

従って、本発明は、上記したような現状を鑑み、糖尿病の予防または改善に有効な新規なインスリン分泌促進剤並びにこれを用いた血糖値改善剤、糖尿病改善剤及び食品を提供することを課題とする。
上記課題を解決するために、本発明者らが鋭意検討を行った結果、ペンタデカン酸トリグリセリド(PdATG)が膵臓β細胞に働きかけて、インスリン分泌を促進する効果を発見した。そして、このインスリン分泌促進作用を有するペンタデカン酸トリグリセリドを有効成分として用いることにより、糖尿病の予防または改善に有効な医薬品及び食品が得られることを見出し、本発明を完成するに至った。
すなわち、上記課題を解決する本発明の第一の観点は、下記式(I):

本発明に係るインスリン分泌促進剤の一実施形態においては、式(I)のトリグリセリドは、R1とR2またはR1とR3がペンタデカン酸残基であることが好ましい。
本発明に係るインスリン分泌促進剤の別の一実施形態においては、R1、R2およびR3の何れか1つが、トリデシル酸(C13)、ミリスチン酸残基(C14)、パルミチン酸残基(C16)またはマルガリン酸残基(C17)であってもよい。
また、本発明に係るインスリン分泌促進剤の別の好ましい実施形態においては、R1、R2およびR3のすべてがペンタデカン酸残基である上記式(I)のトリグリセリドと、R1、R2およびR3の何れか2つがペンタデカン酸残基であり、他の1つがミリスチン酸またはパルミチン酸残基である式(I)のトリグリセリドと、を含むものであってもよい。
さらに、本発明に係るインスリン分泌促進剤のさらに別の好ましい実施形態においては、式(I)のトリグリセリドが、オーランチオキトリウム属又はシゾキトリウム属藻類由来であり、式中、R1、R2およびR3は、それぞれ飽和脂肪酸残基であって、その少なくとも1つはペンタデカン酸残基である、トリグリセリドであってもよい。
本発明の第二の観点は、上記式(I)で表されるトリグリセリドを有効成分として含む血糖値改善剤である。
本発明の第三の観点は、上記式(I)で表されるトリグリセリドを有効成分として含む糖尿病改善剤である。
本発明の第二の観点は、上記式(I)で表されるトリグリセリドを有効成分として含む血糖値改善剤である。

さらに本発明の第四の観点は、上記式(I)で表されるトリグリセリドを有効成分として含む食品である。この食品は、例えば、健康食品、機能性表示食品、あるいは特定保健用食品等として血糖値の高めの人々の改善のために使用されることが好ましい。
本発明に係るペンタデカン酸トリグリセリドは、高めの血糖値を改善するために用いることができ、そのための健康食品および医薬品として有用である。
以下、本発明の有効成分であるペンタデカン酸トリグリセリドについて最初に説明し、続いてその製造方法および各種用途について説明する。

本明細書において、「ペンタデカン酸トリグリセリド」とは、少なくとも1つのペンタデカン酸とグリセロールとのエステルを意味し、下記式(I)に示すR1、R2およびR3の少なくとも1つ、好ましくは何れか2つ、例えば、R1とR2またはR1とR3が、さらに好ましくはR1、R2およびR3の3つがペンタデカン酸残基であるトリグリセリドを含む。ペンタデカン酸のグリセリドへの結合位置は、1~3位のいずれであってもよい。
式中、R1、R2およびR3で表される各飽和脂肪酸残基のうちの少なくともいずれか1つの残基がペンタデカン酸残基であれば他の2つは、それぞれ独立に、ペンタデカン酸残基以外の飽和脂肪酸残基であってもよい。「飽和脂肪酸」とは、分子内に二重結合、三重結合を持たない脂肪酸の総称であり、CnH2n+1COOHの化学式で示される。特に限定されるものではないが、この飽和脂肪酸の化学式において、nは例えば4~26の整数、好ましくは11~19の整数、さらに好ましくは14または16の整数である。また、R1、R2およびR3で表される各飽和脂肪酸残基のうちの1つがペンタデカン酸残基で、残りの2つがペンタデカン酸残基以外の飽和脂肪酸残基である場合、これらの飽和脂肪酸は、同一のものであってもまた相互に異なるものであっても良い。この飽和脂肪酸は、直鎖状または分枝状の飽和脂肪酸であり、酪酸(C4)、吉草酸(C5)、カプロン酸(C6)、エナント酸(C7)、カプリル酸(C8)、ペラルゴン酸(C9)、カプリン酸(C10)、ラウリン酸(C12)、トリデシル酸(C13)、ミリスチン酸(C14)、ペンタデカン酸(C15)、パルミチン酸(C16)、マルガリン酸(C17)、ステアリン酸(C18)、アラキジン酸(C20)、ベヘン酸(C22)、リグノセリン酸(C24)およびセロチン酸(C26)等の直鎖状飽和脂肪酸や、2-ヘキシルデカン酸(C16)、13-メチルペンタデカン酸(C16)、16-メチルヘプタデカン酸(C18)等の分枝状飽和脂肪酸が挙げられる。
