Description
米国特許6218182号明細書は、3D組織の培養方法を説明しており、特に、細胞が植えられ、細胞の異なる側部に接触する2つの媒質流れとともに提供されるバイオリアクタにおいて、生体外肝臓支援装置としての使用のための肝臓組織の培養方法を説明している。
哺乳類の身体における、流れ、栄養分の運搬、ガス、および水の除去の規制は、体内の多くの複雑な機能を含む自動化されるプロセスである。血液は複雑なシステムであり、これは、身体全体を通して細胞へ、または細胞からの大量のガスおよび栄養分を輸送する能力を支援する。流れは、血液の流れを高い要求のある領域へ再分配するように体積および圧力を自動的に変化させる複雑なシステムにより管理される。この分配システムは、数千の分枝を含み、また、それぞれの分枝は、最終的に細胞に養分が付与される場所の寸法レベルに到達するまで内部直径が小さくなる。数値流体力学(Computational Fluid Dynamics, CFD)ソフトウェアの使用は、複雑な構造内およびその容器内の流れの解析を可能にする。適切な特性が同定される場合、材料の使用速度および排出物の生成の両方が、典型的な安全領域内にあることを確認するために、代謝パラメータを検討することができる。一例は、全ての細胞が有気性であることを確保するために、最大細胞密度および酸素消費速度を計算することである。

成長領域の入口および出口における流体分配手段は、各成長表面への十分な流れを可能にする。流体の分配は、数値流体力学および主要な代謝産物利用解析(key metabolite utilization analysis)を用いて解析され、細胞が有害な成長条件にさらされないことを確保する。
本発明の実際的な好ましい実施形態において、台座は、中心指示部の周りで規則的な間隔が隔てられた相互接続された成長表面のマトリックスにより形成される。
好ましくは、培養装置は、大きな開放領域を形成するために均一に構成される、多数の相互接続される成長表面を含み、これは、立方体容積辺りの細胞の最大数を制限し、成長領域の容易な脈管化を促進する。

例
実験プロトコル
本発明による、等しい寸法の4つの部片に切断される2層台座(11mm×11mm×5mm)がマウスでの細胞成長実験に用いられた。
4つの連続培養装置は、免疫欠損NOD/SCIDマウスにインプラントされた。
インプラントから1週間後の刺激反応の解析は、典型的な刺激反応、すなわち、腫れ(swelling)、赤み(redness)、滲出物(exudates)等のサインを示さなかった。コントロール材料HA(Hydroxy Apatite)、すなわち、標準サンプルとして使用される市販されている生物医学的材料の組織学的解析は、炎症(たとえば、リンパ球浸潤)を示さず、逆に、多孔セラミック材料の組織への同化が示された(材料の孔の繊維芽細胞の増殖)。
