発明の分野
[0003] 本発明は、光学顕微鏡の分野に関する。特に、本発明は、生体分子の撮像改良のために、2種類の流体間にある薄い固相界面を利用する。
実施例
実施例1
サンプルの外形形状
[0089] 中央に厚さが20〜50nmの自立窒化シリコン・ウィンドウ[50μm×50μm]を有するシリコン・チップ[5mm×5mm×300μm]を、標準的なフォトリソグラフ法によって、シリコン・ウェア上のLPCVD被覆SiN層上に作成した。ガラス・カバースリップ上にこれらのチップを実装し、図3に示したように、チップとガラス・カバースリップとの間に、厚さ2〜10μmのマイクロチャネルを形成するために、二部PTFE/CTFE流体セルを設計した。このマイクロチャネル、即ち、トランス・チェンバ204を、屈折率がより高い流体(70%グリセロール[n=1.42])で満たし、シス・チェンバ208を、サンプル緩衝水溶液[n=1.33]で満たした。
TIRFの設定および光路図
[0090] 全内部反射[TIR]顕微鏡検査を、図4に示したように設定した。レーザ・ビーム・サイズを0.7mmに縮小し、市販の倒立顕微鏡(Olympus IX-71)の特別設計したバック・ポートに発射し、外部に取り付けられた200mm焦点長のレンズを通じて、60×1.45NA油浸入TIRF対物レンズの背後焦点面において合焦させた。レーザ・ファイバ・カプラをX−軸に沿って動かすことによって、レーザ点を光軸から外れるようにずらした。光が窒化シリコン・メンブレンにおいてグリセロール−水界面に達するまで、光が油、ガラス・カバースリップ、および70%グリセロールを通ってトランス・チェンバ内で屈折するように、ガラス[n=1.52]、70%グリセロール[n=1.42]、およびサンプル緩衝剤[n=1.33]の屈折率を選択した。この界面におけるTIRモードの臨界角は、スネルの法則によって記述されるように、ガラス−水界面と同じである。
サンプル不動化
[0091] 5’末端にビオチン共役があり、5’末端から20番目の位置にアミン修飾チミン塩基があるDNA分子[57bp]をIDT Techから購入した。DNA分子に、販売業者のプロトコルを用いて、ATTO647N染料分子[ATTO−tech]で標識を付けた。ストレプトアビジン・ビオチン(streptavidin-biotin)化学薬品によって、窒化シリコン上でDNA分子を不動化した。新たに用意したピラニア溶液(Piranha solution)において表面を浄化し(硫酸および過酸化水素の7:3v/v溶液において15分)、次いでDI水において徹底的に洗浄した。0.1mg/ml BSA−ビオチンにおいて一晩表面を培養し、結合緩衝剤[10mM Tris−CL、pH8.5]で洗浄し、ストレプタビディンにおいて30〜60分培養し、結合緩衝剤において洗浄し、次いでビオチンで標識を付けたDNAで15〜30分培養した。次いで、流体セルに表面を装填した。70%グリセロールを、ガラス・カバースリップとシリコン・チップとの間にあるトランス・マイクロチャネルに充填し、結合緩衝剤をシス・チェンバに充填した。次いで、TIRFMによって窒化シリコン界面において表面を撮像した。
結果
[0092] 図4に示したように、窒化シリコン・メンブレン(例えば、サイズが20×40μm2、厚さが20nm)を、ガラス・カバースリップ上に装填した。トランス・チェンバおよびシス・チェンバにおける2種類の流体を、それぞれ、グリセロール70%(v/v)(n=1.42)および水(n=1.33)に選択した。
実施例2
TIRFの設定
[0097] 実験において、ナノポア600を有する自立SiNメンブレン(20×20μm2)1124を内蔵するシリコン・チップ1128を、界面として用いた。シリコン・チップ1128をガラス・カバースリップ1140上に装填し、ガラス・カバースリップ1140を、特注で製作したクロロトリフルオロエチレン(CTFEポリマー)流体セル1138上に装填し、図11に示したような、微小流体トランス・チェンバ1100を作成した。流体セル1138は、シリコン・チップ1128を保持するインサート1138aと、流体チェンバを形成する外側セル1138bとを含む。高速硬化ポリジメトリシロキサン(PDMS:polydimethlysiloxane)を用いて、シリコン・チップ1128をCTFEインサート1138aに接合し、ガラス・カバースリップ1140を外側セル1138bに接合した。インサート1138aを有する流体チェンバはシス・チェンバであり、シリコン・チップ1128とガラス・カバースリップ1140との間の空間がトランス・チェンバとなる。流入−流出フロー・チャネルを用いて、トランス・チェンバに、屈折率緩衝剤1112を充填した。電気的測定のために、フロー・チャネルの側面開口内にトランス電極1140aを設け、インサート(Ag/AgCl双方)1116において緩衝剤の中にシス電極1140bを埋没させた。ナノポア600を用いて、図11Aに示すように、光学可視化および測定のために、流体セル1100を倒立顕微鏡1136に対して一直線上に揃えた。
[00102] 光学信号および電気信号の同期検出のために、ハードウェア(例えば、Microsoft WindowsまたはLinux互換パーソナル・コンピュータ)とLABVIEWソフトウェアとの組み合わせを設計した。図13は、取り込みハードウェアを模式的に示す。Axopath200B増幅器1386(Molecular Devices, Inc., Sunnyvale, CA, USA)を、Ag/AgCl電極にヘッドステージ1388を介して接続し、ナノポアを横切るイオン電流信号を増幅する。外部四極バターワース・フィルタ1390を用いて、このイオン電流信号に50KHzでロー・パス・フィルタをかけ、画像取り込みボード1394(Andor iXon取り込みボード)を介してCCDカメラから画像データを受け取った 同じPC内にある多機能データ取り込みボード1392に入力する。多機能データ取り込みDAQボード1392(National Instruments, PCI-6154)を用いて、16ビット・アナログ/デジタル変換分解能で、イオン電流信号を取り込んだ。EM−CCDカメラ1384からの「発火」パルス(各露出の開始を印すTTLパルス)が、イオン電流の取り込みをトリガし、カウンタ・ボード(National Instruments, PCI-6602)上において高精度のタイム・スタンプを生成するために用いられた。カウンタ・ボードは、クロック・レートが250KHzのRTSIバスを用いて、内部でDAQボードと同期を取られていた。したがって、組み合わされたデータ・ストリームは、イオン電流のサンプリングと同期を取られているCCDフレームの各々の開始時における一意のタイム・スタンプを含んでいた。転座イベントが、イオン電流の落下によって検出されたとき、ソフトウェア1396は、カウンタ情報の中で対応するフレーム番号を検索し、この番号に対応する実際の画像をセーブした。カメラ・フレーム・レートは、約1KHz(発火パルス・レート)に設定した。
実施例3
[00105] 4nmポアを介して転座する蛍光標識付きdsNDAを検出するために、同時光学および電気的測定を行った。この実験におけるサンプル濃度は、0.1nM〜0.2nMであった。図15Aおよび図15Bは、ポアの形状、および約4nmのポア例のTEM画像を模式的に示す。421bp断片(DNA−A1647)に、Alexa647蛍光体で標識を付け、重合連鎖反応(PCR)反応、それに続くアミン反応染料との共役の間、低濃度のアミノ修飾チミン塩基の合体によって用いられる。

前記メンブレンの第2側に配置された第2流体チェンバであって、第2屈折率を有する第2流体を収容し、前記第1屈折率が前記第2屈折率よりも高い、第2流体チェンバと、