好ましい実施形態におけるペンタデカン酸トリグリセリドは、R1、R2およびR3のすべてがペンタデカン酸残基である上記式(I)のトリグリセリドと、R1、R2およびR3の何れか2つがペンタデカン酸残基であり、他の1つがミリスチン酸またはパルミチン酸残基であるトリグリセリドとの両方を含む。この混合物中における両者の含有比率は特に限定されないが、質量比で1:2~2:1であることが好ましく、ほぼ1:1であることがさらに好ましい。また、トリグリセリドの総量に対してこれらのそれぞれが10質量%以上、好ましくは20質量%以上含まれる。さらに、ペンタデカン酸を2残基以上含むトリグリセライドの混合物が、油脂中の50質量%以上含まれていることがより好ましい。
さらに好ましい実施形態においては、ペンタデカン酸トリグリセリドは、以下の一般式(II)または(III)で表される。

ペンタデカン酸を2残基以上含むトリグリセリドの混合物が、油脂中の50質量%以上含まれていることがより好ましいが、ペンタデカン酸を2残基以上含むトリグリセリドの含量が50質量%以下であっても、摂取量を多くすることで、目的が達成できる。したがって、本発明の有効成分は、ペンタデカン酸を2残基以上含むトリグリセリドの混合物の状態で存在してもよく、トリグリセリドの総量に対して、少なくとも1質量%、好ましくは50質量%以上、より好ましくは90質量%以上の純度で含まれていれば混合物自体で有効成分としての機能を発揮することができる。
本発明の有効成分は、式Iの化合物以外のトリグリセリドとともに混合物の状態で存在してもよく、トリグリセリドの総量に対して、少なくとも1質量%、好ましくは50質量%以上、より好ましくは90質量%以上の純度で含まれていれば混合物自体で有効成分としての機能を発揮することができる。
(作用効果)
本発明の有効成分であるペンタデカン酸トリグリセリドは、分子内に少なくとも1つのペンタデカン酸を含むが、ペンタデカン酸のような奇数鎖脂肪酸は、TCAサイクルを正常に維持する機能がある。脂肪酸は体内で酸化され、炭素数2個(C2)のアセチル-CoAとなってクエン酸サイクル(TCAサイクル)に入り、補酵素NAD(ニコチンアミドアデニンジヌクレオチド)やFAD(フラビンアデニンジヌクレオチド)をNADH2やFADH2に還元し、電子伝達系によりATPを産生する。ここで、炭素数が偶数の脂肪酸は、全ての炭素鎖がC2のアセチル-CoAに分解されてTCAサイクルに利用されるが、ペンタデカン酸のような奇数鎖脂肪酸の場合は、分解の最後にC3のプロピオニル-CoAが残る。
プロピオニル-CoAは、C4のメチル-マロニル-CoAに変換され、ビタミンB12を補酵素とする酵素メチルマロニル-CoAムターゼによって、TCAサイクルの一員であるスクシニル-CoAに変換される。TCAサイクルに導入されたスクシニル-CoAは、酵素スクシニルCoAシンセターゼによってコハク酸になる。この反応で、シグナル伝達物質であるGTP(グアノシン三リン酸)が生産される。TCAサイクルにおいて、GTPが生産されるのはこの反応だけである。なお、TCAサイクルにおいてATPは生産されない。

プロピオニル-CoAは、C4のメチル-マロニル-CoAに変換され、ビタミンB12を補酵素とする酵素メチルマロニル-CoAムターゼによって、TCAサイクルの一員であるスクシニル-CoAに変換される。TCAサイクルに導入されたスクシニル-CoAは、酵素スクシニルCoAシンセターゼによってコハク酸になる。この反応で、シグナル伝達物質であるGTP(グアノシン三リン酸)が生産される。TCAサイクルにおいて、GTPが生産されるのはこの反応だけである。なお、TCAサイクルにおいてATPは生産されない。
GTPはG-タンパク質と呼ばれる膜タンパク質に結合してシグナルを伝達する。GTPが結合した活性型Gタンパク質は、細胞の様々な生理機能を活性化させる重要な役割を担っている。したがって、奇数鎖脂肪酸は、C2のアセチル-CoAから出発するATP生産に加えて、C3のプロピオニル-CoAから出発する細胞の生理機能活性化にも関与することが示唆される。
TCAサイクルを構成する分子は他の様々な代謝経路にも関与するものであるため、構成分子のいずれかが不足してTCAサイクルが充分に機能しなくなくなる場合がある。そのような場合、奇数鎖脂肪酸が上記のように分解されて生成されたスクシニル-CoAがTCAサイクルに補充される(この現象を補充反応(anaplerosis)と呼ぶ)。また、逆にTCAサイクルの分子が過剰に存在する場合は、いずれかの構成分子を抜き取る反応(cataplerotic reaction)が生じ、TCAサイクルが正常に保たれる。奇数鎖脂肪酸は、プロピオニル-CoAを経由してスクシニル-CoAを補充することにより、TCAサイクルを正常に維持する機能を担っている。
本発明の有効成分は、このような奇数鎖脂肪酸、特に、ペンタデカン酸を分子内に少なくとも1個、好ましくは2個以上有することからこれを摂取することで後述する糖訴求時のインスリン分泌を促進し、血糖値を正常に導く作用を発揮すると考えられる。一般に、遊離脂肪酸は細胞に対して毒性を有するが、本発明の有効成分はトリグリセリドの状態で存在するため安定性が高く、特に、後述するようにラビリンチュラ類藻類のオーランチオキトリウム属またはシゾキトリウム属藻類のオーランチオキトリウム属から得られるペンタデカン酸トリグリセリドは、天然由来で安定性が高い形態で、毒性がない。このため遊離脂肪酸としての、奇数鎖(C15)飽和脂肪酸であるペンタデカン酸とは異なる作用を示し、大量に投与しても毒性がなく、血糖値の低下に有効な効果をもたらす健康食品や医薬品として適していると考えられる。
また、奇数鎖(C15)飽和脂肪酸であるペンタデカン酸は、後述する比較例1において示すように、膵β細胞由来の細胞株に加えインスリン分泌量を測定したところ、本発明に係るペンタデカン酸トリグリセリドとは異なり、インスリン分泌促進効果が見られなかった。このことから、インスリン分泌値を促進し、血糖値を正常に戻せる効果は、遊離脂肪酸であるペンタデカン酸にはなく、ペンタデカン酸トリグリセリドにおいて、初めて認められた効果であることが判った。
(ペンタデカン酸トリグリセリドの製造方法)
本発明の有効成分であるペンタデカン酸トリグリセリドは、化学的に合成されたものであっても、天然に存在するものであってもよい。天然に存在するペンタデカン酸トリグリセリドの供給源としては、生物が体内で生産する脂質、例えば家畜や家禽の脂肪、魚介類の油脂、植物油または脂質生産性の微生物が挙げられる。特に、ラビリンチュラ類藻類のオーランチオキトリウム属またはシゾキトリウム属藻類は、汽水域に生息する従属栄養性藻類であり、水中の栄養分を同化して脂質を生産し、細胞内に蓄積する特徴を有するため好ましい。
本発明の有効成分であるペンタデカン酸トリグリセリドは、化学的に合成されたものであっても、天然に存在するものであってもよい。天然に存在するペンタデカン酸トリグリセリドの供給源としては、生物が体内で生産する脂質、例えば家畜や家禽の脂肪、魚介類の油脂、植物油または脂質生産性の微生物が挙げられる。特に、ラビリンチュラ類藻類のオーランチオキトリウム属またはシゾキトリウム属藻類は、汽水域に生息する従属栄養性藻類であり、水中の栄養分を同化して脂質を生産し、細胞内に蓄積する特徴を有するため好ましい。
オーランチオキトリウム属またはシゾキトリウム属藻類は、所望のトリグリセリドを生産する能力の優れた株を用いるのが好ましい。そのような藻類株は、天然に採取および分離されたものであっても、突然変異誘導およびスクリーニングを経てクローニングされたものであっても、あるいは遺伝子組み換え技術を利用して樹立されたものであってもよい。例えば、オーランチオキトリウム種[Aurantiochytrium Sp.]SA-96株、NIES-3737株、オーランチオキトリウムNB6-3株、オーランチオキトリウムmh0192株またはオーランチオキトリウムmh1959株は、奇数鎖脂肪酸のペンタデカン酸(PDA)を含有するトリグリセリドと、高度不飽和脂肪酸のドコサヘキサエン酸(DHA)やドコサペンタエン酸(DPA)を含有するトリグリセリドを細胞内に大量に蓄積する性質を有する。また、オーランチオキトリウムSp.など自然界で採取したワイルド株には、これらの特異性を超えた脂肪産生株も見出しており、これらの株を有効利用することが、本発明のペンタデカン酸トリグリセリドの製造に用いる微生物として、特に好ましい。
上記オーランチオキトリウム属藻類の培養は、当該技術分野において確立された方法で行われる。即ち、通常の維持培養は、適切に成分調製した培地に藻類を播種し、定法に従い行われる。オーランチオキトリウム属藻類を培養するための培地は、本質的に、塩分、炭素供給源および窒素供給源を含有する。一般的に、微細藻類の培養には、いわゆるGTY培地(人工海水10~40g/L、D(+)グルコース20~100g/L、トリプトン10~60g/L、酵母抽出物5~40g/L)が用いられる。
炭素源としてはグルコース、フルクトース、スクロース等の糖類がある。これらの炭素源を、例えば、培地1リットル当たり20~120gの濃度で添加する。

オーランチオキトリウム属藻類は海洋性藻類であり、培地には適切な量の人工海水が添加される。好ましくは、人工海水は、最終的な培地の塩分濃度が海水(塩分濃度3.4%(w/v))の約10%(v/v)~約100%(v/v)、例えば塩分濃度が約1.0~3.0%(w/v)となるように添加される。
一般的に、微細藻類の培養培地には、グルタミン酸ナトリウム、尿素等の有機窒素、または酢酸アンモニウム、硫酸アンモニウム、塩化アンモニウム、硝酸ナトリウム、硝酸アンモニウム等の無機窒素、または酵母抽出物、コーンスチープリカー、ポリペプトン、ペプトン、トリプトン等の生物由来消化物等の、様々な窒素源が添加され得る。特に、オーランチオキトリウム属藻類の培養に用いる培地に添加する窒素源として、様々な動物の細胞から液体成分を抽出して得られる細胞抽出物が好んで用いられる。培養細胞産物を取得するために細胞を工業的スケールで大量培養しなければならない場合に、細胞由来のアミノ酸、核酸、ビタミン、ミネラル等の栄養素に富み、低コストで入手可能な細胞抽出物の利用は極めて有利である。
しかしながら、上記のように、細胞抽出物をベースに調製した培地を使用すると、培養藻類が生産するトリグリセリド中の奇数脂肪酸の割合が著しく低下してしまうため、本発明の目的物を効率的に生産する場合、培地の窒素源として細胞抽出物を利用することが出来なかった。そこで、本発明者らは、強酸処理した細胞抽出物を添加して調製した藻類培養培地中でオーランチオキトリウム属藻類を培養したところ、当該処理をしない細胞抽出物を添加した場合と比較して、奇数鎖脂肪酸の生産量が劇的に増大することを見出し、奇数鎖脂肪酸を主要成分として含有するトリグリセリドを製造する方法をすでに報告している(特開2017-063633号公報)。
さらに、本発明の好ましい実施形態において、オーランチオキトリウム属藻類を培養するための基本培地は、2%以上のグルコースと0.5~4%のグルタミン酸ナトリウム、0.1~2%の酵母エキス、1~3.3%海水塩、2~20%ホエイ(動物性または植物性)を加えた培地にバリン、またはイソロイシン、メチオニンを0~50mMとプロピオン酸ナトリウム0~50mMを添加する。動物性または植物性のホエイは、豆腐ホエー(大豆ホエー)が好ましい。この基本培地に、2%以上のグルコースと0.5~4%のグルタミン酸ナトリウム、0.1~2%の酵母エキス、1~3.3%海水塩、2~20%ホエイ(動物性または植物性)にて、20~30℃で72時間前培養したオーランチオキトリウムの培養液を2%以上加える。このオーランチオキトリウム添加培養液に、空気を通気させ、穏やかに攪拌する。培養は、20~30℃、pHは5.0~8.5に保持(pH調整には、1.0MのNaOH溶液を用いる)して、48~200時間行う。培養後、遠心分離にてペンタデカン酸トリグリセリドを生産したオーランチオキトリウム細胞を回収することができる(特願2018-171739号出願明細書参照)。
上記のような方法にて得られた培養液から遠心分離または濾過等により回収したペレットを、凍結乾燥または加温による乾燥等により乾燥させる。または、培養後の藻類細胞が懸濁した培地をそのままトリグリセリドの抽出ステップに用いてもよい。抽出は、異なる有機溶媒を用いて複数回行ってもよい。有機溶媒としては、n-ヘキサン・エタノール混合溶媒、クロロホルム・メタノール混合溶媒、またはエタノール・ジエチルエーテル混合溶媒等の極性溶媒と弱極性溶媒の混合液を用いることができる。得られた抽出液は、当業者に既知の方法で精製される。
トリグリセリドを分離する手法は、当業者に既知の分画手法が採用される。分画するトリグリセリド分子の極性、溶媒への溶解度、融点、比重、分子量等の様々な物理化学的特性を利用して分離精製が行われてもよく、好ましくはカラムクロマトグラフィー技術が用いられる。トリグリセリド分離手段の条件は、トリグリセリド混合物の組成および分画すべきトリグリセリドの種類に依存して、当業者による通常の条件検討により設定することが出来る。
オーランチオキトリウム属藻類は、奇数鎖脂肪酸トリグリセリドおよび高度不飽和脂肪酸トリグリセリドのいずれも細胞内で合成して蓄積することが出来る。そのため、得られたオーランチオキトリウム細胞にヘキサンまたは酢酸エチルを加え脂質を抽出後、この脂質溶液に過酸化水素水を加え(必要に応じて水を加える)、室温でオゾンを通気することにより不飽和脂肪酸を酸化分解する。反応終了後、炭酸水素ナトリウムおよび炭酸ナトリウムまたはイオン交換樹脂にて酸化物を取り除き、温度低下と共に析出するペンタデカン酸トリグリセリドを得る。精製されたペンタデカン酸トリグリセリドの組成は、ガスクロマトグラフィー、質量分析法等により分析することができる。
(インスリン分泌促進剤)
本発明の第1の観点に係るインスリン分泌促進剤は、高血糖時にインスリン分泌作用を有する製剤である。

医薬においても、血糖値(グルコース濃度)が高いときに着実に摂取すれば、血糖値に関係なくインスリン分泌し、血糖値を下げることができる。しかし、血糖値が下がることがあっても作用が持続するような場合、インスリン分泌を持続することで、必要以上に血糖値を下げ、低血糖のリスクを招くことになる。本発明のペンタデカン酸トリグリセリドは、食後血糖値が上昇する時にのみ作用し、インスリンの分泌を促進し、食後の血糖値を正常に保つ働きがあり、糖尿病の予防および/または改善することができる。
そして、血糖値上昇による顕在化する障害(疾病)や加齢変化、例えば、細小血管症(網膜症、腎症、神経障害)と大血管症(脳血管障害、冠動脈障害、足の閉塞性動脈障害)などの予防・改善に使用することができる。
ここで糖尿病性網膜症は網膜内の血管障害により出血、白斑、網膜浮腫などを生じて進行すると高度の視力障害を引き起こす疾病である。糖尿病性腎症は血管に富む腎糸球体の障害により蛋白尿と腎機能低下を生じる疾病である。糖尿病性神経障害は糖尿病に最も多い合併症で、主に足の末梢神経と自律神経が障害される疾病である。このような神経障害の予防にも良好な血糖コントロールが重要である。さらに糖尿病は心臓の冠動脈や脳血管などの大きな血管の動脈硬化(大血管症)を起こしやすくし、心筋梗塞や脳卒中など命にかかわる病気の引き金となる。脳卒中、心筋梗塞、足の血行障害は糖尿病患者では発症頻度が2~3倍に増加する。
インスリン分泌促進剤の適用用途も、特に限定されず、目的に応じて設定することができる。例えば、血糖値改善剤や糖尿病改善剤などの医薬品として、使用可能である。
(医薬品)
本発明の第2の観点に係る血糖値改善剤および第3の観点に係る糖尿病改善剤である医薬品は、上記式(I)で表されるペンタデカン酸トリグリセリドを有効成分として含有し、2型糖尿病等で、糖摂取時の血糖値を改善するため、ないし糖尿病の症状を改善するために用いることができ、そのための医薬品として有用である。ここで、「医薬品」とは、インスリン分泌が適時されなくなっている糖尿病患者に対して、血糖値を正常に保つ事で、インスリンの摂取等の治療を必要とすることが無い治療薬を意味する。
本発明の第2の観点に係る血糖値改善剤および第3の観点に係る糖尿病改善剤である医薬品は、上記式(I)で表されるペンタデカン酸トリグリセリドを有効成分として含有し、2型糖尿病等で、糖摂取時の血糖値を改善するため、ないし糖尿病の症状を改善するために用いることができ、そのための医薬品として有用である。ここで、「医薬品」とは、インスリン分泌が適時されなくなっている糖尿病患者に対して、血糖値を正常に保つ事で、インスリンの摂取等の治療を必要とすることが無い治療薬を意味する。
上記式(I)で表されるペンタデカン酸トリグリセリドを配合して医薬品を製造する際には、例えば、デキストリン、デンプン等の糖類;ゼラチン、大豆タンパク、トウモロコシタンパク等のタンパク質;アラニン、グルタミン、イソロイシン等のアミノ酸類;セルロース、アラビアゴム等の多糖類;大豆油、中鎖脂肪酸トリグリセリド等の油脂類等の任意の助剤を添加して任意の剤形に製剤化することができる。
本発明に係る医薬品における上記式(I)で表されるペンタデカン酸トリグリセリドの配合量は、特に限定される訳ではないが、有効性を示す濃度である成人一日当たりペンタデカン酸トリグリセリドの摂取量が1日当たり約10~1000mg程度となるように調整することが好ましい。
(食品)
本発明の第4の観点に係る食品は、上記式(I)で表されるペンタデカン酸トリグリセリドを有効成分として含有し、高めの血糖値を改善するために用いることができ、そのための健康食品として有用である。ここで、「健康食品」とは、食後血糖値が下がりにくくなっている状態の改善を図ることを目的とした飲食物を意味し、国が定めた安全性や有効性に関する基準等を満たした「保健機能食品制度」上での機能性表示食品、あるいは特定保健用食品等を含む広義の「健康食品」を指す。
健康食品は、任意の飲食品に配合したものであってもよいし、本発明の血糖値高めの方用を主成分とする栄養補助食品であってもよい。
本発明の第4の観点に係る食品は、上記式(I)で表されるペンタデカン酸トリグリセリドを有効成分として含有し、高めの血糖値を改善するために用いることができ、そのための健康食品として有用である。ここで、「健康食品」とは、食後血糖値が下がりにくくなっている状態の改善を図ることを目的とした飲食物を意味し、国が定めた安全性や有効性に関する基準等を満たした「保健機能食品制度」上での機能性表示食品、あるいは特定保健用食品等を含む広義の「健康食品」を指す。
健康食品は、任意の飲食品に配合したものであってもよいし、本発明の血糖値高めの方用を主成分とする栄養補助食品であってもよい。
上記式(I)で表されるペンタデカン酸トリグリセリドを配合して食品を製造する際には、例えば、デキストリン、デンプン等の糖類;ゼラチン、大豆タンパク、トウモロコシタンパク等のタンパク質;アラニン、グルタミン、イソロイシン等のアミノ酸類;セルロース、アラビアゴム等の多糖類;大豆油、中鎖脂肪酸トリグリセリド等の油脂類等の任意の助剤を添加して任意の剤形に製剤化することができる。
また本発明の食品における上記式(I)で表されるペンタデカン酸トリグリセリドの配合量は、特に限定される訳ではないが、添加対象飲食品の一般的な摂取量を考慮して成人一日当たりペンタデカン酸トリグリセリドの摂取量が1日当たり約1~100mg程度となるように調整することが好ましい。
上記食品の具体例としては、例えば、清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料等の飲料(これら飲料の濃縮液および調整用粉末を含む);アイスクリーム、アイスシャーベット、かき氷等の氷菓;そば、うどん、はるさめ、ぎょうざの皮、シュウマイの皮、中華麺、即席麺等の麺類;飴、キャンディー、ガム、チョコレート、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、焼き菓子等の菓子類;かまぼこ、ハム、ソーセージ等の水産・畜産加工食品;加工乳、発酵乳等の乳製品;サラダ油、天ぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂および油脂加工食品;ソース、たれ等の調味料;錠剤状、顆粒状等の種々の形態の健康・栄養補助食品類;その他スープ、シチュー、サラダ、惣菜、漬物などを例示することができる。
本発明に係る食品には、種々の食品添加物、例えば、酸化防止剤、香料、各種エステル類、有機酸類、有機酸塩類、無機酸類、無機酸塩類、無機塩類、色素類、乳化剤、保存料、調味料、甘味料、酸味料、果汁エキス類、野菜エキス類、花蜜エキス類、pH調整剤、品質安定剤などの添加剤を単独、あるいは併用して配合してもよい。
本発明に係る食品におけるペンタデカン酸トリグリセリドの含有濃度は、固形分として、0.00001~100質量%程度(以下、%で表わす)、好ましくは0.0005~50%程度含有していると使用性および良好な効果が得られる。
次に実施例を挙げ、本発明を更に詳しく説明するが、本発明はこれら実施例に制約されるものではない。
(製造例1) オーランチオキトリウムを用いたペンタデカン酸トリグリセリドの製造
オーランチキトリウムmh1959株(国立大学法人 宮崎大学農学部 林 雅弘教授より購入)を、3.6%のグルコースと0.5%のグルタミン酸ナトリウム、0.2%の酵母エキス、1%海水塩、10%ホエイを含む培地を用いて、25℃で72時間前培養した。これを以下の基本培地に対し2%となるように加え、空気を通気させ、穏やかに攪拌した。基本培地1kgは、3.6%のグルコースと0.5%のグルタミン酸ナトリウム、0.2%の酵母エキス、1%海水塩、10%ホエイを加えた培地にバリンを50 mMとプロピオン酸ナトリウム25mMを添加して調製した。培養は、25℃、pHは7.40~7.75に保持(pH調整には、1.0 M NaOH溶液を用いる)して、72~96時間培養した。
オーランチキトリウムmh1959株(国立大学法人 宮崎大学農学部 林 雅弘教授より購入)を、3.6%のグルコースと0.5%のグルタミン酸ナトリウム、0.2%の酵母エキス、1%海水塩、10%ホエイを含む培地を用いて、25℃で72時間前培養した。これを以下の基本培地に対し2%となるように加え、空気を通気させ、穏やかに攪拌した。基本培地1kgは、3.6%のグルコースと0.5%のグルタミン酸ナトリウム、0.2%の酵母エキス、1%海水塩、10%ホエイを加えた培地にバリンを50 mMとプロピオン酸ナトリウム25mMを添加して調製した。培養は、25℃、pHは7.40~7.75に保持(pH調整には、1.0 M NaOH溶液を用いる)して、72~96時間培養した。
培養後、3000rpmで15分間遠心分離して、藻体約20gを回収した。得られたオーランチキトリウムの藻体20gに、ヘキサンまたは酢酸エチルを加え、脂質を抽出した。抽出した脂質溶液に過酸化水素水を加え(必要に応じて水を加える)、室温でオゾンを通気した。反応終了後、炭酸水素ナトリウムおよび炭酸ナトリウムまたはイオン交換樹脂にて酸化物を取り除き、温度低下と共に析出するペンタデカン酸トリグリセリド2gを得た。
(ペンタデカン酸トリグリセリドの組成分析)
製造例1で得られたペンタデカン酸トリグリセリドを含む脂質に14%BF3-メタノール0.50mLと酢酸メチル0.25mLとを加え、70℃、30分間加熱して脂肪酸のメチルエステル(FAME)を得た。反応液にn-ヘキサンを正確に1.0mLと5mLの生理食塩水を加え、激しく混合した。混合液を2800rpm、10分間遠心分離し、n-ヘキサン層をガスクロマトグラフィーの試料とした。
製造例1で得られたペンタデカン酸トリグリセリドを含む脂質に14%BF3-メタノール0.50mLと酢酸メチル0.25mLとを加え、70℃、30分間加熱して脂肪酸のメチルエステル(FAME)を得た。反応液にn-ヘキサンを正確に1.0mLと5mLの生理食塩水を加え、激しく混合した。混合液を2800rpm、10分間遠心分離し、n-ヘキサン層をガスクロマトグラフィーの試料とした。
島津製作所社製ガスクロマトグラフ装置GC-2025を用いて、上記試料を分析した。分析条件は、Agilent J&W GCカラムDB-23(60m×0.25mm)を用い、1μLの試料をインジェクトして、キャリヤーガス(He、14psi)にてFID(水素炎イオン化型検出器)で検出した。FAMEの分子種は脂肪酸メチルエステル標準品(GLサイエンス社製)の保持時間をもとに同定した。脂肪酸組成は面積比から求めた。求めた組成は質量比である。奇数鎖脂肪酸の割合は、総脂肪酸量に奇数鎖脂肪酸(C13、C15、C17)の割合(%)を掛けて求めた。得られた結果を以下の表1に示す。
表1に示した結果より、製造例1で得られたトリグリセリド中の奇数鎖脂肪酸の含量は、質量比で68.3%であった。また、脂肪酸としては、主として、ペンタデカン酸残基(C15)とパルミチン酸残基(C16)からなるトリグリセリドであることがわかった。
(ペンタデカン酸トリグリセリドの質量分析)
製造例1で得られたペンタデカン酸トリグリセリドを含む脂質を、Thermo Fischer社製Orbitrap質量分析計Exactive Plus(AMR社製DARTイオン源)を用いて質量分析法にて解析した。その結果を以下の表2に示す。

表2の結果より、製造例1で得られたペンタデカン酸トリグリセリドは、主要なマススペクトルピーク(分子量750)のフラグメント組成から、ペンタデカン酸残基(C15)のみで形成されるトリグリセライドとペンタデカン酸残基(C15)2単位にミリスチン酸残基(C14)を1単位含むトリグリセリドを主として含むことがわかった。
また、マススペクトルピーク(分子量764)のフラグメント組成から、ペンタデカン酸残基(C15)のみで形成されるトリグリセライドを主として含むことがわかった。
さらに、マススペクトルピーク(分子量778)のフラグメント組成から、ペンタデカン酸残基(C15)のみで形成されるトリグリセライドとペンタデカン酸残基(C15)2単位にパルミチン酸残基(C16)を1単位含むトリグリセリドを主として含むことがわかった。
(実施例1) ペンタデカン酸トリグリセリドの安全性の検証
ペンタデカン酸トリグリセリドの安全性を検証するために以下の実験を行った。

ペンタデカン酸トリグリセリドを1,10,100μg/mLとなるように、マウス膵臓β細胞由来の細胞株へin vitroにて添加した。
対照として0.01%、0.1%、1%(v/v)となるようにエタノールを細胞へ添加した。
24時間後に、細胞生存率をWST試薬により測定した。
ペンタデカン酸トリグリセリドを1,10,100μg/mLとなるように、マウス膵臓β細胞由来の細胞株へin vitroにて添加した。
対照として0.01%、0.1%、1%(v/v)となるようにエタノールを細胞へ添加した。
24時間後に、細胞生存率をWST試薬により測定した。
・試験結果
得られた結果を図1に示す。
図1に示すように、ペンタデカン酸トリグリセリドは添加量100μg/mLまでは全く毒性が見られなかった。
またペンタデカン酸トリグリセリドは10μg/mLと100μg/mLの濃度では、細胞増殖がむしろ促進された。
これらの点から、ペンタデカン酸トリグリセリドは、機能を発現する細胞に対する安全性の高い成分であることが証明できた。
得られた結果を図1に示す。
図1に示すように、ペンタデカン酸トリグリセリドは添加量100μg/mLまでは全く毒性が見られなかった。
またペンタデカン酸トリグリセリドは10μg/mLと100μg/mLの濃度では、細胞増殖がむしろ促進された。
これらの点から、ペンタデカン酸トリグリセリドは、機能を発現する細胞に対する安全性の高い成分であることが証明できた。
(実施例2) ペンタデカン酸トリグリセリドによるインスリン分泌促進への効果の検討
ペンタデカン酸トリグリセリドのインスリン分泌促進への効果を検討するために以下の実験を行った。

ペンタデカン酸トリグリセリドを10μg/mL存在下で、低ブドウ糖リンガー液と高ブドウ糖リンガー液をマウス膵臓β細胞由来の細胞株へin vitroにて添加し、30分間刺激した。
対照として0.1%(v/v)となるようにエタノールを細胞へ添加した。
培養液中に分泌されたインスリンはELISA法により測定した。

図2に示すように、低ブドウ糖液では、ペンタデカン酸トリグリセリドによるインスリン分泌促進効果が見られなかったが、高ブドウ糖液では、ペンタデカン酸トリグリセリドによるインスリン分泌促進効果が見られた。
この結果より、ペンタデカン酸トリグリセリドは、血糖値が高いときに、インスリン分泌を促進する効果のある、理想的な血糖値抑制剤であることが分かった。
得られた結果を図2に示す。
図2に示すように、低ブドウ糖液では、ペンタデカン酸トリグリセリドによるインスリン分泌促進効果が見られなかったが、高ブドウ糖液では、ペンタデカン酸トリグリセリドによるインスリン分泌促進効果が見られた。
この結果より、ペンタデカン酸トリグリセリドは、血糖値が高いときに、インスリン分泌を促進する効果のある、理想的な血糖値抑制剤であることが分かった。
なお、単に血糖値を下げる薬剤では、正常血糖値を下げると集中力がなくなる、無気力になる、イライラして落ち着きがなくなる、などの病的症状が出じる。さらに、進行すると、頭痛や吐き気、めまい、冷や汗などの症状が現れ、さらに進むと昏睡状態に陥るなど、危険な状態になる場合もある。
これに対し、ペンタデカン酸トリグリセリドの場合には、このようなリスクが起こらない、理想的な高血糖改善剤になることが分かってきた。したがって、ペンタデカン酸トリグリセリドは、理想的な血糖値改善剤、糖尿病改善剤及び食品であることが分かった。
(比較例1)ペンタデカン酸によるインスリン分泌促進への効果の検討
比較のために、ペンタデカン酸のインスリン分泌促進への効果を検討するために以下の実験を行った。

実施例1と比較例1との結果から、血糖値が高い状態のモデルにおいて、インスリン分泌を促進し、血糖値を正常に戻せる効果は、ペンタデカン酸トリグリセリドにおいて、初めて認められた効果であることが示された。
Claims (8)
- 式(1)におけるR1とR2またはR1とR3がペンタデカン酸残基であるトリグリセリドを有効成分として含む請求項1に記載のインスリン分泌促進剤。
- 式(1)におけるR1、R2およびR3の何れか1つが、トリデシル酸(C13)、ミリスチン酸残基(C14)、パルミチン酸残基(C16)またはマルガリン酸残基(C17)であるトリグリセリドを有効成分として含む請求項1または2に記載のインスリン分泌促進剤。
- 式(1)におけるR1、R2およびR3のすべてがペンタデカン酸残基であるトリグリセリドと、式(1)におけるR1、R2およびR3の何れか2つがペンタデカン酸残基であり、他の1つがミリスチン酸またはパルミチン酸残基であるトリグリセリドと、を含む請求項1または2に記載のインスリン分泌促進剤。
- 式(I)のトリグリセリドが、オーランチオキトリウム属又はシゾキトリウム属藻類由来である請求項1~4のいずれかに記載のインスリン分泌促進剤。
- 請求項1~5のいずれか1つに記載のトリグリセリドを有効成分として含む血糖値改善剤。
- 請求項1~5のいずれか1つに記載のトリグリセリドを有効成分として含む糖尿病改善剤。
- 請求項1~5のいずれか1つに記載トリグリセリドを有効成分として含む食品。
